Tannin-protein complexes as radical scavengers and radical sinks   总被引:8,自引:0,他引:8  
The 2,2'-azinobis(3-ethylbenzothiazoline 6-sulfonic acid) radical cation (ABTS(*)(+)) decolorization assay has been used to determine the antioxidant activity of the polyphenol epicatechin(16) (4 --> 8) catechin (procyanidin, PC) alone or in complex with the model proteins bovine serum albumin (BSA) or gelatin. PC had a molar antioxidant capacity of approximately 54, 92, or 108 radicals at pH values of 3.0, 4.9, or 7.4, respectively. Radical scavenging occurred via a rapid step followed by a slow step. Interaction with gelatin reduced the rate of rapid scavenging by 50% (PC-BSA mixtures reduced by 15%). Inhibition paralleled formation of precipitable PC-protein complexes over a range of protein/PC ratios. However, inhibition was virtually overcome in 10 min. Reaction with ABTS(*)(+) converted the PC-protein complexes from a dissociable form to a form resistant to dissociation by strong denaturants such as SDS. This study demonstrates that PC is a potent ABTS(*)(+) scavenger even when bound to protein and that the complexes may act as a radical sink within the gastrointestinal tract.  相似文献

Milled wood and milled wood lignin (MWL) samples were subjected to DFRC and thioacidolysis. Despite the fact that both methods selectively cleave aryl ether bonds, substantial differences in results were obtained. Lignin thioacidolysis gave total molar yields of degradation monomer products in the range of 3.5-7 mol % higher than DFRC. GPC analysis showed that the thioacidolysis-treated lignin was degraded to a lower average molecular weight than that treated by DFRC. Contrary to results reported for lignin model compounds, these results indicate that the DFRC method does not completely or efficiently degrade the lignin polymer. In fact, the DFRC-degraded lignin retained much of the characteristics of the original MWL. Elemental analysis revealed the presence of bromine in the DFRC-treated lignin, and two-dimensional (1)H-(13)C HMQC NMR spectroscopy showed the presence of beta-O-4 linkages in the DFRC-treated lignin. No beta-O-4 interunit linkages were detected in the thioacidolysis-treated lignin. These results are consistent with the lower monomer yields and the higher average molecular weight of the DFRC-treated lignin and indicate inefficiency in the chemistry of the method, probably due to steric constraints of the polymeric nature of lignin.  相似文献

A procedure has been developed and validated for measuring the concentration of pentobarbital residues in dry, extruded animal feed in the range of 3-200 ng/g (ppb) with an estimated limit of quantitation of 2 ppb. The method was developed for surveillance purposes: to measure the concentration of euthanizing agent which might be present in feeds incorporating rendered products which themselves might include some fraction of euthanized animals. A previously published qualitative procedure was modified by adding isotopically labelled pentobarbital as an internal standard. Dry feed was ground and extracted with methanol. The extract was loaded on a mixed-mode (C-18, anion exchange) solid-phase extraction cartridge designed for barbiturate residues. Pentobarbital was eluted and derivatized for gas chromatography/mass spectrometry in positive ion chemical ionization mode. Quantitation was based on the ratio of dimethyl-pentobarbital MH+ (m/z 255) vs dimethyl-pentobarbital-d(5) (m/z 260) in standards and extracts. Accuracy ranged from 112% at 3 ppb to 96% at 200 ppb, with relative standard deviations ranging from 4% at 3 ppb to 2% at 200 ppb.  相似文献

High-energy x-ray fluorescence measurements were used to make elemental maps and qualitative chemical analyses of individual Pseudomonas fluorescens strain NCIMB 11764 cells. Marked differences between planktonic and adhered cells were seen in the morphology, elemental composition, and sensitivity to Cr(VI) of hydrated cells at spatial scales of 150 nm. Polar ice-core records suggest that an arctic or boreal source was responsible for more than 30% of the large increase in global atmospheric methane (CH4) concentration during deglacial climate warming; however, specific sources of that CH4 are still debated. Here we present an estimate of past CH4 flux during deglaciation from bubbling from thermokarst (thaw) lakes. Based on high rates of CH4 bubbling from contemporary arctic thermokarst lakes, high CH4 production potentials of organic matter from Pleistocene-aged frozen sediments, and estimates of the changing extent of these deposits as thermokarst lakes developed during deglaciation, we find that CH4 bubbling from newly forming thermokarst lakes comprised 33 to 87% of the high-latitude increase in atmospheric methane concentration and, in turn, contributed to the climate warming at the Pleistocene-Holocene transition.  相似文献
