Tyrosine hydroxylase (TH) is the rate-limiting enzyme of dopamine (DA) biosynthesis, which is up-regulated by vitamin C administration. Nurr1 gene is highly expressed in brain and important for midbrain DAergic cell development and survival. But, the role of vitamin C and/or vitamin E during Nurr1 expression is yet to be known. Further, the synergistic effect of vitamins C and E on TH expression has not yet been explored clinically. Therefore, we studied the effects of single or co- administration of vitamin C (0.5 mM) and vitamin E (1 mM) for 72 hr, on both TH and Nurr1 expression in in vitro primary cultured gestational days (GD) 13.5 rat ventral mesencephalon (VM) by Western blot and immunocytochemistry. Our study revealed highest expressions of both TH and Nurr1 in the vitamin C + vitamin E treated group. TH expression was also increased in the vitamin C treated group than that of the control group, but the vitamin E treated group did not show any statistically significant effect. However, both the vitamin C and the vitamin E treated groups revealed increased expression of Nurr1 protein as compared with the control group. The present experimental data suggest that vitamin C can up-regulate the protein expression of TH, but Nurr1 level was elevated either by vitamin C or by vitamin E administration. Further, vitamin E acts synergistically with vitamin C to elevate TH and Nurr1 expression, which is the most novel finding of our study and for the first time; we reported this result, since there have been no published data on the synergistic effect of both the antioxidant vitamins on TH and Nurr1 expression in VM. As the motor function defect due to the progressive loss of DAergic neuron is the major reason of Parkinsonism, therefore, the results of our study finally suggest the effective role of vitamin C and vitamin E during early treatment of Parkinsonism.  相似文献

We identified a potential molecular marker associated with meat quality traits in the myosin heavy chain 4, MYH4 gene of Landrace pigs. Sequencing revealed a single nucleotide polymorphism (SNP; g.‐1398G>T) in the 5' upstream region of MYH4. It was significantly associated with the number of type IIa muscle fibers and water‐holding capacity based on filter‐paper fluid uptake. The GG genotype groups had a greater number of type IIa fibers and a larger area composed of type IIa fibers than the other genotype group (P = 0.004 and P = 0.061, respectively). Expression level of MYH4 gene in the genotype TT or GT was higher than in genotype of GG (P < 0.0001). The T allele may enhance expression level of MYH4 gene and then the portion of IIb type fiber in the muscle be increased by the T allelle. Therefore, we suggest that the g.‐1398G>T in the 5' upstream region of the porcine MYH4 may be used as a molecular marker for meat quality traits, although its functional effect is not defined yet.  相似文献

The effects of corn particle size on nutrient digestibility and energy utilization in pigs were determined under optimal (experiment 1, 25?±?1 °C) or heat stress (experiment 2, 37?±?1 °C) conditions. In Exp. 1 and 2, five experimental diets were tested using a 5?×?5 Latin square design involving five barrows (Landrace × Yorkshire × Duroc, average initial body weight of 30?±?1 kg and 45.0?±?1.8 kg, respectively, in individual metabolic cages). Dietary treatments were as follows: 200-, 300-, 400-, 600-, 800-μm corn particle sizes obtained by mesh screens. Under optimal thermal conditions, digestibility of dry matter (DM) and crude fiber (CF) from 200-μm diet was higher (P?<?0.05) compared to that from the 300-μm and 400-μm diets. The digestibility of crude protein (CP) and ether extract (EE) was the highest (P?<?0.05) at the 200-μm particle size. The apparent total tract digestibility of energy was significantly higher (P?<?0.05) on the 200-μm diet. Under heat stress, digestibility of CF when corn was ground to 600 μm was higher (P?<?0.05) compared to 300 and 400 μm. Digestibility of NDF and ADF was the highest (P?<?0.05) at 600-μm corn particle size. In conclusion, grinding corn to 200-μm corn particles had a positive effect on DM, CP, EE, and CF under optimal thermal condition, while the 600-μm corn particle size had positive effects on digestibility of CF, NDF, and ADF than 200-μm corn particle size under heat stress.

This study was performed to elucidate the anticancer mechanism of a lipid-soluble ginseng extract (LSGE) by analyzing induction of apoptosis and arrest of cell cycle progression using the NCI-H460 human lung cancer cell line. Proliferation of NCI-H460 cells was potently inhibited by LSGE in a dose-dependent manner. The cell cycle arrest at the G0/G1 phase in NCI-H460 cells was induced by LSGE. The percentage of G0/G1 phase cells significantly increased, while that of S phase cells decreased after treatment with LSGE. The expression levels of cyclin-dependent kinase2 (CDK2), CDK4, CDK6, cyclin D3 and cyclin E related to G0/G1 cells progression were also altered by LSGE. In addition, LSGE-induced cell death occurred through apoptosis, which was accompanied by increasing the activity of caspases including caspase-8, caspase-9 and caspase-3. Consistent with enhancement of caspase activity, LSGE increased protein levels of cleaved caspase-3, caspase-8, caspase-9, and poly-ADP-ribose polymerase (PARP). These apoptotic effects of LSGE were inhibited by the pan-caspase inhibitor Z-VAD-fmk. These findings indicate that LSGE inhibits NCI-H460 human lung cancer cell growth by cell cycle arrest at the G0/G1 phase and induction of caspase-mediated apoptosis.  相似文献

H9N2 influenza viruses are endemic in many Asian countries. We demonstrated that H9N2 influenza viruses isolated from poultry in Korean live bird markets are genetically changing and could cause the clinical signs in layers. Genetic analysis showed that Korean avian H9N2 influenza viruses are distinct from H9N2 influenza viruses circulating in poultry in China and Hong Kong. When we infected layers with H9N2 isolates, layers showed about 30% mortality and the reduction of egg productions. Considering that H9N2 influenza virus is one of potential pandemic candidates, the continuous surveillance is needed to monitor avian H9N2 influenza viruses for the poultry industry and humans.  相似文献

PVC and SAN are often mixed to compensate for the disadvantages of each polymer. Miscibility and thermal stability of PVC/SAN blend were investigated in this study by blending SAN polymer having 20, 24, 28, 32 % of acrylonitrile contents. Two polymers were mixed using a melt blending method with a single screw extruder. DSC them ogram was used to evaluate miscibility of the two polymers. SAN having 24 % of acrylonitrile showed the best miscibility with PVC. In order to evaluate degradation behavior, blended polymer was heat treated in DSC furnace and glass transition temperature was measured consecutively. Glass transition temperature increased continuously with annealing time due to degradation and cross-linking of polymer chains. Melt index of blended polymer was always higher than that of PVC.  相似文献

Objective  To compare the effect of three different administration rates of one dose of propofol on the depth and duration of anaesthesia and cardiopulmonary function during induction of anaesthesia in rats using electroencephalogram (EEG) and clinical signs.
Study design  Prospective, randomized experimental trial.
Animals  Twenty-one, adult, male Sprague-Dawley rats weighing 341 ± 26 g (mean ± SD) (325 to 480 g).
Methods  Animals were randomly divided into three groups to receive 20 mg kg−1 propofol as a bolus injection over 1, 2 or 3 minutes (groups P1, P2 and P3 respectively) intravenously (IV). The total duration and number of burst suppression (BS) episodes in the EEG, the time to loss of righting reflex, reflex score from electrical stimulation, respiratory rate, mean arterial pressure and pulse rate were measured from the beginning of propofol injection.
Results  While loss of reflex to electrical stimulus and time to loss of righting reflex in group P3 were slower than in other groups, the total duration and number of BS episodes in group P3 were significantly higher than in groups P1 and P2 and cardiopulmonary depression was less prominent in group P3 than in groups P1 and P2 up to 2 minutes after the start of administration.
Conclusions  Twenty milligram per kg propofol administration IV for 3 minutes increased the duration of anaesthesia and decreased cardiopulmonary depression in rats.
Clinical relevance  Slower infusion of propofol produced surgical anaesthesia with less cardiopulmonary depression in rats.  相似文献

Objective  To investigate the effects of atipamezole administered before butorphanol, on tail-flick latency (TFL) and also following medetomidine–ketamine anaesthesia in rats.
Study design  Prospective, randomized experimental study.
Animals  Thirty-four adult male Sprague–Dawley rats weighing 260–390 g.
Methods  TFL in 50 °C water was used to measure antinociception. In the first experiment, rats received saline ( n  = 5) or atipamezole ( n  = 5) followed by butorphanol treatments. In the second experiment, three groups of rats received saline ( n  = 8), atipamezole ( n  = 8) or atimpamezole ( n  = 8) followed by butorphanol 60 minutes after medetomidine–ketamine administration.
Results  In the first experiment, butorphanol significantly increased TFL compared to baseline. Atipamezole significantly decreased this effect. In the second experiment, TFL was significantly increased after recovery from medetomidine–ketamine anaesthesia compared to baseline. This was almost completely blocked by atipamezole. Atipamezole with butorphanol after recovery from the anaesthesia significantly reduced TFL compared to saline but still significantly increased TFL compared to the baseline.
Conclusion and clinical relevance  Atipamezole attenuated the analgesic effects of butorphanol. When postoperative atipamezole is used to hasten recovery from anaesthesia in rats, it may interfere with the postoperative analgesic effect of butorphanol.  相似文献

A series of N-substituted phthalimide derivatives were synthesized based on a pharmacophore study of paecilocin A (a natural PPAR-γ agonist) and synthetic leads. The introduction of hydrophilic and hydrophobic groups to the phthalimide skeleton yielded compounds 3–14. Compound 7 showed significant PPAR-γ activation in a luciferase assay using rat liver Ac2F cells. Docking simulations showed that a free hydroxyl group on the phthalimide head and a suitable hydrophilic tail, including a phenyl linker, were beneficial for PPAR-γ activation. Compound 7 and rosiglitazone concentration-dependently activated PPAR-γ with EC50 values of 0.67 μM and 0.028 μM, respectively. These phthalimide derivatives could be further investigated as a new class of PPAR-γ ligands.  相似文献
