The problem of mercury (Hg) accumulation in the aquatic food chain is widespread in Sweden. The methylmercury (MeHg) in runoff from catchments may be an important component of the MeHg load in surface waters. The spring flood in northern Sweden constitutes a major portion of the annual catchment runoff. This brief, but large event, therefore, may be of significance for the annual output of MeHg from soils to surface waters in this region. Methylmercury, total mercury (Hg-tot) and other chemical parameters were measured in spring flood runoff during April and May 1993 from two tributaries as well as the headwater mire of the 50 ha Svartberget Catchment. Snow cores from April 1993 and April 1994 prior to the onset of spring snowmelt were also analyzed. Stable isotope techniques were used to estimate the proportion of snowmelt in runoff. During the spring flood, Hg-tot and TOC were diluted in output from the headwater mire compared to the concentrations observed prior to the flood. Over half of the runoff from the mire was snowmelt according to the isotope Hydrograph separation. In runoff from the two forested tributaries, however, TOC and Hg-tot concentrations increased. About a third of this runoff was snowmelt. MeHg concentrations in the spring flood declined at all locations to the lowest levels recorded during 1993 (<0.2 ng l?1). The runoff concentrations of MeHg were less than the average snow core concentration of 0.3 4+/- 0.17 ng l?1. The differences in MeHg dynamics in comparison to TOC or Hg-tot suggest that there are factors independent of the availability of Hg-tot or TOC, and even contemporary MeHg deposition (in this case the snowpack MeHg concentrations) which determine the concentrations and output of MeHg during spring flood. 相似文献
Whey protein isolate (WPI) was chemically modified by vanillic acid in order to enhance its cross-linkability by laccase enzyme. Incorporation of methoxyphenol groups created reactive sites for laccase on the surface of the protein and improved the efficiency of cross-linking. The vanillic acid modified WPI (Van-WPI) was characterized using MALDI-TOF mass spectrometry, and the laccase-catalyzed cross-linking of Van-WPI was studied. Furthermore, the vanillic acid modification was compared with the conventional approach to improve laccase-catalyzed cross-linking by adding free phenolic compounds. A small extent of the vanillic acid modification significantly improved the cross-linkability of the protein and made it possible to avoid color formation in a system that is free of small phenolic compounds. Moreover, the potential application of Van-WPI as emulsifier and the effect of cross-linking on the stability of Van-WPI emulsion were investigated. Effects of a Trichoderma reesei tyrosinase (TYR) and a Trametes hirsuta laccase (LAC) on breadmaking performance of gluten-free oat flour were investigated by SDS-PAGE analysis of oat protein fractions, large deformation rheology, and microscopy of the doughs, as well as on the basis of specific volume and firmness of the gluten-free breads. TYR induced the formation of higher molecular weight proteins in the SDS-PAGE assay. Microscopical analysis showed more intensive protein aggregation in the TYR-treated dough than in the dough without TYR. TYR also increased the firmness of the dough, which was assumed to be because of the cross-linking of oat globulins. LAC did not affect the oat globulins. TYR alone, or together with a commercial Thermomyces lanuginosus xylanase (XYL), increased significantly the specific volume of the gluten-free oat bread. A combination of TYR and XYL also increased the softness of the bread, whereas a combination of LAC and XYL improved the specific volume but did not affect the softness of oat bread. The results thus indicate that cross-linking of oat globulins by TYR, especially with the addition of XYL, was beneficial for improving the texture of gluten-free oat bread. 相似文献
ABSTRACT: Avian influenza virus can be divided into two groups, highly pathogenic avian influenza virus (HPAI) and low pathogenic avian influenza virus (LPAI) based on their difference in virulence. To investigate if the difference in clinical outcome between LPAI and HPAI in chickens is due to immunological host responses in the lung within the first 24 hours post infection (hpi), chickens were infected with LPAI or HPAI of subtype H7N1. Virus was found in the caudal and cranial part of the lung. With LPAI, virus was localised around the intrapulmonary bronchus and secondary bronchi. In sharp contrast, HPAI was detected throughout the whole lung. However, based on viral RNA levels, no quantitative difference was observed between LPAI and HPAI infected birds. In infected areas of the lungs, an influx of CD8α+ cells as well as KUL01+ macrophages and dendritic cells (DC) occurred as fast as 8 hpi in both infected groups. No major difference between LPAI and HPAI infected birds in the induction of cytokines and interferons at mRNA level in lung tissue was found.In conclusion, the differences in lethality for chickens infected with LPAI or HPAI could be ascribed to difference in location of the virus. However similar amounts of viral RNA, similar cytokine mRNA levels, and similar influxes of CD8α+ and KUL01+ macrophages and DC were found between HPAI and LPAI in the lungs. A cytokine storm at mRNA level as described for mammals was not observed in the lungs of HPAI infected birds within 24 hpi. 相似文献
The influence of acute exposure to zearalenone (ZEN) on porcine oocyte maturation, fertilization or sperm penetration ability during both in vitro maturation and fertilization was evaluated. First, oocytes were cultured in ZEN-containing (0-1000 μg/l) maturation medium and then fertilized. The oocytes maturing in vitro without ZEN were then fertilized in ZEN-containing fertilization medium. The maturation rates of oocytes and penetration ability of sperm decreased significantly in the presence of 1000 μg/l of ZEN. However, neither increases in the rates of degeneration and DNA fragmentation of oocytes nor reductions in normal and polyspermic fertilization were observed. ZEN did not affect the sperm penetration rates; however, 1000 μg/l ZEN had positive effects on normal and polyspermic fertilization rates. Therefore, it can be suggested that an acute exposure of porcine oocytes during maturation and of oocytes and sperm during fertilization to ZEN up to 1000 μg/l may not affect the fertility of the oocytes. 相似文献
Poultry are important for many poor households in developing countries, but there are many constraints to poultry production, including disease. One of the most important diseases of chickens is Newcastle disease (ND). Even though there are effective vaccines against this disease available in most countries, uptake by small-scale poultry keepers is often low. In this study, two areas in Kenya and Tanzania were studied, where some villages had received additional support to get vaccination and other villages had not. In Kenya, 320 households from 10 villages were interviewed, of which half of the villages had active promotion of vaccination through village-based advisors. In Tanzania, 457 households were interviewed, of which 241 came from villages that have had active support through either a project or government extension services. Knowledge about vaccines and the attitudes towards vaccinating against ND was evaluated using mixed multivariable logistic models. Results indicate that in Kenya, the most important determinants for understanding the function of a vaccine were having had support in the village and to have knowledge about ND signs, while in Tanzania gender and previous vaccine use were important in addition to having had support. Attitudes towards vaccination were mainly determined by knowledge, where more knowledge about how vaccines work in general or about ND contributed to more positive attitudes. Among Kenyan farmers that had never used the vaccine before, the amount of birds they lost to disease and predators also influenced attitudes. In conclusion, this study supports the notion that knowledge is a very important component of extension support and that simply making vaccines available may not be sufficient for high levels of uptake.
Foliar deposits and persistence of azadirachtin were investigated after spraying ‘Margosan-O’® formulation at three dosage and volume rates on to balsam fir and oak seedlings in a laboratory chamber. Droplet-size spectra and deposits were assessed using artificial samplers, and foliar residues were measured by high-performance liquid chromatography (HPLC) at different intervals of time after treatment. Disappearance of azadirachtin from both types of foliage was rapid, and the DT50 (the time required for 50% of the initial concentration to disappear) values ranged from 17 to 22 h. The initial azadirachtin concentrations varied widely from 4 to 96 μg g?1 (fresh weight), but the rate constants 0.0311 to 0.0414, were not significantly different. The data thus indicated a poor relationship between initial concentrations and the rate of loss. The influence of spray droplet size and cuticular wax content of foliage on persistence of azadirachtin was investigated using both foliar types, and glass microscope slides without and with the foliar wax coating. Neither the type of surface nor the droplet size influenced the persistence. The rate of azadirachtin loss from the glass slides was faster than from both types of foliage, and was unaffected by the presence of wax coating. The fir foliage had higher cuticular wax content than the oak foliage, but the persistance of azadirachtin was similar in both foliar types. 相似文献
