This paper describes a simple methodology for evaluating the bacterial binding of ciprofloxacin labelled with technetium Tc 99m. Using this methodology, the binding of 99mTc-ciprofloxacin by live Escherichia coli was compared with the binding of 99mTc-ciprofloxacin by killed E. coli and the binding of 99mTc-pertechnetate (99mTcO4-) by live E. coli. The antimicrobial effect of 99mTc-ciprofloxacin on E. coli was evaluated. Four groups were defined: live E. coli with 99mTc-ciprofloxacin, live E. coli with 99mTcO4 , killed E. coli with 99mTc-ciprofloxacin, and killed E. coli with 99mTcO4-. After 0, 2, and 4 h of incubation of 1 x 10(8) colony-forming units of E. coli suspended in 5 mL of sterile distilled water with 1.85 MBq of 99mTc-ciprofloxacin or 99mTcO4, 1 mL from each sample was centrifuged. The radioactivity of the bacterial pellet and that of the supernatant were measured separately, and the percentage of sample radioactivity attributable to bacterial binding was calculated. Of the 99mTc-ciprofloxacin, 3.6% to 5.9% was bound to live or killed E. coli; only 0.1% to 0.2% of the 99mTcO4- was bound to live E. coli (P < 0.0001). No significant difference in 99mTc-ciprofloxacin binding was found between live and killed E. coli (P = 0.887). An antimicrobial effect on E. coli was seen with 99mTc-ciprofloxacin: colony counts were reduced after 4 h. The small amount of 99mTc-ciprofloxacin binding and the lack of difference in binding between live and killed E. coli may limit the utility of this methodology in evaluating the presence of E. coli infection. 相似文献
For canine and feline patients with head tumors, simultaneous irradiation of the primary tumor and mandibular and retropharyngeal lymph nodes (LNs) is often indicated. The purpose of this study was to assess the repeatability of a planning target volume (PTV) expansion protocol for these LNs. Two CT image sets from 44 dogs and 37 cats that underwent radiation therapy for head tumors were compared to determine LN repositioning accuracy and precision; planning‐CT (for radiation therapy planning) and cone‐beam CT (at the time of actual treatment sessions). Eleven percent of dogs and 65% of cats received treatment to their LNs. In dogs, the mandibular LNs were positioned more caudally (P = 0.0002) and the right mandibular and right retropharyngeal LNs were positioned more to the left side of the patient (P = 0.00015 and P = 0.003, respectively). In cats, left mandibular LN was positioned higher (toward roof) than the planning‐CT (P = 0.028). In conclusion, when the patient immobilization devices and bony anatomy matching are used to align the primary head target and these LNs are treated simultaneously, an asymmetrical PTV expansion that ranges 4–9 mm (dogs) and 2–4 mm (cats), depending on the directions of couch movement, should be used to include the LNs within the PTV at least 95% of the time. 相似文献
A 3-year-old Wirehaired Fox Terrier was presented to the University Veterinary Hospital, University College Dublin, for evaluation of chronic cough of 8-months duration. Bronchoscopy showed a severely dilated collapsed left principal bronchus filled with highly viscous white mucus. Cytologically, globular lipid-like material and round concentrically laminated crystalline structures were evident within the proteinaceous mucus. These findings resembled the calcospherites and granular caseous debris often observed in human tuberculous patients. A Ziehl-Neelsen-stained cytocentrifuged preparation of material obtained by bronchoalveolar lavage revealed a few acid-fast rods within macrophages, suggestive of tuberculosis. At necropsy, granulomas with caseous necrosis were present in the lung parenchyma, bronchial and mediastinal lymph nodes, liver, pancreas, and mesentery. Granulomas were adherent to both kidney capsules and to the diaphragm. Histologically, there was evidence of mild calcification within caseous granulomas, which was confirmed by von Kossa's stain. Using Ziehl-Neelsen stain, acid-fast rods were identified within granulomas; bacterial culture was positive for Mycobacterium bovis. The cytologic findings in this case have not been reported previously in dogs and demonstrate a possible correlation between tuberculosis and calcospherite-like bodies with caseous, globular material in bronchial mucus, similar to that described in human patients. 相似文献
Using a cross-sectional survey, we determined the prevalence and risk factors associated with bovine brucellosis in herds under extensive production system in southwestern Nigeria. Antibodies to Brucella species in serum samples were tested using the Rose Bengal test (RBT) and competitive enzyme-linked immunosorbent assay (cELISA); for milk, the milk ring test (MRT) and indirect-ELISA (i-ELISA) were used. Questionnaire was administered to cattle herdsmen to determine factors predisposing the animals to bovine brucellosis. Data were analyzed using STATA 12. From 513 serum and 635 milk samples tested among 120 herds, overall animal-level prevalence of 10.1% (95% CI 7.5–12.7%) and 20.2% (95% CI 17.1–23.3%) were recorded by RBT and MRT, respectively; while 9.4% (95% CI 6.9–11.9%) and 17.8% (95% CI 14.8–20.8%) were obtained using cELISA and i-ELISA, respectively. In all, from the 120 herds tested, 29.2% and 43.3% were positive by RBT and MRT, respectively. Multivariable logistic regression revealed that herd location (OR?=?8.12, 95% CI 1.68–38.90) and improper disposal of placenta/fetus (OR?=?17.33, 95% CI 4.81–62.33) were predictors for a seropositive herd using RBT; while herd location (OR?=?5.13, 95% CI 1.27–20.28), large herd size (OR?=?2.62, 95% CI 1.15–5.85), and occurrence of abortion for a year or more (OR?=?4.62, 95% CI 1.53–13.71) were predictors of seropositivity to antibodies to Brucella spp. using MRT. We found high prevalence of brucellosis in cattle herds under extensive management system in southwestern Nigeria. Urgent and coordinated control strategies are required to mitigate this problem.
Objective— To report use of a locking compression plate (LCP) for fetlock arthrodesis in Thoroughbred racehorses after catastrophic disruption of the suspensory apparatus. Study Design— Retrospective case study. Animals— Racing Thoroughbreds (n=6) with a catastrophic breakdown injury of the suspensory apparatus. Methods— Medical records (2004–2006) of horses that had fetlock arthrodesis using an LCP were reviewed. Preoperative recorded variables were: age, gender, affected limb, injury type and occurrence, limb support used and time to surgery. Recorded surgical variables were: implants used and cost, surgery time, and anesthetic recovery method. Postoperative recorded variables included: treatment, physical status, complications, and outcome. Results— Of 6 Thoroughbred racehorses that had LCP arthrodesis, 4 were sound for breeding purposes the next year and 2 horses were euthanatized (1 at 16 days, 1 at 68 days) because of proximal interphalangeal joint (PIJ) luxation. No surgical complications were associated with implant application. Postoperative complications were similar to those reported for other arthrodesis techniques. Conclusions— LCP is a viable option for fetlock arthrodesis in Thoroughbred racehorses after catastrophic suspensory apparatus disruption. Clinical Relevance— Adequate fetlock stability can be achieved with an LCP, and although the repair is likely more stable it is ∼3 times more expensive than using a limited contact dynamic compression plate (LC-DCP) with cortical screws. 相似文献
Chromium (Cr) is an essential mineral element and has been used in pig diets to improve growth performance, insulin sensitivity, immune response and carcase traits and to reduce heat or other stress responses. The aims of thiss study were to determine the impact of nano-sized chromium tripicolinate (nCrPic) on growth performance, feed efficiency and carcase characteristics of finisher gilts during the summer period. A total of 60 finisher Large White x Landrace gilts were stratified on initial weight and then within strata randomly allocated into two treatment groups in three replicates during mid-summer for 28 days. All pigs were housed in individual pens and had ad libitum access to feed and water. Pigs were fed either a control finisher diet (wheat-based diet containing 13.8 MJ digestible energy (DE) per kilogram and 0.56 g available lysine/MJ DE) or a control diet containing 400 ppb Cr as nCrPic. Dietary nCrPic supplementation increased feed intake by 6 % over the entire study (P?=?0.05). In particular, dietary nCrPic increased average daily feed intake (ADFI) by 8 % (P?=?0.02) during the final 2 weeks of the study. Moreover, dietary nCrPic tended to improve average daily feed (ADFI) over the entire study (P?=?0.09). However, there were no significant effects of nCrPic on feed conversion ratio (FCR), final weight, hot standard carcase weight (HCWT), P2 depth or dressing percentage. Plasma cortisol was decreased by 25 % (P?=?0.06) by dietary nCrPic supplementation. However, there were no effects of nCrPic on plasma glucose, insulin and nonesterified fatty acids (NEFA), might because of the higher feed intake. In conclusion, this study demonstrates that dietary nCrPic supplementation at 400 ppb can increase feed intake in finisher gilts during mid-summer, suggesting that nCrPic can ameliorate some of the negative effects of heat stress in pigs, possibly via decreased of circulating cortisol. 相似文献
