This study addresses development and validation of a composite multifactorial pain scale (CPS) in an experimental equine model of acute orthopaedic pain. Eighteen horses were allocated to control (sedation with/without epidural analgesia - mixture of morphine, ropivacaine, detomidine and ketamine) and experimental groups: amphotericin-B injection in the tarsocrural joint induced pain and analgesia was either i.v. phenylbutazone administered post-induction of synovitis, or pre-emptive epidural mixture, or a pre-emptive combination of the 2. Eighty-four olive accessions obtained from the National Conservatory of Boughrara-Sfax (Tunisia), previously evaluated for morphological traits, were analysed with 47 random amplified polymorphic DNA (RAPD) markers. They were compared with other olive genotypes originated from Eastern or Western Mediterranean. The highest and lowest similarities between genotypes, estimated by simple matching algorithm, were 0.98 and 0.40, respectively. A dendrogram based on Ward's method and a factorial correspondence analysis (FCA) showed that most of Tunisian accessions are closely related to olive genotypes originating from the Eastern Mediterranean and some are clustering with genotypes originated from the Western Mediterranean. These findings suggested multiple and complex origin of Tunisian olive. A comparative study between a previous morphological analysis and current RAPD assay was carried out and discussed.  相似文献

Four Pseudomonas strains were evaluated for their intrinsic properties conferring their ability to protect long English cucumber against Pythium aphanidermatum in hydroponic culture. Two of the strains, BTP1 and its siderophore-negative mutant M3, increased plant yield as compared with the non-inoculated control plants. Strain BTP7 was intermediate in its biocontrol activity while strain ATCC 17400 failed to reduce disease development. The role of pyoverdines could not be confirmed since treatment with either BTP1 or its siderophore-negative mutant M3 provided similar suppression of Pythium disease. In addition, no siderophores were detected in the nutrient solution. BTP1 did not inhibit pathogen growth in vitro on several media, suggesting that antibiosis was not a mechanism of suppression. Quantification of root bacterial populations did not indicate differences among the strains. On the other hand, roots treated with either BTP1 or its sid mutant M3 contained more antifungal phenolics than roots from any other treatments including controls. These results suggest that antifungal compounds induced by inoculation of cucumber roots with the fluorescent Pseudomonas strains BTP1 and M3 participate actively in the protection of cucumber plants against P. aphanidermatum  相似文献

The aim of the study was to elucidate the spatial distribution of soil microorganisms and enyzme activities in a long‐term wastewater treated soil. Soil was sampled from a plough layer of the Ah horizon of a sandy Haplic Luvisol which was either (1) irrigated with municipal wastewater for almost 100 years, or (2) no more irrigated since 20 years, or (3) never received wastewater. The samples were fractionated by wet sieving to obtain seven size fractions of organic and mineral soil particles, and a separate silt+clay fraction. The individual soil samples contained between 1.2% (never irrigated) and 4.1% (long‐term irrigated) organic particles by weight, but these particles harboured up to 47.8% of the total soil carbon and 41.7% of nitrogen, and thus represented an important storage of energy and nutrient for microorganisms. In total, however, the highest C and N amounts were accumulated in the silt+clay fraction, whereas coarser mineral particles which dominanted by weight in the Haplic Luvisol were low in C and N. The highest numbers of bacteria, actinomycetes and fungi per gram of the individual soil fractions were found in organic particles of the long‐term irrigated soil. Less nutrient‐dependent oligotrophic bacteria were for the most part associated with the silt+clay fraction, irrespective of the soil treatment with wastewater. Similar to microbial counts, also the ATP content, as a measure of active microbial biomass, and the activities of β‐glucosidase, β‐acetylglucosaminidase, and proteinase were higher in the long‐term irrigated soil than in that which was never irrigated. In most cases slightly enhanced values of microbiological and biochemical parameters were still detectable 20 years after the wastewater irrigation was terminated. The values of the individual parameters decreased in all soil samples under testing in general gradually with decrease in size of the organic soil particles. In conclusion, the coarse soil organic particles > 5 mm and the silt+clay fraction < 0.05 mm represent the sites with the highest microbial inhabitance, ATP contents and enzyme activities in the Ah horizon of an Haplic Luvisol. Long‐term wastewater irrigation resulted in an increase of microbial counts, total biomass and soil enzyme activities.  相似文献

Microbial degradation, organic synthesis and ecotoxicology were used to investigate the fate of diuron after spreading on soils. Quantitative biodegradation assays were performed with fungal strains, showing that diuron was degraded but not entirely mineralized. The modifications observed consisted in demethylation of the terminal nitrogen atom. The identified metabolites were synthesized in sufficient amounts to confirm their structures and determine their non‐target toxicity using four biotests. The two metabolites exhibited higher effects than parent diuron. This limited biodegradability and potential aquatic toxicity suggest that diuron is of higher environmental concern than previously recognized. © 2000 Society of Chemical Industry  相似文献

Resveratrol and its oligomers: ε-viniferine, gnetin C, Pallidol and gnetin E, as well as three new dehydrodimers, cyphostemmines A–C, have been isolated from the roots of Cyphostemma crotalarioides (Ampelidaceae). Such compounds have not been reported previously in the family Ampelidaceae. Cis ε-viniferin has also been characterized as a minor component of the extract; it may have undergone partial transformation in solution into trans ε-viniferin. ©1999 Society of Chemical Industry  相似文献

A methanol extract of roots of Lasiosiphon kraussianus (Meisn) (Thymelaeaceae) showed potent insecticidal activity against Aphis gossypii (Glov) and Drosophila melanogaster (Meig). Bioassay-driven fractionation of this extract led to the isolation and characterisation of two known daphnane diterpenoids: Excoecaria toxin (1) and wikstrotoxin D (2) . The two natural products were inferior to methomyl in activity against A gossypii and Myzus persicae (Sulz) in contact assays but were superior in ingestion assays against D melanogaster. This is the first report on insecticidal activities of compounds 1 and 2 . AChE was insensitive to the two natural products. © 1999 Society of Chemical Industry  相似文献

As conventional or chemical pesticides have negative impact on environment and health of both farmer and consumers, it becomes relevant to develop alternative solutions to limit their use. In this context, innovative strategies to accelerate the development of biocontrol agents are welcome. For a decade of years, it has been demonstrated that lipopeptides are very efficient weapons against fungi responsible for crop diseases. Lipopeptides are secondary metabolites, produced by many microorganisms including beneficial rhizobacteria. The lipopeptide biosynthetic pathways include nonribosomal peptide synthetases. These modular enzymatic complexes work as assembly lines to build the peptides step by step, leading to the production of original peptide compounds with specific features as the presence of non proteinogenic monomers and cyclic and branched structures. In this paper, Florine and Norine bioinformatics tools, especially dedicated to non-ribosomal synthetases and their products are presented. Their use is mainly focused on the discovery of lipopeptides produced by Bacillus or Pseudomonas because they seem to represent a versatile reservoir of active secondary metabolites with promising activities for applications in phytosanitary area.  相似文献
