A new rice mutant Suweon 464 (S-464), which has extreme contrast in cooking quality and physicochemical properties from those of its mother variety Ilpumbyeo (IP), revealed striking differences in ultrastructure in in situ, fractured whole grain, and isolated starch preparation. In scanning electron microscopy (SEM), compound starch granules (CSG) in whole grains of IP were readily split during fracturing, whereas those in S-464 were structurally intact and were enclosed within a sac-like structure tolerant of fracturing. In isolated preparation, IP starch consisted entirely of individual starch granules, whereas S-464 starch consisted of mostly large CSG enclosed within the sac, preventing the release of the individuals. In transmission electron microscopy (TEM), S-464 starch granules were smaller, solidly "condensed", and highly contrasted, whereas those of IP were larger, loosely "diffused", and less contrasted. The boundaries of amyloplasts and starch granules in S-464 were coated with a thin proteinaceous layer, presumed to be the counterpart of the sac that enclosed the CSG observed in SEM.  相似文献

A one-dimensional mathematical model, termed sediment transport rate-based model, is developed for determining rainfall-induced soil erosion and sediment transport. The model is comprised of (1) the kinematic-wave equation for overland flow, (2) a transport rate-based advection equation for rainfall-induced soil erosion and sediment transport, and (3) a semi-Lagrangian algorithm for numerical solution of the soil erosion and sediment transport equation. A series of soil flume experiments under simulated rainfalls were conducted to simulate the overland flow and sediment transport and to test the sediment transport rate-based model. Numerical results of sediment transport rate-based model indicate that (i) hydrographs display an initial rising limb, followed by a constant discharge and then a recession limb; (ii) sediment transport rate graphs exhibit the distributions similar to the hydrographs; and (iii) sediment concentration graphs show a steep-receding limb followed by a constant distribution and a receding tail. The numerically simulated hydrographs, sediment transport rate and concentration distributions are in good agreement with those measured in laboratory experiments, demonstrating the efficacy of the transport rate-based model.  相似文献

Active compounds with antidiabetic potential were isolated from silk peptide E5K6 by consecutive ultrafiltration and gel filtration using Biogel P-2 and RS-HPLC using a YMC-Pack Pro C18 column. The highest α-glucosidase inhibitory activity of silk peptide E5K6 resulted from fractions with MW <1 kDa. The activities of gel-filtered fractions from silk peptide E5K6 of <1 kDa were assayed in vitro, demonstrating that the fourth peak (F4) had the highest α-glucosidase inhibitory activity (IC(50) = 37.1 mg/mL). F4 of silk peptide E5K6 was separated by HPLC into two peaks. Moreover, the purified compounds were identified as Gly-Glu-Tyr (GEY, MW = 367 Da) and Gly-Tyr-Gly (GYG, MW = 295 Da) according to amino acid sequences, and their α-glucosidase inhibitory activities (IC(50)) were 2.7 and 1.5 mg/mL, respectively.  相似文献

Muffins containing different amounts and molecular weights (MW) of β‐glucan were evaluated for the effect of β‐glucan on the physical characteristics of the muffins and on in vitro bile acid binding and fermentation with human fecal flora. Wheat flour muffins were prepared with the addition of β‐glucan extracts with high‐, medium‐, or low‐MW. For oat flour muffins, the native oat flour contained high‐MW β‐glucan; the oat flours were treated to create medium‐ and low‐MW β‐glucan within the prepared muffin treatments. For each 60‐g muffin, the amounts of β‐glucan were 0.52, 0.57, and 0.59 g for high‐, medium‐, and low‐MW β‐glucan wheat flour muffins, and 2.38, 2.18, and 2.23 g for high‐, medium‐, and low‐MW β‐glucan oat flour muffins, respectively. The lower the MW of the β‐glucan in muffins, the lower the height and volume of the muffins. The oat flour muffins were less firm and springy than the wheat flour muffins as measured on a texture analyzer; however, MW had no effect on muffin texture. The oat flour muffins bound more bile acid than did the wheat flour muffins. The muffins with high‐MW β‐glucan bound more bile acid than did those with low‐ and medium‐MW β‐glucan. Muffin treatment affected the formation of gas and total short‐chain fatty acids (SCFA) compared with the blank without substrate during in vitro fermentation. There were no differences in pH changes and total gas production among muffin treatments. The high‐MW β‐glucan wheat flour muffins produced greater amounts of SCFA than did the wheat flour muffin without β‐glucan and the oat flour muffins; however, there were no differences in SCFA production among muffins with different MW. In general, the β‐glucan MW affected the physical qualities of muffins and some potential biological functions in humans.  相似文献

Phytoremediation is an attractive, economic alternative to soil removal and burial methods to remediate contaminated soil. However, it is also a slow process. The effect of humic acid in enhancing B and Pb phytoextraction from contaminated soils was studied (pot experiment) using transplanted vetiver grass (Vetiveria zizanioides (L.) Nash). Boron was applied at 0, 45, 90 and 180 kg B ha?1 soil (as H3BO3) in 16 replicates. Of the 64 pots, four pots each were treated with 0, 100, 200 and 400 kg ha?1 humic acid (HA) solution. In a separate experiment, Pb was applied (as Pb(NO3)2) at 0, 45, 90 and 180 kg Pb ha?1 prior to addition of HA solutions at levels identical to the B experiment. Experiments were conducted using a randomized complete block design with four replicates. Vetiver grass was harvested 90 days after planting. Lead addition beyond 45 kg Pb ha?1 decreased Pb uptake mostly due to a yield decline. Humic acid application increased Pb availability in soil and enhanced Pb uptake while maintaining or enhancing yield. An application of 200 kg HA ha?1 was optimal for maintaining yield at elevated Pb levels. Boron application did not impact yield but greatly increased B content of roots and shoot. Boron uptake was greatest upon addition of 400 kg HA ha?1. We conclude that HA addition to vetiver grass can be an effective way to enhance phytoremediation of B and Pb but optimum rates differ depending on soil B and Pb contamination levels.  相似文献

The precipitation and adsorption of pentachlorophenol (PCP) on allophane was confirmed by adsorption experiments carried out at various pll values. The precipitation of PCP took place around base-unsaturated clay particles when the concentration of PCP exceeded solubility. The adsorption of PCP involves anion exchange reaction as well as physical adsorption due to van der Waals' force, and the PCP adsorbed as anions is released with greater when washed with deionized water.

Allopbane of Andosols appears to adsorb PCP largely as anions, while humus, balloysite, and allophane (molar SiO2/Al2O3 ratio about 2) seem to adsorb PCP mostly as molecules. The precipitation of PCP in mlcell might participate in the adsorption. PCP mixed with layer silicate day minerals such as illite, montmorillonite, and kaolinite sublimated by about 200°C. In contrast to this, PCP mixed or adsorbed on allophane did not sublimate by 200°C but burnt out between 250 and 500°C showing a strong exothermic reaction.  相似文献

In this study, the applications of mesoporous materials based on silica, and those with modifications, namely post-synthetic grafting, co-condensation, and pure SBA-15, were investigated for the removal of phosphate from sewage. The mesostructures were confirmed by X-ray diffraction, Brunauer?CEmmett?CTeller, Fourier transform infrared spectroscopy, and transmission electron microscopy. The absorption of phosphate by the mesoporous adsorbents was examined, using different adsorption models to describe the equilibrium and kinetic data. The maximum adsorption capacities of the mesostructured adsorbents were found to be 69.970, 59.890, and 2.018?mg/g for the co-condensation, post-synthetic grafting, and pure SBA-15, respectively. The kinetic data showed that the adsorption of phosphate onto three different mesostructures followed the pseudo-first-order kinetic model.  相似文献

This study was conducted to determine the lipolytic effects of eight kinds of citrus peel oils and their components. All of the citrus peel oils revealed lipolytic effects on olive oil model solution ranging from 10.9 to 73.8%. Hakyul (Citrus natsudaidai Hayata) showed the highest lipolytic effect (73.8%), followed by yuza (Citrus junos Sieb. ex Tanaka, 68.1%) and lemon (Citrus limonium, 63.4%), and their effects were comparable with or stronger than that of 5 mM raspberry ketone (p < 0.05). Among 17 authentic compounds relating to citrus peel oils, octanal (78.6%) showed the highest lipolytic effect, followed by gamma-terpinene (76.3%), limonene (75%), terpinen-4-ol (70.7%), nerol (69.9%), p-cymene (67.7%), and geranyl acetate (67.2%), and their effects were stronger than that of 5 mM raspberry ketone (p < 0.05). Ethyl acetate, alpha-pinene, myrcene, citronellal, linallyl acetate, and citronellol exhibited poor lipolytic effect in the model solution. Lipolytic effect was found to be high when the oils included a higher content of gamma-terpinene and p-cymene. Limonene showed potential lipolytic effect, and its effect is likely to be enhanced by the presence of gamma-terpinene and p-cymene. It is considered that monoterpene hydrocarbons consisting of one or two double bonds would have stronger lipolytic effect than those having three double bonds.  相似文献

Three major oat components, β-glucan, starch, and protein, and their interactions were evaluated for the impact on viscosity of heated oat slurries and in vitro bile acid binding. Oat flour from the experimental oat line "N979" (7.45% β-glucan) was mixed with water and heated to make oat slurry. Heated oat slurries were treated with α-amylase, lichenase, and/or proteinase to remove starch, β-glucan, and/or protein. Oat slurries treated with lichenase or lichenase combined with α-amylase and/or proteinase reduced the molecular weight of β-glucan. Heat and enzymatic treatment of oat slurries reduced the peak and final viscosities compared with the control. The control bound the least amount of bile acids (p < 0.05); heating of oat flour improved the binding. Heated oat slurries treated with lichenase or lichenase combined with α-amylase and/or proteinase bound the least amount of bile acid, indicating the contribution of β-glucan to binding. Oat slurries treated with proteinase or proteinase and α-amylase together improved the bile acid binding, indicating the possible contribution of protein to binding. These results illustrate that β-glucan was the major contributor to viscosity and in vitro bile acid binding in heated oat slurries; however, interactions with other components, such as protein and starch, indicate the importance of evaluating oat components as whole system.  相似文献
