Recently, isolation and in vitro culture of putative spermatogonial stem cells (SSCs) in the domestic cat have been conducted. However, the cellular niche conditions that facilitate the establishment and long‐term maintenance of feline SSCs (FSSCs) have not been described. Therefore, we investigated the type of feeder cells used to stimulate colony formation and growth of FSSCs among the various factors in the FSSC niche. Spermatogonial stem cells isolated from feline testes were cultured on mitotically inactivated testicular stromal cells (TSCs) derived from cats, dogs and mice, and mouse embryonic fibroblasts (MEFs). The formation and growth of colonies derived from SSCs cultured on each type of feeder cell were identified at passage 0, and the morphology, alkaline phosphatase (AP) activity and expression of SSC‐specific genes in surviving colonies were investigated at passage 4. Among these diverse feeder cells, TSCs from cat showed the greatest colony formation, growth and maintenance of FSSCs, and SSC colonies cultured by passage 4 showed a typical dome‐shaped morphology, AP activity and expression of SSC‐specific genes (NANOG, OCT4, SOX2 and CD9). Accordingly, these results demonstrate that feline TSCs could be used as feeder cells to support the establishment and maintenance of SSCs from domestic cats.  相似文献

The gate bias dependence of the field-effect mobility in pentacene-based insulated gate field-effect transistors (IGFETs) was interpreted on the basis of the interaction of charge carriers with localized trap levels in the band gap. This understanding was used to design and fabricate IGFETs with mobility of more than 0.3 square centimeter per volt per second and current modulation of 10(5), with the use of amorphous metal oxide gate insulators. These values were obtained at operating voltage ranges as low as 5 volts, which are much smaller than previously reported results. An all-room-temperature fabrication process sequence was used, which enabled the demonstration of high-performance organic IGFETs on transparent plastic substrates, at low operating voltages for organic devices.  相似文献

The impact of rising atmospheric carbon dioxide (CO2) may be mitigated, in part, by enhanced rates of net primary production and greater C storage in plant biomass and soil organic matter (SOM). However, C sequestration in forest soils may be offset by other environmental changes such as increasing tropospheric ozone (O3) or vary based on species-specific growth responses to elevated CO2. To understand how projected increases in atmospheric CO2 and O3 alter SOM formation, we used physical fractionation to characterize soil C and N at the Rhinelander Free Air CO2-O3 Enrichment (FACE) experiment. Tracer amounts of 15NH4+ were applied to the forest floor of Populus tremuloides, P. tremuloides-Betula papyrifera and P. tremuloides-Acer saccharum communities exposed to factorial CO2 and O3 treatments. The 15N tracer and strongly depleted 13C-CO2 were traced into SOM fractions over four years. Over time, C and N increased in coarse particulate organic matter (cPOM) and decreased in mineral-associated organic matter (MAOM) under elevated CO2 relative to ambient CO2. As main effects, neither CO2 nor O3 significantly altered 15N recovery in SOM. Elevated CO2 significantly increased new C in all SOM fractions, and significantly decreased old C in fine POM (fPOM) and MAOM over the duration of our study. Overall, our observations indicate that elevated CO2 has altered SOM cycling at this site to favor C and N accumulation in less stable pools, with more rapid turnover. Elevated O3 had the opposite effect, significantly reducing cPOM N by 15% and significantly increasing the C:N ratio by 7%. Our results demonstrate that CO2 can enhance SOM turnover, potentially limiting long-term C sequestration in terrestrial ecosystems; plant community composition is an important determinant of the magnitude of this response.  相似文献

Two methyl anthranilate formulations, ReJex‐iT® TP‐40 and AG‐36, were tested as bird‐repelling agents for animal feed and vegetable sprouts, respectively. Feral pigeons (Columba livia) and house sparrows (Passer domesticus) are important pests of animal husbandry and field crops in Israel. In each 4‐day experiment, four birds were held together in an aviary, and were offered four trays or dishes with animal feed or cauliflower sprouts in four corners of the cage. The trays and dishes were transposed daily in a Latin square sequence. Pigeons consumed significantly more untreated feed than treated feed. When no alternative untreated food was offered, there was no significant repellency. Treatment of animal feed with TP‐40 to give 2.0 ml methyl anthranilate kg?1 or less had no repellent effect on sparrows. Concentrations of 4.0 ml kg?1 or more repelled the sparrows significantly. When no untreated feed was presented, pigeons and sparrows preferred the lowest concentration of methyl anthranilate presented. When AG‐36 was tested on vegetable sprouts, the sparrows tended significantly to avoid the highest concentration used—14.5 ml methyl anthranilate litre?1 spray. The inter‐specific differences between the birds indicate that the efficacy of methyl anthranilate formulations as a repellent has to be specified for every species. This study did not determine (1) the possibility of developing habituation, (2) the behaviour of birds under natural conditions in the field, or (3) the influence of the feeding behaviour of a sympatric bird species on the repellency of methyl anthranilate to other species. © 2001 Society of Chemical Industry  相似文献

OBJECTIVE: To report on the first case of congenital heart defects in pigs in Australia. DESIGN: Retrospective analysis of case records from an inbred herd of "Westran" pigs at the University of Sydney, between January 2001 and December 2004. Detailed gross and histological examination of 15 hearts from pigs that had died or were euthanased in 2004. CASE DETAILS: The necropsy records from a population of 471 pigs that had died (106 pigs) or were euthanased for research purposes (365 pigs) were analysed and the incidence of heart defects recorded, together with basic demographic data. No attempts were made to diagnose the condition in live pigs. RESULTS: Congenital heart defects were diagnosed in 6.4% of pigs but this is likely to be an underestimate of the incidence of the deformity. Eighteen pigs died on the farm as a result of the defect, and 12 pigs were diagnosed with the defect as an incidental finding. The most common abnormality seen at necropsy was a sac-like dilatation on the right lateral surface of the right atrium. This was associated with secondary deformity and hypoplasia of the adjacent left ventricle, interventricular region and part of the right ventricle. All hearts showed atrial septal defects of varying size. CONCLUSION: This is the first reported case of congenital heart defects in pigs in Australia, and one of less than five reported cases of atrial septal defects in pigs in the world. The authors conclude that there may be an element of genetic predisposition to the malformation, since it has only been reported in this inbred line of pigs.  相似文献
