Abstract.— In South Carolina, studies have been conducted to develop rearing techniques for southern flounder Paralichthys lethostigma a candidate for aquaculture development and stock enhancement programs. To help define environmental tolerances, a variety of salinity studies were conducted with the early life stages of this species. Eggs were buoyant at 32 ppt and sank at 29 ppt with salinities of 30–31 ppt providing varying levels of suspension in the water column. Eggs incubated at 0 and 5 ppt all died, whereas 82.5% hatched at 10 ppt but larvae died shortly thereafter. At 63 h post-fertilization, there were no differences in hatch level for eggs incubated at salinities of 15 to 35 ppt (mean hatch level 98.5%). In a 72-h study, fish 3 wk post-metamorphosis (13.7 mm TL, 50-d-old) were acclimated to seven salinities ranging from 0–30 ppt. Fish held at 0 ppt salinity exhibited a statistically (P < 0.05) lower survival (20.0%) than those exposed to 5–30 ppt salinity concentrations. No differences were detected in survival (mean 99.1%) among fish held in the higher salinities. A second study examined the tolerance of older juveniles to lower salinities. Juvenile flounder (95.2 mm TL, 220-d-old) were acclimated to 0, 1,5 and 10 ppt salinities and reared for 2 wk. Results showed that fish could tolerate salinities of 0–10 ppt (100% survival). These data indicate that salinity tolerance of southern flounder increases with age. In addition to the short duration studies, a replicated 11-mo duration tank grow-out study was conducted at mean salinity 5.4 ppt and mean temperahue 22.6 C with an all male population. Flounder grew from a mean length of 100 mm to 213 mm TL and weight from 8.9 to 104.3 g. Growth of the cultured fish approximated that observed among male flounders in the wild.  相似文献

Increasingly knowledge is shared using new web‐based channels. The use of these channels is relatively new in the realm of soil science. However, they may prove to be very useful for connecting time‐poor, geographically dispersed audiences in an efficient and cost‐effective way. There has been limited study of this form of knowledge sharing for natural science (let alone soil science). In New South Wales (NSW), Australia, the NSW Department of Primary Industries (NSW DPI) has coordinated the Soil Network of Knowledge (SNoK) monthly webinar over 3 years, with over 40 webinars targeting an audience of next users of soil information. Aggregated data collected from these webinars indicated that the majority of information is used for professional development by participants. Whether this will eventually lead to on‐ground practice change because of the improved professional and technical knowledge of participants is still a moot point and will require further investigation over time. The information collected on attendance and participation showed that webinars are a good way to connect with some target audiences such as other government bodies that have a historical relationship with NSW DPI. However, other target audiences that NSW DPI has always found difficulty in engaging, such as private advisers, require more active overtures to drive participation. The growing subscriber base is encouraging.  相似文献

Gossypium barbadense L. cotton has significantly better fiber quality than Upland cotton (G. hirsutum L.); however, yield and environmental adaptation of G. barbadense is not as wide as Upland. Most cotton in the world is planted to Upland cultivars. Many attempts have been made, over a considerable number of years, to introgress fiber quality alleles from G. barbadense into Upland. However, introgression barriers, primarily in the form of interspecific incompatibility, have limited these traditional approaches. The use of chromosome substitution lines (CSL) as a bridge should provide a more efficient way to introgress alleles from G. barbadense into Upland. We crossed 18 G. barbadense CSL to three cultivars and developed a random mated population. After five cycles of random mating followed by one generation of self-pollination to increase the seed supply, we grew the random mated population and used 139 G. barbadense chromosome specific SSR markers to assess a random sample of 96 plants for introgression. We recovered 121 of 139 marker loci among the 96 plants. The distribution of the G. barbadense alleles ranged from 10 to 28 alleles in each plant. Among the 96 plants we found individual plants with marker loci from 6 to 14 chromosomes or chromosome arms. Identity by descent showed little relatedness among plants and no population structure was indicated by a heat map. Using CSL we were able to develop a mostly Upland random mated population with considerable introgression of G. barbadense alleles which should be useful for breeding.  相似文献

Verticillium wilt (VW, caused by Verticillium dahliae Kleb) is a destructive fungal soil-borne disease in Upland cotton (Gossypium hirsutum L.). High levels of VW resistance can be transferred into Upland from Pima cotton (G. barbadense L.) through interspecific introgression breeding. In this greenhouse study, VW resistance was evaluated in a multi-parent advanced generation inter-cross (MAGIC) introgressed line (IL) population, derived from a random mated Barbadense Upland population with five generations of intermating (called RMBUP-C4) between three Upland cotton cultivars and 18 CS-B Upland lines each carrying a pair of G. barbadense chromosome or arm in the TM-1 background. The objectives of this study were to, (1) evaluate VW resistance of 530 MAGIC ILs in the greenhouse; and (2) to identify lines with VW resistance in the MAGIC population based on a total of three replicated greenhouse tests. Approximately 8 plants for each line in each replicate were grown and screened for VW resistance using three parameters i.e., disease leaf severity rating, percentage defoliated leaves, and percentage infected plants, with a total of ~?25,190 plants evaluated. A correlation analysis indicated that the three parameters were significantly and positively correlated with one another in each test. The disease leaf severity rating was the best parameter to assess VW resistance due to its relatively low coefficient of variation and its higher resolution to differentiate resistant genotypes from susceptible ones. Of the 530 genotypes, 5 showed resistance to VW, namely, NMIL348, NMIL518, NMIL405, NMIL290, NMIL307 and had higher levels of resistance to VW with mean disease leaf severity ratings, percentage of defoliated leaves, and percentage of infected plants across three tests ranging from 0.58–1.46, 9.46–26.74, and 25–95%, respectively. These lines can be used as parental lines to improve VW resistance in cotton breeding programs.  相似文献

The purpose of the present study was to evaluate the species composition and salinomycin sensitivity of Eimeria oocysts isolated from commercial broiler farms that differed by means of coccidiosis control (anticoccidial drugs [ACD] vs. live oocyst vaccines [VAC]). A comparison of Eimeria species composition and salinomycin sensitivity was also made before and after a producer switched from salinomycin to live oocyst vaccines. In general, no significant difference was observed in the concentration of Eimeria spp. oocysts in litter from VAC-utilizing farms compared to litter from ACD-utilizing farms. Application of PCR-based methods to detect coccidia found that Eimeria species distribution in litter from VAC operations more closely resembled the species composition in the live oocyst vaccines. Drug sensitivity testing found that Eimeria oocysts from VAC operations displayed greater salinomycin sensitivity as measured by weight gain and feed conversion efficiency compared to oocysts from ACD farms. These findings provide additional evidence for the usefulness of live oocyst vaccines to restore ionophore sensitivity in poultry operations that contain an ionophore-resistant population of Eimeria spp. oocysts.  相似文献

The study is based on 141 pregnant Bos indicus cows, from days 20 to 70 post‐insemination. First, special attention was given to the macroscopically observable phenomena of attachment of the conceptus to the uterus, i.e. the implantation, from about days 20 to 30 post‐insemination up to day 70, and placentome development by growth, vascularization and increase in the number of cotyledons opposite to the endometrial caruncles. Secondly, as for the conceptuses, semiquantitative, statistical analyses were performed of the lengths of chorio‐allantois, amnion and yolk sac; and the different parts of the centre and two extremes of the yolk sacs were also analysed. Thirdly, the embryos/foetuses corresponding to their membranes were measured by their greatest length and by weight, and described by the appearance of external developmental phenomena during the investigated period like neurulation, somites, branchial arcs, brain vesicles, limb buds, C‐form, pigmented eye and facial grooves. In conclusion, all the data collected in this study from days 20 to 70 of bovine pregnancy were compared extensively with corresponding data of the literature. This resulted in an ‘embryo/foetal age‐scale’, which has extended the data in the literature by covering the first 8 to 70 days of pregnancy. This age‐scale of early bovine intrauterine development provides model for studies, even when using slaughtered cows without distinct knowledge of insemination or fertilization time, through macroscopic techniques. This distinctly facilitates research into the cow, which is now being widely used as ‘an experimental animal’ for testing new techniques of reproduction like in vitro fertilization, embryo transfer and cloning.  相似文献

Soluble fractions of human intestinal cancer and fetal intestinal cell membranes produced delayed hypersensitivity reactions in patients with intestinal cancer. These soluble fractions and perchloric acid extracts of intestinal cancer cells were fractionated by polacrylamide-gel electrophoresis.
