Assessment of urine levels of luteinizing hormone (LH) for predicting the reproductive status of animals is in practice. The aim of this study was to predict the period of ovulation based on the urine levels of LH for timed‐artificial insemination to increase the conception rate in buffaloes, which are naturally silent‐oestrous animals. Level of LH in urine was assessed using ELISA, and a cut‐off LH concentration for prediction of ovulation period was obtained using receiver operating characteristic analysis. Artificial insemination was performed before‐ and after ‐positive prediction of ovulation period adopting this method, and the rates of conception were assessed. Urine LH level of 105 mIU/ml (n = 14) was derived as a cut‐off concentration which predicts the ovulation period. The buffaloes in the positively predicted group (day 1 or 2) inseminated via intracervical route had an increase in the conception rate (83.33%); however, the insemination in the before‐positive‐prediction group resulted in poor conception rates (day 0; 16.66%) compared to that of the naturally inseminated group (day 0; 75.0%). In conclusion, the urinary LH would possibly be a fairly reliable predictor of the ovulation period. The day when cut‐off LH concentration is obtained may be taken as the most favourable time for artificial insemination, so as to attain a much better rate of conception in the buffalo. 相似文献

Frozen-thawed (FT) boar sperm have a reduced fertile life, due in part to a capacitation-like status induced by cooling. Reversal of this cryocapacitation in vitro by exposure to boar seminal plasma (SP) has been demonstrated. The objective of these studies was to determine the effect of SP on the ability of FT sperm to create an oviductal sperm reservoir following artificial insemination (AI). In Experiment one, 35 pre-pubertal gilts were injected (IM) with 400 IU eCG plus 200 IU hCG to induce oestrus. At detection of oestrus, gilts were inseminated with 3 x 10(9) live sperm, either fresh (FS; n = 13), FT (n = 10), or FT supplemented with 10% v/v SP (n = 12). Gilts were killed 8 h later, their reproductive tracts recovered and the uterotubal junctions (UTJs) flushed to recover sperm. Fewer (p < 0.01) sperm were recovered following FT, compared to FS, inseminations, and there was no evident effect of SP. In Experiment two, 30 pre-pubertal gilts received IM injections of 1000 IU eCG followed by 5 mg pLH 80 h later to control time of ovulation. Gilts were inseminated with 3 x 10(9) live FS sperm (n = 6), FT sperm (n = 15) or FT sperm plus 10% SP (n = 9) at 12 h before ovulation and then sacrificed 8 h later. The UTJs were dissected and flushed for sperm recovery. Fewest (p < 0.001) sperm were recovered following FT insemination and there was no evident effect of SP. These data demonstrate that the size of the sperm reservoir is markedly reduced in gilts inseminated with FT sperm. However, the lack of effect of SP suggested that either it did not reverse cryocapacitation or that such a reversal does not impact the in vivo ability to create a sperm reservoir. 相似文献

Fertility of primiparous lactating does in the early postpartum (pp) period is very low mainly due to pronounced deficient energy intake, influencing oocyte and embryo developmental competence. The hypothesis used in this work was that high‐lignin fibre diet supplied during the rearing period could increase feed intake and, consequently, improve the reproductive physiology and metabolic status of primiparous does in the early pp period. Diets with high‐lignin [HL: 15.8% dry matter (DM)] or standard‐lignin content (SL: 4.9% DM) were supplied until parturition time. No diet effects in serum oestradiol, progesterone concentrations and follicle categories were found in the histological study. Metaphase II rate of in vitro‐matured oocytes was significantly higher in the SL vs the HL group (p < 0.001). Cytoplasmically degenerated oocytes (in terms of abnormal distribution of cortical granules) and follicular atresia rate were significantly lower in the SL group than in the HL group (p < 0.05 and p < 0.005 respectively). In addition, HL‐fed does showed lower number of viable embryos and higher rate of retarded in vivo‐recovered embryos compared with the SL group (p < 0.05). Neither in vitro embryo development of viable embryos nor conception rate was significantly different between groups. Feed intake increased during the first pregnancy in the HL group (p < 0.05), but not during early lactation. Serum protein, non‐esterified fatty acid and leptin concentrations, as well as estimated body composition were similar in does fed with both diets. In conclusion, the enhancement of reproductive management by using highly lignified products in rearing diets does not seem to report physiological reproductive benefits affecting oocyte maturation rate and embryo viability in primiparous lactating does. 相似文献

In a laboratory rainfall simulator study soil surface roughness was measured using contact (roller chain, pin meter) and noncontact devices (laser scanner, photogrammetry). Soil surfaces with two initial roughness conditions (aggregates < 20 mm and < 63 mm) were investigated before and after 90 mm of simulated rainfall. Measured plot area was 50 by 55 cm. A comparison of soil roughness measurement techniques was undertaken with regard to data acquisition and computation efforts, resolution, precision and capability to represent soil surface features.As for the contact methods, resolution (cm range) and precision (mm range) is limited which constrains their application to calculation of simple surface parameters. Resolution and precision in the sub millimeter range could be obtained with the laser scanner, while for the photogrammetric method the measurement uncertainty was approximately 1–2 mm. Measurement time was highest (90 min) for the pin meter technique, though data were ready to use for analyses. Laser scanner measurements took 34 min. Several steps of data post-processing required 30 more minutes. Data acquisition was fastest for photogrammetry (5 min), but expert knowledge as well as special hard- and software were necessary for time-consuming photo analyses, taking about 120 min.The chain and pin meter were compared using a profile index. Profile lengths matched well for smooth surfaces; on rough surfaces, the chain meter gave shorter profile lines. Between profile index from chain measurements and the random roughness derived from pin meter data a polynomial regression could be found. Parameters of distributions of elevations as well as inclinations and depressions were used to compare the laser scanner and the photogrammetric technique. Generally the laser scanner was able to reproduce small aggregates as well as voids in between them, while DEM from stereophotos was smoothed between major aggregates. This led to skewed distributions of elevations and inclinations, as well as to a lower surface area (up to 39%), and a lower depression volume (up to 68%). Shapes of depressions were significantly different as well. The used photogrammetric technique is supposed to be successful in producing adequate DEMs for already smoothed surfaces, e.g. after rainfall events.The study revealed different fields of application and limitations of the compared devices. Using a non-adequate technique for certain situations will definitely have implications on further analyses concerning connectivity of runoff pathways, surface protection from raindrop impact or runoff detachment and sediment transport. 相似文献
