Fatty acid patterns of phospholipids and lipopolysaccharides in the characterisation of microbial communities in soil: a review 总被引:45,自引:0,他引:45

This review discusses the analysis of whole-community phospholipid fatty acid (PLFA) profiles and the composition of lipopolysaccharides
in order to assess the microbial biomass and the community structure in soils. For the determination of soil microbial biomass
a good correlation was obtained between the total amount of PLFAs and the microbial biomass measured with methods commonly
used for determinations such as total adenylate content and substrate-induced respiration. Generally, after the application
of multivariate statistical analyses, whole-community fatty acid profiles indicate which communities are similar or different.
However, in most cases, the organisms accounting for similarity or difference cannot be determined, and therefore artefacts
could not be excluded. The fatty acids used to determine the biomass vary from those which determine the community structure.
Specific attention has to be paid when choosing extraction methods in order to avoid the liberation of fatty acids from non-living
organic material and deposits, and to exclude the non-target selection of lipids from living organisms, as well. By excluding
the fatty acids which were presumed to be common and widespread prior to multivariate statistical analysis, estimates were
improved considerably. Results from principal component analysis showed that determining the levels of fatty acids present
in both low and high concentrations is essential in order to correctly identify microorganisms and accurately classify them
into taxonomically defined groups. The PLFA technique has been used to elucidate different strategies employed by microorganisms
to adapt to changed environmental conditions under wide ranges of soil types, management practices, climatic origins and different
perturbations. It has been proposed that the classification of PLFAs into a number of chemically different subgroups should
simplify the evaluating procedure and improve the assessment of soil microbial communities, since then only the subgroups
assumed to be involved in key processes would be investigated.

The effect of termites on the breakdown of Pennisetum pedicellatum straw under Sahelian conditions was studied in northern Burkina Faso during the dry and wet seasons of 1995. Litterbags were
laid on plots treated with drieldrin at a rate of 500 g a.i. (active ingredient) ha–1 to exclude termite activity and on untreated plots. Termites were the only macroscopically visible consumers which were observed
in the litterbags during the dry and wet season. In litterbags with no termites present the rate of breakdown of straw was
lower during both the dry or wet seasons than in litterbags with termites. In the litterbags with termites, much of the mulch
was taken off and replaced by termite-made sheeting. From April to September over 60% of the added material disappeared from
termite litterbags, while only 18% of the added straw disappeared from litterbags without termites. From October to December,
28% of the straw disappeared from termite-infested litterbags versus 8% from litterbags without termites. The contribution
of termites to the breakdown of straw was estimated to be over 70% for each of the two experiments. Under Sahelian conditions,
the breakdown of organic residues is strongly influenced by termites.

Fresh strawberries (Fragaria x ananassa Duch.), raspberries (Rubus idaeus Michx.), highbush blueberries (Vaccinium corymbosum L.), and lowbush blueberries (Vaccinium angustifolium Aiton) were stored at 0, 10, 20, and 30 degrees C for up to 8 days to determine the effects of storage temperature on whole fruit antioxidant capacity (as measured by the oxygen radical absorbing capacity assay, Cao et al., Clin. Chem. 1995, 41, 1738-1744) and total phenolic, anthocyanin, and ascorbate content. The four fruit varied markedly in their total antioxidant capacity, and antioxidant capacity was strongly correlated with the content of total phenolics (0.83) and anthocyanins (0.90). The antioxidant capacity of the two blueberry species was about 3-fold higher than either strawberries or raspberries. However, there was an increase in the antioxidant capacity of strawberries and raspberries during storage at temperatures >0 degrees C, which was accompanied by increases in anthocyanins in strawberries and increases in anthocyanins and total phenolics in raspberries. Ascorbate content differed more than 5-fold among the four fruit species; on average, strawberries and raspberries had almost 4-times more ascorbate than highbush and lowbush blueberries. There were no ascorbate losses in strawberries or highbush blueberries during 8 days of storage at the various temperatures, but there were losses in the other two fruit species. Ascorbate made only a small contribution (0.4-9.4%) to the total antioxidant capacity of the fruit. The increase observed in antioxidant capacity through postharvest phenolic synthesis and metabolism suggested that commercially feasible technologies may be developed to enhance the health functionality of small fruit crops. 相似文献

A new, simple, precise, and rapid ion chromatography (IC) method has been developed to determine chloride in mustard sauces using a mixture of phthalic acid, acetone, and water adjusted to pH 5.0 as eluent. Conductometric detection was carried out. The retention time for chloride was 1.5 min. Linearity was obtained up to a concentration level of 100 mg/L NaCl. The method was statistically evaluated for accuracy and precision after being used to assay the chloride from mustard sauces. Within the same samples, the chloride levels obtained by IC were compared with the sodium concentrations quantified by atomic absorption spectrophotometry. 相似文献

An ethyl acetate-soluble extract of Chorizanthe diffusa was found to exhibit significant antioxidant activity, as judged by scavenging stable 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals and inhibition of 12-O-tetradecanoylphorbol 13-acetate (TPA)-induced free radical formation with cultured HL-60 cells. Bioassay-directed fractionation of this extract using the DPPH antioxidant assay as a monitor led to the isolation of five structurally related flavonoids (1-5), including the novel compound 5,8,3',4',5'-pentahydroxy-3, 7-dimethoxyflavone (1). Isolates 1-5 demonstrated varying degrees of antioxidant or antimutagenic activity. Two of the compounds, 5,7,3', 4'-tetrahydroxy-3-methoxyflavone (2) and quercetin (4), were subsequently found to inhibit carcinogen-induced preneoplastic lesions in a mouse mammary organ culture model. Palm oil carotenoids are a mixture of alpha- and beta-carotenes, which are used as food colorants. They may also be applied as a functional food ingredient because of the provitamin A activity of alpha- and beta-carotenes and their proposed beneficial roles in the prevention of chronic diseases. This paper discusses the results of an incomplete balanced crossover study with 69 healthy adult volunteers to compare palm oil carotenoids with synthetic beta-carotene in their efficacies to increase plasma levels of carotenoids. Four days of supplementation with natural palm oil carotenoids (7.6 mg/day of alpha-carotene, 11.9 mg/day of all-trans-beta-carotene, 7.5 mg/day of cis-beta-carotene) or synthetic beta-carotene (23.8 mg/day of all-trans-beta-carotene, 4.4 mg/day of cis-beta-carotene), added to a mixed meal, resulted in significant increases in plasma levels of the supplied carotenoids as compared to consumption of a low-carotenoid meal (i.e., 7.2-fold increase in alpha-carotene and 3.5-fold increase in all-trans-beta-carotene following palm oil carotenoids; 6.9-fold increase in all-trans beta-carotene following synthetic beta-carotene). As the carotenoid content differed between the treatments, the relative plasma responses were calculated per milligram of beta-carotene intake. These were similar for the two supplements, suggesting that the presence of alpha-carotene does not affect the bioavailability of beta-carotene from palm oil. It was concluded that 4 days of supplementation with palm oil carotenoids or synthetic beta-carotene improves the plasma beta-carotene status substantially, whereas alpha-carotene is additionally delivered by the palm oil supplement. 相似文献

The relative mobility of pyrithiobac [sodium 2-chloro-6-(4, 6-dimethoxypyrimidin-2-ylthio)benzoate], a new herbicide used for postemergence control of broadleaf weeds in cotton (Gossypium hirsutum), was evaluated and compared against that of bromide (Br(-)) tracer on four soils representative of cotton-growing regions using intact soil columns under saturated flow conditions. Pyrithiobac breakthrough curves were asymmetrical in shape with significant tailing and displaced to the left of 1 pore volume in the Houston Black clay (fine, montmorillonitic, thermic Udic Pellustert), Orelia fine sandy clay loam (fine-loamy, mixed, hyperthermic Typic Ochraqualfs), and Ships silty clay (very-fine, mixed, thermic Udic Chromustert) soils. Breakthrough of pyrithiobac in the Hidalgo sandy loam soil (fine-loamy, mixed, hyperthermic Typic Calciustoll) was delayed and more symmetrical, with peak pyrithiobac concentration reached after 1.2 pore volumes. The immobile pore water (IPW) fractions estimated from the Br(-) breakthrough curves ranged from 20 to 87% of total pore water. The IPW values demonstrated that soils with the greatest amount of IPW (Ships with IPW = 87.3%) exhibited the most rapid movement of pyrithiobac (peak concentration after 0.04 pore volume). The experimentally determined pyrithiobac breakthrough curves confirmed the high mobility of this herbicide in these alkaline and predominantly smectitic soils. Milled oat groat pearlings, trichomes, flour, and bran were extracted with methanol and the fractions tested in vitro for antioxidant capacity against low-density lipoprotein (LDL) oxidation and R-phycoerythrin protein oxidation in the oxygen radical absorbance capacity (ORAC) assay. The oxidative reactions were generated by 2,2'-azobis(2-amidinopropane) HCl (AAPH) or Cu(2+) in the LDL assay and by AAPH or Cu(2+) + H(2)O(2) in the ORAC assay and calibrated against a Trolox standard to calculate Trolox equivalents (1 Trolox equivalent = 1 TE = activity of 1 micromol of Trolox). The antioxidant capacity of the oat fractions was generally consistent with a potency rank of pearlings (2.89-8.58 TE/g) > flour (1.00-3.54 TE/g) > trichome (1.74 TE/g) = bran (1.02-1.62 TE/g) in both LDL and ORAC assays regardless of the free radical generator employed. A portion of the oat antioxidant constituents may be heat labile as the greatest activity was found among non-steam-treated pearlings. The contribution of oat tocols from the fractions accounted for <5% of the measured antioxidant capacity. AAPH-initiated oxidation of LDL was inhibited by the oat fractions in a dose-dependent manner, although complete suppression was not achieved with the highest doses tested. In contrast, Cu(2+)-initiated oxidation of LDL stimulated peroxide formation with low oat concentrations but completely inhibited oxidation with higher doses. White grape seeds were subjected to sequential supercritical fluid extraction. By increasing the polarity of the supercritical fluid using methanol as a modifier of CO(2), it was possible to fractionate the extracted compounds. Two fractions were obtained; the first, which was obtained with pure CO(2), contained mainly fatty acids, aliphatic aldehydes, and sterols. The second fraction, obtained with methanol-modified CO(2), had phenolic compounds, mainly catechin, epicatechin, and gallic acid. The fractions were bioassayed. Antimicrobial activities were checked on human pathogens, and a high degree of activity was obtained with the lipophilic fraction. Agrochemical activities on phytopathogenic fungi and activities on the etiolated wheat coleoptile bioassay were also checked. The more polar fraction was active in the latter bioassay. 相似文献

Nutrient release in clinoptilolite-phosphate rock (Cp-PR) systems occurs through dissolution and cation-exchange reactions. Investigating the kinetics of these reactions expands our understanding of nutrient release processes. Research was conducted to model transport kinetics of nutrient release in Cp-PR systems. The objectives were to identify empirical models that best describe NH4, K, and P release and define diffusion-controlling processes. Materials included a Texas clinoptilolite (Cp) and North Carolina phosphate rock (PR). A continuous-flow thin-disk technique was used. Models evaluated included zero order, first order, second order, parabolic diffusion, simplified Elovich, Elovich, and power function. The power-function, Elovich, and parabolic-diffusion models adequately described NH4, K, and P release. The power-function model was preferred because of its simplicity. Models indicated nutrient release was diffusion controlled. Primary transport processes controlling nutrient release for the time span observed were probably the result of a combination of several interacting transport mechanisms. 相似文献
