Lupus-Type "Anticoagulant" in a Dog With Hemolysis and Thrombosis

A circulating anticoagulant was detected in a 2-year-old Chesapeake Bay Retriever with hemolytic anemia, nephrotic syndrome, thrombocytopenia, polyarthropathy, and pulmonary thromboembolism. A persistent prolongation of the activated partial thromboplastin time (aPTT) was detected, and it did not correct with repeated administration of fresh frozen plasma. The aPTT was still prolonged, with a 1:1 mixture of patient's plasma and normal dog plasma in vitro, suggesting the presence of a circulating inhibitor. Results of assays to characterize the inhibitor were compatible with those described for the lupus anticoagulant in human patients with systemic lupus erythematosus. Paradoxically, patients having the lupus anticoagulant are at increased risk for thrombosis. Pulmonary thromboembolism has been described as a frequent complication of immune-mediated hemolytic anemia in the dog, and the presence of a circulating anticoagulant should be considered as a potential mechanism.     

Compost Research Runs “Hot” and “Cold” at La Trobe University

? Careful monitoring of the composting process — physically, chemically and biologically — enables managers to determine key nutritional and ecological factors vital to production of a quality compost. At La Trobe University, trials compared composts which achieve high temperature (80° C) with those controlled at moderate thermophilic temperatures (45 — 55° C) to demonstrate whether the hot phase is essential or whether composts of equal nutritional status can be produced by processing at lower temperatures.     

Nitrogen requirement of wide-spaced row crops in the presence of starter fertilizer

Abstract. There is a need to develop sustainable nitrogen (N) management systems that minimize environmental losses by maximizing the use efficiency of applied fertilizers, particularly with wide-row annual crops that are often poor at utilizing N. A key approach is to match nitrogen supply with crop demand using improved methods of fertilizer application and timing. One technique is to target liquid 'starter' fertilizers close to the seed, or around the roots of transplants, and to omit or reduce conventional broadcast applications. This paper examines the effects of starter fertilizer combined with various rates of seedbed and/or top-dressed N on the growth and yield of bulb onion ( Allium cepa L.), crisp lettuce ( Lactuca sativa L.), forage maize ( Zea mays Bonaf.) and sugarbeet ( Beta vulgaris L.). Starter fertilizer improved early growth and, in combination with reduced rates of supplementary N, gave yields comparable with higher rates of base N with each crop, except sugarbeet. The use of top-dressed N was as effective as base N in supplementing starter fertilizer and had the benefit that it allowed a top-dressing requirement to be estimated accurately using a simple nitrogen balance equation. These results, taken with earlier work, show that starter fertilizers offer clear opportunities for reducing N inputs, while maintaining yield and quality of these crops.     

The use of soil data in natural heritage planning and management

Abstract. Native woodland restoration is a conservation priority within the UK and there is an increasing awareness that a long-term strategy is required to guide this process. A GIS-based modelling approach has been developed, which links site conditions as expressed in an integrated soil and land cover dataset with the site requirements for different woodland types. There are three important aspects to the model – the parameter weightings, the added value of the integrated dataset, and the woodland categories which are described and predicted. The initial quantitative and qualitative validation has been encouraging although more is required and planned. The results indicate that for broad strategic planning purposes, predictions of woodland potential are not improved significantly when climatic factors are incorporated. The concept that soil acts as an integrator of other environmental variables is discussed alongside the practical application of the model by a range of users.     

Toll-like receptor 3 (TLR3): a new marker of canine monocytes-derived dendritic cells (cMo-DC)

Toll-like receptors (TLRs) are a family of functionally important receptors for recognition of pathogen-associated molecular pattern (PAMP) since they trigger the pro-inflammatory response and upregulation of costimulatory molecules, linking the rapid innate response to adaptative immunity. In human leukocytes, TLR3 has been found to be specifically expressed in dendritic cells (DC). This study examined the expression of TLR3 in canine monocytes-derived DC (cMo-DC) and PBMC using three new anti-TLR3 mAbs (619F7, 722E2 and 713E4 clones). The non-adherent cMo-DC generated after culture in canine IL-4 plus canine GM-CSF were labelled with the three anti-TLR3 clones by flow cytometry, with a strong expression shown for 619F7 and 722E2 clones. By contrast, TLR3 expression was low to moderate in canine monocytes and lymphocytes. These results were confirmed by Western blot using 619F7 and 722E2 clones and several polypeptide bands were observed, suggesting a possible cleavage of TLR3 molecule or different glycosylation states. In addition, TLR3 was detectable in immunocytochemistry by using 722E2 clone. In conclusion, this first approach to study canine TLR3 protein expression shows that three anti-TLR3 clones detect canine TLR3 and can be used to better characterize canine DC and the immune system of dogs.
