We investigated the effects of a low crude protein (CP) diet and a low CP diet supplemented with synthetic essential amino acids (EAA) on the meat quality of broiler chickens. Twenty‐one‐day‐old chickens were assigned to one of three diets: control, low CP (LCP), or low CP supplemented with EAA (ELCP). The chickens received these diets for 10 days. The shear force value (SFV) and free glutamate content of the Pectoralis major muscle were measured as indicators of the meat toughness and taste. The collagen and crude fat content of the muscle and the cross‐sectional area of myofibers were measured to evaluate the effects of the LCP and ELCP diets on meat toughness. The SFV of the ELCP group was 47% lower than that of the control group (P < 0.01). However, the LCP diet did not affect the SFV. The collagen and crude fat content were not affected by the dietary treatment. The cross‐sectional area was lower in the LCP and ELCP groups (P < 0.05) than the control group. The free glutamate content of muscle was not affected by the dietary treatment. Thus, a low CP diet supplemented with EAA is an effective means of producing tender meat. 相似文献

Canine non-rabies combined vaccines are widely used to protect animals from infectious agents, and also play an important role in public health. We performed a large-scale survey to investigate vaccine-associated adverse events (VAAEs), including anaphylaxis, in Japan by distributing questionnaires on VAAEs to veterinary hospitals from April 1, 2006 through May 31, 2007. Valid responses were obtained for 57,300 vaccinated dogs at 573 animal hospitals; we obtained VAAEs information for last 100 vaccinated dogs in each veterinary hospital. We found that of the 57,300, 359 dogs showed VAAEs. Of the 359 dogs, death was observed in 1, anaphylaxis in 41, dermatological signs in 244, gastrointestinal signs in 160, and other signs in 106. Onset of VAAEs was mostly observed within 12h after vaccination (n=299, 83.3%). In this study, anaphylaxis events occurred within 60 min after vaccination, and about half of these events occurred within 5 min (n=19, 46.3%). Furthermore, where anaphylaxis was reported, additional information to support the diagnosis was obtained by reinvestigation. Our resurvey of dogs with anaphylaxis yielded responses on 31 dogs; 27 of these demonstrated collapse (87.1%), 24 demonstrated cyanosis (77.4%), and both signs occurred in 22 (71.0%). Higher rates of animal VAAEs, anaphylaxis, and death were found in Japan than in other countries. Further investigations, including survey studies, will be necessary to elucidate the interaction between death and vaccination and the risk factors for VAAEs, and thus develop safer vaccines. Yeso sika deer captured in winter around Lake Akan in Hokkaido were reared for 8–10 months at Tokyo University of Agriculture in Abashiri. Six 1‐year‐old females and males and six 2‐year‐old or older (adult) females and males were slaughtered and their carcasses were processed. The chemical composition, mineral contents and fatty acid composition of the loin were measured. No marked influence of gender or age was noted in the chemical composition of loin. In the mineral contents, significant differences were noted. The potassium and sulfur contents were lower and the sodium content was higher in adult deer meat (P < 0.05, respectively) and the potassium content was higher in male deer meat (P < 0.05). Arsenic, cadmium or lead were not detected. In the unsaturated fatty acid, a significant interaction was detected (P < 0.05), and it was high in 1‐year‐old female deer meat and low in 1‐year‐old male deer meat. Significant gender or age differences were noted only in the mineral contents in the loin of deer reared for a short period after capture. 相似文献

To reduce the availability of soil cadmium (Cd) to soybeans (Glycine max (L.) Merr.), we employed a liming by partial mixing (PM) technique in two drained paddy fields on Gray Lowland soils, which had 0.1 mol L–1 hydrochloric acid-extractable Cd concentrations as high as 1.08 and 1.40 mg kg–1. Among the different application methods tested, PM application (PM2) using a width of 20 cm and a depth of 20 cm was found to be most appropriate for reducing the seed Cd concentration and to obtain the optimum yield at Site A. Under PM2, a liming rate of 38% of that for broadcast incorporated into the surface 15 cm layer (Bc) was suitable to reduce the seed Cd concentration at Site A, whereas the lime rate with PM2 was set at 50% of that for Bc (PM2-50) at Site B due to the higher availability of soil Cd. The root system was limited within the range of lime and fertilizer application for PM2 as well as PM2-50; thus, the lime and fertilizer were supplied successfully to the rooting zone. The soil pH value was lower under PM2 at Site A and PM2-50 at Site B compared with Bc, whereas the seed Cd concentration was lower for PM2 and PM2-50. This may be explained by the intensive uptake of calcium and magnesium with PM2 as well as PM2-50. The seed Cd concentration in the cultivar “Ryuhou” at the target pH of 6.5 was approximately 30% lower with PM2-50 than Bc at Site B. In addition, the average seed Cd concentrations in one cultivar and two lines, characterized by the lower Cd uptake with higher retention in roots and higher accumulation in leaves, were approximately 40% lower compared with “Ryuhou.” Thus, the combination of liming with PM2-50 at the target pH of 6.5 and a low-Cd cultivar (or lines) minimized the seed Cd concentration. The highest seed Cd concentration was found in the first year of soybean cultivation, which was considered to be caused by the release of Cd from organic nitrogen compounds during the nitrogen mineralization process. 相似文献

Intramuscular fat content is increased by feeding of low lysine diets in pigs. Reduction in dietary lysine intake results in low plasma lysine concentration and low cytosolic lysine concentration in skeletal muscles. From these observations, we hypothesized that low plasma lysine concentration in pigs fed on low lysine diets reduced supply of lysine from blood circulation to preadipocytes, and this limited supply of lysine might promote adipocyte differentiation in porcine muscles. In order to verify the hypothesis, we investigated the effects of low concentrations of lysine in culture medium on differentiation of 3T3‐L1 preadipocytes. Low concentration of lysine suppressed lipid accumulation and messenger RNA (mRNA) expression and enzyme activity of fatty acid synthase. mRNA expressions of peroxisome proliferator‐activated receptor γ (PPARγ) and CCAAT/enhancer binding protein α (C/EBPα) were lower in cells cultured in low lysine medium. On the other hand, mRNA and protein expressions of C/EBPβ and C/EBPδ were not inhibited by low concentrations of lysine in culture medium. These results indicate that low lysine concentrations in culture medium inhibit differentiation of 3T3‐L1 preadipocytes through inhibiting the mRNA expressions of PPARγ and C/EBPα. 相似文献

Glycoconjugates are known to be involved in many physiological events in vertebrates. Sialidase is one of the glycosidases, which removes sialic acid from glycoconjugates. In mammals, the properties and physiological functions of sialidases have been investigated, while there is little understanding of fish sialidase. Here, to investigate the significance of fish neu4 sialidase, neu4 gene was cloned from medaka brain mRNA and identified. Sialidase-specific motifs (GPG, YRVP and Asp-Box) were well conserved in the medaka neu4 polypeptide. Optimal pH of medaka neu4 sialidase was 4.6, but its activity was sustained even at neutral and weak alkaline pH. The neu4 considerably cleaved sialic acid from 4-methylumbelliferyl-N-acetyl-α-d-neuraminic acid and sialyllactose, but not from ganglioside and fetuin, which are good substrates for human NEU4. neu4 activity was mostly detected in mitochondria/lysosome fraction after biochemical fractionation, and indirect immunofluorescence assays revealed neu4 localization in lysosome in neu4 overexpressed cells. Next, developmental change in medaka neu4 and other sialidase mRNA levels were estimated by real-time PCR. Each sialidases showed different expression patterns in embryonic development: neu4 was up-regulated at late developmental stage in embryo, and neu3a mRNA level was quite high in 0.5 dpf. On the other hand, neu3b expression was drastically increased after hatching, suggesting that each sialidase may play a different role in embryonic development. 相似文献

It is desirable to produce beef with high levels of monounsaturated fatty acids (MUFA), as this is related to fat softness and palatability. However, the physiology of MUFA synthesis in bovine fat during the fattening process remains to be established. In this study, in order to elucidate the relationship between plasma components and the fatty acid composition of intramuscular fat, we investigated the effect of plasma obtained from fattening cattle on the messenger RNA (mRNA) expressions of the adipogenesis‐related gene in a clonal bovine intramuscular preadipocyte line (BIP cells). The mRNA expressions of stearoyl‐CoA desaturase, adipocyte Protein 2, peroxisome proliferator‐activated receptor gamma and sterol regulatory element‐binding protein 1 in BIP cells were significantly higher following treatment with those plasma samples collected from the cattle with the highest diaphragmatic unsaturated fatty acids to saturated fatty acids ratio (US/S). Furthermore, the concentration of nonesterified fatty acid (NEFA) in the plasma samples had an inverse correlation with carcass diaphragmatic US/S. These results indicate that cattle with a low ratio of US/S in fat may be discriminated from the population of fattening cattle before slaughter by measuring the effect of their plasma on gene expression in BIP cells as well as their plasma concentration and composition of NEFA. 相似文献

Excessive lipid peroxidation negatively affects the physiological response and meat quality of chickens. Delaying post‐hatch feeding was previously found to increase lipid peroxidation in the skeletal muscle of finishing broiler chickens. The aims of this study were to investigate the effects of delayed post‐hatch feeding on lipid peroxidation and the mRNA expressions of antioxidant enzymes in the pectoralis major muscle of broiler chicks during the post‐hatching period. Newly hatched chicks either had immediate free access to feed (freely‐fed chicks) or had no access to feed from 0 to 2 days old (delayed‐fed chicks), after which both groups were fed ad libitum until 4 or 13 days old. The lipid peroxidation level was higher in the delayed‐fed than freely‐fed chicks at 2, 4, and 13 days old. At 2 days old, the mRNA expressions of Cu/Zn‐SOD, Mn‐SOD, and GPX7 were lower in the delayed‐fed than freely‐fed chicks, while catalase mRNA levels did not differ. Furthermore, at 4 and 13 days old, lower mRNA expressions of Cu/Zn‐SOD and Mn‐SOD were observed in the delayed‐fed than freely‐fed chicks. These results suggest that delaying post‐hatch feeding reduces the mRNA levels of Cu/Zn‐SOD and Mn‐SOD, consequently affecting muscle lipid peroxidation in chicks during subsequent growth. 相似文献
