A triploid hybrid with an ABC genome constitution, produced from an interspecific cross between Brassica napus (AACC genome) and B. nigra (BB genome), was used as source material for chromosome doubling. Two approaches were undertaken for the production of hexaploids: firstly, by self-pollination and open-pollination of the triploid hybrid; and secondly, by application of colchicine to axillary meristems of triploid plants. Sixteen seeds were harvested from triploid plants and two seedlings were confirmed to be hexaploids with 54 chromosomes. Pollen viability increased from 13% in triploids to a maximum of 49% in hexaploids. Petal length increased from 1.3 cm (triploid) to 1.9 cm and 1.8 cm in the two hexaploids and longest stamen length increased from 0.9 cm (triploid) to 1.1 cm in the hexaploids. Pollen grains were longer in hexaploids (43.7 and 46.3 μm) compared to the triploid (25.4 μm). A few aneuploid offsprings were also observed, with chromosome number ranging from 34 to 48. This study shows that trigenomic hexaploids can be produced in Brassica through interspecific hybridisation of B. napus and B. nigra followed by colchicine treatment.  相似文献

Recent time-series measurements of atmospheric O2 show that the land biosphere and world oceans annually sequestered 1.4 +/- 0.8 and 2.0 +/- 0.6 gigatons of carbon, respectively, between mid-1991 and mid-1997. The rapid storage of carbon by the land biosphere from 1991 to 1997 contrasts with the 1980s, when the land biosphere was approximately neutral. Comparison with measurements of delta13CO2 implies an isotopic flux of 89 +/- 21 gigatons of carbon per mil per year, in agreement with model- and inventory-based estimates of this flux. Both the delta13C and the O2 data show significant interannual variability in carbon storage over the period of record. The general agreement of the independent estimates from O2 and delta13C is a robust signal of variable carbon uptake by both the land biosphere and the oceans.  相似文献

The objective of this research was to determine the effect of stocking density and inclusion of subtherapeutic chlortetracycline in the diet on Salmonella fecal prevalence and antimicrobial resistance in growing swine. A 2 x 2 factorial design was employed on a privately owned commercial swine farm. Four finisher rooms were included in the study. Two of the rooms received 50 g/tonnes of chlortetracycline in the ration, two rooms received no antimicrobials in the feed. In each room, alternate pens were assigned to either high stocking density (0.60 m2/pig) or low stocking density (0.74 m2/pig). Pigs were placed in the finisher rooms at 10 weeks of age and followed for 6 weeks. Individual fecal samples were collected from the floors of each pen and cultured once weekly. Antimicrobial resistance phenotypes were determined. Data were analyzed using multilevel, multivariable logistic regression. Pigs fed chlortetracycline were at increased odds (OR 6.88, 95% CI 2.77-17.12) to shed Salmonellae. No other associations between treatments (CTC and stocking density) and Salmonella prevalence or reduced susceptibility to antimicrobials were identified. Variance in the odds of a fecal sample to be positive was distributed mostly at the lowest level, the individual fecal sample. The increased risk of shedding associated with inclusion of subtherapeutic chlortetracycline in swine diets is discordant with previous results by our group, suggesting farm or strain specific factors may impact this association. Understanding this risk may provide a potential intervention for controlling Salmonella pre-harvest.  相似文献

To avoid loss of yield, crops must maintain tissue potassium (K) concentrations above 5–40 mg K (g DM)–1. The supply of K from the soil is often insufficient to meet this demand and, in many agricultural systems, K fertilisers are applied to crops. However, K fertilisers are expensive. There is interest, therefore, in reducing applications of K fertilisers either by improving agronomy or developing crop genotypes that use K fertilisers more efficiently. Agronomic K fertiliser use efficiency is determined by the ability of roots to acquire K from the soil, which is referred to as K uptake efficiency (KUpE), and the ability of a plant to utilise the K acquired to produce yield, which is referred to as K utilisation efficiency (KUtE). There is considerable genetic variation between and within crop species in both KUpE and KUtE, and chromosomal loci affecting these characteristics have been identified in Arabidopsis thaliana and several crop species. Plant traits that increase KUpE include (1) exudation of organic compounds that release more non‐exchangeable soil K, (2) high root K uptake capacity, (3) early root vigour, high root‐to‐shoot ratios, and high root length densities, (4) proliferation of roots throughout the soil volume, and (5) high transpiration rates. Plant traits that increase KUtE include (1) effective K redistribution within the plant, (2) tolerance of low tissue K concentrations, and, at low tissue K concentrations, (3) maintenance of optimal K concentrations in metabolically active cellular compartments, (4) replacement of K in its non‐specific roles, (5) redistribution of K from senescent to younger tissues, (6) maintenance of water relations, photosynthesis and canopy cover, and (7) a high harvest index. The development of crop genotypes with these traits will enable K fertiliser applications to be reduced.  相似文献

Wheat class identification by bulk sample analysis using a machine vision method would be helpful for automation of grain handling, binning and shipping operations in grain elevators. A machine vision system with a monochrome camera was used to identify eight western Canadian wheat classes at four moisture levels (11%, 14%, 17% and 20% wet basis) by bulk sample analysis (n=100 images for each group of samples). Grayscale images (1024×768 pixels) of the grain bulk were captured by the monochrome camera, and stored on a data acquisition system. Algorithms were developed to extract 32 textural features automatically from the grayscale images. The mean gray values of the western Canadian wheat classes ranged between 106 and 143, and it was the highest for Canada Prairie Spring Red and the lowest for Canada Western Extra Strong and Canada Western Red Winter. The mean gray values of the wheat samples were significantly higher at 17% moisture content and lower at 11% moisture content among the tested moisture levels (α=0.05). The overall classification accuracies of a quadratic discriminant function were 93.8%, 92.5%, 92.0% and 94.4% when the wheat classes were at 11%, 14%, 17% and 20% moisture contents, respectively. Similarly, the accuracies of a linear discriminant function were 96.1%, 95.0%, 95.4% and 96.3% at 11%, 14%, 17% and 20% moisture contents, respectively. When the wheat classes were identified irrespective of moisture levels (images of the four moisture level grains in each class were mixed together), the accuracy was 89.8% and 85.4% for quadratic and linear discriminant functions, respectively. A monochrome image analysis system has the potential to use for online identification of classes in wheat handling facilities. However, further research is required to determine the performance of the developed method for impurities in bulk grain such as foreign material and dockage.  相似文献

Nitrogen fertilization in the nursery, along with altering the configuration of forest gaps, may improve the reforestation success of longleaf pine seedlings. During the very droughty 1998 growing season in Florida and Georgia, survival was higher under the forest canopy than in small (0.10 ha, 36 m diameter) and large (1.6 ha, 144 m diameter) canopy gaps. In the large gaps, survival of containerized seedlings was higher along the edges, particularly the SW edge. Shade from adult trees and the nurse effect of shrubs increased survival, while grass competition reduced survival. During dry years part of the “exclusionary zone” along the edge of canopy gaps (SW sector) may serve as a “survival zone”, at least in the short term. A model using oval-shaped gaps oriented from NW to SE, with an area of 0.25 ha is proposed to maximize the survival and growth of artificially regenerated longleaf pine seedlings.  相似文献

Sapflow sensors were used to investigate variation in sapflow velocity at different positions in the sapwood of three-year-old Eucalyptus globulus ssp. globulus Labill. trees. Sapflow velocity was measured at 5-mm intervals across the sapwood by moving two probe sets simultaneously on two opposite radii. Another probe set placed in a fixed position at right angles to the first two sets acted as a control. A sapflow velocity ratio was defined as the velocity given by each moving sensor divided by that given by the static sensor. Correlation between observations of sapflow velocity at different positions exceeded 95%, and the ratio of velocity between any pair of sensors was constant. We observed radial variation in sapflow velocity across the sapwood with the lowest velocities at the center of the tree. Variation due to sensor position was high implying the need for large numbers of sensors for accurate estimates of sap flux. To overcome this need, we used a correction coefficient, namely a simple weighted average of the sapflow ratios with depth in the sapwood, for each fixed sensor. We recommend the use of three probe sets to estimate the correction coefficient. Subsequently, two probe sets can be placed at two fixed positions for routine measurements of sap flux.  相似文献
