The parthenogenetic Haemaphysalis longicornis larvae engorged on cattle naturally infected with Theileria sergenti were reared at 24 degrees C. The resultant nymphal ticks were incubated at 37 degrees C to clear the effect of incubation on the development and maturation of sporozoites. The sporozoites in the salivary glands of the nymphal ticks exposed to 37 degrees C for 16 days were observed by the methyl green pyronin staining method. The ticks exposed to 37 degrees C were ground up in a mortar and the supernatant of the tick suspension in PBS was inoculated into cattle. The cattle showed parasitemia and specific antibody response 18 days after inoculation. Consequently, the parasites in the tick salivary glands became infective to cattle by incubating infected. ABSTRACT:   A ghrelin gene has been cloned and sequenced in common carp Cyprinus carpio . Ghrelin cDNA is composed of 461 bp [with a 36-bp 5'-untranslated region (UTR) and a 113-bp 3'-UTR], which translates into a protein of 103 amino acid residues. Carp ghrelin (preproghrelin) contained a predicted signal peptide of 26 amino acid residues, the ghrelin domain ( Gly 27– Val 45) and C-terminal peptide ( Gly 46– Phe 103). Homology analysis of the ghrelin domain of carp with that of other known ghrelin in vertebrates showed good similarity to teleost ghrelin (50–81.8%). Hydropathy analysis based on the deduced amino acid sequence of ghrelin domains in teleosts showed a similar profile. Carp ghrelin clustered with ghrelin of goldfish Carassius auratus and other teleosts, away from mammalian, reptilian, avian, amphibian and chondrichthian ghrelin, by phylogenetic analysis. Genomic organization of carp ghrelin gene was composed of four exons and three introns, which was the same as that of other teleosts and human ghrelin genes. The carp ghrelin gene was expressed in unstimulated tissues such as foregut, hindgut, spleen and brain. In spleen cells, expression of the ghrelin gene increased upon stimulation with lipopolysaccharide (LPS), phytohemagglutinin (PHA) or imiquimod. The identification of carp ghrelin gene and the analysis of the modulation of its expression in immune-activated conditions will allow a more complete analysis of the roles of ghrelin in teleosts.  相似文献

BACKGROUND: The pyrethroid resistance of the diamondback moth Plutella xylostella (L.) is conferred by increased gene expression of cytochrome P450 to detoxify the insecticide and/or through gene mutation of the sodium channel, which makes the individual insensitive to pyrethroids. However, no information is available about the correlation between the increased metabolic detoxification and the target insensitivity in pyrethroid resistance. RESULTS: Frequencies of pyrethroid‐resistant alleles (L1014F, T929I and M918I) and two resistance‐related mutations (A1101T and P1879S) at the sodium channel and expression levels of the cytochrome P450 gene CYP6BG1 were examined individually using laboratory and field strains of P. xylostella. Real‐time quantitative PCR analysis using the laboratory strains revealed that levels of larval expression of the resistant strain, homozygous for the pyrethroid‐resistant alleles other than the M918I, are significantly higher than those of the susceptible strain. In the field strains, the expression levels in insects having the same resistant alleles as those of the resistant strains varied greatly among individuals. The expression levels were not significantly higher than those in the heterozygotes. CONCLUSION: Significant correlation between the target insensitivity and the increased metabolic detoxification in pyrethroid resistance of P. xylostella was observed in the laboratory but not in the field. Interspecific hybridization is a useful technique to introduce characteristics from wild species into crops. Garden asparagus (Asparagus officinalis, 2n = 2x = 20) is an economically important vegetable that is native to the Mediterranean region but widely cultivated in many countries. The genus Asparagus is comprised of over 100 species. Asparagus kiusianus (2n = 2x = 20) is a wild asparagus species endemic to Japan. This species occurs on the coast and is likely to be a salt-resistant species. Although the geographic distribution of these two species is not close, molecular phylogenetic analysis demonstrated that these two species are closely related. In this study, a reciprocal cross between A. officinalis and A. kiusianus was carried out by hand pollination, and progeny were obtained from both crossings. These progeny exhibited a morphologically intermediate phenotype in terms of flower shape, and restriction fragment length polymorphism analysis confirmed that these were indeed interspecific hybrids. The interspecific hybrids were fertile, and backcross progeny with garden asparagus was also generated. These interspecific hybrids are expected to supply novel traits to garden asparagus.  相似文献

There is increasing evidence that global warming affects the development of rice. High temperatures during ripening increase the ratio of undesirable chalky grains followed by deteriorating grain appearance quality. In order to detect quantitative trait loci (QTLs) controlling the occurrence of white-back and basal-white chalky grains of brown rice, QTL analysis was performed using recombinant inbred lines derived from a cross between two strains, ‘Tsukushiroman’ (sensitive to heat stress) and ‘Chikushi 52’ (tolerant of heat stress). The F7 and F8 lines were exposed to heat stress during the ripening period in two locations, Fukuoka and Kagoshima, in Japan. QTLs for white-back grains and basal-white grains were detected on chromosomes 1, 3, and 8, and those for basal-white grains were detected on chromosomes 2, 3, and 12. QTLs on chromosome 8 for white-back grains were shared in the plants grown in both locations. Near-isogenic lines (NILs), which harbored a segment from ‘Chikushi 52’ on chromosome 8 with the genetic background of ‘Tsukushiroman’, showed relatively lower ratios of white-back grains than ‘Tsukushiroman’. Therefore, insertion of the ‘Chikushi 52’ genomic region of the QTL on chromosome 8 can improve the quality of rice when it is grown under heat stress conditions.  相似文献

Asparagus (Asparagus officinalis L.) is a dioecious plant. In general, male and female plants are used for open-field culture and intensive cultivation, respectively. Farmers distinguish between the sexes by observing the form of the flower organs. However, because flowering begins 2?C3 years after planting, the sexes cannot be differentiated at transplantation by using this method, and planting of an all-male population is not possible. In this study, the usefulness of loop-mediated isothermal amplification (LAMP), a simple method of gene amplification, for sex distinction at the DNA level was determined. In addition, the phenotypic differences in seeds and seedlings of male and female plants were investigated for application as a method of early sex distinction. By using the LAMP method, the sex could be correctly identified in 100% of the seedlings, suggesting that this method is effective for sex distinction at the gene level. Principal component analysis was conducted with 11 selected parameters after investigating the seeds and seedlings of both male and female plants. The results revealed that male plants tend to have many stalks or cladophylls and female plants tend to have large plant forms, suggesting that the sexes can be distinguished by the external appearance of the seedlings before planting. LAMP and observation of the seedling phenotypes could be useful methods of sex distinction for increasing the efficiency of asparagus breeding.  相似文献

Abstract

Cabbage plants were grown at graded levels of nitrogen supply and light intensity, and the combined effects on cabbage-head development were studied during the spring and autumn seasons.

The cabbage-head yield (i.e. dry weight of inner head leaves) was the highest at 50 ppm N in the nutrient solution, followed by 500 and 5 ppm N at full light intensity, and decreased with decrease in the, light intensity by shading. The decrease in cabbage-head yield caused by the shading was greater at high nitrogen levels during the autumn (low solar energy) season than during the spring (high solar energy) season. As the nitrogen supply increased or the light intensity decreased during both the spring and autumn seasons, nitrogen, especially soluble N, accumulated and the carbohydrate (sugar and starch) content decreased in the plants. Cabbage-head development occurred efficiently when the total N content in the outer leaves remained at about 2–3% and the carbohydrate production was large at full light intensity.  相似文献

The International Maize and Wheat Improvement Center (CIMMYT) acts as a catalyst and leader in a global maize and wheat innovation network that serves the poor in the developing world. Drawing on strong science and effective partnerships, CIMMYT researchers create, share, and use knowledge and technology to increase food security, improve the productivity and profitability of farming systems and sustain natural resources. This people-centered mission does not ignore the fact that CIMMYT’s unique niche is as a genetic resources enhancement center for the developing world, as shown by this review article focusing on wheat. CIMMYT’s value proposition resides therefore in its use of crop genetic diversity: conserving it, studying it, adding value to it, and sharing it in enhanced form with clients worldwide. The main undertakings include: long-term safe conservation of world heritage of both crop resources for future generations, in line with formal agreements under the 2004 International Treaty on Plant Genetic Resources for Food and Agriculture, understanding the rich genetic diversity of two of the most important staples worldwide, exploiting the untapped value of crop genetic resources through discovery of specific, strategically-important traits required for current and future generations of target beneficiaries, and development of strategic germplasm through innovative genetic enhancement. Finally, the Center needs to ensure that its main products reach end-users and improve their livelihoods. In this regard, CIMMYT is the main international, public source of wheat seed-embedded technology to reduce vulnerability and alleviate poverty, helping farmers move from subsistence to income-generating production systems. Beyond a focus on higher grain yields and value-added germplasm, CIMMYT plays an “integrative” role in crop and natural resource management research, promoting the efficient use of water and other inputs, lower production costs, better management of biotic stresses, and enhanced system diversity and resilience.  相似文献

The most reliable method for determining carbon in soils is the dry combustion method, the technique of which was established for the first time by JUSTUS von LIEBIG in 1831 and for nearly a century his combustion technique, essentially unmodified save for improvements in materials, constituted the standard method for determination of carbon and hydrogen. PREGL (1), in his publication on organic microanalyses in 1916, reported an excellent method in which a carbon determination could be carried out in an hour with a sample of only a few milligrams. After that, his method was improved in many respects by many investigators (2,3). Since these methods require, however, both complicated and expensive apparatus as well as trained personnel and are time-consuming, they are not considered to be suitable for practical use in soil research laboratories at all.  相似文献

The speciation of mercury (Hg) in Minamata Bay (Japan) was studied over a 2-year period (2006?C2008). Concentrations of dissolved total Hg, dissolved methylmercury (MeHg), particulate total Hg, and suspended solids were 0.43?±?0.14 ng/l (mean?±?standard deviation), 0.10?±?0.06 ng/l, 3.04?±?2.96 ng/l, and 5.94?±?2.10 mg/l, respectively. Correlations between concentrations of particulate total Hg and suspended solids at four depths (surface: 0 m; mid-depth: ?6 m, ?10 m; and bottom +1 m layer) were only significant in the bottom +1 m layer. The mean dissolved MeHg concentration and the ratio of dissolved MeHg to dissolved total Hg were considerably higher in summer compared to other seasons. The data suggest that bottom sediment was not the sole source of MeHg, and that MeHg may be produced in the water column by the conversion of divalent Hg eluted from resuspended bottom sediment. The correlation between seawater characteristics such as salinity, temperature, dissolved oxygen (DO), and dissolved MeHg concentration indicates that Hg methylation could be influenced by the heterotrophic activity of microorganisms in the seawater. In particular, inverse correlations were observed between DO, salinity, and MeHg concentration. However, dissolved MeHg concentrations did not correlate with seawater characteristics such as pH or chlorophyll-a.  相似文献
