Reasons for performing study: The pathophysiological events inhibited by prophylactic digital hypothermia that result in reduction of the severity of acute laminitis are unknown. Objectives: To determine if digital hypothermia inhibits lamellar inflammatory signalling during development of oligofructose (OF) induced laminitis. Methods: Fourteen Standardbred horses were given 10 g/kg bwt OF by nasogastric tube with one forelimb (CRYO) continuously cooled by immersion in ice and water and one forelimb (NON‐RX) at ambient temperature. Lamellae were harvested prior to the onset of lameness (24 h post OF administration, DEV group, n = 7) or at the onset of lameness (OG1 group, n = 7). Lamellar mRNA was purified and cDNA produced for real time‐quantitative PCR analysis of mRNA concentrations of cytokines (IL‐6, IL‐1β, IL‐10), chemokines (CXCL1, CXCL6, CXCL8/IL‐8, MCP‐1, MCP‐2), cell adhesion molecules (ICAM‐1, E‐selectin), COX‐2 and 3 housekeeping genes. Data were analysed (NON‐RX vs. CRYO, NON‐RX vs. archived control [CON, n = 7] lamellar tissue) using nonparametric tests. Results: Compared with CON, the OG1 NON‐RX had increased (P<0.05) lamellar mRNA concentrations of all measured mediators except IL‐10, IL‐1β and MCP‐1/2, whereas only CXCL8 was increased (P<0.05) in DEV NON‐RX. Within the OG1 group, CRYO limbs (compared with NON‐RX) had decreased (P<0.05) mRNA concentrations of the majority of measured inflammatory mediators (no change in MCP‐1 and IL‐10). Within the DEV group, mRNA concentrations of CXCL‐1, ICAM‐1, IL‐1β, CXCL8 and MCP‐2 were decreased (P<0.05) and the anti‐inflammatory cytokine IL‐10 was increased (compared with NON‐RX limbs; P<0.05). Conclusions: Digital hypothermia effectively blocked early lamellar inflammatory events likely to play an important role in lamellar injury including the expression of chemokines, proinflammatory cytokines, COX‐2 and endothelial adhesion molecules. Potential relevance: This study demonstrates a potential mechanism by which hypothermia reduces the severity of acute laminitis, and may help identify molecular targets for future laminitis intervention. 相似文献
The use of marker assisted selection in the beef cattle industry to date has involved using traditional EPD in tandem with molecular test information. In the current study, a multiple-trait simulation was carried out to create a beef cattle data set using genetic parameter estimates from the literature to identify the best procedure for combining both sources of information and to assess the added benefit of the procedure. To reach these objectives, the following simulation/ analysis steps were implemented: (1) varying percentages (100, 5, or 0) of available records for the trait of interest, (2) varying percentages (100, 50, 25, or 0) of animals with molecular information, (3) scenarios where the favorable (F) or the unfavorable (U) allele was more frequent, and (4) analysis of the response due to selection over 5 generations. The data sets included 3 correlated traits in which 2 of them, birth weight and postweaning gain, had complete recording and the availability of records for the third trait (marbling score) varied. It was further assumed that molecular information was available for the third trait for a causative gene that explained 10% of the genetic variation. Estimates of Pearson correlations between true and predicted breeding values for marbling score declined as the amount of information declined, and instances in which the molecular information was recorded were always closer to the true values than in the case in which the molecular information was absent. When the U allele was more frequent, rank correlation estimates were increased among top sires, low accuracy sires, and high accuracy sires by approximately 24.9, 12.1, and 4.7% with limited marbling score records and complete genotyping compared with limited marbling score records and no genotyping. Similar results were seen when the F allele was more frequent. When there was a complete absence of recording for the trait of interest, the same trends in correlations were observed and were lower than when the trait of interest was recorded. Jointly considering molecular and phenotypic information showed a greater long-term response compared with tandem selection, showing that discrimination of candidates for selection based solely on molecular information is not optimal. 相似文献
A three-phase study was conducted in high-potential farming and peri-urban area in Kikuyu Division central Kenya to obtain farm and management data and to monitor health and productivity of pigs in smallholder farms. The first phase was a cross-sectional study in which 87 farms (that had been selected from a total of 179 farms using a simple random selection) were visited once and data on important farm and management factors were gathered using semi-structured questionnaires. The second phase was a pilot study that was conducted in the 87 study farms for a period of 3 months to pretest the data-collection tools and to evaluate the general research methodology for the longitudinal study. The third phase was a prospective 12-month observational study in which health and productivity of pigs were monitored monthly in 76 herds that were still active and had participated in the previous studies.
The initial voluntary enrolment among the eligible farms was 99%. The median farm size was 1 acre. All the farms kept crossbreed pigs of Large White or Landrace (median nine pigs per farm). The median number of sows per herd was one. Most farmers engaged in farrow-to-finish pig-production system and most (60%) did not keep a breeding boar. The pigs were stall-fed the year round. Guard rails/piglet devices were present in 22% of the herds. Few (8%) farmers disinfected pig pens (especially the farrowing area). None of the farmers reported the use of vaccination against pig diseases. Most farmers (84 and 96%) indicated that they controlled for mange and worm infestations, respectively. To control mange, 50% of the farmers used acaricides, 34% used engine oil and 12% used both. Anthelmintics were used to control worms. No farmer had a particular control programme in place for both worms and mange. Artificial heating for piglets was not used in any of the farms. High costs of feeds (which were of variable qualities) lack of credit and genetically high-quality breeding boars and diseases were ranked highly by the farmers as the main production constraints. Thirteen percent (11/87) of farmers withdrew during the pilot study; 10 farmers had sold their pigs and one had died. Thirty-three percent (25/76) of the farms withdrew during the longitudinal study for various reasons that included death of pigs (3%; 2/76) and sale of the pigs (30%; 23/76)—mainly because of financial need. 相似文献
This study investigated the effect of exogenous insulin-like growth factor (IGF)-1 on the proliferation and differentiation of osteoblastic cells from Chinese Holstein cattle and the resultant bone nodule formation and mineralisation in vitro. The osteoblastic cells were isolated and cultured, then identified using Giemsa and alkaline phosphatase (ALP) staining methods. The effect of different concentrations of IGF-1 on cell growth was assessed by MTT assay. The ALP activity and osteocalcin (OC) concentration in the osteoblastic cells were measured by a colorimetric assay and a radioimmmunoassay, respectively. Calcium nodules were observed using alizarin red S stain, while the content of matrix calcium was determined by atomic absorption spectrophotometry. Cell proliferation in the cultures was stimulated by IGF-1 at concentrations ranging from 1 to 200 ng/mL, with the maximum effect observed at 100 ng/mL. This effect was observed from day 1 and peaked at day 5, but decreased at day 7. At concentrations of 10 ng/mL and 100 ng/mL, IGF-1 significantly induced ALP activity, OC level, matrix calcium content, and nodule formation of the osteoblastic cells by 20–180% (P < 0.05 or P < 0.01), compared to controls. The results suggested that IGF-1 is an anabolic agent for the proliferation, differentiation, mineralisation and calcium content of dairy cow osteoblasts, and could therefore act as a potential treatment for the metabolic bone diseases in these animals. 相似文献
Chemokines play a vital role in leukocyte activation and emigration that reportedly plays a central role in laminar injury in equine laminitis. The purpose of this study was to evaluate the pattern of laminar chemokine expression in horses in the classical carbohydrate overload (CHO)-model of laminitis. Laminar samples were obtained 24h following water administration in the control group (CON, n=8), and at the onset of fever (≥ 102°F, 12-22 h post CHO, DEV group, n=8) and at the onset of lameness (20-48 h post CHO, LAM group, n=8) in induced horses. Real time quantitative PCR was performed on all samples in order to determine laminar mRNA concentrations of both CXC chemokines (CXCL1, CXCL6, CXCL8) and CC chemokines (CCL2 [MCP-1], CCL3 [MIP-1α], and CCL8 [MCP-2]). Data were subjected to ANOVA followed by Student-Newman-Keuls (P<0.05). Laminar mRNA concentrations for all CXC chemokines were increased (P<0.05) at both the DEV and LAM horses when compared to the control horses, whereas mRNA concentrations of CCL2 and CCL8 were only increased in the LAM horses when compared to controls and the DEV horses. When taken in context with our previous studies, CXCL1, CXCL6 and CXCL8 increases precede peak laminar leukocyte accumulation. Additionally, CCL2 and CCL8 expression corroborate previous reports of monocyte/macrophage accumulation in affected laminae. Compared with previous studies, our findings demonstrate that increased laminar CXC chemokine expression consistently precedes peak leukocyte accumulation and onset of lameness in CHO laminitis models. Chemokine antagonists may be considered as possible therapeutic targets to decrease the influx of leukocytes that occurs during the development of equine laminitis. 相似文献
