With the aim of genotyping Echinococcus granulosus cysts found in Mexican livestock, we collected hydatid cysts from the livers and lungs of pigs in slaughterhouses in the state of Morelos, Central Region of Mexico. DNA was extracted from the parasites and examined by polymerase chain reaction (PCR) of rDNA internal transcribed spacer 1 (ITS1-PCR), Eg9-PCR, Eg16-PCR, and PCR-restriction fragment length polymorphism (PCR-RFLP). In addition, fragments of the genes coding for mitochondrial cytochrome c oxidase subunit 1 (CO1) and NADH dehydrogenase 1 (ND1) were sequenced. Two different genotypes of E. granulosus were unequivocally identified, the common sheep genotype, G1, and the common pig genotype, G7. The G1 genotype of E. granulosus has not been previously demonstrated in Mexico. Because of its recognized infectivity in humans, G1 genotype is a direct threat to human health and its presence in Mexico is consequently of immediate public health importance and epidemiological relevance.  相似文献

BACKGROUND: Pest resurgence following a pesticide application may occur owing to a stimulatory (hormetic) response to sublethal insecticide concentrations. The objective of the present study was to examine the potential for a greenhouse‐derived red clone of Myzus persicae to exhibit resurgence owing to a hormetic response following a systemic imidacloprid treatment in a bell pepper greenhouse. RESULTS: No differences in mortality and fecundity were observed among apterous adults exposed to sublethal imidacloprid concentrations on excised pepper leaves fed aqueous solutions of imidacloprid. Survival of first‐generation progeny was negatively affected by imidacloprid exposure, yet surviving progeny exhibited no differences in development rates or fecundity from progeny of adults unexposed to imidacloprid. Aphid mortality declined most rapidly in clip cages on pepper leaves at the top of the pepper canopy as compared with leaves present at the middle or bottom of the pepper canopy. CONCLUSION: Imidacloprid decays rapidly in mature pepper plants, resulting in sublethal concentrations in the upper canopy in as little as 4 weeks. Sublethal insecticide concentrations have been implicated in the resurgence of pest populations; however, exposure to sublethal doses of imidacloprid are unlikely to result in pesticide‐induced resurgence of the M. persicae aphid clone examined in this study. Spread of Verticillium wilt into newly established olive orchards in Andalucía, southern Spain, has caused concern in the olive industry in the region. This spread may result from use of Verticillium dahliae-infected planting material, which can extend distribution of the highly virulent, defoliating (D) pathotype of V. dahliae to new areas. In this study, a molecular diagnostic method for the early in planta detection of D V. dahliae was developed, aimed especially at nursery-produced olive plants. For this purpose, new primers for nested PCR were designed by sequencing a 992-bp RAPD marker of the D pathotype. The use of the specific primers and different nested-PCR protocols allowed the detection of V. dahliae pathotype D DNA in infected root and stem tissues of young olive plants. Detection of the pathogen was effective from the very earliest moments following inoculation of olive plants with a V. dahliae pathotype D conidia suspension as well as in inoculated, though symptomless, plants.  相似文献

We conducted two experiments to evaluate the effects of added choice white grease on performance and carcass merit of barrows and gilts reared under commercial conditions. Pigs were housed either 20 (Exp. 1) or 25 (Exp. 2) per pen and were provided 0.67 m2 of pen space per pig. Diets were based on corn and soybean meal and fed in a meal form. The proportion of soybean meal was increased in diets with added fat to maintain the same calorie:lysine ratio in all diets within a weight phase. In Exp. 1, 480 pigs were fed diets with 0, 2, 4, or 6% fat. Total lysine contents of the control diets were 1.21, 0.88, and 0.66% during the weight phases 36 to 59, 59 to 93, and 93 to 120 kg, respectively. Gain:feed was increased linearly (P < 0.01) due to fat addition in all weight intervals and over the total experiment. The effect of added fat on ADG was not consistent among the weight phases; a linear (P < 0.01) improvement was found from 36 to 59 kg, but no effect was found during the heavier weight phases. Over the total experiment, however, ADG was improved (P < 0.01) linearly. Carcass traits were not affected by treatment. Experiment 2 used 900 pigs to evaluate possible carryover effects on performance and carcass merit from feeding 6% fat. The experiment was divided into four phases: 25 to 45, 45 to 70, 70 to 90, and 90 to 115 kg; lysine contents of the control diets fed in each phase were 1.23, 1.05, 0.81, and 0.63%, respectively. The six treatments consisted of no added fat throughout the experiment or 6% added fat fed from 25 to 45 kg, 25 to 70 kg, 25 to 90 kg, 25 to 115 kg, or 45 to 70 and 90 to 115 kg. Carryover effects for ADG and G:F (P < 0.07) were found for the 90- to 115-kg interval and for ADFI and ME intake (P < 0.05) for the 45- to 70- and 70-to 90-kg intervals. When fat was added in the previous weight interval, ADG and G:F were improved and ADFI and ME intake were decreased in the subsequent weight interval. Pigs fed fat from 25 to 115 kg had more (P < 0.05) backfat and lower (P < 0.05) carcass leanness than pigs on the other treatments. These data suggest that fat can be added or removed from diets of growing-finishing pigs without any detrimental carryover effects. In fact, the positive carryover effect on ADG and G:F from 95 to 115 kg suggests that feeding fat from 25 to 95 kg will maximize performance over the total growing-finishing period but minimize any detrimental effects of added fat on carcass leanness.  相似文献

We have examined the effect of Pen, an aqueous extract of the dry mycelium of Penicillium chrysogenum, on plant–pathogen interactions. Pen controlled a broad range of pathogens on several crop plants under greenhouse and field conditions. Pen protected grapevine from downy and powdery mildew (caused by Plasmopara viticola and Uncinula necator), tomato from early blight (caused by Phytophthora infestans), onion from downy mildew (Peronospora destructor) and apple trees from apple scab (caused by Venturia inaequalis) to a similar extent as fungicides such as copper and sulphur or well-known inducers such as benzothiadiazole or β-aminobutyric acid. Pen had no major direct fungicidal effect and is thus supposed to protect plants by activating their defense mechanisms. The raw material for extraction of Pen was available in constant quality, a prerequisite for commercial application. Under certain conditions, Pen caused phytotoxic side effects. The symptoms mostly consisted of small necrotic spots or, more rarely, of larger necrotic areas. The development of the symptoms was dependent on several parameters, including concentration of Pen, the number of applications, the persistence on the plant tissue, the plant species and variety and environmental conditions. In grapevine, a partially purified fraction of Pen was much less toxic than the crude Pen extract, but protected the plants to a similar extent against P. viticola. Our data show that Pen has interesting and unique properties as a plant protection agent, but more research is needed to further reduce its phytotoxic side effects.  相似文献

The relative effect of metaphylactic ceftiofur crystalline free acid (CCFA) versus metaphylactic tilmicosin was evaluated in beef calves under commercial feedlot conditions in Nebraska. At feedlot arrival, 11,605 animals at ultrahigh risk of developing bovine respiratory disease (BRD) were allocated to one of three experimental groups: CCFA-3 (6.6 mg/kg SC), CCFA-7 (6.6 mg/kg), or TILM-3 (tilmicosin, 10 mg/kg SC). Animals were eligible for subsequent BRD treatment 3 (CCFA-3 and TILM-3 groups) or 7 (CCFA-7 group) days later. Compared with the TILM-3 group, overall chronicity, overall mortality, BRD mortality, and metabolic mortality rates were significantly (P < .05) lower in the CCFA-3 and CCFA-7 groups; average daily gain was significantly (P < .05) higher in the CCFA-3 group; the proportion of quality grade No Roll carcasses was significantly (P < .05) lower in the CCFA-3 and CCFA-7 groups; and there were per-animal advantages of 22.05 dollars and 18.98 dollars in the CCFA-3 and CCFA-7 groups, respectively. In beef calves at ultrahigh risk of developing BRD, it is more cost effective to administer metaphylactic CCFA than tilmicosin at feedlot arrival.  相似文献

We conducted two experiments to evaluate the effects of dietary energy density and lysine:calorie ratio on the growth performance and carcass characteristics of growing and finishing pigs. In Exp. 1, 80 crossbred barrows (initially 44.5 kg) were fed a control diet or diets containing 1.5, 3.0, 4.5, or 6.0% choice white grease (CWG). All diets contained 3.2 and 2.47 g of lysine/Mcal ME during growing (44.5 to 73 kg) and finishing (73 to 104 kg), respectively. Increasing energy density did not affect overall ADG; however, ADFI decreased and feed efficiency (Gain:feed ratio; G:F) increased (linear, P < .01). Increasing energy density decreased and then increased (quadratic, P < .06) skinned fat depth and lean percentage. In Exp. 2, 120 crossbred gilts (initially 29.2 kg) were used to determine the effects of increasing levels of CWG and lysine:calorie ratio fed during the growing phase on growth performance and subsequent finishing growth. Pigs were fed increasing energy density (3.31, 3.44, or 3.57 Mcal ME/kg) and lysine:calorie ratio (2.75, 3.10, 3.45, or 3.80 g lysine/Mcal ME). No energy density x lysine:calorie ratio interactions were observed (P > .10). Increasing energy density increased ADG and G:F and decreased ADFI of pigs from 29.5 to 72.6 kg (linear, P < .05). Increasing lysine:calorie ratio increased ADG and ADFI (linear, P < .01 and .07, respectively) but had no effect on G:F. From 72.6 to 90.7 kg, all pigs were fed the same diet containing .90% lysine and 2.72 g lysine/Mcal ME. Pigs previously fed with increasing lysine:calorie ratio had decreased (linear, P < .02) ADG and G:F. Also, pigs previously fed increasing CWG had decreased (linear, P < .03) ADG and ADFI. From 90.7 to 107 kg when all pigs were fed a diet containing .70% lysine and 2.1 g lysine/Mcal ME, growth performance was not affected by previous dietary treatment. Carcass characteristics were not affected by CWG or lysine:calorie ratio fed from 29.5 to 72.6 kg. Increasing the dietary energy density and lysine:calorie ratio improved ADG and G:F of growing pigs; however, pigs fed a low-energy diet or a low lysine:calorie ratio from 29 to 72 kg had compensatory growth from 72 to 90 kg.  相似文献

The aim of this study was to identify genomic regions associated with 305-day milk yield and lactation curve parameters on primiparous (n = 9,910) and multiparous (n = 11,158) Holstein cows. The SNP solutions were estimated using a weighted single-step genomic BLUP approach and imputed high-density panel (777k) genotypes. The proportion of genetic variance explained by windows of 50 consecutive SNP (with an average of 165 Kb) was calculated, and regions that accounted for more than 0.50% of the variance were used to search for candidate genes. Estimated heritabilities were 0.37, 0.34, 0.17, 0.12, 0.30 and 0.19, respectively, for 305-day milk yield, peak yield, peak time, ramp, scale and decay for primiparous cows. Genetic correlations of 305-day milk yield with peak yield, peak time, ramp, scale and decay in primiparous cows were 0.99, 0.63, 0.20, 0.97 and −0.52, respectively. The results identified three windows on BTA14 associated with 305-day milk yield and the parameters of lactation curve in primi- and multiparous cows. Previously proposed candidate genes for milk yield supported by this work include GRINA, CYHR1, FOXH1, TONSL, PPP1R16A, ARHGAP39, MAF1, OPLAH and MROH1, whereas newly identified candidate genes are MIR2308, ZNF7, ZNF34, SLURP1, MAFA and KIFC2 (BTA14). The protein lipidation biological process term, which plays a key role in controlling protein localization and function, was identified as the most important term enriched by the identified genes.  相似文献

Feedlots in southern Alberta apply composted (CM) or stockpiled (SM) manure with straw (ST) or wood-chip (WD) bedding to cropland, but few studies have examined the effect of manure type and bedding material on carbon composition of these feedlot amendments prior to land application using solid-state 13C NMR-DPMAS (nuclear magnetic resonance-direct polarization, magic angle spinning). The particulate organic matter fraction was extracted from four amendment treatments (CM-ST, CM-WD, SM-ST, SM-WD) to avoid possible paramagnetic interference from considerable mineral soil in the manure from the unpaved feedlot. The hypothesis was that O-alkyl C of POM should be lower for more decomposed manure types (CM than SM) and bedding materials (ST than WD), and that alkyl C, aromatic C, carboxyl C, aromaticity, and alkyl:O-alkyl (A:O-A) ratio should be greater for the more decomposed amendments. The C composition of all feedlot amendments was dominated by aromatic C (8%–14%) and O-alkyl (7%–14%) C and had considerable less contribution from carboxyl (2%–4%) and alkyl C (1%–3%). The manure type hypothesis was supported for O-alkyl C (but not for the other three C groups), aromaticity, but not the A:O-A ratio. The bedding hypothesis was supported for O-alkyl C, aromaticity, and A:O-A ratio, but not for alkyl C, aromatic C, and carboxyl C. A decrease in O-alkyl C, increase in aromaticity, and increase in A:O-A ratio (bedding only) with more decomposed manure types or bedding materials suggested that these 13C NMR parameters may have potential to evaluate the maturity and stability of composted feedlot manures.  相似文献
