A field strain of Cooperia oncophora resistant to oxfendazole was isolated from a commercial cattle rearing property in Waikato, New Zealand. Resistance to oxfendazole was assessed by means of a faecal egg count depression test and an in vitro egg hatch test. This is the first documented case of anthelmintic resistance in Cooperia spp. and the first report of anthelmintic resistance in cattle in New Zealand. 相似文献

Although burbot ( Lota lota Gadidae) are widespread and abundant throughout much of their natural range, there are many populations that have been extirpated, endangered or are in serious decline. Due in part to the species' lack of popularity as a game and commercial fish, few regions consider burbot in management plans. We review the worldwide population status of burbot and synthesize reasons why some burbot populations are endangered or declining, some burbot populations have recovered and some burbot populations do not recover despite management measures. Burbot have been extirpated in much of Western Europe and the United Kingdom and are threatened or endangered in much of North America and Eurasia. Pollution and habitat change, particularly the effects of dams, appear to be the main causes for declines in riverine burbot populations. Pollution and the adverse effects of invasive species appear to be the main reasons for declines in lacustrine populations. Warmer water temperatures, due either to discharge from dams or climate change, have been noted in declining burbot populations at the southern extent of their range. Currently, fishing pressure does not appear to be limiting burbot populations world-wide. We suggest mitigation measures for burbot population recovery, particularly those impacted by dams and invasive species. 相似文献

Immunotherapy to prevent recurrence of clinical signs of atopic dermatitis (AD) is based on intradermal or serological tests that assist in identifying allergen-specific immunoglobulin E hypersensitivities. Unfortunately, the results of such tests can be negatively influenced by several factors, which include the age of the patients, the season of testing and the administration of anti-allergic drugs. Screening to predict when these expensive tests will be useful would benefit owners of dogs with AD. The objectives of this study were to determine whether a point-of-care allergen-specific immunodot assay (Allercept E-Screen, Heska Corp., Ft Collins, CO, USA) could predict results of either intradermal or Allercept full panel serological tests in atopic dogs. Thirty dogs living in the south-eastern USA were diagnosed with AD in accordance with current standards. Allergen-specific intradermal, serological and E-Screen tests were performed in all subjects. For flea, house dust mite and pollen allergens altogether, results of the E-Screen assay agreed with those of intradermal and serological tests in 26/30 dogs (87%) and 25/30 dogs (83%), respectively. In this group of dogs, the probabilities of obtaining intradermal or serological tests positive for these allergens were 70 and 67%, respectively. If either skin or serum tests were performed only in dogs with positive E-Screen tests, the probability of obtaining positive results would be increased from 70 to 95% and from 67 to 90%, respectively. In this population of dogs with AD, results of the E-Screen point-of-care immunodot assay was found to often agree with those of allergen-specific intradermal or Allercept tests for selected allergen groups. 相似文献

OBJECTIVE: To evaluate a group of vaccine site-associated sarcomas (VSS) for the presence of feline foamy virus (FeFV) DNA, using polymerase chain reaction (PCR) methods. SAMPLE POPULATION: 50 formalin-fixed paraffin-embedded (FFPE) tissue blocks from VSS of cats. PROCEDURE: DNA was extracted from FFPE sections of each tumor, and regions of the gag and pol genes of FeFV were amplified by use of PCR methods, using 1 primer set for each region. Sensitivity of the method was compared between fresh and FFPE cells, using mouse kidney tissue that was injected with FeFV-infected cultured cells and using agarose-cell pellets. Results-Feline foamy virus DNA was not detected in VSS tissues. Sensitivity of the method was 10 times greater in fresh versus FFPE mouse tissues. Sensitivity of the method in fresh FeFV-infected cultured cells versus FFPE agarose-cell pellets was equal when fixation was 24 or 48 hours and 10 times greater when fixation was 72 hours or 1 week. CONCLUSIONS AND CLINICAL RELEVANCE: A PCR-based method can be successfully applied to FFPE tissues for FeFV DNA detection. Bacterial canker disease is a major limiting factor in the growing of cherry and other Prunus species worldwide. At least five distinct clades within the bacterial species complex Pseudomonas syringae are known to be causal agents of the disease. The different pathogens commonly coexist in the field. Reducing canker is a challenging prospect as the efficacy of chemical controls and host resistance may vary against each of the diverse clades involved. Genomic analysis has revealed that the pathogens use a variable repertoire of virulence factors to cause the disease. Significantly, strains of P. syringae pv. syringae possess more genes for toxin biosynthesis and fewer encoding type III effector proteins. There is also a shared pool of key effector genes present on mobile elements such as plasmids and prophages that may have roles in virulence. By contrast, there is evidence that absence or truncation of certain effector genes, such as hopAB, is characteristic of cherry pathogens. Here we highlight how recent research, underpinned by the earlier epidemiological studies, is allowing significant progress in our understanding of the canker pathogens. This fundamental knowledge, combined with emerging insights into host genetics, provides the groundwork for development of precise control measures and informed approaches to breed for disease resistance. 相似文献

Plants can play an important role in wastewater treatment and water reuse in terrestrial and space systems. Chive growth in biologically treated graywater, simulating the anticipated early planetary base graywater, was evaluated in this study for NASA. Phytotoxicity due to physiochemical parameters such as ammonium-nitrogen (NH4 +-N), nitrite-nitrogen (NO2 ?-N), pH, and sodium (Na+) was assessed using a series of hydroponic experiments in an environmentally controlled growth chamber. Nitrification in wastewater was observed in all graywater treatments, which converted NO2 ?-N (a toxic form of nitrogen) and NH4 +-N (toxic at high concentrations) to nitrate-nitrogen (NO3 ?-N) (preferred N form for plant uptake). Irrespective of the increase in the NO3 –-N concentration due to nitrification, chives in the wastewater treatments typically had poor or no growth. The high levels of Na+ present in the graywater treatments affected potassium uptake and may have affected other nutrient uptake. The impact of nitrification on wastewater pH and NO2 ?-N toxicity is believed to be the critical factor affecting chive growth and may hinder the use high nitrogen waste streams for plant growth unless NO2 ?-N concentrations are controlled during biological treatment of graywater. 相似文献

ABSTRACT Thirty-eight bacterial strains isolated from hazelnut (Corylus avellana) cv. Tonda Gentile delle Langhe showing a twig dieback in Piedmont and Sardinia, Italy, were studied by a polyphasic approach. All strains were assessed by fatty acids analysis and repetitive sequence-based polymerase chain reaction (PCR) fingerprinting using BOX and ERIC primer sets. Representative strains also were assessed by sequencing the 16S rDNA and hrpL genes, determining the presence of the syrB gene, testing their biochemical and nutritional characteristics, and determining their pathogenicity to hazelnut and other plants species or plant organs. Moreover, they were compared with reference strains of other phytopathogenic pseudomonads. The strains from hazelnut belong to Pseudomonas syringae (sensu latu), LOPAT group Ia. Both fatty acids and repetitive-sequence-based PCR clearly discriminate such strains from other Pseudomonas spp., including P. avellanae and other P. syringae pathovars as well as P. syringae pv. syringae strains from hazelnut. Also, the sequencing of 16S rDNA and hrpL genes differentiated them from P. avellanae and from P. syringae pv. syringae. They did not possess the syrB gene. Some nutritional tests also differentiated them from related P. syringae pathovars. Upon artificial inoculation, these strains incited severe twig diebacks only on hazelnut. Our results justify the creation of a new pathovar because the strains from hazelnut constitute a homogeneous group and a discrete phenon. The name of P. syringae pv. coryli is proposed and criteria for routine identification are presented. 相似文献

Temporal depositional rates are important in order to understand the production and occurrence of perchlorate (ClO4−) as limited information exists regarding the impact of anthropogenic production or atmospheric pollution on ClO4− deposition. Perchlorate concentrations in discrete ice core samples from the Eclipse Icefield (Yukon Territory, Canada) and
Upper Fremont Glacier (Wyoming, USA) were analyzed using ion chromatography tandem mass spectrometry to evaluate temporal
changes in the deposition of ClO4
− in North America. The ice core samples cover a time period from 1726 to 1993 and 1970 to 2002 for the Upper Fremont Glacier
(UFG) and Eclipse ice cores, respectively. The average ClO4
− concentration in the Eclipse ice core for the time period from 1970 to 1973 was 0.6 ± 0.3 ng L−1, with higher values of 2.3 ± 1.7 and 2.2 ± 2.0 ng L−1 for the periods 1982–1986 and 1999–2002, respectively. All pre-1980 ice core samples from the UFG had ClO4
− concentrations <0.2 ng L−1, and the post-1980 samples ranged from <0.2 ng L−1 to a maximum of 2.6 ng L−1 for the year 1992. A significant positive correlation (R = 0.75, N = 15, p < 0.001) of ClO4− with SO42− was found for the annual UFG ice core layers and of ClO4
− with SO42− and NO3− in sub-annual Eclipse ice samples (R > 0.3, N = 121, p < 0.002). The estimated yearly ClO4− depositional flux for the Eclipse ice core ranged from 0.6 (1970) to 4.7 μg m−2 year−1 (1982) and the UFG from <0.1 (pre-1980) to 1.4 μg m−2 year−1 (1992). There was no consistent seasonal variation in the ClO4− depositional flux for the Eclipse ice core, in contrast to a previous study on the Arctic region. The presence of ClO4− in these ice cores might correspond to an intermittent source such as volcanic eruptions and/or any anthropogenic forcing
