Disease prevalence studies are one of the most valuable tools to demonstrate the risk or impact of certain infections in local and global economies. The data obtained in these studies contribute to develop strategies for disease control. The present study aims to provide information about the prevalence of babesiosis and anaplasmosis in the northern regions of Sudan. Blood samples from four different states of Sudan were collected from apparently healthy cattle (n=692), DNA was extracted and the prevalence of Babesia and Anaplasma species was analyzed by PCR. The results confirmed the presence of Babesia bigemina, Babesia bovis and Anaplasma marginale in cattle in northern Sudan with overall prevalence rates of 4.0%, 1.9% and 6.1%, respectively. Statistical analysis revealed that the prevalence of B. bigemina, B. bovis and A. marginale varies significantly between Sudanese states as well as in different age groups, while gender seems not to have a significant effect on the prevalence of these pathogens among Sudanese cattle. The highest prevalence for B. bigemina was found in the Aljazirah State while the highest number of A. marginale positive samples was reported in River Nile.  相似文献

Phylogeny is an evolutionary reconstruction of the past relationships of DNA or protein sequences and it can further be used as a tool to assess population structuring, genetic diversity and biogeographic patterns. In the microbial world, the concept that everything is everywhere is widely accepted. However, it is much debated whether microbes are easily dispersed globally or whether they, like many macro-organisms, have historical biogeographies. Biogeography can be defined as the science that documents the spatial and temporal distribution of a given taxa in the environment at local, regional and continental scales. Speciation, extinction and dispersal are proposed to explain the generation of biogeographic patterns. Cyanobacteria are a diverse group of microorganisms that inhabit a wide range of ecological niches and are well known for their toxic secondary metabolite production. Knowledge of the evolution and dispersal of these microorganisms is still limited, and further research to understand such topics is imperative. Here, we provide a compilation of the most relevant information regarding these issues to better understand the present state of the art as a platform for future studies, and we highlight examples of both phylogenetic and biogeographic studies in non-symbiotic cyanobacteria and cyanotoxins.  相似文献

Vascular endothelial growth factor (VEGF) is a major regulator of angiogenesis and a potential autocrine growth factor for neoplastic cells in various malignancies. In the present study, we have investigated expression of VEGF and VEGF receptors in canine mastocytomas and the canine mastocytoma cell line C2. As assessed by immunostaining of tissue sections and cytospin slides, primary neoplastic mast cells (MC) and C2 cells were found to express the VEGF protein. In Northern blot and RT-PCR experiments, C2 cells expressed VEGF mRNA in a constitutive manner. VEGF mRNA expression in C2 cells was counteracted by LY294002 and rapamycin, suggesting involvement of the PI3-kinase/mTOR pathway. Moreover, C2 cells were found to express VEGF receptor-1 (Flt-1) and VEGF receptor-2 (KDR). However, recombinant VEGF failed to promote (3)H-thymidine uptake in C2 cells, and a neutralizing anti-VEGF antibody (bevacizumab) failed to downregulate spontaneous proliferation in these cells. In addition, rapamycin decreased the expression of VEGF in C2 cells at the mRNA and protein level without suppressing their proliferation. Together, canine mastocytoma cells express VEGF as well as VEGF receptors. However, despite co-expression of VEGF and its receptors, VEGF is not utilized as an autocrine growth regulator by canine mastocytoma cells.  相似文献

The objective of this study was to investigate the effects of acute exposure to copper, chromium and mixtures of these metals on reproductive parameters of female Carcinus maenas. The parameters studied were ovarian esterase, acid and alkaline phosphatase activity, ovarian protein content, gonadosomatic index (GSI) and hepatosomatic index (HSI). Metals induced significant decreases on ovarian alkaline phosphatase activity, esterase activity and protein content. Ovarian acid phosphatase activity significantly increased in crabs exposed to chromium, while only a slightly increase was found in females exposed to copper and to mixtures of these metals. Furthermore, a dose-dependent reduction of both GSI and HSI were observed in crabs exposed to metals. These observations suggest that copper and chromium may interfere with the ovarian cycle in Carcinus maenas and, therefore, with the reproduction of this species. In general, the mixture had more pronounced effects than individual metals. This subject requires further investigation because in polluted estuaries, crabs may be simultaneously exposed to several metals.  相似文献

We present a segmented partial least squares (PLS) prediction model for firmness of ‘Rocha’ pear (Pyrus communis L.) during fruit ripening under shelf-life conditions. Pears were collected from three different orchards. Orchard I provided the pears for model calibration and internal validation (set 1). These were transferred to shelf-life in the dark at 20 ± 2 °C and 70% RH, immediately after harvest. External validation was performed on the pears from the other two orchards (sets 2 and 3), which were stored under different conditions before shelf-life. Fruit was followed in the shelf-life period by visible/near infrared reflectance spectroscopy (Vis/NIRS) in the range 400–950 nm. The correlation between firmness and the reflectance at some wavelength bands was markedly different depending on ripening stage. A segmented partial least squares model was then constructed to predict firmness. This PLS model has two segments: (1) unripe and ripening/ripe pears (high firmness); (2) over-ripe pears (low firmness). The prediction is done in two steps. First, a full range model (full model) is applied. When the full model prediction gives a low firmness value, then the over-ripe model is applied to refine the prediction. The full model is reasonably significant in regression terms, robust, but allows only a coarse quantitative prediction (standard deviation ratio, SDR = 2.48, 1.50 and 2.40 for sets 1, 2 and 3, respectively). Also, RMSEP% = 139%, 91% and 56%, indicating large relative errors at low firmness values. The segmented model improved moderately the correlation, and the values of RMSEC, RMSEP and SDR; it improved significantly the RMSEP% (29%, 55% and 31%), providing an improvement of the relative prediction errors at low firmness values. This method improves the ordinary PLS models. Finally, we tested whether chlorophyll alone was enough for a predictive model for firmness, but the results showed that the absorption of chlorophyll alone does not explain the performance of the PLS models.  相似文献

The denitrification potential of the soil horizons between 0- and 90-cm depth of 20 agricultural fields, representative of the most frequent combinations of agricultural crops and soil textures in Flanders (Belgium), and the factors affecting the denitrification potential were studied in the laboratory under controlled conditions. The denitrification potential in the presence of an added soluble C and N source was measured at 15°C after saturation of air-dried soil samples with water. The denitrification potential of the lower horizons was generally negligible compared to the upper horizons. The lower denitrification potential of the deeper horizons could partially be explained by their limited C availability. The denitrification potential of the upper horizons strongly depended on texture. Based on this parameter the soils could be divided into three groups: soils with a high clay content (>30% clay) were characterised by a high denitrification potential (>8.33 µg N g-1 dry soil day-1); soils with medium texture had a medium denitrification potential, between 0.41 and 7.25 µg N g-1 dry soil day-1; and soils with a high sand content (>80% sand) had a low denitrification potential (<2.58 µg N g-1 dry soil day-1). In most cases, extending the saturation period during pre-incubation increased the denitrification potential. Comparison of the denitrification potential of the upper horizons with and without addition of a soluble C source showed that the denitrification potential of the upper horizons of these soils was limited by their percentage of endogenous C. The measured denitrification potentials indicate that denitrification losses in soils high in clay content can be important when NO3 - concentrations are high.  相似文献

Purpose

Standard assays for phytotoxicity provide a reductionist view on the performance of plants under toxic stress. To address two of the most prominent issues in plant toxicity studies, our aims were (1) to assess how well physiological and biochemical parameters complement standard toxicological endpoints when testing natural soils and (2) to assess the suitability of three commonly used control soils as comparative references.

Material and methods

We compared the performance of Zea mays and Helianthus annuus in three control soils (artificial Organisation for Economic Co-operation and Development (OECD) soil, standard LUFA 2.2 soil, and turf-perlite) against three natural soils representing a gradient of contamination (from a deactivated uranium mine). Standard endpoints (emergence and biomass) were estimated, along with pigment content, photosynthetic parameters, cellular injury, and proline content.

Results and discussion

The toxicological profile of natural soils was highly dependent on the control soil used as reference; also, plant physiological performance was influenced by the soils’ properties. We discuss the need to interpret and combine multiple lines of evidence as a way to increase the degree of confidence one classifies soils based on their ecotoxicity, and this is where the integration of physiological and biochemical parameters bring added value.

Conclusions

When facing large variability in soil characteristics, it is best to collect and integrate as much information possible to strengthen conclusions about phytotoxicity of natural soils. Obviously, this refutes reductionist views and places the final conclusion in the hands of expert judgment.

Chitosan is widely used in the biomedical field due its chemical and pharmacological properties. However, intake of chitosan results in renal tissue accumulation of chitosan and promotes an increase in calcium excretion. On the other hand, the effect of chitosan on the formation of calcium oxalate crystals (CaOx) has not been described. In this work, we evaluated the antioxidant capacity of chitosan and its interference in the formation of CaOx crystals in vitro. Here, the chitosan obtained commercially had its identity confirmed by nuclear magnetic resonance and infrared spectroscopy. In several tests, this chitosan showed low or no antioxidant activity. However, it also showed excellent copper-chelating activity. In vitro, chitosan acted as an inducer mainly of monohydrate CaOx crystal formation, which is more prevalent in patients with urolithiasis. We also observed that chitosan modifies the morphology and size of these crystals, as well as changes the surface charge of the crystals, making them even more positive, which can facilitate the interaction of these crystals with renal cells. Chitosan greatly influences the formation of crystals in vitro, and in vivo analyses should be conducted to assess the risk of using chitosan.  相似文献

This study investigated the in vivo genotoxicity of piquiá pulp (Caryocar villosum) and its potential antigenotoxicity on doxorubicin (DXR)-induced DNA damage by comet assay and micronucleus test. In addition, the phytochemicals present in piquiá pulp were determined. Piquiá fruit pulp (75, 150 or 300?mg/kg b.w.) was administered by gavage to Wistar rats for 14?days, and the animals received an injection of saline or DXR (15?mg/kg b.w., i.p.) 24 h before they were euthanized. The phytochemical analysis revealed the presence of carotenoids; phenolic compounds, including flavonoids; tannins and α-tocopherol in piquiá pulp. No statistically significant differences were observed in the evaluated parameters, demonstrating the absence of cytotoxic and genotoxic effects of piquiá pulp at all tested doses. In liver, kidney, cardiac and bone marrow cells, piquiá significantly reduced the DNA damage induced by DXR. Our results showed that the lowest piquiá dose caused the largest decrease in DNA damage and the highest dose caused the smallest decrease, demonstrating an inverse dose-response of piquiá pulp. Furthermore, we observed a difference in the potential antigenotoxic effects in several tissues. In conclusion, our results demonstrated that piquiá pulp was not genotoxic and inhibited the genotoxicity induced by DXR, but some of the protective effects that were observed depended on the doses and experimental conditions. Therefore, further investigations are needed to clarify how piquiá pulp positively affects human health.  相似文献
