Interest in the consumption of fresh fruits and vegetables is, to a large extent, due to its content of bioactive nutrients and their importance as dietary antioxidants. Among all of the selected fruits and vegetables, strawberries and black grapes have relatively high antioxidant capacities associated with high contents of total phenolic compounds, ascorbic acid, and flavonols. More interesting, the results of this study indicated that in most fruits and vegetables storage did not affect negatively the antioxidant capacity. Better, in some cases, an increase of the antioxidant capacity was observed in the days following their purchase, accompanied by an increase in phenolic compounds. In general, fruits and vegetables visually spoil before any significant antioxidant capacity loss occurs except in banana and broccoli. When ascorbic acid or flavonoids (aglycons of flavonols and anthocyanins) were concerned, the conclusions were similar. Their content was generally stable during storage. 相似文献

The anticarcinogenic effect of garlic has been demonstrated in both epidemiologic and experimental studies. In this study, possible mechanisms involved in the anticarcinogenic effect of garlic consumption were assessed by determining its capacity to alter drug metabolizing enzymes, in relation with its alliin content. Rats were fed a diet for 2 weeks containing 5% garlic powders produced from bulbs grown on soils with different levels of sulfate fertilization and therefore containing differing amounts of alliin. Activities of several hepatic enzymes, which are important in carcinogen metabolism such cytochromes P450 (CYP) and phase II enzymes, were determined. Garlic consumption slightly increased ethoxyresorufin O-deethylase and CYP 1A2 levels. In contrast, garlic consumption decreased CYP 2E1 activity and the level of the corresponding isoform. UDP glucuronosyl transferase and glutathion S-transferase activities were increased by garlic powders. The alliin content of the garlic powders was positively correlated with UGT activity although not with other activities. Effects produced by garlic consumption were qualitatively similar to that of diallyl disulfide, a sulfur compound that has been extensively studied. The aim of the study was to elucidate the spatial distribution of soil microorganisms and enyzme activities in a long‐term wastewater treated soil. Soil was sampled from a plough layer of the Ah horizon of a sandy Haplic Luvisol which was either (1) irrigated with municipal wastewater for almost 100 years, or (2) no more irrigated since 20 years, or (3) never received wastewater. The samples were fractionated by wet sieving to obtain seven size fractions of organic and mineral soil particles, and a separate silt+clay fraction. The individual soil samples contained between 1.2% (never irrigated) and 4.1% (long‐term irrigated) organic particles by weight, but these particles harboured up to 47.8% of the total soil carbon and 41.7% of nitrogen, and thus represented an important storage of energy and nutrient for microorganisms. In total, however, the highest C and N amounts were accumulated in the silt+clay fraction, whereas coarser mineral particles which dominanted by weight in the Haplic Luvisol were low in C and N. The highest numbers of bacteria, actinomycetes and fungi per gram of the individual soil fractions were found in organic particles of the long‐term irrigated soil. Less nutrient‐dependent oligotrophic bacteria were for the most part associated with the silt+clay fraction, irrespective of the soil treatment with wastewater. Similar to microbial counts, also the ATP content, as a measure of active microbial biomass, and the activities of β‐glucosidase, β‐acetylglucosaminidase, and proteinase were higher in the long‐term irrigated soil than in that which was never irrigated. In most cases slightly enhanced values of microbiological and biochemical parameters were still detectable 20 years after the wastewater irrigation was terminated. The values of the individual parameters decreased in all soil samples under testing in general gradually with decrease in size of the organic soil particles. In conclusion, the coarse soil organic particles > 5 mm and the silt+clay fraction < 0.05 mm represent the sites with the highest microbial inhabitance, ATP contents and enzyme activities in the Ah horizon of an Haplic Luvisol. Protein of endosperm of maize grains originating from three wild-type inbreds and their opaque-2 versions were solubilized in diverse extracts (E) by the sequential use of 0.5 M NaCl, water (E(1,2)), alcohol plus a reducing agent (E(3)), and salt plus a reducing agent (E(4)). Zeins were isolated in extracts E(3) and E(4) obtained by using 55% (w/w) isopropyl alcohol (i-PrOH) + 0.2% dithiothreitol (DTT) followed by 0.5 M NaCl + 0.2% DTT buffered at pH 10 or 60% tert-butyl alcohol (t-BuOH) + 0.2% DTT followed by 0.5% sodium acetate + 0.2% DTT in 30% t-BuOH. For a given genotype the percentage of extracted zeins was independent of the nature of the alcohol. The latter had a slight effect on the respective magnitude of E(3) and E(4): E(3) increased at the expense of E(4) when t-BuOH was substituted to i-PrOH for their isolation. The percentage of the total endosperm nitrogen present in E(3) + E(4) was identical to that of fractions F(II) + F(III) + F(IV) isolated according to the classical Landry-Moureaux extraction procedure. SDS-PAGE analysis revealed the presence of all types of zeins (alpha, beta, gamma, and delta) in E(3) and F(III), residual zeins in E(4) isolated with t-BuOH, and streaking only in E(4) and F(IV) isolated with NaCl at pH 10. The data together with those of the literature were discussed with regard to the influence of procedure on the yield of zeins using alcoholic extraction. 相似文献

Casein proteolysis can occur in milk during storage leading to its gelation. The two main proteolytic systems suspected to be involved are the plasmin and the proteases produced by psychrotrophic bacteria. The latter have been shown to cleave kappa-casein at the Phe105-Met106 bond. Although several techniques allow the determination of plasmin in milk, few rapid and easy-to-perform analytical techniques are available to check for bacterial proteolytic activity. This study presents the development of an inhibition ELISA allowing for the quantification of the kappa-casein intact at the Phe105-Met106 bond. It uses a monoclonal antibody specifically directed against this peptide bond that binds to the protein as long as the molecule's cleavage site is intact but not when it is cleaved. This simple technique allows for the rapid analysis of more than 20 samples within 3 h. Applied to commercial milks, this assay allowed for the detection of unstable milk. 相似文献

Hydroponics culture generates large amounts of wastewater that are highly concentrated in nitrate and phosphorus but contains almost no organic carbon. Constructed wetlands (CWs) have been proposed to treat this type of effluent, but little is known about the performance of these systems in treating hydroponic wastewater. In addition, obtaining satisfactory winter performances from CWs operated in cold climates remains a challenge, as biological pathways are often slowed down or inhibited. The main objective of this study was to assess the effect of plant species (Typha sp., Phragmites australis, and Phalaris arundinacea) and the addition of organic carbon on nutrient removal in winter. The experimental setup consisted of 16 subsurface flow CW mesocosms (1 m2, HRT of 3 days) fed with 30 L?d1 of synthetic hydroponics wastewater, with half of the mesocosms fed with an additional source of organic carbon (sucrose). Carbon addition had a significant impact on nitrate and phosphate removal, with removal means of 4.9 g m-2?d-1 of NO3-N and 0.5 g m-2 d-1 of PO4-P. Planted mesocosms were generally more efficient than unplanted controls. Furthermore, we found significant differences among plant treatments for NO3-N (highest removal with P. arundinacea) and COD (highest removal with P. australis/Typha sp.). Overall, planted wetlands with added organic carbon represent the best combination to treat hydroponics wastewater during the winter. 相似文献

Worldwide genetic diversity in 200 individuals comprising 41 castor bean accessions was assessed using amplified fragment
polymorphisms (AFLPs) and simple sequence repeats (SSRs). We found that, despite surveying five continents and 35 countries,
genetic diversity in castor bean germplasm is relatively low (overall H
e = 0.126 for AFLPs and 0.188 for SSRs) compared to estimates of genetic diversity in other plant species. Our data also show
no geographic structuring of genotypes across continents or countries within continents. An assessment of the congruence between
AFLP and SSRs indicates a low correlation (R
2 = 0.19) between the two data sets, but each marker class nonetheless shows similar patterns of low-genetic diversity and
a lack of geographic structure. Our data do suggest that SSRs yield a higher percentage of polymorphic loci, higher heterozyosity
and a greater range of genetic distances, and are therefore more informative than are AFLPs on a locus-by-locus basis. Based
on comparisons with numerous other plant species, we suggest that the lower genetic variation in this worldwide collection
may be due to one or more factors including: sampling strategies that have not captured the full extent of genetic variation
in the species; artifactual variation due to long-term germplasm storage and seed regeneration; or intense selection followed
by domestic cultivation of a limited number of castor bean genotypes, which are widely propagated for their horticultural

The assessment of withdrawal periods for milk is affected by the occurrence of data below the lower analytical quantification limit (BLQ data) and the resulting uncertainty. The current regulatory approach for dealing with BLQ residues is simple and easy: BLQ data (and missing data) are arbitrarily reassigned a value of one‐half the LOQ before any calculation on the data with one of the three currently applicable methods. Here, we reconsider the determination of the withdrawal period of milk with data below the limit of quantification. Theoretical background on analytical limits and pharmacometric considerations will be established. Then, we analyze the uncertainty problems caused by the current approach and propose a calculation solution (maximum‐likelihood estimation handling left‐censored data) included in nonlinear mixed‐effects modeling.
