The dynamics of blood neutrophil acyloxyacyl hydrolase (AOAH) activity, the appearance of endotoxin (lipopolysaccharide, LPS) in blood and the role of blood neutrophil AOAH in the severity of Escherichia coli and endotoxin mastitis were investigated in early postpartum dairy cows experimentally challenged with either endotoxin (n = 6) or E. coli (n = 6). The AOAH activity of blood neutrophils started to decrease significantly at post challenge hours (PCH) 6-24 and 12-24 in the endotoxin and E. coli-challenged groups, respectively; it returned to pre-challenged values at PCH 48 in both endotoxin- and E. coli-challenged groups. The cows were classified as moderate and severe responders according to milk production loss in the non-challenged quarters at PCH 48. There were no severe responders in the endotoxin-challenged group. In the E. coli-challenged group, only 1 severe responder was identified. The pre-challenge neutrophil AOAH activity of the severe responder was approximately 30% lower than that of moderate responders. No LPS was detected in the plasma of endotoxin-challenged cows; neither was it found in the plasma of moderate responders in the E. coli-challenged group at any PCH. However, at PCH 6, a remarkable amount of LPS was detected in the plasma of the severe responder from the E. coli-challenged group. Furthermore, neutrophil AOAH activity was increased by approximately 70% in the severe responder at PCH 6, but it increased by only approximately 15% in moderate responders. This was followed by a decreased neutrophil AOAH activity at PCH 12-24 and 24-72 in moderate and severe responders, respectively; the decreased AOAH activity at those PCH was more pronounced in the severe responder. The pronounced decreased neutrophil AOAH activity during mastitis often coincided with extreme leukopenia, neutropenia and a maximal number of immature neutrophils in the blood. Our results demonstrate that a decrease in neutrophil AOAH activity results in the appearance of LPS in the blood, and low blood neutrophil deacylation activity could be considered as a risk factor for severe clinical coliform mastitis.  相似文献

Intramammary infections of dairy cows with Gram-positive bacteria such as Staphylococcus aureus (major cause of mastitis) have received a lot of attention because of their major economic impact on the dairy farm through production losses induced by an increase in somatic cell count. Management strategies, including greater awareness for efficient milking and hygienic measures, have limited the spread of Gram-positive bacteria and resulted in a significant decrease of proportion of S. aureus isolates and subclinical mastitis worldwide. Other organisms such as coliform subspecies and Streptococcus uberis, both environmental bacteria that cause clinical mastitis, have received less attention. Escherichia coli causes inflammation of the mammary gland in dairy cows around parturition and during early lactation with striking local and sometimes severe systemic clinical symptoms. This disease affects many high producing cows in dairy herds and may cause several cases of death per year in the most severe cases. It is well known that bacterial, cow and environmental factors are interdependent and influence mastitis susceptibility. Many studies, executed during the last decade, indicate that the severity of E. coli mastitis is mainly determined by cow factors rather than by E. coli pathogenicity. During E. coli mastitis, the host defense status is a cardinal factor determining the outcome of the disease. Today, we know that the neutrophil is a key factor in the cows' defense against intramammary infection with E. coli. Effective elimination of the pathogen by neutrophils is important for the resolution of infection and the outcome of E. coli mastitis. This review is a compilation of some major findings over the last 15 years concerning mainly host factors that modulate and influence neutrophil function and the mammary inflammatory reaction. The individual chapters address: virulence factors of E. coli strains, how neutrophils kill E. coli, connection between endotoxins, tumor necrosis factor-alpha and nitric oxide, severity classification of E. coli mastitis, lifespan of neutrophils, host factors that influence severity, tissue damage and production loss.  相似文献

Zagros forests are mainly covered byQuercus brantii L. coppices and oak sprout clumps occupy the forest area like patches. We investigated post-fire herbaceous diversity in the first growing season after fire. For this purpose neighboring burned and unburned areas were selected with the same plant species and ecological conditions. The data were collected from areas subjected to different fire severities. Overall 6 treatments were considered with respect to fire severity and the mi-crosites of inside and outside of oak sprout clumps including: unburned inside and outside of sprout clumps (Ni and No), inside of sprout clumps that burned with high fire severity (H), inside of sprout clumps that burned with moderate fire severity (M), outside of sprout clumps that burned with low fire severity (OH and OM). Different herbaceous com-position was observed in the unburned inside and outside of oak sprout clumps. The species diversity and richness were increased in treatments burned with low and moderate fire severity. However, in treatment burned with high fire severity (H), herbaceous cover was reduced, even-ness was increased, and richness and diversity were not significantly changed. We concluded that besides the microsites conditions in forest, fire severity is an inseparable part of the ecological effect of fire on her-baceous composition.  相似文献

Microbial indices and their spatial patterns are strongly affected by environmental factors. Spatial variability of soil properties is one of the most important causes of variability in soil microbial indices. This research was conducted in the Caspian forest to assess spatial variabilities and frequency distributions of microbial properties.Ninety soil samples were taken using a grid sampling design 40 9 40 m. Microbial indices, organic carbon,nitrogen and pH were determined. Soil variable distributions showed that microbial indices had abnormal distributions. Logarithmic transformation produced normal distribution. Spatial continuity using geostatistical(variogram) was studied and maps obtained by point kriging.The variograms revealed the presence of spatial autocorrelation. The results indicate that spatial dependence of soil microbial indices was affected by non-intrinsic factors and forest management procedures. The maps show that soil microbial indices and soil properties have spatial variability. The spatial pattern of microbial indices was correlated to organic carbon and nitrogen.  相似文献

The deleterious effects of heat stress on animal health are being increasingly recognized. This study aimed to determine hair cortisol (HC) and serotonin levels in lactating Holstein cows under heat stress conditions with different coat and hair‐cut color. Forty‐five multiparous lactating Holstein cows (days in milk = 130 ± 47, body weight = 753 ± 85 kg) were divided to two main groups of over 80% black coat color (BC) and over 85% white coat color (WC) visually observed based on registry certificates and subdividing to black hair sample (BH) and white hair samples (WH) in 2 × 2 factorial arrangements. Hair samples were taken from the forehead of the individuals. Higher HC levels were observed in BC than WC cows (P < 0.05). No differences were found in HC levels between BH and WH groups (P > 0.05). Serotonin levels showed no difference between BC and WC (P > 0.05). Interaction between coat color and hair color was not significant (P > 0.05). The cortisol levels in hair are not affected by pigmentation. However, pigmentation within the coat alters cortisol levels. In conclusion, white coat color retains less cortisol than the black coat. Therefore, white coats are preferable for dairy cows under heat stress conditions.  相似文献

Wide and discriminate use of antibiotics has resulted in serious biological and ecological concerns, especially the emergence of antibiotic resistance. Probiotics, known as beneficial microbes, are being proposed as an effective and eco-friendly alternative to antibiotics. They were first applied in aquaculture species more than three decades ago, but considerable attention had been given only in the early 2000s. Probiotics are defined as live or dead, or even a component of the microorganisms that act under different modes of action in conferring beneficial effects to the host or to its environment. Several probiotics have been characterized and applied in fish and a number of them are of host origin. Unlike some disease control alternatives being adapted and proposed in aquaculture where actions are unilateral, the immense potential of probiotics lies on their multiple mechanisms in conferring benefits to the host fish and the rearing environment. The staggering number of probiotics papers in aquaculture highlights the multitude of advantages from these microorganisms and conspicuously position them in the dynamic search for health-promoting alternatives for cultured fish. This paper provides an update on the use of probiotics in finfish aquaculture, particularly focusing on their modes of action. It explores the contemporary understanding of their spatial and nutritional competitiveness, inhibitory metabolites, environmental modification capability, immunomodulatory potential and stress-alleviating mechanism. This timely update affirms the importance of probiotics in fostering sustainable approaches in aquaculture and provides avenues in furthering its research and development.  相似文献

A 6-mo-old, male western lowland gorilla (Gorilla gorilla gorilla) was evaluated because of tetany of both hands. The gorilla had alternating periods of constipation, diarrhea, and bloating since birth. A diagnosis of idiopathic hypocalcemia was based on severe hypocalcemia, a normal vitamin D level, response to oral calcium and vitamin D therapy, and eventual resolution. Idiopathic hypocalcemia, an uncommon disease in neonatal humans, should be considered in young gorillas with persistent gastrointestinal problems or acute tetany.  相似文献

Tree growth equations are an important and common tool used to effectively assess the yield and determine management practices in forest plantations. Increasingly, they are being developed for urban forests, providing tools to assist urban forest managers with species selection, placement, and estimation of management costs and ecosystem services. This study describes the development of allometric equations for Fraxinus americana and F. pennsylvanica growing in Oakville, Canada. With data collected from 103 ash trees, five allometric models were tested to develop equations estimating diameter-at-breast-height (dbh), tree height, crown width and crown height, using age and dbh as explanatory variables. Mean annual growth rates are presented to demonstrate species growth performance and were not significantly different over the first 40 years of growth for the two species. Of all the tested random coefficient models for both species, the cubic with weight 1/x provided the best fit for estimating dbh from age. The best models for other parameters were the loglog for crown height from dbh, the quadratic for crown diameter from dbh, and the linear for tree height from dbh for F. americana. Model types showed more consistency for F. pennsylvanica with linear providing the best fit for crown diameter, crown height and tree height from dbh. The number of model types suggests the difficulty of fitting any single model to the vast array of conditions affecting plant growth in urban areas where management practices and environment can significantly influence tree size and growth. These models may be used to estimate the growth of ash tree populations in Oakville and communities with similar climate, soil, planting, and management environments.  相似文献

Water absorption and water vapor transition are significant mechanisms in many industrial processes. Nanocomposite polymers are appropriate materials for water transition process due to their specific properties. The present work was aimed to enhance the water absorption and water vapor transition by high loading incorporation of silica nanoparticles (NPs) into polyacrylonitrile (PAN) and Pebax 1657 polymers. PAN was used to prepare both the nanocomposite films (NCFs) and support layers of nanocomposite membranes (NCMs). Pebax 1657 was used to fabricate the selective layers of NCMs. Incorporation of NPs into the NCFs were carried out using ex-situ and in-situ (sol-gel) methods. NCFs were fabricated by electrospinning and casting methods. The fabricated NCFs of fibers and polymers were examined through water absorption tests. For fabrication of NCMs, the electrospinning and dip-coating methodologies and the incorporation of NPs into the selective layers by in-situ method were used. Besides, the water vapor permeation tests have been designed and constructed to examine the NCMs for dehydration of methane gas. In order to provide efficient NPs dispersion into NCFs, the surface functionalization of silica NPs with ethylene glycol (EG) was also considered. The functionalized silica NPs were used in the fabrication of NCFs and into the selective layer of NCMs. It is found that in applying the ex-situ method, there is a lot of NPs agglomeration, while the dispersion quality of NPs is higher with the sol-gel method. In addition, the dispersion is improved greatly with functionalization of silica NPs. The water uptake ratio of electrospun mats is about 400 % higher than the cast layers for the same loadings of NPs. The water uptake ratio of the polymeric layers containing EG is about 50 % higher than the samples without EG. The permeance of water vapor for the NCM (15 wt.% of SiO2/Pebax 1657) fabricated by sol-gel method was enhanced to 54.4 % higher than the membranes without NPs. Functionalization of silica NPs also enhanced the water vapor permeation process. For instance, the water vapor of the NCM containing 30 wt.% of EG/Pebax1657 was increased by 68.7 %.  相似文献

Biscogniauxia mediterranea (De Not.) Kuntze, is a widespread fungus, causes outbreaks of Charcoal disease on cork oak and dieback thousands oak trees in recent years in Iran. Yet no efficient and effective management methods have been found to control the disease. The present study aimed to provide a suitable method for charcoal disease management through silviculture operations, systemic pesticides and biological compounds. The results showed that, simultaneous implementation of silvicultural practices (scarification+sanitation), use of systemic fungicides and Trichoderma compounds, reduce the number and depth of cankers, increase callus thickness, and ultimately improve health of the diseased trees. But disease severity and fungi activity were increased when scarification operation implements alone. The greatest effectiveness of the treatments and health condition improvement of the diseased trees was observed 12 months after applying the treatments. Then, preventive and therapeutic effectiveness of the treatments reduced over time. Sanitation and scarification practices can increase soil permeability and also destroy the larvae, pupa or adult insects and oak borer beetles that play an important role in disease outbreaks. Therefore, simultaneous implementation of sanitation and scarification practices with application of biological and systemic compounds can reduce primary source of pathogen inoculum and help trees to recover their health.  相似文献
