Identification of candidate genes required for susceptibility to powdery or downy mildew in cucumber
Powdery mildew (PM, caused by Podosphaera fusca) and downy mildew (DM, caused by Pseudoperonospora cubensis) are important diseases of cucumber (Cucumis sativus). Breeding for resistance has been undertaken since the 1940s, but underlying resistance genes have not been functionally analysed yet. The published genome sequence of cucumber catalyses the search for such genes. Genetic studies have indicated that resistances to PM and DM in cucumber are often inherited recessively, which indicates the presence of susceptibility genes (S-genes). Therefore we analyzed the cucumber genome for homologs of functionally proven S-genes known from other plant species. We identified 13 MLO-like genes in cucumber, three of which cluster in Clade V, the clade that contains all known MLO-like susceptibility genes to powdery mildews in other dicots. The expression of one of these three genes, CsaMLO1, located on chromosome 1, was upregulated after PM inoculation. It co-localizes with a QTL for PM resistance previously identified. Also homologs of the susceptibility genes PMR4 and PMR5 are located at this QTL. The second MLO-like gene from Clade V (CsaMLO8) resides in a recessively inherited major QTL for PM resistance at the bottom of chromosome 5, together with a PMR6-like gene. Two major QTL for DM recessive resistance at the top of chromosome 5 co-localize with CsaDMR6-2, which is homologous to the DMR6 susceptibility gene in Arabidopsis. Foot-and-mouth disease virus (FMDV) is the causative agent of a highly contagious acute vesicular disease affecting cloven-hoofed animals, including cattle, sheep and pigs. The current vaccine induces a rapid humoral response, but the duration of the protective antibody response is variable, possibly associated with a variable specific CD4+ T cell response. We investigated the use of heat shock protein 70 (Hsp70) as a molecular chaperone to target viral antigen to the Major Histocompatibility Complex (MHC) class II pathway of antigen presenting cells and generate enhanced MHC II-restricted CD4+ T cell responses in cattle. Monocytes and CD4+ T cells from FMDV vaccinated cattle were stimulated in vitro with complexes of Hsp70 and FMDV peptide, or peptide alone. Hsp70 was found to consistently improve the presentation of a 25-mer FMDV peptide to CD4+ T cells, as measured by T cell proliferation. Complex formation was required for the enhanced effects and Hsp70 alone did not stimulate proliferation. This study provides further evidence that Hsp70:peptide complexes can enhance antigen-specific CD4+ T cell responses in vitro for an important pathogen of livestock. 相似文献

Global change is exacerbating the need for more effective mechanisms and approaches for working towards economic, social/cultural
and environmental sustainability. It is now well recognised that science for sustainability will require integrated problem-focussed
research that is by nature trans-disciplinary. Resolutions to both global and regional scale issues must involve participation
of a diverse number of stakeholders. One region that would benefit greatly from such an integrated approach is the Northern
Territory of Australia. This area is home to some of the most pristine savanna landscapes in the world and it is also occupied
by one of the oldest living human cultures which are still in existence today. However, in recent years there has been increasing
pressure to develop this region. The Northern Territory is also facing problems associated with having a growing Aboriginal
population with deepening health and social problems. So as well as needing to facilitate adaptation in response to global
change and development for economic prosperity of the region, there is an obligation to alter management practices to reduce
social disadvantage. In light of this, it will be important that future landscapes are multifunctional and designed to ensure
the preservation of biological and cultural diversity, as well as having provision for the livelihoods of the people that
live within them. This article recommends the adoption of a landscape ecological approach for strategic development and sustainable
planning, which captures and incorporates the values and identities of the different stakeholders, as well as engaging them
in a continuous, adaptive process of planning and management, and in doing so discusses the importance of human ecological

The effects of dietary flax oil and antioxidants on ascites and pulmonary hypertension in broilers using a low temperature model 总被引:3,自引:1,他引:3
1. Three experiments were conducted using a low temperature model to induce pulmonary hypertension (PH) and ascites in broiler chickens. Diets containing 25 g or 50 g flax oil/kg food and control diets with an equivalent amount of animal/vegetable (A/V) blend oil, with and without supplemental antioxidants (vitamin C and vitamin E) were used. The amount of PH was assessed by the ratio of right ventricle weight to total ventricle weight (RV/TV ratio). Birds were considered to suffer from pulmonary hypertension syndrome (PHS) if the RV/TV ratio was greater than 0.299. 2. In experiment 1, the test diets contained 50 g oil/kg food and were given during the grower period only. Birds fed on the flax oil diet tended to have a lower incidence of PHS, ascites and lower RV/TV ratios than birds fed on the control diet. However, when the flax oil diet was supplemented with antioxidants, the incidence of ascites, PHS, haematocrit and whole blood and plasma viscosity increased compared with birds fed on the flax oil diet without antioxidants. These effects were not seen in experiment 2, when the test diets containing 30 g oil/kg food (25 g flax oil plus 5 g A/V blend oil/kg food compared to 30 g A/V blend oil/kg food) were given during the grower period. However, in experiment 3, when the test diets containing 30 g oil/kg food were given from day 1 to week 8, birds fed on the control diet supplemented with antioxidants had a higher incidence of PHS than those fed on the control diet alone. 3. In all 3 experiments, there was no significant effect of dietary fat source or supplemental antioxidants on total food intake or food conversion. 4. We conclude that diets containing 50 g flax oil/kg food tend to reduce the incidence of PHS and ascites in broilers using a low temperature model but the results were not statistically significant. In some cases, supplementing diets with a combination of vitamin E and vitamin C increased the incidence of ascites and PHS. 相似文献

We have provided a detailed structural analysis of porcine alveolar macrophage TLR3 extracellular domain (ECD). The porcine TLR3-ECD contains 18 leucine-rich repeats (LRRs) consisting of blocks of consensus motifs and non-consensus motifs containing insertions. Excluding the N-terminal and C-terminal LRRs, porcine TLR3 has two LRRs with insertions, resulting in one LRR of 39 amino acids and another LRR of 34 amino acids. Furthermore, we have conducted the first examination of the regulated expression of porcine alveolar macrophage TLR3 during in vivo co-infection with influenza virus and Bordetella bronchiseptica. There was a bi-phasic upregulation of porcine TLR3 during influenza virus infection (day 1 and day 10 post-infection). Co-infection resulted in an enhanced expression of porcine TLR3 only at day 1 post-infection. Interestingly, B. bronchiseptica induced an upregulation in alveolar macrophage TLR3 expression at day 10 post-infection. Based on our work and that of others, TLR3 likely plays a key role in the immune response of lung cells to influenza virus infection in several mammalian species. 相似文献
