We examined the impact of the effluent discharged from a freshwater (trout and related species) fish hatchery on the presence of antibiotic-resistant microorganisms in a small stream. There had been no documented use of antibiotics in the hatchery for at least 6 months prior to our study, although a variety of biocides were employed routinely for cleaning. Heterotrophic bacteria and Escherichia coli were isolated from both water column and sediment samples at sites above and below the discharge of the hatchery effluent as well as from the hatchery effluent itself. Randomly chosen isolates (≥96 isolates per site) were tested for their resistance to ampicillin, cephalexin, erythromycin, and tetracycline. Resistance to at least one antibiotic was found in greater than 30% of both the heterotrophic isolates and the E. coli isolates from each of the sites. There were no significant differences among the sites in the proportion of the heterotrophic isolates resistant to any specific antibiotic. The proportion of E. coli isolates resistant to tetracycline in the hatchery effluent and in both the downstream water and sediment samples was significantly higher than in either the upstream water or sediment. These results support the possibility of the hatchery as a source of tetracycline-resistant microorganisms even in the absence of recent use of this antibiotic.  相似文献

Reducing the current U.K. curing temperature from 28 to 20 °C would help to reduce energy costs; however, onion skin appearance and consumer acceptability may be detrimentally affected. The aim of this study was to elucidate the compounds responsible for the difference in color between brown and red onions cured at 20 and 28 °C by monitoring dynamic biochemical changes in the skin at set intervals during curing and after storage from two years' data. Sugar concentrations appeared to play no role in the difference in onion skin appearance when cured at different temperatures. Using regression, principal component, and partial least-squares discriminant analyses, the decrease in skin H° after the curing of brown onion cultivars at 28 °C was linked to a decrease in individual flavonol concentrations, possibly due to their oxidation at higher temperatures into brown pigmented compounds. Red onion cultivars cured at lower temperatures and for a shorter curing period had higher concentrations of individual anthocyanins as well as a darker skin color. Skin water content was reduced significantly in only the first 6 days of curing. Taken together, this suggests that current U.K. curing practice could be carried out at a lower temperature (20 °C) and/or for a shorter duration, resulting in reduced curing costs and possibly improved skin appearance.  相似文献

Soil microinvertebrates are closely associated with soil decomposition and nutrient cycles and may be particularly responsive indicators for soil management practices. However, identification of appropriate bioindicator species for many systems has been severely limited by a lack of information on species taxonomy, distribution, and functional role. We evaluated Indicator Species Analysis (ISA) as an objective method for assessing the indicator potential of different taxa without regard to their ecological role or expected management response. Restricting ordination and site classification to significant indicator morphotaxa reduced the dimensionality of the community data matrix by 69% while only slightly decreasing the efficiency of unsupervised classification (from 87.2 to 84.4%); the percentage of total variability explained by first two PCA axes increased following ISA. When these same indicator morphotaxa were used to classify an independent set of samples, the percentage of total variability explained by the first two PCA axes increased from 55.3 to 65.2%; cluster analysis of the test dataset correctly classified 47 out of 50 plots by cover type (94% accuracy). However, restriction of analysis to indicator morphotaxa alone reduced detection of differences between sampling dates relative to the complete dataset. Although care needs to be taken to ensure that the dataset used for indicator selection is fully representative of underlying temporal and spatial variability, ISA appears to overcome many of the limitations associated with parametric and multivariate approaches for identifying indicator morphotaxa and has the potential to greatly reduce the taxonomic expertise and labor costs associated with sorting and identification of soil microarthropods.  相似文献

A homogenous enzyme immunoassay (EIA) for measurement of serum thyroxine (T4) concentration was evaluated for use with canine and feline serum. Although Anaplasma marginale was known to be endemic in Italy, the diversity of Anaplasma spp. from this area have not been characterized. In this study, the prevalence of Anaplasma spp. antibodies in randomly selected farm animals collected on the island of Sicily was determined by use of a MSP5 cELISA for Anaplasma spp. and an immunofluorescence test specific for Anaplasma phagocytophilum. Genetic variation among strains of Anaplasma spp. from animals and ticks was characterized using the A. marginale msp1alpha and the Anaplasma spp. msp4 genes. Eight species of ticks were collected and tested by PCR. Seropositivity for Anaplasma spp. and A. phagocytophilum was detected in bovine and ovine samples. All the donkeys were seropositive for A. phagocytophilum but not for Anaplasma spp. Four A. marginale genotypes were identified by msp4 sequences from bovine and tick samples. Two new genotypes of Anaplasma ovis were characterized in sheep. The sequences of A. phagocytophilum from three donkeys proved to be identical to the sequence of the MRK equine isolate from California. Six A. marginale genotypes were found in cattle and one tick using the A. marginale msp1alpha sequences. All genotypes had four repeated sequences in the N-terminal portion of the MSP1a, except for one that had five repeats. The Italian strains of A. marginale contained three repeat sequences that were not reported previously. Definition of the diversity of Anaplasma spp. in Sicily reported, herein is fundamental to development of control strategies for A. marginale, A. ovis and A. phagocytophilum in Sicily.  相似文献

Anaplasma marginale infection in Europe has been limited to the Mediterranean and eastern countries, to Austria and to very sporadic cases in Switzerland. There are no reports of its occurrence in the countries north of Switzerland. A severe outbreak of anaplasmosis in August 2002 in a cattle farm in the canton Grisons, Switzerland, north of the Alps, with more than 300 cattle that had to be culled, came unexpected and gave reason to hypothesize presence of an increased yet undetected prevalence of A. marginale in Switzerland. Randomly selected bovine serum samples collected in 1998 and 2003 were tested using a competitive inhibitory ELISA (cELISA) to test the hypothesis. Our validation of the diagnostic sensitivity and specificity of this test, done in the outbreak herd, yielded 99.2 and 83.3%, respectively, probably underestimating the true specificity. The true seroprevalence of anaplasmosis in Swiss cattle determined by cELISA was likely to be zero with upper 95% confidence limits of 2.49% in the canton Grisons and 1.17% in the rest of Switzerland, respectively, in 1998. For 2003, these estimates were even lower. There was no significant difference in apparent prevalences between 1998 and 2003. In search of a possible reservoir, three chamoises out of 46 free ranging wild ruminants from the Swiss National Park, Grisons, tested positive in the cELISA. This reaction is in accordance with A. marginale or a cross reacting agent such as Anaplasma ovis. From our results we conclude that the hypothesis of an increased prevalence of anaplasmosis in cattle in Switzerland must be rejected.  相似文献

OBJECTIVE: To evaluate the long-term risks and benefits of early-age gonadectomy, compared with traditional-age gonadectomy, among cats adopted from a large animal shelter. DESIGN: Retrospective cohort study. ANIMALS: 1,660 cats. PROCEDURE: Cats underwent gonadectomy and were adopted from an animal shelter before 1 year of age; follow-up was available for as long as 11 years after surgery (median follow-up time, 3.9 years). Adopters completed a questionnaire about their cats' behavior and medical history. When possible, the cats' veterinary records were reviewed. Statistical analyses were conducted to identify any associations between the occurrence of 47 medical and behavioral conditions and the cats' age at gonadectomy. RESULTS: Among male cats that underwent early-age gonadectomy (< 5.5 months of age), the occurrence of abscesses, aggression toward veterinarians, sexual behaviors, and urine spraying was decreased, whereas hiding was increased, compared with cats that underwent gonadectomy at an older age. Among male and female cats that underwent early-age gonadectomy, asthma, gingivitis, and hyperactivity were decreased, whereas shyness was increased. CONCLUSIONS AND CLINICAL RELEVANCE: Gonadectomy before 5.5 months of age was not associated with increased rates of death or relinquishment or occurrence of any serious medical or behavioral condition and may provide certain important long-term benefits, especially for male cats. Animal shelters can safely gonadectomize cats at a young age, and veterinarians should consider recommending routine gonadectomy for client-owned cats before the traditional age of 6 to 8 months.  相似文献
