Point‐of‐care testing (POCT) refers to any laboratory testing performed outside the conventional reference laboratory and implies close proximity to patients. Instrumental POCT systems consist of small, handheld or benchtop analyzers. These have potential utility in many veterinary settings, including private clinics, academic veterinary medical centers, the community (eg, remote area veterinary medical teams), and for research applications in academia, government, and industry. Concern about the quality of veterinary in‐clinic testing has been expressed in published veterinary literature; however, little guidance focusing on POCT is available. Recognizing this void, the ASVCP formed a subcommittee in 2009 charged with developing quality assurance (QA) guidelines for veterinary POCT. Guidelines were developed through literature review and a consensus process. Major recommendations include (1) taking a formalized approach to POCT within the facility, (2) use of written policies, standard operating procedures, forms, and logs, (3) operator training, including periodic assessment of skills, (4) assessment of instrument analytical performance and use of both statistical quality control and external quality assessment programs, (5) use of properly established or validated reference intervals, (6) and ensuring accurate patient results reporting. Where possible, given instrument analytical performance, use of a validated 13s control rule for interpretation of control data is recommended. These guidelines are aimed at veterinarians and veterinary technicians seeking to improve management of POCT in their clinical or research setting, and address QA of small chemistry and hematology instruments. These guidelines are not intended to be all‐inclusive; rather, they provide a minimum standard for maintenance of POCT instruments in the veterinary setting. 相似文献

Abstract Our objectives were to determine whether sedation with metomidate hydrochloride (hereafter, “metomidate”) during transportation of threespot gourami Trichogaster trichopterus would prevent an increase in blood glucose levels and improve fish marketability (i.e., based on appearance and behavior) in comparison with unsedated controls. Threespot gourami are obligate air-breathers that possess a labyrinth organ, enabling the fish to respire air above the water surface; these fish should be lightly sedated during transport. Fish were transported for approximately 24 h via truck and domestic airline. Blood was sampled at 0, 2, 6, and 12 h posttransportation, and appearance and behavior were observed at 0, 2, 4, 6, and 12 h and 7 d posttransportation. Metomidate concentrations tested were 0.0 (control), 0.1, 0.2, 0.3, and 0.4 mg/L. At the concentrations tested, metomidate neither inhibited elevations in blood glucose nor improved marketability. Fish that were transported with 0.3-mg/L metomidate were less marketable based on behavioral indices, and fish that were transported with 0.4-mg/L metomidate had higher glucose levels than control fish. Use of metomidate as a transport sedative for threespot gourami should be considered with caution and may be problematic at the concentrations tested; however, further research examining additional indices of stress may clarify metomidate use for this species. Enzymatic transformation of humic acids (HA), fulvic acids (FA) and indole was examined using naphthalene 1,2-dioxygenase (NDO). NDO was used as a model for dioxygenase enzymes found in various microbial species. Indole was used as a model substrate for NDO-catalyzed reactions resulting in condensation products. Although NDO is not classified as a soil enzyme, all HA and FA tested were susceptible to NDO-induced transformation. The extent of NDO-specific NADH oxidation in solutions containing HA and FA paralleled the percent aromaticity of the HA and FA. Furthermore, the UV–Vis absorptive properties of NDO-treated HA and FA were altered in a manner suggesting condensation reactions similar to the formation of indigo from indole. Condensation reactions were enhanced in NDO-treated mixtures containing indole and an FA. NDO retained activity for 2 weeks under ambient conditions, and retained some enzymatic activity for 9 days based on detection of specific metabolites by HPLC, suggesting prolonged extracellular activity. Humic substances have not previously been known to be substrates for dioxygenases; even more significant was that dioxygenase enzymes can facilitate condensation reactions between indole-like functional groups well-known to be present in HA and FA. These results illustrate how dioxygenases can be potential humic-modifying enzymes when released into the environment upon microbial death and concurrent cell lysis which could alter the bioavailability of organic contaminants associated with dissolved organic matter through specific modulation of enzyme activity involving substrate competition. 相似文献

This study tested the hypothesis that feeding an identically formulated, low sugar and starch concentrate in three forms (5-mm extruded [E], 4-mm pellet [P], and 19-mm oval [O]) would affect consumption rate and glucose or insulin responses, or both. Horses received 1.8 kg treatment feed in a randomized, crossover design, with samples taken every 30 minutes for 6 hours for blood glucose and insulin response. Pearson's correlation compared consumption time, insulin and glucose peak, and time to peak insulin and glucose. The pellet (P) elicited a lower (P = .01) glucose concentration at 2.5 hours than O. The pellet also elicited a lower (P = .03) insulin concentration at 5.5 hours than E and O. There were no differences (P > .05) in area under the curve (AUC) insulin, peak insulin, and time to peak insulin for the three treatments. Average insulin concentration was lower (P = .01) for P versus O. There were no differences (P > .05) in average insulin between P and E, nor between O and E. There were no differences (P > .05) in AUC and peak glucose concentration. Time to peak glucose was longer (P = .04) for P versus E. Average glucose concentration was lower (P = .02) for P versus O. Consumption time was longer (P = .03) for O versus P. There was a positive correlation between consumption time and time to peak insulin (r = 0.46, P = .029). Further research on feeding practices, feed forms, and consumption times that affect glycemic response is necessary. 相似文献

Differences in the cytological interpretation of bronchoalveolar lavage fluid (BALF) after cytospin preparation (CP) or manual smearing of pelleted cells preparation (MSP) were investigated in client-owned dogs and cats with inflammatory or infectious lower respiratory disease. Bronchoalveolar lavage fluid from healthy cats was also examined. With MSP, cell lysis was more frequently observed, and cellular distribution was more heterogeneous throughout the slide. When samples from healthy and diseased animals were considered together, a significantly greater percentage of neutrophils was seen on CP than on MSP slides (P<0.002). Cytospin preparations were considered of better quality in all individual comparisons. Cytospin preparation is advised in the evaluation of BALF with low total cell count. When only MSPs are evaluated, clinicians should be aware that differential neutrophil counts may underestimate the counts found on CP slides. 相似文献

Two sibling foxhounds born to a Leishmania seropositive bitch were presented after testing seropositive for Leishmania. Leishmania infantum infection was detected via histopathology, culture, and quantitative polymerase chain reaction (q-PCR).
