Energy is an essential nutrient for all horses, and it is especially important in performance horses, pregnant and lactating mares, and young growing horses. A negative energy balance in horses such as these may result in unsatisfactory performance, decreased fertility, or slow growth. Therefore, ensuring adequate energy intake is an important aspect of the nutritional management of the equine. This study was undertaken to investigate the effects of feeding large, carbohydrate-rich, concentrate meals on the satiety-inducing hormone, leptin. Three groups of yearling horses were rotated through 3 feeding schedules in a replicated 3x3 Latin square design. Horses were fed 2, 3, or 4 times per day (2x, 3x, and 4xfeeding schedules, respectively), each for a period of 11 d, with the total amount of daily feed held constant. Horses were weighed and BCS was determined on the first day of each period. Blood samples were collected before the morning meal on d 1, 4, and 7 of each period. Additionally, blood was sampled for the last 24 h of the 2xand 4xdietary periods. Neither weight nor BCS changed during the study (P = 0.99 and P = 0.28, respectively). Both mean and peak plasma glucose were greatest in 2xhorses (P < 0.05), as were mean areas under the curve. Serum leptin concentration increased in 2xhorses (P < 0.05), but not in horses fed 3 or 4 times daily. Leptin was elevated in horses with greater BCS (P < 0.05) and increased steadily throughout the study (P < 0.05). Data from the 24-h collection indicated that 2xhorses had fluctuations in leptin production throughout the day (P < 0.05), whereas horses fed 4 times daily did not. Overall, this study indicates that feeding horses 2 large concentrate meals daily can increase mean serum leptin concentrations and may cause fluctuations in leptin production over a 24-h period. Bovine mastitis caused by strains of S. aureus is the most economically important disease affecting the dairy industry worldwide. Commercially available vaccines show various degrees of success and work in research laboratories with experimental vaccines suggests that in part, the failure of these vaccines lies in the limited antigenic repertoire contained in the vaccine formulations. Since it seems impractical to produce a vaccine containing antigens from all major S. aureus mastitis isolates, we took the approach of using two surface antigens GapB and GapC that appear to be conserved and constructed a GapC/B chimera as the basis for a vaccine. The humoral and cellular immune responses to GapC/B were compared to the responses to the individual proteins, alone or in combination. The GapC/B protein elicited strong humoral and cellular responses in mice as judged by the levels of total IgG, IgG1, IgG2a, and number of IL-4- and IFN-gamma-secreting cells. These results suggest that this chimeric protein could be an attractive target for further vaccine efficacy studies. 相似文献

Determining suitable locations for seed transfer under climate change: a global quantitative method 总被引:1,自引:1,他引:0
Changing climate conditions will complicate efforts to match seed sources with the environments to which they are best adapted. Tree species distributions may have to shift to match new environmental conditions, potentially requiring the establishment of some species entirely outside of their current distributions to thrive. Even within the portions of tree species ranges that remain generally suitable for the species, local populations may not be well-adapted to altered local conditions. To assist efforts to restore forests and to maximize forest productivity in the face of climate change, we developed a set of 30,000 quantitatively defined seed transfer “ecoregions” across the globe. Reflecting current and future conditions, these were created by combining global maps of potentially important environmental characteristics using a large-scale statistical clustering technique. This approach assigns every 4?km2 terrestrial raster cell into an ecoregion using non-hierarchical clustering of the cells in multivariate space based on 16 environmental variables. Two cells anywhere on the map with similar combinations of environmental characteristics are located near each other in this data space; cells are then classified into relatively homogeneous ecoregion clusters. Using two global circulation models and two emissions scenarios, we next mapped the predicted environmentally equivalent future locations of each ecoregion in 2050 and 2100. We further depicted areas of decreasing environmental similarity to given ecoregions, both in current time and under climate change. This study was conducted to study absorption of glucosamine (GlucN) and chondroitin sulfate (CS) in horses immediately after feeding. Six mature mares were used in a replicated 3 × 3 Latin-square-designed experiment. The experiment consisted of three 15-day periods, which included 10 days of diet adaptation followed by a 5-day sampling period. Blood was drawn after feeding on one day during each sampling period. Horses were fed a control diet (40% hay, 60% concentrate) balanced to meet NRC requirements for maintenance of mature horses (NRC, Nutrient requirements of horses, 1989). In one experimental diet, 2.0 g CS and 5.5 g GlucN were added to the basal ration at each feeding. In the other experimental diet, 3.5 g CS and 8.5 g GlucN were added to the basal ration at each feeding. After collections, blood was centrifuged and plasma was harvested and stored until analyzed for the presence of each compound. Analyses for plasma GlucN were performed using high-performance liquid chromatography. CS in the plasma was analyzed using a color reagent, dimethylmethylene blue, followed by ultraviolet spectrophotometry. There were no significant differences (P < .05) in the concentration of either CS or GlucN in plasma, when comparing the three different diets. This leads to a conclusion that these compounds were not absorbed intact through the intestinal wall into the bloodstream. This poses a question as to whether oral forms of these compounds are absorbed and are able to migrate to joints through the blood to improve joint function. 相似文献

This study quantifies the effects of marked weight gain on glucose and insulin metabolism in 16 cats which increased their weight by an average of 44.2% over 10 months. Significantly, the development of feline obesity was accompanied by a 52% decrease in tissue sensitivity to insulin and diminished glucose effectiveness. In addition, glucose intolerance and abnormal insulin response occurred in some cats. An important finding was that normal weight cats with low insulin sensitivity and glucose effectiveness were at increased risk of developing impaired glucose tolerance with obesity. High basal insulin concentrations or low acute insulin response to glucose also independently increased the risk for developing impaired glucose tolerance. Male cats gained more weight relative to females and this, combined with their tendency to lower insulin sensitivity and higher insulin concentrations, may explain why male cats are at greater risk for diabetes. Results suggest an underlying predisposition for glucose intolerance in some cats, which is exacerbated by obesity. An experiment was performed to determine the effects of Ca and P supplementation, inactivity, and subsequent aerobic exercise on bone metabolism in young, mature, and aged horses. Twelve stock-type geldings were blocked into three groups: 2 and 3, 7 to 11, and 15 to 21 years of age. Horses within age groups were evenly divided and randomly assigned to one of two dietary treatments; control (C) or Ca and P supplementation (T). Horses consuming C and T received about 133% and 275% of National Research Council (NRC) requirements for Ca and P, respectively. The trial consisted of eight 21-day periods. During periods I, II, and III, all horses were inactive and received no forced exercise. During periods IV, V, VI, VII, and VIII, all horses were placed on an ascending aerobic exercise regimen. Blood samples were taken at the beginning of period I and at 21-day intervals thereafter to determine serum concentrations of osteocalcin (OST) and type I carboxyterminal telopeptide (ICTP). Dorsal-palmar and lateral-medial radiographs were taken of the left third metacarpal at the beginning of period I and at 21-day intervals thereafter to monitor changes in bone densities, as indicated by radiographic bone aluminum equivalencies (RBAE). Mean serum OST was affected by age (P < .004) and the interaction of time and age (P < .006). Mean serum ICTP was affected by age (P < .001), time (P < .001), the interaction of time and age (P < .001); and the interaction of time and treatment (P < .05). There was an effect of treatment on overall mean medial (MRBAE; P < .04) and total (TRBAE; P < .04) RBAE. Overall mean dorsal RBAE (DRBAE; P < .001), palmar RBAE (PRBAE; P < .003), lateral RBAE (LRBAE; P < .001), MRBAE (P < .001), normalized MRBAE (P < .001), TRBAE (P < .05), and normalized TRBAE (P < .05) were affected by time. There was an effect of the interaction of time and age on overall mean DRBAE (P < .05), LRBAE (P < .002), MRBAE (P < .02), normalized MRBAE (P < .02), TRBAE (P < .005), and normalized TRBAE (P < .005). Overall mean DRBAE (P < .02) and PRBAE (P < .001) were affected by the interaction of time, treatment, and age. Biochemical and radiographic data from this experiment indicate that dietary Ca and P supplementation may affect bone remodeling in aerobically exercised horses of varying ages following a period of inactivity. CASE HISTORY A 21-year-old male sulphur-crested cockatoo (Cacatua galerita) was presented following the sudden appearance of blood associated with the passage of faeces and urates. CLINICAL AND PATHOLOGICAL FINDINGS: There was fresh blood-staining of the feathers around the vent. The dorsal mucosal wall of the proctodeum was erythematous and roughened in appearance. An endoscopic biopsy was performed, and histological examination revealed multiple fronds of epithelium; the mucosa varied from simple to pseudostratified columnar epithelium, with diffuse hyperplasia of goblet cells. The underlying connective tissue stroma was well vascularised and was infiltrated with mixed inflammatory cells, comprising granulocytic cells and macrophages. PCR testing for both herpesvirus and papillomavirus, using consensus primers, was negative. DIAGNOSIS: Cloacal papillomatosis. CLINICAL RELEVANCE: This case manifested typical clinical signs and histological lesions of cloacal papillomatosis in the absence of demonstrable herpesvirus or papillomavirus. Veterinarians need to consider this disease in the differential diagnosis of blood in the droppings of parrots and cockatoos even in countries where psittacine herpesviruses are exotic diseases. 相似文献
