Seventy-two Holstein steers averaging 182 kg were assigned randomly to one of six treatment groups: 1) nonimplanted controls (C); 2) implanted with 36 mg of zeranol (Z); 3) implanted with 20 mg of estradiol benzoate and 200 mg of progesterone (EP); 4) implanted with 140 mg of trenbolone acetate (TBA); 5) implanted with 140 mg of trenbolone acetate plus 20 mg of estradiol benzoate and 200 mg of progesterone (TBA + EP); and 6) implanted with 140 mg of trenbolone acetate plus 36 mg of zeranol (TBA + Z). Each treatment group consisted of three replications of four animals per pen, which were implanted on d 0, 56, 112, and 168. Masculinity and muscling scores were assigned at 24 h preslaughter. Hide removal difficulty was scored by a plant supervisor. Quality and yield grade data were obtained at 24 h postmortem. Longissimus muscle (LM) steaks were removed and cooked for Warner-Bratzler shear (WBS) determinations and sensory panel (SP) evaluations. Over the entire feeding period (249 d), TBA + EP steers had higher (P less than .05) ADG than TBA + Z, TBA, and C steers. All treatments had higher (P less than .05) ADG than C, with the exception of TBA. The only feed efficiency differences were those following the 168-d implant time, when TBA steers were more (P less than .05) efficient than TBA + Z or C steers. The TBA + EP and TBA + Z steers were more (P less than .05) masculine and their hides were more (P less than .05) difficult to remove than those of EP and C steers. Carcass weights of TBA + EP steers were heavier (P less than .05) than those of TBA or C steers. The TBA + EP steers had larger (P less than .05) LM areas than Z, TBA, and C steers. Also, TBA + EP steers tended (P = .07) to have lower numerical yield grades than EP, Z, or C steers. Even though mean marbling scores and quality grades were similar (P greater than .05) among treatment groups, only 50% of TBA + EP carcasses graded low Choice or higher, compared with 100, 75, 82, 90, and 83% for C, TBA, Z, EP, and TBA + Z carcasses, respectively. The only meat palatability differences were that myofibrillar and overall tenderness scores tended to be lower (P = .07) for steaks from EP and TBA + Z than for steaks from Z and C groups. 相似文献

Postpartum Dysgalactia Syndrome (PDS) represents a considerable health problem of postpartum sows, primarily indicated by mastitis and lactation failure. The poorly understood etiology of this multifactorial disease necessitates the use of the porcine mammary epithelial cell (PMEC) model to identify how and to what extent molecular pathogen defense mechanisms prevent bacterial infections at the first cellular barrier of the gland. PMEC were isolated from three lactating sows and challenged with heat-inactivated potential mastitis-causing pathogens Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) for 3 h and 24 h, in vitro. We focused on differential gene expression patterns of PMEC after pathogen challenge in comparison with the untreated control by performing microarray analysis. Our results show that a core innate immune response of PMEC is partly shared by E. coli and S. aureus. But E. coli infection induces much faster and stronger inflammatory response than S. aureus infection. An immediate and strong up-regulation of genes encoding cytokines (IL1A and IL8), chemokines (CCL2, CXCL1, CXCL2, CXCL3, and CXCL6) and cell adhesion molecules (VCAM1, ICAM1, and ITGB3) was explicitly obvious post-challenge with E. coli inducing a rapid recruitment and activation of cells of host defense mediated by IL1B and TNF signaling. In contrast, S. aureus infection rather induces the expression of genes encoding monooxygenases (CYP1A1, CYP3A4, and CYP1B1) initiating processes of detoxification and pathogen elimination. The results indicate that the course of PDS depends on the host recognition of different structural and pathogenic profiles first, which critically determines the extent and effectiveness of cellular immune defense after infection.

The presence of recent dichlorodiphenyltrichloroethane (DDT) inputs is established for Paranaguá Bay biota, i.e. bivalves, fish and one sponge. Values ranged from 6.9 to 156.2 ng ??DDT/g dry weight. Three fish species analysed showed ??DDT values from 36.8 to 92.1 ng/g dry weight. The highest contents (up to 156.2 ng ??DDT/g dry weight) were found for mangrove oysters (Crassostrea rhizophorae) at locations affected by sewage discharge from Paranaguá City. Turtles as herbivores were not affected by this input with values of 0.7 and 2.2 ng ??DDT/g dry weight. The areal distribution of samples suggests that usage of DDT is widespread around the bay. Background
Little is known about the effects of gypsum application to remediate saline–sodic soils in the tropics and the role of microbial indicators in soil reclamation.Aims
Our study aimed at (1) remediating a highly weathered, irrigated sodic Lixisol under prolonged urban crop production by clean water and gypsum application and (2) to determine the remediation effects on soil microbial indices.Methods
A three-factorial on-farm experiment with maize (Zea mays L.) was used to study effects on soil microbial biomass of (1) soil degradation at two levels of salinity, (2) irrigation with clean water and wastewater, and (3) the impact of added gypsum during a typical growing season.Results
At the high-degradation site, the 0.5 M K2SO4 extractable carbon (C) content was 40% higher than at the low-degradation site. In addition, microbial biomass carbon (MBC) and microbial biomass nitrogen (MBN) were 20% lower than at the low-degradation site, while fungal ergosterol was even 40% lower, leading to a 33% lower ergosterol/MBC ratio. Wastewater irrigation increased MBN but decreased ergosterol content at the low-degradation site while having no effect at the high-degradation site. Gypsum amendment led to higher MBN at the low-degradation site but to lower MBN at the high-degradation site. Gypsum amendment always increased the ergosterol content whereby this increase was stronger at the low-degradation site, especially in combination with wastewater irrigation.Conclusions
From a microbial perspective, high soil degradation levels should be avoided by treatment of a saline–sodic wastewater prior to its use for irrigation rather than relying on future remediation strategies of affected field sites. 相似文献78.

We investigated central Mediterranean Pinus halepensis plantations under semi-arid climate in order to evaluate the combined effect of soil treatment and afforestation practices on spontaneous plant species composition,richness and evenness,and on the trend and speed of vegetation dynamics.Phytosociological relevés of three different plot typologies,i.e.(1) soil-treatment and plantation,(2) only soil-treatment,(3) no soil-treatment and no plantation,were compared by(a) multivariate analysis and(b) with reference to species richness and evenness.Moreover,in order to compare vegetation dynamics within the plantations with those ones ongoing in semi-natural garrigue communities,we compared life form and syntaxonomic spectra between phytosociological relevés taken at 8 years of distance.DCA showed that floristic species composition and similarity are influenced by the canopy cover of Pine trees as well as by soil-treatment practices.Although species richness and evenness are not clearly related to neither soil treatment nor Pine afforestation,canopy cover clearly plays a major role:in fact,the highest Ph cover rates correspond to the lowest values of understory species richness.This is true also if only species of biogeographical/conservation interest are considered.Regarding vegetation dynamics,sites with dense Pine canopy cover evolve much slower than the adjacent garrigue communities.The same factors invoqued to explain the patterns of floristic composition and similarity(i.e.allelopathy and competition for light,water and nutrients) may also explain the lowering of diversity of therophytes and the strong decline of the cover perfomed by both therophytes and hemicryptophytes underneath the canopy of dense Ph plantations.Thus,in sites where Ph cover exceeds about 80%,thinning is recommended not only in order to accelerate succession,but also to give a natural ’shape’ to afforestations. 相似文献

Tests were carried out to determine the weighing precision of a 2 m deep lysimeter with a 1 m² cross‐sectional area and a total mass of 3500 to 3850 kg, depending on the soil water content. The weighing mechanism consists on three shear‐stress cells laid out for a load capacity of 1320 kg each. Mass changes as small as 20 g, which is equivalent here to a water gain or loss of 0.02 mm, can be measured with good accuracy and stability under favorable environmental conditions (low wind speed, relatively constant temperature). This precision does not depend on the position on the lysimeter where the mass change occurs and is as good as the best values reported in the literature for other lysimeters. To prevent water and debris from entering the cleavage between lysimeter vessel and pit casing, a rubber collar can be placed across the cleavage. It is attached to the casing and extends about 1–2 cm into the vessel. Although the collar is not supposed to touch the vessel, it does at a few points. This seriously lowers weighing precision, because this contact exerts forces on the vessel, which distort the true weight. Hence, one should refrain from using this type of collar and develop another one. Weighing precision decreases with increasing wind speed, because wind exerts forces on the lysimeter vessel and can thus alter its apparent weight. It is temperature‐dependent, too. 相似文献
