Low-field magnetic resonance imaging (MRI) was performed on the stifle joints of four normal adult mongrel dogs using a 0.064 Tesla scanner. Markers were placed on each stifle joint to serve as reference points for comparing gross sections with the images. A T1-weighted sequence was used to image one stifle joint on each dog in the sagittal plane and the other stifle joint in the dorsal plane. The dogs were euthanized immediately following MRI and the stifle joints frozen intact. Each stifle joint was then embedded in paraffin, again frozen, and sectioned using the markers as reference points. On T1-weighted images, synovial fluid had low signal intensity (dark) compared to the infrapatellar fat pad which had a high signal intensity (bright). Articular cartilage was visualized as an intermediate bright signal and was separated from trabecular bone by a dark line representing subchondral bone. Menisci, fibrous joint capsule, and ligamentous structures appeared dark. In the true sagittal plane, the entire caudal cruciate ligament was often seen within one image slice. The patella was visualized as an intermediate bright signal (trabecular bone) surrounded by a low intensity signal (cortical bone). The trochlea and the intercondylar notch were difficult areas to analyze due to signal volume averaging of the curved surface of these areas and the presence of several types of tissues.  相似文献

Infections of the skin or subcutis of the naso-ocular region develop through two mechanisms. Cases with lesions but without concomitant signs of nasal disease probably result from cat scratch injuries. Under certain circumstances, such lacerations result in the introduction of saprophytic microorganisms in such large numbers that host defence mechanisms are overwhelmed. This results in localised, variably invasive, disease in an otherwise immunocompetent host. An unpredictable range of organisms can give rise to such infections including a variety of fungal and bacterial genera. Causal organisms will likely vary from one geography to another as a result of differences in soil type and related environmental factors. Accordingly, procurement of appropriate tissue specimens for culture and susceptibility testing is essential to guide therapy, as these cases require medical and sometimes surgical intervention in order to effect a favourable outcome. In contrast, patients with naso-ocular lesions and concurrent signs of nasal disease have a different pathogenesis. Primary infection of the sinonasal region likely results from the inhalation of infectious propagules, with the infection subsequently penetrating overlying bones to invade the subcutaneous space. These lesions are typically the result of cryptococcosis or aspergillosis and must be distinguished from invasive nasal malignancies. An approach to the investigation and treatment of these patients is presented together with photographs of representative cases.  相似文献

Cyclooxygenase‐2 (COX‐2) is overexpressed in several human and animal neoplasms, including the human endometrial carcinoma. It has been suggested as a prognostic marker and a potential therapeutic target. This study aimed to (i) clarify histological aspects of feline endometrial adenocarcinomas (FEA) of the papillary serous type and (ii) characterize COX‐2 immunohistochemical expression in normal, hyperplastic and neoplastic endometrium in this species. Archived paraffin‐embedded tissue samples of 33 FEA, eight cystic endometrial hyperplasias (CEHs) and 21 samples of normal, healthy endometrium in the follicular (FS; n = 10) and luteal (LS; n = 11) stages were evaluated. Histological evaluation of haematoxylin and eosin‐stained sections of the FEA revealed a papillary proliferation of neoplastic cells of serous type, accompanied by clear and multinucleated cells. Other architectural arrangements mainly included solid and tubular growth. Randomly distributed areas of necrosis within the tumours were commonly observed. Invasion of the myometrium, of the serosa and of the vascular and/or lymphatic vessels was not constant features. The mean number of mitoses was higher in FEA compared to non‐neoplastic endometrium. COX‐2 scores were lower in FEA (p = 0.003) and CEH (p = 0.05) when compared to normal epithelium (NE). The loss of the membrane apical reinforcement in epithelial cells was observed in FEA samples, which was accompanied by the dislocation of COX‐2 labelling into the cytoplasm and the perinuclear area; in contrast, in epithelial cells in the healthy and hyperplastic endometria, the immunoreaction showed the characteristic pattern of apical membrane reinforcement, suggestive of the membrane polarization. COX‐2 epithelial scores were higher in the FS than in the LS. No differences were found in stromal COX‐2 expression between normal, CEH and FEA groups, but it was higher in the LS than in the FS. In summary, loss of COX‐2 compartmentalization in neoplastic epithelial cells might be one of the molecular events underlying endometrial carcinogenesis.  相似文献

This study describes the enzymatic properties of an ecto-5'-nucleotidase in Trichomonas gallinae. The enzyme hydrolyzes nucleoside monophosphates at pH 7.2 and is activated by divalent cations, such as magnesium. Ecto-5'-nucleotidase activity was insensitive to levamisole, tetramisole (alkaline phosphatase inhibitors), and AMPCP (adenosine 5'-[alpha,beta-methylene]diphosphate), an ecto-5'-nucleotidase inhibitor, whereas 0.1mM ammonium molybdate (considered a potent inhibitor of 5'-nucleotidase activity) completely inhibited the enzyme activity. The apparent K(M) (Michaelis constant) and Vmax (maximum velocity) values for Mg2+-AMP were 466+/-57 microM and 3.7+/-0.59 nmolPi/min/10(6) trichomonads, respectively. Considering that trichomonads lack the ability to synthesize purines and pyrimidines de novo, the presence of an ecto-5'-nucleotidase in intact trophozoites of T. gallinae could be important in regulating the extracellular nucleotide levels and generating adenosine, essential for the survival strategies of the parasite.  相似文献

Semiarid ecosystems, also known as drylands, contain small amounts of carbon (C). To date only few studies have evaluated soil C dynamics in these ecosystems. Cyanobacterial soil crusts are considered a major source of organic C in semiarid ecosystems through photosynthesis, increasing soil organic C (SOC) pool as carbohydrates. This study considers cyanobacterial soil crusts as a source of C in the southwest Kalahari. Cyanobacterial soil crusts are well adapted to drylands because extracellular polysaccharide (EPS) secretions provide stabilization of soil and resistance to desiccation. The carbohydrate and chlorophyll a content were evaluated in the upper soil profile on Kalahari Sands and pan soils. Topsoil carbohydrate concentration decreased exponentially with depth. The carbohydrate content produced in cyanobacterial soil crusts in the southwest Kalahari represents up to 75% of the total SOC and is thus an essential component for the fertility of Kalahari Sand soils.  相似文献

Equid herpesvirus 5 (EHV-5) infection was detected in a farm in Italy by the use of a semi-nested polymerase chain reaction (PCR) targeting glycoprotein B of EHV-5 on nasal swabs and blood samples of clinically healthy and randomly selected Lipizzaner horses (n = 55). Twenty-five horses at the age of 4–17 years and 30 at an age of 1–3 years were sampled once. The association of the infection with these age-groups and the gender of the horses was investigated. The apparent prevalence of EHV-5 infection was significantly different between age-cohorts: it was higher in the younger group of horses with 73,3% and 80% positives in nasal swabs and blood respectively, compared to 40% of nasal swabs and 20% of blood in the older horses. An age-dependence therefore was observed: the young age is more frequently associated with EHV-5 infection.  相似文献

Disease foci of the conifer root and butt decay pathogen Heterobasidion irregulare are typically initiated following germination of spores deposited on freshly cut stumps, followed by expansion by tree to tree spread through root contacts and grafts. Although most abundant in autumn, inoculum of H. irregulare is also available during some cold winter periods in northern portions of the eastern United States. In both laboratory and field experiments, fresh stem discs of red pine (Pinus resinosa) were inoculated with H. irregulare conidia to test the null hypothesis that the pathogen would not survive prolonged exposure to cold temperatures experienced during winters in those regions. After exposure to cold for various periods of time, discs were incubated and then observed for presence of the Spiniger state of the pathogen. In each of two laboratory trials, the fungus survived on red pine discs incubated at ≤?20°C for 90 days. The field experiment was conducted during each of two winters at two infested red pine plantations. Inoculated discs were deployed periodically beginning in early winter and retrieved after various periods of time until spring. The Spiniger state of the pathogen was frequently observed on discs after laboratory incubation, even after field exposure of up to 6 months, during which temperatures ≤?25°C were sometimes recorded. Whether viable due to growth from inoculum initially placed on the discs, or natural inoculum deposited on discs exposed in the infested plantations, winter temperatures did not eliminate survival of the pathogen on this natural substrate. The potential for survival on stump surfaces during sub‐freezing temperatures during which application of stump protectants in water is impractical should be considered in evaluating the potential risk from H. irregulare to economically and environmentally valuable conifer forests of northcentral and northeastern North America.  相似文献
