We evaluated the curative efficacy of a gelatin β-tricalcium phosphate (β-TCP) sponge loaded with mesenchymal stem cells (MSC), bone morphogenic protein-2 (BMP-2), and platelet-rich plasma (PRP) by insertion into an experimentally induced osteochondral defect. A hole of 10 mm diameter and depth was drilled in the bilateral medial femoral condyles of 7 thoroughbred horses, and into each either a loaded sponge (treatment) or a saline-infused β-TCP sponge (control) was inserted. After 16 weeks, defects were examined by computed tomography, macroscopic analyses, and histological analyses. The median subchondral bone density and macroscopic subscores for joint healing were significantly higher in the treatment legs (P < 0.05). Although there was no significant difference in total histological scores between groups, hyaline cartilaginous tissue was observed across a wider area in the treatment group. Equine joint healing can be enhanced by inserting a BMP-2-, MSC-, and PRP-impregnated β-TCP sponge at the lesion site.  相似文献

ABSTRACT

Sowing using a rotary tiller rotating in reverse (up-cut) is a labor-saving technology for soybean production because it can perform tillage and sowing, make ridges, and bury winter crop residues in one operation. However, the power requirement of up-cut rotation is higher than of normal rotation (down-cut), resulting in lower working speeds. Shallow tillage sowing methods may be a solution to this problem. Here, we develop a shallow tillage sowing method using an up-cut rotary and a side-disk which could perform the aforementioned works in one operation and conducted field experiments in 2015 and 2016 to study the effect of our sowing method (up-cut shallow tillage; UST) on ridge shape, soil volumetric water content (VWC) and soybean growth and yield, compared to conventional sowing methods (up-cut conventional tillage; UCT, and down-cut conventional tillage; DCT). Ridge shape did not differ among sowing methods. The VWC at 10 and 20 cm depths in DCT was always higher than in UST and UCT, indicating poorer drainage. The VWC at 10 cm depth decreased quickly after rain in UST and UCT, indicating good surface-layer drainage, but at 20 cm depth in UCT, VWC was lower than in UST, especially during dry spells, indicating higher water-holding capacity in UST than in UCT. Shoot and root growth and seed yield did not differ among sowing methods, indicating that UST did not negatively affect soybean growth and yield. These results suggest that UST can be used for soybean production.  相似文献

An experiment was carried out to analyse the effects of substituting fish‐meal (FM) with soya bean meal and shrimp protein hydrolysate (SPH) on the percent survival, growth rate, feed efficiency and molt frequency of redclaw crayfish hatchlings, Cherax quadricarinatus. The experiment was performed in a closed recirculating‐water system over a 56‐day period. Five diets were tested: a control diet (SPH0) comprising 100% FM without SPH, and four diets with 2.5%, 5.0%, 10.0% and 15.0% SPH substitution (diets SPH2.5, SPH5, SPH10 and SPH15 respectively). Analysis of water quality indicated that it was adequate to maintain a good growth and survival of hatchlings. The results showed that 100% survival was obtained for hatchlings fed with SPH5, SPH10 and SPH15 diets. Higher mean values of final weight were obtained for the SPH5 diet. The highest weight gain was observed in SPH2.5 and SPH5 diets. Hatchlings showed similar specific growth rates for all diets. The efficiency rate of the SPH15 diet was lower compared to the other diets. Overall, individual feed consumption was higher in hatchlings fed diets of SPH5 and SPH15. It is recommended that 5.0% of SPH could be included in diets of hatchling redclaw crayfish to improve their growth efficiency.  相似文献

Platelet-rich plasma (PRP) was administered to cattle with hoof defects (10-mm drill hole) to promote sole defect healing. Two types of gels were prepared. One was a mixed gel [compound of gelatin microspheres (GM) incorporated with PRP and alginate] and was used for the test group, while the other was an alginate gel used for the control group. Each hole was filled with one of these gels. The drill hole depth, pressure pain, and hardness of the regenerated tissue were measured for 3 weeks after treatment. These measurements improved significantly in the test group than in the control group. The results suggest that a compound of GM incorporated with PRP and alginate had a hoof-regenerating effect in cattle.  相似文献

This study evaluated the therapeutic effects of a gelatin-β-TCP sponge (sponge) incorporating BMP-2 (BMP-2/sponge) on bone regeneration in equines. Six bone defects were created in third metacarpals of five thoroughbred horses, and a total of six treatments were applied in a randomized manner. The treatments were BMP-2/sponge, BMP-2/gelatin hydrogel sheet (sheet), free BMP-2, bFGF/sheet, plain sponge, and plain sheet. The defects were monitored for 16 weeks by radiography and then examined by histological analysis. Radiographic evaluation scores of bone regeneration revealed significantly greater bone regeneration of defects treated with BMP-2/sponge than defects treated with plain sponge or BMP-2 sheet (P<0.05). In histological analysis, compact bone was observed over a wide area in the BMP-2/sponge treatment. We concluded that the treatment with BMP-2/sponge accelerated bone regeneration in the equines of this study.  相似文献

Nematodes of the family Heligmonellidae (Heligmosomoidea; Trichostrongylina) reside in the digestive tracts of rodents and lagomorphs. Although this family contains large numbers of genera and species, genetic information on the Heligmonellidae is very limited. We collected and isolated adult worms of three species in Japan that belong to the family Heligmonellidae, namely Heligmonoides speciosus (Konno, 1963) Durette-Desset, 1970 (Hs) from Apodemus argenteus, Orientostrongylus ezoensis Tada, 1975 (Oe) from Rattus norvegicus and Lagostrongylus leporis (Schulz, 1931) (Ll) from Pentalagus furnessi, and sequenced the entire internal transcribed spacer regions, ITS-1 and ITS-2 of ribosomal DNA. ITS-1 of Hs, Oe and Ll was 426, 468 and 449 bp in length, and had a G+C content of about 41, 41 and 37 %, respectively. ITS-2 of Hs, Oe and Ll was 297, 319 and 276 bp in length and had a G+C content of about 38, 40 and 28%, respectively. The data of Hs, Oe and Ll were compared with those of two other known species within the family Heligmonellidae, Calorinensis minutus (Dujardin, 1845) (Cm) and Nippostrogylus brasiliensis (Travassos, 1914) (Nb), and with those of two species of Heligmosomidae (Heligmosomoidea), Heligmosomoides polygyrus bakeri and Ohbayashinema erbaevae. Phylogenetic analysis placed Hs, Oe and Ll in the same clade with Cm and Nb, forming a Heligmonellidae branch in both ITS-1 and ITS-2, separate from the Heligmosomoidea branch. These results demonstrated that the ITS-1 and ITS-2 sequences are useful for differentiating the Heligmonellidae nematode species. This study is the first to describe the ITS-1 and ITS-2 sequences of Hs, Oe and Ll.  相似文献

The effect of moisture content on the stress wave propagation velocity was investigated in order to estimate the Young’s modulus of full-scale timbers in an air-drying state using the measurement of stress wave propagation velocity above the fiber saturation point. Using Japanese cedar lumber, the velocity and the density under high-moisture condition and air-drying states were measured respectively; after measuring the modulus of elasticity in an air-drying state, the moisture content of each condition was measured. By performing numerical analysis on these data, the relationship between the moisture content and the rate of change of velocity of full-scale timbers was derived. This relationship was used to estimate the Young’s modulus of the timber in the air-drying state from the velocity in high-moisture condition. First, the velocity and the Young’s modulus in an air-drying state were estimated accurately from its density, moisture content and velocity under high-moisture condition. In cases where the density could not be measured, using the database of mechanical properties with the Monte Carlo simulation method, the Young’s modulus of the full-scale timber in an air-drying state might be estimated within 20% accuracy from its moisture content and velocity under high-moisture condition.  相似文献

In 2016, in Miyakojima, Okinawa, Japan, banana plants (Musa?×?paradisiaca) ‘Shima-banana’ developed yellowing and wilt associated with vascular discoloration of the pseudostems. Fusarium oxysporum, identified based on morphological characters, was frequently isolated from the vascular tissue of the infected plant and reproduced the original symptoms on ‘Shima-banana’ after drench inoculation with a spore suspension. Thereby, we determined that the disease is Panama disease caused by F. oxysporum. This is the first official report of Panama disease (Panama-byo in Japanese) of banana in Japan.  相似文献

The present study was undertaken to know the effect of supplementation of fish oil with high n-3 polyunsaturated fatty acids (PUFA) on oxidative stress-induced DNA damage of rat liver in vivo. Male Wistar rats were fed a diet containing fish oil or safflower oil with high n-6 PUFA at 50 g/kg of diet and an equal amount of vitamin E at 59 mg/kg of diet for 6 weeks. Livers of rats fed fish oil were rich in n-3 PUFA, whereas those of rats fed safflower oil were rich in n-6 PUFA. Ferric nitrilotriacetate was intraperitoneally injected to induce oxidative stress. The degree of lipid peroxidation of the liver was assessed by the levels of phospholipid hydroperoxides and thiobarbituric acid-reactive substances (TBARS), and the degree of oxidative DNA damage was assessed by comet type characterization in alkaline single-cell gel electrophoresis and 8-hydroxy-2'-deoxyguanosine levels. The levels of TBARS of the livers of the fish oil diet group increased to a greater extent than those of the safflower oil diet group, whereas the levels of the hydroperoxides of the livers of both diet groups increased to a similar extent. The vitamin E level of livers of the fish oil diet group was remarkably decreased. The degree of DNA damage of both diet groups was increased, but the increased level of the fish oil diet group was remarkably lower than that of the safflower oil diet group. The above results indicate that fish oil supplementation does not enhance but appears to protect against oxidative stress-induced DNA damage and suggest that lipid peroxidation does not enhance but lowers the DNA damage.  相似文献
