When the influence of host species, inoculum density, temperature, leaf wetness duration, and leaf position on the incidence of gentian brown leaf spot caused by Mycochaetophora gentianae, was examined, the fungus severely infected all seven Gentiana triflora cultivars, but failed to infect two cultivars of G. scabra and an interspecific hybrid cultivar. Inoculum density correlated closely with disease incidence, and a minimum of 102 conidia/mL was enough to cause infection. In an analysis of variance, temperature and leaf wetness duration had a significant effect upon disease incidence, which increased with higher temperature (15–25°C) and longer duration of leaf wetness (36–72 h). No disease developed at temperatures lower than 10°C or when leaf wetness lasted <24 h. At 48-h leaf wetness, disease incidence was 0, 28, 77, and 85% at 10, 15, 20, and 25°C, respectively. Middle and lower leaves on the plant were more susceptible than upper leaves. In microscopic observations of inoculated leaves, >50% of conidia germinated at temperatures >15°C after 24-h leaf wetness. More appressoria formed at higher temperatures (15–25°C) with extended duration of leaf wetness (24–72 h). At 48-h leaf wetness, appressorium formation was 0, 8, 26, and 73% at 10, 15, 20, and 25°C, respectively. These results suggest that temperature and leaf wetness duration were important factors for infection of gentian leaves.  相似文献

The uncertainty in pesticide residue levels (UPRL) associated with sampling size was estimated using individual acetamiprid and cypermethrin residue data from preharvested apple, broccoli, cabbage, grape, and sweet pepper samples. The relative standard deviation from the mean of each sampling size (n = 2(x), where x = 1-6) of randomly selected samples was defined as the UPRL for each sampling size. The estimated UPRLs, which were calculated on the basis of the regulatory sampling size recommended by the OECD Guidelines on Crop Field Trials (weights from 1 to 5 kg, and commodity unit numbers from 12 to 24), ranged from 2.1% for cypermethrin in sweet peppers to 14.6% for cypermethrin in cabbage samples. The percentages of commodity exceeding the maximum residue limits (MRLs) specified by the Japanese Food Sanitation Law may be predicted from the equation derived from this study, which was based on samples of various size ranges with mean residue levels below the MRL. The estimated UPRLs have confirmed that sufficient sampling weight and numbers are required for analysis and/or re-examination of subsamples to provide accurate values of pesticide residue levels for the enforcement of MRLs. The equation derived from the present study would aid the estimation of more accurate residue levels even from small sampling sizes.  相似文献

Chiyoda basin is located in Saga Prefecture in Kyushu Island, Japan, and lies next to the tidal compartment of the Chikugo River to which the excess water in the basin is drained away. Chiyoda basin has a total area of about 1,100 ha and is a typical flat and low-lying paddy-cultivated area. The main environmental issue in this basin is total nitrogen (TN) and total phosphorus (TP) load management because TN and TP, which loaded from farmlands, degrade surface water as a result of anthropogenic eutrophication. This paper presents a mathematical model of TN and TP runoff during an irrigation period in Chiyoda basin in order to elucidate the pollutant fluxes that accompany water transportation in paddy fields and drainage canals, and to evaluate pollutant removal from the study area to the Chikugo River. First, the water flow and the algorithm of gate operation were simulated by a continuous tank model and the accuracy of the model was then evaluated by comparing the simulated water levels with observed ones during an irrigation period. The observed and simulated water levels were in good agreement, indicating that the proposed model is applicable for drainage and water supply analyses in flat, low-lying paddy-cultivated areas. Second, the TN and TP runoff during an irrigation period was simulated based on the TN and TP loads that were determined by observed data in paddy fields. For TN runoff, the simulated results and observed data were in good agreement whereas for TP runoff, the simulated results were higher than the observed data. However, if the settled TP within the paddy tank was calculated as 6%, then the simulated results and the observed data were in good agreement. We concluded that TN runoff from paddy field to the drainage canal system was not affected much by the sediment related process. The present study could provide farmers and managers with a useful tool for controlling the water distribution in an irrigation period, and the TN and TP loads in the downstream area as well as the Chikugo River.  相似文献

We compared the hair cortisol levels of lactating dairy cows in a cold‐ and a warm‐temperate region out of four climatic zones in Japan. We simultaneously investigated the effects of calving number, lactation period and month of hair sampling. Hair of nine Holstein lactating cows chosen from each region (i.e. 18 cows per sampling) was sampled in March, June, September and December. Number of calvings (1, 2, ≥3) and lactation duration (early: <100, middle: 101–200, and late: >201 days) were balanced between regions. Cortisol was extracted from hair by methanol, and its level was determined with a cortisol immunoassay kit. A multi‐way analysis of variance revealed that the effects of month of hair sampling (P < 0.001) and its combination with region (P < 0.05) were significant. In a multiple comparison test, significant differences (P < 0.01) in hair cortisol level (pg/mg of hair) were found between June (13.0 ± 1.0) and the other 3 months, and between September (1.6 ± 0.2) and December (4.5 ± 0.3). The rise in cortisol level from March to June was more intense in the cold‐temperate region. These results demonstrate the necessity of considering seasonal variations in each climatic region when we use hair cortisol level as an indicator of stress.  相似文献

ABSTRACT:   The timing of molting and metamorphosis was monitored for Panulirus japonicus phyllosoma larvae cultured in the laboratory. Larvae cultured under natural light–dark cycle molted synchronously within approximately 1.0 h before and after sunrise, except for a short period after hatching; the timing of molting changed with time of sunrise. When three artificial light–dark cycles were introduced, larvae molted around the start of lighting, irrespective of the light–dark cycles. In the event of a sudden change in the start or end of lighting, molting was regulated by an endogenous rhythm. The start of lighting had a greater impact on the timing of molting than the end of lighting, suggesting that sunrise is probably the critical signal for phase-setting in molting rhythm. Metamorphosis to the puerulus stage occurred within 0.4 h before and 1.2 h after sunset under a natural light–dark cycle, and the timing of metamorphosis was also changed artificially by regulating the end of lighting.  相似文献

We excised surgically a feline granulomatous lesion and performed histopathological, mycological and molecular examinations. As a result, it was diagnosed as sporotrichosis, which was the second recorded case of a cat so afflicted in Japan. After the operation, we recognized another nodule on the lymph node. Histopathological examination was therefore performed, but no fungi were detected. To prevent recurrence, the cat was administered a antimycotic drug, itraconazole. As a result, no recurrence was found. Excision of the lesion is the treatment of choice for feline sporotrichosis.  相似文献

Portal vein aneurysm (PVA) is a rare abnormal dilatation of the portal vein, which has not been reported in dogs. We describe the findings of ultrasound and computed tomography in a case of PVA in a young male toy poodle, with the final diagnosis established by explorative surgical observation. The dog had an aneurysmal fusiform dilatation in the extrahepatic portal vein with portal hypertension and multiple portsystemic shunts. This is the first report of canine PVA.  相似文献

Somatic cell nuclear transfer (SCNT) is a useful technique for creating pig strains that model human diseases. However, production of numerous cloned disease model pigs by SCNT for large-scale experiments is impractical due to its complexity and inefficiency. In the present study, we aimed to establish an efficient procedure for proliferating the diabetes model pig carrying the mutant human hepatocyte nuclear factor-1α gene. A founder diabetes transgenic cloned pig was generated by SCNT and treated with insulin to allow for normal growth to maturity, at which point epididymal sperm could be collected for cryopreservation. In vitro fertilization and intrafallopian insemination using the cryopreserved epididymal sperm resulted in diabetes model transgenic offspring. These results suggest that artificial reproductive technology using cryopreserved epididymal sperm could be a practical option for proliferation of genetically modified disease model pigs.  相似文献
