The aim of this study was to evaluate single and double centrifugation tube methods for concentrating equine platelets. Whole blood samples were collected from clinically normal horses and processed by use of single and double centrifugation tube methods to obtain four platelet concentrates (PCs): PC-A, PC-B, PC-C, and PC-D, which were analyzed using a flow cytometry hematology system for hemogram and additional platelet parameters (mean platelet volume, platelet distribution width, mean platelet component concentration, mean platelet component distribution width). Concentrations of transforming growth factor beta 1 (TGF-beta(1)) were determined in all the samples. Platelet concentrations for PC-A, PC-B, PC-C, and PC-D were 45%, 44%, 71%, and 21% higher, respectively, compared to the same values for citrated whole blood samples. TGF-beta(1) concentrations for PC-A, PC-B, PC-C, and PC-D were 38%, 44%, 44%, and 37% higher, respectively, compared to citrated whole blood sample values. In conclusion, the single and double centrifugation tube methods are reliable methods for concentrating equine platelets and for obtaining potentially therapeutic TGF-beta(1) levels.  相似文献

Natural phenolic antioxidants have been tested in hake (Merluccious merluccious) microsomes as inhibitors of lipid oxidation promoted by fish muscle prooxidants: hemoglobin (Hb), enzymatic NADH-iron and nonenzymatic ascorbate-iron. The phenolics selected were as follows: (a) a grape phenolic extract (OW), (b) a fraction (IV) with isolated grape procyanidins with a medium-low degree of polymerization and galloylation percentage, (c) hydroxytyrosol obtained from olive oil byproducts, and (d) a synthetic phenolic antioxidant, propyl gallate. All compounds delayed lipid oxidation activated by Hb, enzymatic NADH-iron, and nonenzymatic ascorbate-iron, excluding hydroxytyrosol that was not an effective antioxidant on oxidation promoted by nonenzymatic iron. The relative antioxidant efficiency was independent of the prooxidant system, IV > propyl gallate > OW > hydroxytyrosol, and showed a positive correlation with their incorporation into microsomes (p < 0.05). The reducing capacity or ability for donating electrons and the chelating properties may also contribute to the antioxidant activity of phenolics, although these factors were less decisive than their affinity for incorporating into the microsomes. Conversely, the inhibition of Hb oxidation by phenolics and their polarity did not seem to play an important role on antioxidant mechanism. These results stressed the importance of incorporating the exogenous antioxidants into the membranes where are located key substances for fish lipid oxidation (highly unsaturated phospholipids, iron-reducing enzymes, and endogenous alpha-tocopherol).  相似文献

Particles emanating from comet 81P/Wild 2 collided with the Stardust spacecraft at 6.1 kilometers per second, producing hypervelocity impact features on the collector surfaces that were returned to Earth. The morphologies of these surprisingly diverse features were created by particles varying from dense mineral grains to loosely bound, polymineralic aggregates ranging from tens of nanometers to hundreds of micrometers in size. The cumulative size distribution of Wild 2 dust is shallower than that of comet Halley, yet steeper than that of comet Grigg-Skjellerup.  相似文献

Dimethylsulfoniopropionate (DMSP) accounts for most of the organic sulfur fluxes from primary to secondary producers in marine microbial food webs. Incubations of natural communities and axenic cultures with radio-labeled DMSP showed that dominant phytoplankton groups of the ocean, the unicellular cyanobacteria Prochlorococcus and Synechococcus and diatoms, as well as heterotrophic bacteria take up and assimilate DMSP sulfur, thus diverting a proportion of plankton-produced organic sulfur from emission into the atmosphere.  相似文献

Pythium isolates were recovered from endive plants (Cichorium endivia) showing vascular necrosis collected from commercial fields located in Castellón province (eastern Spain). They were identified as Pythium tracheiphilum on the basis of their phenotypical and molecular profile. Pathogenicity tests conducted with two P. tracheiphilum isolates, obtained from endive and lettuce (Lactuca sativa), respectively, in this region, confirmed that both isolates were pathogenic to endive, with no significant differences in virulence between them. This is the first report of vascular necrosis caused by P. tracheiphilum on endive in Spain.  相似文献

Despite considerable current interest in biological invasions, the common life-history characteristics of successful invaders remain elusive. The widely held hypothesis that successful invaders have high reproductive rates has received little empirical support; however, alternative possibilities are seldom considered. Combining a global comparative analysis of avian introductions (>2700 events) with demographic models and phylogenetic comparative methods, we show that although rapid population growth may be advantageous during invasions under certain circumstances, more generally successful invaders are characterized by life-history strategies in which they give priority to future rather than current reproduction. High future breeding expectations reduce the costs of reproductive failure under uncertain conditions and increase opportunities to explore the environment and respond to novel ecological pressures.  相似文献

In preparing this document the Authors aimed to pool current information on canine and feline mast cell disease. The information was gathered from international studies and a emphasis was placed on material and opinion with a strong evidence base. We intend it to form the basis of our understanding in this disease at the current time and we anticipate that it will be particularly useful for the general practitioner. The present study describes the glycosidic content of the interstitial tissue in testes from healthy boars and from unilateral and bilateral abdominal cryptorchid boars using lectin histochemistry. The Leydig cells of healthy boars contained glycans with fucosyl, mannosyl, glucosyl, neuraminic acid and galactosyl residues, which have structural and transport functions, and participate in androgen synthesis and in cell regulation. Unilateral cryptorchidism induced high glucosyl and low galactosyl content in the Leydig cells of scrotal testes, resulting in impaired androgen production. In abdominal testes, the Leydig cells exhibited increased amounts of glucosyl and reduced amounts of galactosyl and neuraminic acid residues, resulting in defective cell regulation and lack of androgen synthesis. In healthy boars, the extracellular glycans contained fucosyl, galactosyl, glucosyl and neuraminic acid residues, which confer viscoelasticity on the interstitial tissue and participate in substrate transport, hormone binding and cell-cell interaction. Unilateral cryptorchidism did not induce anomalies in extracellular glycans in scrotal testes, but unilateral and bilateral cryptorchidism resulted in an increased content of fucosyl and galactosyl, and a decreased content of glucosyl and neuraminic acid residues in abdominal testes, leading to reduced viscoelasticity and defective substrate transport across the extracellular matrix.  相似文献
