Toxins that are secreted by cone snails are small peptides that are used to treat several diseases. However, their effects on parasites with human and veterinary significance are unknown. Toxoplasma gondii is an opportunistic parasite that affects approximately 30% of the world’s population and can be lethal in immunologically compromised individuals. The conventional treatment for this parasitic infection has remained the same since the 1950s, and its efficacy is limited to the acute phase of infection. These findings have necessitated the search for new drugs that specifically target T. gondii. We examined the effects of the synthetic toxin cal14.1a (s-cal14.1a) from C. californicus on the tachyzoite form of T. gondii. Our results indicate that, at micromolar concentrations, s-cal14.1a lowers viability and inhibits host cell invasion (by 50% and 61%, respectively) on exposure to extracellular parasites. Further, intracellular replication decreased significantly while viability of the host cell was unaffected. Our study is the first report on the antiparasitic activity of a synthetic toxin of C. californicus. 相似文献

It has been argued that large ungulates play a key role in natural forest dynamics, but in Britain, the largest native ungulates
(aurochs and elk) are extinct. Cattle could have some similar effects, and are widely used, but rarely tested, for nature
conservation management. Here, we test conservation management with cattle at a native Scots pine Pinus sylvestris forest in Scotland. Our hypotheses were that cattle impacts would (a) increase the abundance of an understorey shrub of conservation
importance, bilberry Vaccinium myrtillus; and (b) increase Scots pine seedling establishment. Two cattle trials were established, one for each objective, based on
a 14-ha plot within the forest, and a 6.1-ha plot at the forest edge. Cattle were introduced for 2 months to both plots, giving
an approximately 100- to 150-fold increase in ungulate biomass per hectare when compared to background deer abundance. The
ground layer vegetation in both treatment and matched control plots was monitored before and after treatment, and subsequently
over a 4-year period. At the within-forest trial, bilberry percent-cover, 4 years after treatment, was 1.9 times higher in
the cattle area than in the control (95% confidence limits 1.6–2.3). Bilberry percent-cover increases were directly related
to the degree of trampling impact on heather Calluna vulgaris, as recorded immediately after the trial. At the forest-edge trial, there were vegetation changes in the cattle plot that
were considered favourable to pine seedling establishment: reduced moss/litter depth and vegetation percent-cover, and increased
ground-level light incidence. However, too few new pine seedlings were found for formal analysis, partly due to unexpectedly
low seed-fall. Nevertheless, the potential for cattle to create conditions that would subsequently promote pine regeneration
was illustrated by the observation of large numbers of new, young pine seedlings at the within-forest cattle plot. We recommend
that managers and researchers collaborate to develop further trials like the ones reported here, to give an improved understanding
of the conditions under which cattle can yield nature conservation benefits in woodlands. 相似文献

There are still no effective means to control Dutch elm disease (DED), caused by the vascular fungi Ophiostoma ulmi and O. novo-ulmi. Plant phenolics may provide a new strategy for DED control, given their known antifungal activity against pathogens and
their involvement in plant defence mechanisms. The in vitro antifungal activity of salicylic acid, carvacrol, thymol, phenol,
o-cresol, m-cresol, p-cresol, and 2,5-xylenol against the DED pathogens was tested. Also, the protective effect of watering Ulmus minor seedlings with these compounds was tested against O. novo-ulmi. Salicylic acid, carvacrol, and thymol showed the strongest antifungal in vitro activity, while carvacrol and salicylic acid
provided the strongest in vivo protection against O. novo-ulmi (63 and 46% reduction of leaf wilting symptoms with respect to controls, respectively). The effect of the treatments on tree
phenology was low, and a significant negative relation was observed between the number of days to bud burst and the leaf wilting
symptoms after inoculation, probably determined by genetic differences among the elm tree progenies used. The treatments with
salicylic acid, carvacrol and thymol induced the highest shift in phenolic metabolite profile with respect to control trees.
The protective effect of carvacrol and salicylic acid is discussed in terms of their combined activity as antifungal compounds

Woody plant encroachment is a threat to savanna ecosystems worldwide. By exploiting differences in the physiology and seasonality of herbaceous species and encroaching hardwoods, herbicides can be used to control woody shrubs in savannas without causing lasting harm to desirable vegetation. We applied three herbicides and one tank mix to control shrubs following removal of the slash pine (Pinus elliottii Engelm.) canopy and replanting with container-grown longleaf pine (Pinus palustris Mill.) seedlings in a mesic-wet savanna in the southeastern USA. The herbicides tested were imazapyr, sulfometuron methyl, hexazinone, and a hexazinone + sulfometuron methyl tank mix. 4 years after application, no negative effects on understory species richness, diversity, evenness, or community composition were evident in any of the herbicide treatments. Oaks (Quercus spp.), one of the dominant shrub genera on the study site, were resistant to sulfometuron methyl, and this herbicide was therefore ineffective both as a pine release treatment and for enhancing herbaceous species cover. Imazapyr was the most effective treatment overall, leading to significant improvements in longleaf pine seedling growth and also enhancing herbaceous species cover. Both hexazinone and the hexazinone + sulfometuron methyl tank mix provided some seedling growth and understory enhancement as well. In particular, the tank mix significantly increased wiregrass cover relative to the control. Shrubs resprouted quickly following a dormant-season prescribed fire in the fifth year after treatment, indicating that herbicide-related increases in herbaceous cover may be lost if an aggressive prescribed fire program is not implemented. 相似文献

Ultrasound evaluation of the thyroid gland was performed in healthy, hypothyroid, and euthyroid Golden Retriever dogs with nonthyroidal illness (NTI) to determine the diagnostic usefulness of ultrasound for differentiating between euthyroid and hypothyroid dogs. Thirty-six healthy, 11 hypothyroid, and 35 euthyroid dogs with NTI were evaluated. Each thyroid lobe was examined ultrasonographically for size, shape, echogenicity, and homogeneity. Thyroid lobe volume was estimated by using the equation for an ellipsoid: pi/6(length X height x width). No differences were found between healthy dogs and euthyroid dogs with NTI. In the majority of euthyroid dogs, the thyroid lobes were fusiform and triangular in shape in longitudinal and transverse planes, respectively. The thyroid capsule appeared smooth. The thyroid parenchyma had a homogeneous echogenic pattern and usually was hyperechoic or isoechoic compared with the surrounding musculature. Ultrasound findings in hypothyroid dogs were more variable, including a greater frequency of round to oval-shaped thyroid lobes in the transverse imaging plane (P < .05), hypoechogenicity of the thyroid parenchyma compared with surrounding musculature (P < .001), and a decrease in the size and volume of the thyroid lobes and total volume of the thyroid gland (P < .05) compared with euthyroid dogs. Other findings in hypothyroid dogs included an irregular surface to the thyroid capsule, a heterogeneous pattern to the thyroid parenchyma, and differences in the echogenic pattern between the left and right thyroid lobes. Results suggest that determination of thyroid size and volume by ultrasound may be a useful adjunctive test for differentiating between hypothyroid and euthyroid dogs with NTI. 相似文献

Zearalenone (ZEA) is a resorcylic acid lactone derivative produced by various Fusarium species that are widely found in food and feeds. The structure of zearalenone is flexible enough to allow a conformation able to bind to mammalian oestrogen receptors, where it acts as an agonist. Using oestrogen-dependent Human Breast Cancer (MCF-7) cells, the oestrogenic activity of zearalenone and its derivatives were compared using 17 beta-oestradiol as a positive control. The results obtained demonstrate that the oestrogenic potency of ZEA derivatives could be ranked in the following order: alpha-zearalenol > alpha-zearalanol > zearalenone > beta-zearalenol. Since pigs have been reported to be among the most sensitive animal species, biotransformation studies with pig liver subcellular fractions were conducted. These studies indicated that alpha-zearalenol is the main hepatic metabolite of zearalenone in pigs, and it is assumed that 3 alpha- and 3 beta-hydroxysteroid dehydrogeneases are involved in the hepatic biotransformation, since the formation of alpha-zearalenol and beta-zearalenol could be inhibited by prototypic substrates for either enzyme. The bioactivation of ZEA into the more active alpha-zearalenol seems to provide a possible explanation for the observed high sensitivity of pigs towards feeding-stuffs contaminated with the mycotoxin. 相似文献

Two-dimensional and M-mode echocardiograms were recorded from 41 horses before they were successfully treated for atrial fibrillation. In addition, these examinations were performed in a subgroup of 20 horses after treatment, and the results were compared with pretreatment values. Atrial fibrillation in this group of horses was associated with a reduction of mean left ventricular fractional shortening (mean 31 %± 5.24%), and 22 of the 41 horses were below the reference range. The remaining mean M-mode variables were within the normal reference range, although 12 horses had increased left ventricular lumen dimensions in systole, and 8 horses had decreased left ventricular ejection times. Abnormal motion of the mitral valve was present in all horses and was characterized by the absence of A peaks, which were replaced by small diastolic undulations in 55% of the horses. In horses 1 to 20, after conversion to sinus rhythm, the mean fractional shortening increased (35.34%± 5.4%, P = .004), but there were no significant differences in heart rate or left ventricular lumen diameters in systole or diastole. These results suggest that ventricular function may be compromised by the presence of atrial fibrillation. However, this improved after correction of the arrhythmia. 相似文献

OBJECTIVE: To compare the effects of administration of 2 volumes of a calcium solution (calcium oxide and calcium gluconate) on plasma ionized calcium concentration (PICaC) and clinical recovery from naturally occurring hypocalcemia (NOHC; milk fever) in lactating dairy cows. ANIMALS: 123 cows with NOHC (PICaC < 0.95 mmol/L [3.81 mg/dL]) and 20 clinically normal control cows. PROCEDURES: Affected cows were treated IV once or repeatedly with 450 (n = 56) or 750 mL (67) of calcium solution (1.65 g of calcium/100 mL) until clinical recovery was achieved. The PICaC was assessed 48 hours after the first treatment or after the treatment that achieved clinical recovery. Biochemical recovery was defined as PICaC > or = 0.95 mmol/L. Plasma from control cows was used for PICaC reference range determination. Plasma samples from both groups were assessed after storage for 20 days at 20 degrees C. RESULTS: The PICaC reference range derived from blood collected in tubes containing lithium heparin was 1.02 to 1.29 mmol/L (4.09 to 5.17 mg/dL). Following storage, plasma samples were suitable for PICaC assessment. All cows treated with > or = 1 volume of 450 and 750 mL of calcium solution recovered clinically; however, 31 of 83 (37%) evaluated cows were not biochemically recovered at 48 hours following treatment. Only cows with PICaC < 0.48 mmol/L (1.92 mg/dL) before the first treatment had to be treated > or = 3 times. CONCLUSIONS AND CLINICAL RELEVANCE: Results did not support the need to increase the administered volume of calcium solution from 450 to 750 mL for treatment of NOHC in dairy cows. 相似文献
