Brown (BT) and rainbow trout (RT) in freshwater (FW) were treated with ovine growth hormone (GH), GH + iopanoic acid (IOP), and GH + IOP plus triiodothyronine (T3) for RT only. After 1 week of treatment, trout were transferred to 30 o/oo SW and treatment continued. In FW, GH treatment increased significantly plasma T3 level (BT) and T3/T4 ratio (BT and RT) by stimulating T4 to T3 deiodination. In the GH + IOP group, the plasma T3 levels and T3/T4 ratio fell significantly as T4 to T3 deiodination was inhibited. In GH + IOP + T3-treated RT, plasma T3 and T3/T4 ratios increased significantly relative to other groups. No mortality occurred and plasma osmolarity (PO) was not altered by any treatment in FW. After transfer to SW, all IOP + GH trout died within 2 (BT) or 3 days (RT). All GH-treated or control BT survived to the end of the experiment (6 days). RT survival rates tended to be improved in GH and GH + IOP + T3 groups relative to controls. Correlatively on day 1 the PO increase was significantly higher in IOP + GH groups (BT and RT) than in the other groups and significantly lower in GH and GH + IOP + T3 treated RT than in controls from days 1 to 6. These data confirm the requirement of T3 and deiodination of T4 to T3 for the development of hypoosmoregulatory mechanisms in SW as previously shown (Lebel and Leloup 1992). Furthermore, the suppression of the hypoosmoregulatory effect of GH, when conversion of T4 to T3 was inhibited by IOP and the reversal when T3 was added to IOP + GH treatment suggests that GH osmoregulatory action in SW acts via the simulation of T4-5′ monodeiodination which increases T3 production.  相似文献

A large set of genes was identified in the phytopathogenic fungus Botrytis cinerea by using an expressed sequence tag approach. The fungus was grown in axenic culture and a cDNA library was produced. From this library, 6559 ESTs were obtained. The combined sequences represent 3026 unisequences that corresponds to approximately one-quarter of the estimated total number of genes in B. cinerea. Approximately 18% of the ESTs showed significant similarities with genes coding for proteins with known functions,~56% were similar to genes coding for proteins with unknown functions and ~26% were orphans. A substantial B. cinerea gene inventory including putative virulence factors was therefore obtained and is now available at the Génoplante-Info Database interface (http://urgi.infobiogen.fr///Projects/GPiDB/Interface/).  相似文献

Two deep-working soil tillage tools, one which inverts soil (plough) and one which does not (chisel), were used before sowing wheat after various crop successions combining eyespot host and non-host crops. Soil structure was nearly the same and crop residues were located in the different soil layers. Eyespot sporulation was estimated by visually assessing pot plants which had been on the trial plots for a fixed length of time. Field plants were also assessed for disease at several wheat growth stages. A kinetic equation expressing disease level as a function of degree-days was fitted to the disease levels observed on the field plants. This equation is based on eyespot epidemiology and depends on two parameters reflecting the importance of the primary and the secondary infection cycles respectively. Pot plant and early field plant disease levels and primary infection were closely correlated to the presence of crop residues in the top layer. The amount of residues depended on both crop succession and soil tillage. Where the previous crop was a host crop preceded by a non-host crop, soil inversion buried host residues, thus decreasing the primary infection risk. Where however the previous crop was a non-host crop preceded by a host crop, soil inversion carried the host residues back to soil surface, thus increasing the primary infection risk. Secondary infection was not correlated to either crop succession or soil tillage.  相似文献

The importance of carotenoid content in apricot (Prunus armeniaca L.) is recognized not only because of the color that they impart but also because of their protective activity against human diseases. Current methods to assess carotenoid content are time-consuming, expensive, and destructive. In this work, the application of rapid and nondestructive methods such as colorimeter measurements and infrared spectroscopy has been evaluated for carotenoid determination in apricot. Forty apricot genotypes covering a wide range of peel and flesh colors have been analyzed. Color measurements on the skin and flesh ( L*, a*, b*, hue, chroma, and a*/ b* ratio) as well as Fourier transform near-infrared spectroscopy (FT-NIR) on intact fruits and Fourier transform mid-infrared spectroscopy (FT-MIR) on ground flesh were correlated with the carotenoid content measured by high-performance liquid chromatography. A high variability in color values and carotenoid content was observed. Partial least squares regression analyses between beta-carotene content and provitamin A activity and color measurements showed a high fit in peel, flesh, and edible apricot portion (R(2) ranged from 0.81 to 0.91) and low prediction error. Regression equations were developed for predicting carotenoid content by using color values, which appeared as a simple, rapid, reliable, and nondestructive method. However, FT-NIR and FT-MIR models showed very low R(2) values and very high prediction errors for carotenoid content.  相似文献

Initiation and maintenance of mitosis require the activation of protein kinase cyclin B-Cdc2 and the inhibition of protein phosphatase 2A (PP2A), which, respectively, phosphorylate and dephosphorylate mitotic substrates. The protein kinase Greatwall (Gwl) is required to maintain mitosis through PP2A inhibition. We describe how Gwl activation results in PP2A inhibition. We identified cyclic adenosine monophosphate-regulated phosphoprotein 19 (Arpp19) and α-Endosulfine as two substrates of Gwl that, when phosphorylated by this kinase, associate with and inhibit PP2A, thus promoting mitotic entry. Conversely, in the absence of Gwl activity, Arpp19 and α-Endosulfine are dephosphorylated and lose their capacity to bind and inhibit PP2A. Although both proteins can inhibit PP2A, endogenous Arpp19, but not α-Endosulfine, is responsible for PP2A inhibition at mitotic entry in Xenopus egg extracts.  相似文献

Carbon dioxide, water vapour and energy fluxes were measured above and within a maritime pine forest during an atypical year with long-lasting reduced soil water availability. Energy balance closure was adequately good at both levels. As compared with what is usually observed at this site the ecosystem dissipated less energy via latent heat flux and more via sensible heat flux. The understorey canopy was responsible for a variable, significant component of the whole canopy fluxes of water vapour and carbon dioxide. The annual contribution of the understorey was 38% (154 mm) of the overall evaporation (399 mm) and 32% (89 mm) of the overall sensible heat flux (274 mm). The participation of the understorey reached 45% of the overall evaporation and 30% of the daytime overall assimilation during significant soil water deficit periods in summertime. Even during winter, understorey photosynthesis was consistent as it compensated soil and understorey respiration. The ecosystem behaved as a sink of carbon, with a negative annual carbon budget (−57 g C m−2). However, due to high soil water deficit, the annual ecosystem GPP was 40% less than usually observed at this site. This budget resulted from a sink of −131 g C m−2 for the overstorey and a source of +74 g C m−2 for the understorey. Moreover, on an annual basis the overstorey layer contributed to almost two-thirds of the ecosystem respiration. Finally, the effect of long-lasting soil water deficit on the maritime pine forest was found more important than the effect of the heat wave and drought of summer 2003.  相似文献

The ex ante assessment of innovative agro-ecological innovations is a key step in the development of more sustainable crop management systems. To this end, models are useful tools because they make it possible to rapidly assess numerous innovations in different contexts. Whereas many farm optimisation models focusing on the farmer’s strategic decision to adopt new crop management systems have been published, little attention has been given to the ex ante modelling of the dynamic operational impacts of innovation adoption at the farm level. BANAD, a mechanistic model for such applications, is proposed. It allows the ex ante assessment of innovative management systems including new agro-ecological techniques, while taking into account different farming contexts and policy and market conditions. It includes three components: (i) a crop management system model, (ii) a crop model (SIMBA) and (iii) a farming system model. Our results applied to the ex ante assessment of six innovative banana management systems for three contrasted farm types in Guadeloupe showed that the impacts of agro-ecological innovations, which include rotations, improved fallow, intercropping, pest-resistant cultivar, and an integrated organic system, can vary considerably according to (i) the farm type in which the innovation is integrated, (ii) the nature of the agro-ecological innovations, and (iii) the criteria considered and the temporal horizon of the assessment. Innovative intercropping systems that were effective at the field level in terms of the yield improvement and decreased pesticide use could be problematic at the farm level because they increased the workload and decreased income. The adoption of rotations or improved fallow seemed to be relevant for smallholders but could induce a critical period of 1.5-2.5 years during which income decreased drastically. Under certain conditions of markets and subsidies, very environmentally friendly innovations that are less productive can however be economically effective.  相似文献

Most of the monthly catches and CPUE of 13 studied commercial species in the coastal waters of the north-western Mediterranean were significantly positively correlated with run-off of local rivers (Rhône and Muga) and the wind mixing index during the spawning season, with time lags of less than a year (transfer function analyses). Rhône and Ebre interannual fluctuations in run-off were synchronous and were related to the North Atlantic Oscillation (NAO), river run-offs being negatively correlated to high NAO episodes. The fluctuations of river discharges and the wind mixing index were cyclic but not related. The results showed that enhanced hydroclimatic conditions in the NW Mediterranean were favourable for the productivity of the fish and invertebrate stocks, and suggest the presence of linkage between recruitment of Mediterranean species and local (river discharges, wind conditions) and global (NAO) environmental conditions.  相似文献
