Soil samples were collected from rhizosphere of litchi-growing areas of North-Western Himalayan Region (NWHR) of India, for finding qualitative and quantitative differences in arbuscular mycorrhizal (AM) fungi and Azotobacter chroococcum. These samples were taken from plants being grown in different cultivation types namely, weed control with weedicides or tillage; orchard floor either clear or with cover crops; intercropping with cereals and legumes. Qualitative and quantitative differences were noticed with different cultivation types and a marked reduction in the AM fungi was observed in orchards where chemicals were used for weed control and intensive farming system was used on the orchard floor. AM fungi were generally abundant in the soils with range pH 5.5–6.6. Among different AM fungi retrieved from the soils, Glomus spp. was most dominant. Fifteen AM fungal species were isolated, identified and characterized and along with their ability to colonize the roots. In the soil samples, a marked variation in viable bacterial count of A. chroococcum was also noticed due to varied physico-chemical characteristics of the orchard soils. The changes in AM fungal species composition can be attributed to changes in soil chemical properties resulting from cultural practices such as ploughing, application of chemical fertilizers and weedicides. An experiment was also conducted to study the comparative efficacy of four dominant and frequently occurring indigenous AM species namely, Glomus fasciculatum (Thaxter sensu Gerdemann), G. magnicaulis (Hall), G. mosseae (Nicol. & Gerd.), Gigaspora heterogamma (Nicol. & Gerd.) and two A. chroococcum strains viz., AZ1 and AZ2 singly and in dual combination to evaluate their effect in air-layers system. Dual inoculation of G. fasciculatum and AZ1 increased total root length of air-layered shoots by 81.39% over uninoculated control. These studies indicated that indigenously isolated AM fungal species and A. chroococcum strains can be used for air-layering for better adaptation under specific agro-climatic and ecological zone conditions.  相似文献

Background:Gut microbiota plays a key role in health,immunity,digestion,and production in layers.Factors such as environment,diet,diseases,stress,and flock management significantly affect gut microbiota;however,it is not known how potential stressors such as intramuscular injections or feed withdrawal alter the composition of gut microbiota that result in increased the shedding level of foodborne pathogens.In the current study,the effects of intramuscular corticosterone injection and feed withdrawal were evaluated to understand their role in Salmonella Typhimurium shedding and changes in the composition of gut microbiota in layers.Results:Salmonella shedding was observed for 8 weeks post-infection.There was a significant increase in Salmonella Typhimurium count after intramuscular injection and feed withdrawal.The Salmonella infected and the negative control groups showed significant differences in the abundance of different genera in gut microbiota at week 1 and up to week 7 post infection.The infected group showed a significant reduction in alpha diversity of gut microbiota.Firmicutes reduced significantly(P<0.05)after intramuscular injection,while the feed withdrawal groups did not cause any significant changes in Proteobacteria-Firmicutes ratio.Furthermore,intramuscular injection resulted in a significant change in alpha diversity of gut microbiota.Conclusions:Exposure of chicks to relatively low dose of Salmonella Typhimurium can lead to persistent shedding in pullets.The Salmonella Typhimurium infection disrupted the gut microbiota composition immediately after infection.The potential stress of intramuscular injection and feed withdrawal significantly increased the Salmonella Typhimurium count in faeces.The intramuscular injection also resulted in a significant alteration of the Proteobacteria-Firmicutes ratio,which could increase the risk of dysbiosis.  相似文献

The present study reports the first survey for the detection of antirabies antibodies in street and household dogs in India. We aimed to check the efficacy of control programs for the disease in the Union territory of Chandigarh. The serum samples were collected from 100 street and 50 household dogs and tested for the presence of antirabies antibodies by ELISA. As per WHO criteria, a titre of >0.5 IU/ml of antirabies antibody in serum samples was taken as the protective level. Protective antirabies antibody titre was found only in 1% of the street dogs and 16% of the pet dogs. The awareness among the pet dog owners about the antirabies vaccination schedule was low as 18% did not know the vaccination status of the dog and 66% had got the initial immunization done with only three doses and annual boosters were not given. A National Rabies Elimination Program needs to be launched as a collaborative venture by both medical and veterinary practitioners to curb this deadly disease. Also periodic surveys to test the status of antirabies IgG among dogs need to be carried out to ascertain the attainment of WHO protective levels  相似文献

Context

East Indian sandalwood (Santalum album L.) in commercial markets is highly prone to adulteration. A number of cases were registered with regard to the adulteration of East Indian sandalwood, but the lack of technical tools for the precise species identification of the source wood stalled most of the court cases.

Aims

The standard DNA barcode regions, the rbcL, matK and trnH-psbA chloroplast genomic sequences recommended by the Consortium of Barcode of Life (COBOL) were analysed to distinguish wood adulterants of East Indian sandalwood.

Methods

Standard polymerase chain reactions with COBOL recommended primers were performed for all three barcode loci. The PCR products after gel elution were sequenced and alignments were performed using CLUSTALX.

Results

Single nucleotide polymorphisms (SNPs) identified with rbcL and trnH-psbA sequences of Erythroxylum monogynum Roxb. as well as with matK sequences of Osyris wightiana Wall ex. Wight could be efficiently utilized for the detection/monitoring of East Indian sandalwood adulterants. Among the two common adulterants O. wightiana and E. monogynum, the former was more similar to S. album and grouped together in the dendrogram.

Conclusion

The study recommends the exploitation of DNA barcoding technique using standard barcodes to trace sandalwood timber adulterants.  相似文献

Phytochemical investigation of Annona squamosa twigs, resulted in isolation and identification of twelve known (1-12) compounds among them one 1-(4-β-D-glucopyranosyloxyphenyl)-2-(β-D-glucopyranosyloxy)-ethane (11) is synthetically known but first time isolated from natural sources. Their structures were elucidated using 1D and 2D NMR spectroscopic analysis. The isolated compounds (2-8, 11) were evaluated for H+ K+-ATPase activity. Three of these compounds (+)-O-methylarmepavine (2), N-methylcorydaldine (3), isocorydine (6) showed promising anti-secretory activity. Activity of these compounds, comparable to the standard drug omeprazole is novel to our finding. Moreover, there is no information accessible regarding the pharmacological effect of A. squamosa on the gastrointestinal system. This study is the first of its kind to show the significant anti-ulcer effect of A. squamosa. The present study aimed to evaluate the gastroprotective effect of A. squamosa (AS) and to identify its active constituents. Anti-ulcer activity was evaluated against cold restraint (CRU), pyloric ligation (PL), aspirin (ASP), alcohol (AL) induced gastric ulcer and histamine (HA) induced duodenal ulcer model and further confirmed through in vitro assay of H+ K+-ATPase activity and plasma gastrin level. AS and its chloroform and hexane fraction attenuated ulcer formation in CRU, PL, HA model and displayed anti-secretory activity in vivo through reduced free, total acidity and pepsin in PL, confirmed by in vitro inhibition of H+ K+-ATPase activity with corresponding decrease in plasma gastrin level. Cytoprotection of AS was apparent with protection in AL, ASP models and enhanced mucin level in PL.  相似文献

A case of an enteric coronavirus infection in a 6-week-old dromedary calf is described. The animal had diarrhea for 5 days and died despite symptomatic treatment. Numerous viral particles, approximately 140 nm in diameter, with club-like projections were detected in the feces by electron microscopy. These characteristics were consistent with a coronavirus. Immunohistochemical reactivity with 2 antigenic group II coronavirus-specific antibodies confirmed the presence of viral antigen in colonic epithelial cells. The death of the animal was attributed to a neutrophilic and emphysematous colitis that likely was caused by an infection with a Clostridium sp.  相似文献

Populations of wild Beta L. species exist as weeds in commercial sugar beet (Beta vulgaris L. subspecies vulgaris) fields in the Imperial Valley, California. Significant losses to sugar yield and quality result if these wild plants are not removed. In cases of extreme infestation, fields are abandoned without harvest. No selective chemicals are available to differentiate conventional sugar beet from wild relatives and hand removal is labor intensive and expensive. Planting sugar beet varieties with tolerance to glyphosate is a potential solution for infested fields, but risk of gene flow to adjacent wild relatives must be determined. Previous research identified these populations as either Beta vulgaris L. subspecies maritima (L.) Arcang. or Beta macrocarpa Guss. This distinction is critical because B. v. subsp. maritima will readily cross hybridize with cultivated sugar beet while B. macrocarpa rarely will. In April 2011, we collected herbarium samples, mature seed, and leaf tissue from wild Beta populations in 25 infested sugar beet fields throughout the Imperial Valley. Bolting cultivated beets were identified at two locations. Taxonomy of whole plant herbarium samples was unclear due to wild beet stem elongation when under competition with sugar beet plants for canopy light. Morphology of plants from collected seed grown in non-competitive conditions assigned taxonomy of these populations to B. macrocarpa. We used molecular tools to determine the genetic structure of wild Beta populations throughout the Imperial Valley. Extracted DNA was genotyped with 22 simple sequence repeat molecular markers and evaluated for population structure. The bolting beet samples were clearly separated from the majority of B. macrocarpa samples, except for two. The remaining wild populations were further divided into two subgroups suggesting exchange of genetic information or a common ancestor.  相似文献

The genetic correlations for age at first egg, part‐year egg production, egg weight and body weight at 20 weeks were high and positive in pure‐breds and their cross‐bred half‐sibs. This indicates that the performance of the cross between White Leghorn and Rhode Island Red can be further improved through intra‐population selection for these characters in the two parental breeds.  相似文献

Genetic analysis of sodium and sodium/potassium (Na/K) ratios in leaf and stem was carried out through diallel analysis involving two tolerant, one moderately tolerant, and two sensitive genotypes as parents. Three-week-old seedlings were subjected to a critical level of salinization (17 decisiemens per meter of electrical conductivity). Leaf and stem sampling (from 5 parents and 20 hybrids arranged in a randomized block design with three replications of 10 each) was done 30 days after salinization when susceptible parents were severely effected. Predominance of non-additive gene action for stem sodium and dominance component for leaf Na/K were noticed while both additive and non-additive components played a significant role for stem Na/K. A single group of genes seems to be operative for these characters. Hybrids produced by crossing the two sensitive parents were tolerant suggesting genetic complementation and involvement of different loci in the two parents for salinity tolerance. The overall dominant nature of tolerance and the additive gene action for these salinity related characters suggested the possibility of breeding pearl millet lines through hybridization and selection to pyramid the favorable genes.  相似文献
