Over 60 different samples comprising 35 distinct honeys were evaluated for their ability to induce mammalian phase 2 detoxication enzymes using a microtiter plate assay of quinone reductase (QR) induction with murine hepatoma cells in microtiter plates. This assay has been used extensively to identify and isolate a variety of natural and synthetic inducers from plants. All 35 honeys examined induced elevations of mammalian QR activity ranging from 153 to 2155 units/g with a mean of 630 and a median of 417 units/g. The concentrations for doubling the QR activity (CD) of certain of the prominent flavonoids found in honey were also assessed (pinostrobin, 0.5 microM; pinocembrin, 110 microM; chrysin, 25 microM) and compared to those of related, more commonly described flavonoids such as quercetin (2.7 microM) and myricetin (58 microM). On the basis of the extremely high QR inducing potency of one of these compounds, pinostrobin (5-hydroxy-7-methoxyflavanone), a bioassay-guided search was conducted which revealed a dietary source of pinostrobin, Boesenbergia pandurata (fingerroot), with extraordinarily high ability to induce mammalian phase 2 detoxication enzymes. Although the QR inducing activity of buckwheat honeys was 2155 +/- 951 units/g (n = 8 samples), which is less than 10% of the average values obtained from fresh broccoli, the potency of fingerroot rhizomes (ca. 110,000 units/g) is even higher than that of broccoli and the potencies of fingerroot oil and powdered rhizome (ca. 500,000 units/g) rival that of broccoli sprouts.  相似文献

We compared four approaches to conservation site selection to protect forest biodiversity in the Triangle Region of North Carolina, USA. Using biological inventory data and an inventory-based conservation plan as benchmarks, we evaluated the potential effectiveness of a focal species plan and three “simple” plans (large forested patches, close to wetlands and riparian areas, diverse forest types). Effectiveness was measured in three ways: the number of inventory elements captured at least once by the plan (representation), the total number of inventory elements captured (completeness), and the proportion of land in the inventory-based plan included (overlap). We further examined the potential effectiveness of the simple plans by calculating their overlap with land identified by the focal species approach. The simple and focal species plans did not differ markedly in terms of representation, but diverged when completeness and overlap were considered. Although representation rates for all four plans were relatively high, lower rates for completeness and overlap raise concerns about long-term viability. The simple plans did not identify the same lands as the focal species plan, and are thus unlikely to provide appropriate habitat for the focal species. Each approach we tested failed to capture some subset of species and communities, highlighting the importance of explicit conservation targets and consideration of ecological processes. Forced to act quickly and with little data, our findings suggest using initially a set of complementary simple plans, each focused on a different habitat type. This should be considered a stopgap measure, however, while more sophisticated plans are constructed, defining explicit conservation targets and considering ecological processes.  相似文献

A monoclonal antibody dot-blot assay was used to evaluate detergent lysates of tonsil tissue from mule deer to detect PrP(CWD), the marker for the cervid transmissible spongiform encephalopathy chronic wasting disease (CWD). Samples of formalin-fixed brain and tonsil tissues from mule deer were examined for PrP(CWD) using immunohistochemistry (IHC) with Mab F99/97.6.1, the gold standard for diagnosis of preclinical CWD. The contralateral tonsil from each of the 143 deer was prepared for confirmatory IHC and as a 10% (wt/vol) detergent lysate without purification or enrichment steps for monoclonal antibody dot-blot assay. PrP(CWD) was detected by dot-blot assay in 49 of 50 samples considered positive by IHC. Forty-eight of the positive samples were evaluated with a quantitative dot-blot assay calibrated with recombinant PrP. Tonsillar PrP(CWD) concentrations ranged from 34 to 1,188 ng per 0.5 mg starting wet weight of tissue. The abundant PrP(CWD) in mule deer tonsil will facilitate development and validation of high-throughput screening tests for CWD in large populations of free-ranging deer.  相似文献

We developed a computer model to simulate carbon storage in managed slash pine plantations in Florida. The model is based on ecosystem level carbon dynamics and land use data structured by age class of planted trees. Based on a scenario of constant planting at the average level (1952–1979), Florida slash pine forests would store carbon at the rate of 2.81 × 106 t year?1. However, simulated carbon storage is sensitive to the age class distribution of the forests. There will be a 106 t year?1 decrease in carbon storage between 1980 and 1987, due to reduced levels of planting during the past decade. Above-ground biomass distribution, leaf area, above-ground net primary productivity and foliage characteristics were determined for 90- and 350-year-oldPinus edulis-Juniperus monosperma ecosystems on the Colorado Plateau of northern Arizona. These ecosystems have low biomass, leaf area and primary productivity compared with forests in wetter environments. Biomass of the 350-year-old pinyon-juniper stand examined in this study was 54.1 mg ha−1; that of the 90-year-old stand was 23.7 mg ha−1. Above-ground net primary production averaged 2.12 mg ha−1 year−1 for the young and 2.88 mg ha−1 year−1 for the mature stand; tree production was about 80% of these values for both stands. Projected ecosystem leaf area (LAI) of the stands was 1.72 m2 m−2 and 1.85 m2 m−2, respectively. Production efficiency (dry matter production per unit leaf area) was 0.129 kg m−2 year−1 for the young, and 0.160 kg m−2 year−1 for the mature stand. Production efficiency of the study sites was below the 0.188 kg m−2 year−1 reported for xeric, pure juniper stands in the northern Great Basin. Biomass of pinyon-juniper ecosystems of northern Arizona is generally below the 60–121 mg ha−1 reported for pinyon-juniper stands of the western Great Basin in Nevada. A climatic gradient with summer precipitation decreasing between southeast Arizona and northwest Nevada occurs in the pinyon-juniper region. Great Basin pinyon-juniper ecosystems lie at the dry-summer end of this gradient while pinyon-juniper ecosystems of the Colorado Plateau lie at about the middle of this gradient. In spite of wetter summers, pinyon-juniper ecosystems of northern Arizona are less productive than those of the Great Basin.  相似文献

ABSTRACT The timely and accurate detection of pathogens is a critical aid in the study of the epidemiology and biology of plant diseases. In the case of regulated organisms, the availability of a sensitive and reliable assay is essential when trying to achieve early detection of the pathogen. We developed and tested a real-time, nested polymerase chain reaction (PCR) assay for the detection of Phytophthora ramorum, causal agent of sudden oak death. This technique then was implemented as part of a widespread environmental screen throughout California. The method here described is sensitive, detecting less than 12 fg of pathogen DNA, and is specific for P. ramorum when tested across 21 Phytophthora spp. Hundreds of symptomatic samples from 33 sites in 14 California counties were assayed, resulting in the discovery of 10 new host species and 23 infested areas, including 4 new counties. With the exception of a single host, PCR-based discovery of new hosts and infested areas always was confirmed by traditional pathogen isolations and inoculation studies. Nonetheless, molecular diagnostics were key in early pathogen detection, and steered the direction of further research on this newly discovered and generalist Phytophthora species.  相似文献

Organisms in the genus Anaplasma are obligate intracellular pathogens that multiply in both vertebrate and invertebrate hosts. The type species, Anaplasma marginale, causes bovine anaplasmosis and infects erythrocytes of the vertebrate host and undergoes a complex developmental cycle in ticks which serve as biological vectors. Infected cattle, wild ruminants and ticks can all serve as reservoirs of A. marginale. In this study, hunter killed Iberian red deer (Cervus elaphus hispanicus) from the region of Castilla-La Mancha in southwestern Spain were tested for Anaplasma infection. We found that 10% of the deer examined were seropositive for Anaplasma. Three A. marginale strains were subsequently obtained from salivary glands of Hyalomma marginatum that were removed from these deer, and the sequence of the major surface protein (msp)4 gene was determined for each strain and used for phylogenetic studies. Maximum parsimony analyses of msp4 sequences from H. marginatum ticks in comparison with New World cattle and bison isolates reported previously, suggested different origins for these Spanish A. marginale strains. The results of this study demonstrated that Iberian red deer are naturally infected with Anaplasma, and may therefore serve as a wildlife reservoir of the pathogen. Although the link between deer infection and the strains of A. marginale identified in ticks was not established, H. marginatum and Rhipicephalus bursa were identified as potential biological vectors for A. marginale in this region and may effect transmission of A. marginale between deer and cattle populations.  相似文献
