A nine-year-old female desexed Great Dane presented with a painful, proliferative, soft red putative neoplastic vascular mass on the nictitating membrane. Three 7-day cycles of the topical cytotoxic drug mitomycin C 0.04%, applied four times daily to the lesion using a low-dose alternate-week pulse therapy, brought about rapid remission of the lesion. The MW (moment magnitude) 7.9 Denali fault earthquake on 3 November 2002 was associated with 340 kilometers of surface rupture and was the largest strike-slip earthquake in North America in almost 150 years. It illuminates earthquake mechanics and hazards of large strike-slip faults. It began with thrusting on the previously unrecognized Susitna Glacier fault, continued with right-slip on the Denali fault, then took a right step and continued with right-slip on the Totschunda fault. There is good correlation between geologically observed and geophysically inferred moment release. The earthquake produced unusually strong distal effects in the rupture propagation direction, including triggered seismicity.  相似文献

Oats (Avena sativa L.) have received significant attention for their positive and consistent health benefits when consumed as a whole grain food, attributed in part to mixed‐linkage (1‐3,1‐4)‐β‐d ‐glucan (referred to as β‐glucan). Unfortunately, the standard enzymatic method of measurement for oat β‐glucan is costly and does not provide the high‐throughput capability needed for plant breeding in which thousands of samples are measured over a short period of time. The objective of this research was to test a microenzymatic approach for high‐throughput phenotyping of oat β‐glucan. Fifty North American elite lines were chosen to span the range of possible values encountered in elite oats. Pearson and Spearman correlations (r) ranged from 0.81 to 0.86 between the two methods. Although the microenzymatic method did contain bias compared with the results for the standard streamlined method, this bias did not substantially decrease its ability to determine β‐glucan content. In addition to a substantial decrease in cost, the microenzymatic approach took as little as 6% of the time compared with the streamlined method. Therefore, the microenzymatic method for β‐glucan evaluation is an alternative method that can enhance high‐throughput phenotyping in oat breeding programs.  相似文献

Nature has been instrumental as a source for therapeutics. Despite the fact that we live in an oceanic planet, a number of technical factors have historically hampered the evolution of a marine-based chamanic medicine. With the implementation of scuba diving tools and the development of sophisticated instruments for the isolation and elucidation of structures of natural products from marine organisms, major advances have been made in the discovery of marine derived therapeutics. The availability of ARA-C, a nucleoside analog that is a basic component in the treatment of acute myeloid leukemia, and its fluorinated analog Gemcitabine, an important therapeutic tool in the treatment of pancreatic cancer and in non small cell lung cancer, is a solid proof and validation of the potential of this approach. As a result of our discovery and developmental program, three innovative compounds with novel mechanisms of action: ET-743, AplidinR and Kahalalide F, have been shown to display a positive therapeutic index and activity in resistant solid tumors that supports the ongoing clinical phase III/II trials. ET-743 represents the first active agent against sarcomas developed in the past 25 years and has demonstrated a therapeutic potential in pretreated ovarian cancer. Several chemical entities are under advanced preclinical testing and additional candidates for clinical development are emerging, including compounds hitting a specific target. Moreover, the development of a given marine candidate implies the collaboration of an interdisciplinary team special focused on supply, formulation, pharmacogenetics and preclinical toxicology.  相似文献

Abstract Although neutral molecular markers have long been important tools for describing genetic variation in threatened fish species, many of the most critical questions in conservation relate more to quantitative genetic variation than to neutral markers. Quantitative genetic studies are typically expensive and time-consuming to conduct, especially in some of the long-lived vertebrates of conservation concern. The present review of recent literature in fish conservation genetics examines the traditional role of molecular studies in describing conservation units and providing indirect inference about local adaptation and adaptive potential. Of special interest are approaches that use a combination of molecular and quantitative genetic methods. Such studies are likely to provide important new insights into many conservation-related problems. The review also explores how increasing interest in non-neutral molecular markers is contributing to our understanding of the geographic scale and evolutionary importance of local adaptation in threatened populations. It is increasingly clear that advanced genetic technologies for the exploration of neutral and non-neutral molecular variation are leading to a fundamental shift in the way complex phenotypic traits are studied. This new synthesis of methods will have dramatic implications for fish conservation genetics and biology in general.  相似文献

BACKGROUND: Leaf wettability considerably defines the degree of retention of water and agrochemical sprays on crop and non‐target plant surfaces. Plant surface structure varies with development therefore the goal was to characterise the wettability of soybean leaf surfaces as a function of growth stage (GS). RESULTS: Adaxial surfaces of leaves developed at GS 16 (BBCH) were 10 times more wettable with water than leaves at the lower canopy (GS 13). By measuring contact angles of a liquid having an intermediate surface tension on different leaf patches, an illustrative wetting profile was elucidated, showing to what degree wetting varies (from > 120° to < 20°) depending on leaf patch and GS. While the critical surface tension of leaf surfaces at different GSs did not correlate with the observed changes, the slope of the Zisman plot accurately represented the increase in wettability of leaves at the upper canopy and lateral shoots (GSs 17 to 19, 21 and 24). The discrimination given by the slopes was even better than that by water contact angles. SEM observations revealed that the low wettability observed at early GSs is mainly due to a dense layer of epicuticular wax crystals. The Zisman plot slope does not represent the changes in leaf roughness (i.e. epicuticular wax deposition), but provides an insight into chemical and compositional surface characteristics at the droplet–leaf interface. CONCLUSIONS: The results with different wettability measurement methods demonstrated that wetting is a feature that characterises each developmental stage of soybean leaves. Positional wettability differences among leaves at the same plant and within the same leaf are relevant for performance, selectivity and plant compatibility of agrochemicals. Very-long-baseline interferometry experiments, involving observations of extragalactic radio sources, were performed in 1969 to determine the vector separations between antenna sites in Massachusetts and West Virginia. The 845.130-kilometer baseline was estimated from two separate experiments. The results agreed with each other to within 2 meters in all three components and with a special geodetic survey to within 2 meters in length; the differences in baseline direction as determined by the survey and by interferometry corresponded to discrepancies of about 5 meters. The experiments also yielded positions for nine extragalactic radio sources, most to within 1 arc second, and allowed the hydrogen maser clocks at the two sites to be synchronized a posteriori with an uncertainty of only a few nanoseconds.  相似文献
