【目的】氨酰-tRNA合成酶(aminoacyl-tRNA synthetases, aaRSs)与遗传信息传递密切相关,已发现植物中aaRSs家族蛋白在维持翻译功能之余,还参与配子发生与胚发育、质体的早期发育以及免疫信号的感知与病害防御等生物学过程。本研究利用水稻胚乳发育缺陷突变体,分析水稻色氨酰-tRNA合成酶(WRS1)在胚乳发育中的作用,证明WRS1基因编码一个影响水稻胚乳发育的关键因子。【方法】本研究通过甲烷磺酸乙酯(ethyl methane sulfonate, EMS)诱变籼稻(Oryza sativa subsp. indica)品种N22,筛选到一个稳定遗传的水稻粉质胚乳突变体(wrs1),图位克隆获得目标基因。对wrs1成熟种子进行形态学观察以及淀粉相关理化性质测定,利用细胞学切片分析wrs1发育中胚乳的结构,利用实时荧光定量PCR(quantitative real-time PCR, qRT-PCR)和GUS活性染色分析基因表达模式,通过qRT-PCR比较野生型与突变体花后12 d胚乳中淀粉合成相关基因表达情况,免疫印迹检测野生型与突变体成熟种子中淀粉合成酶蛋白积累情况,使用全自动氨基酸分析仪测定游离氨基酸含量。【结果】 wrs1突变体幼苗表现出明显的发育滞后且逐渐蔫萎死亡,从杂合突变体(WRS1wrs1)中分离到的粉质籽粒呈现明显的腹部皱缩,粒厚、千粒重下降,同时总淀粉含量下降,糊化淀粉的峰值黏度和崩解值均低于野生型。wrs1突变体发育胚乳中复合淀粉颗粒变小,排列疏松。WRS1定位于第12染色体长臂约183 kb的区间内,测序发现编码色氨酰-tRNA合成酶(tryptophanyl-tRNA synthetase, WRS)基因的第6外显子上发生单碱基替换,导致一个保守位置上的甲硫氨酸被替换。wrs1突变体中大部分淀粉合成相关基因表达量下调,且野生型与突变体间基因表达的变化与相应蛋白积累的差异存在不一致的趋势。wrs1突变体籽粒中蛋白质积累降低,而游离氨基酸含量显著升高。【结论】 WRS1编码色氨酰-tRNA合成酶,该基因突变后通过影响氨基酸稳态和蛋白质合成,造成淀粉合成相关基因异常表达从而影响淀粉的合成与积累,导致种子发育缺陷。 相似文献

目的 探明不同类型水稻品种产量和氮素吸收利用对FACE(大气CO2浓度增高)响应的差异。方法 以常规粳稻、杂交籼稻、常规籼稻共6个品种为供试材料,研究FACE对不同类型水稻产量、氮素吸收利用的影响。结果 1)FACE处理极显著提高了水稻产量,平均增加24.17%, 常规籼稻增幅最大,FACE和对照均以杂交籼稻最高;2)FACE处理显著增加了单位面积穗数,常规粳稻增幅最大,并显著增加了杂交籼稻和常规籼稻每穗粒数;3)FACE处理显著提高了成熟期吸氮量和氮素籽粒生产效率,成熟期吸氮量平均增加21.23%,杂交籼稻增幅最大, FACE和对照均以常规籼稻最高;氮素籽粒生产效率平均增加7.33%,杂交籼稻增幅最大,FACE和对照均以杂交籼稻最高。成熟期吸氮量对产量促进作用略大于成熟期氮素籽粒生产效率;4)FACE处理降低了植株含氮率,成熟期平均下降0.105个百分点,常规粳稻降幅最大。FACE处理极显著提高植株干物质量,成熟期平均增加23.95%,常规籼稻增幅最大;FACE处理显著提高常规籼稻和杂交籼稻成熟期单穗吸氮量,分别增加10.79%、13.93%,但常规粳稻下降了9.60%;FACE处理显著提高了成熟期群体吸氮强度,平均增加22.29%,杂交籼稻增幅最大。FACE处理对水稻全生育期天数无显著影响;FACE处理显著提高茎鞘、叶片、穗各器官吸氮量,叶片增幅最大,平均增加51.86%,杂交籼稻增幅最大;FACE处理显著提高了不同生育阶段吸氮量,抽穗-成熟阶段增幅最大,平均增加108.90%,杂交籼稻增幅最大;5)植株干物质量、单穗吸氮量、吸氮强度、穗吸氮量、抽穗-成熟阶段吸氮量对成熟期总吸氮量的促进作用分别大于植株含氮率、单位面积穗数、生育天数、茎鞘叶吸氮量、移栽-分蘖和分蘖-抽穗阶段吸氮量;6)FACE处理显著提高了氮肥偏生产力,降低了每百千克籽粒需氮量,前者平均增加24.16%,常规籼稻增加最多;后者平均降低4.7%,常规籼稻降幅最大。结论 FACE处理可显著提高水稻产量和氮素吸收利用效率,但品种间差异较大。 相似文献

玉米脱粒破碎率关键影响因子及其最优预测模型研究 总被引:1,自引:0,他引:1
子粒破碎率高是当前中国玉米机械收获子粒的重要限制因素,解析破碎率变化原因,建立其简便预测模型是需要解决的重要问题。本文采集7个玉米品种在3个种植密度下组合的果穗,在不同子粒含水率梯度条件下开展单穗脱粒试验。种植密度在6万~9万株/hm2范围内对子粒破碎率没有影响,品种、子粒的含水率、抗侧压碎力和穿刺强度等的影响均达到统计显著水平。品种对破碎率变化的偏贡献率为12.7%,且品种的偏贡献率子粒含水率的偏贡献率抗侧压碎力的偏贡献率穿刺强度的偏贡献率,种植密度的偏贡献率接近于零。破碎率的最优预测因子是穿刺强度,预测模型:破碎率=10.25×0.990穿刺强度,满足破碎率不高于5%约束的穿刺强度值不得低于60 MPa。研究结果可为玉米破碎率预测、宜机收玉米新品种培育与鉴定、脱粒机具设计与制造提供数据支撑和技术参考。 相似文献

为了解不同作物蒸散量估算方法在淮北地区的适用性,利用新马桥实验站称重式蒸渗仪测定了2016-2017年冬小麦全生育期的实际蒸散值,结合Hargreaves-Samani(H-S)、 FAO-56 PM、Turc、Makkind(Mak)、 Priestley-Taylor(P-T)、Mcloud(Mcl)和DeBruin-Keijman(D-K)7个模型,分析了冬小麦田的蒸散特征,将蒸散的估算值(ET_0)和实测值(ET_C)进行了对比。结果表明,相对于ET_C值,7个模型拟合得到的ET_0的RMSE值为0.99~2.29 mm·d~(-1),且H-S FAO-56 PMTurcMakP-TMclD-K; ET_C与ET_0的相关系数为0.74~0.97,其中FAO-56 PM的相关性最高,P-T、 Mak、 D-K、 H-S也表现出较好的相关性。综合来看,H-S法总体表现较好,更适合该地区。对6种主要气象要素与实测蒸散值进行主成分分析发现,温度是影响ET_C的主要因子,湿度、日照时数和平均风速(2 m)对淮北冬小麦田蒸散值的影响不大;H-S模型以温度数据为基础,利用线性订正法和湿度指数项订正法将H-S模型本地化后检验发现,其优化结果良好,RMSE降低(0.68 mm·d~(-1))。 相似文献

AIM: To investigate the effect of Linc00152 on the viability, apoptosis and radiosensitivity of cervical cancer cells. METHODS: RT-qPCR was used to detect the expression levels of Linc00152 and microRNA-376c-3p(miR-376c-3p) in human cervical cancer HeLa cells and SiHa cells, and normal cervical Ect1/E6E7 cells. The cervical cancer HeLa cells with low Linc00152 expression or miR-376c-3p over-expression were established. MTT assay, flow cytometry, colony formation assay and Western blot were used to determine the cell viability, apoptosis, radiosensitivity and related protein expression. The dual-luciferase reporter assay was used to verify the regulatory relationship between Linc00152 and miR-376c-3p in the HeLa cells. RESULTS: Compared with the Ect1/E6E7 cells, Linc00152 was up-regulated in the HeLa cells and SiHa cells, and miR-376c-3p was down-regulated (P < 0.05). Low expression of Linc00152 or over-expression of miR-376c-3p inhibited the viability of HeLa cells, induced apoptosis, enhanced the radiosensitivity, inhibited the protein expression of cyclin D and Bcl-2, and promoted the protein expression of P21 and Bax (P < 0.05). Linc00152 negatively regulated miR-376c-3p expression in the HeLa cells, and inhibition of miR-376c-3p expression reversed the effect of low expression of Linc00152 on HeLa cell viability, apoptosis and radiosensitivity. CONCLUSION: Linc00152 is highly expressed in the cervical cancer cells. Linc00152 affects the viability, apoptosis and radiosensitivity of HeLa cells by targeting miR-376c-3p, which is a potential diagnosis and treatment target for cervical cancer. 相似文献

AIM To investigate the effect of forsythiaside A (FA) on immune function in rats with ulcerative colitis and its related mechanism. METHODS Healthy SD rats were randomly divided into 5 groups: control group (no treatment, normal feeding), model group (establishment of rat ulcerative colitis model), and low, medium and high doses of FA groups (treatment of the model rats with FA at 5 mg/kg, 20 mg/kg and 80 mg/kg, respectively). The malondialdehyde (MDA) content and superoxide dismutase (SOD) activity in rat colon tissues were measured by colorimetry, and the serum levels of tumor necrosis factor-α (TNF-α), interleukin-2 (IL-2) and IL-4 were detected by ELISA. The spleen index and thymus index, the percentages of CD3+, CD4+ and CD8+ T-lymphocytes in peripheral blood mononuclear cells (PBMC), the serum IgA and IgG levels, and the serum complement C3 and C4 levels were also determined. RESULTS The colon tissues of the rats in model group showed obvious inflammation and ulceration, indicating that the animal model was successfully established. Compared with model group, the colonic inflammation and ulceration were significantly attenuated in FA groups, among which the high dose had the best effect. Compared with control group, the spleen index and thymus index of the rars in model group were decreased (P <0.05), MDA content in colon tissues was increased (P <0.05), and SOD activity in colon tissues was decreased (P <0.05). The levels of CD3+, CD4+, CD8+ and CD4+/CD8+ T-lymphocytes in PBMC, and the serum levels of C3, C4 and IL-4 were decreased (P <0.05), while the serum levels of IgA, IgG, TNF-α, and IL-2 were increased in model group as compared with control group. Furthermore, the spleen index and thymus index of the rats in FA groups were increased (P <0.05), the MDA content in the colon tissues was decreased (P <0.05), and the SOD activity in the colon tissues was increased (P <0.05). The levels of CD3+, CD4+, CD8+ and CD4+/CD8+ T-lymphocytes in PBMC, and the serum levels of C3, C4 and IL-4 were increased (P <0.05), while serum IgA, IgG, TNF-α and IL-2 levels were decreased in FA groups as compared with model group (P <0.05). CONCLUSION Forsythiaside A effectively attenuates the colonic lesions in rats with ulcerative colitis, and its mechanism may be related to reinforcement of oxygen free radical scavenging power, alleviation of inflammatory response, and enhancement of immune function. 相似文献

AIM To investigate the protective effect of resveratrol (Res) on cortical neurons in rat bacterial meningitis (BM) model. METHODS Group B hemolytic Streptococcus was injected via the posterior cistern to establish a BM model. Resveratrol was administered intranasally and microRNA-223-3p (miR-223-3p) antagomir was administered by intracerebroventricular injection. HE staining was used to observe the pathological changes of the brain tissue. Loeffler scoring method was used to evaluate the neurobehavioral functions. TUNEL staining was used to detect neuronal apoptosis. The expression of interleukin-1β (IL-1β), IL-18, glial fibrillary acidic protein (GFAP) and ionized calcium-binding adaptor molecule 1 (Iba1) was detected by immunofluorescence staining. The protein levels of nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3), cleaved caspase-1, IL-1β and IL-18 were determined by Western blot. The expression level of miR-223-3p was detected by RT-qPCR. Online software TargetScan was used to search for the complementary nucleotide sequences between miR-223-3p and NLRP3 mRNA. RESULTS Compared with sham group, the thickness of meninges in BM model was increased, the neurological score was decreased (P <0.05), and the number of TUNEL positive neurons was increased significantly (P <0.05). Astrocytes and microglia were activated, the fluorescence intensity of IL-1β and IL-18 was increased (P <0.05), and the expression levels of NLRP3, cleaved caspase-1, IL-1β, IL-18 and miR-223-3p were increased (P <0.05). Compared with BM group, after treatment with resveratrol, the neurological score was increased (P <0.05), the number of TUNEL positive neurons was decreased significantly (P <0.05), and the inflammatory response of astrocytes and microglia was suppressed. The fluorescence intensity of IL-1β and IL-18 was decreased (P <0.05), the protein levels of NLRP3, cleaved caspase-1, IL-1β and IL-18 were decreased (P <0.05), and the expression level of miR-223-3p was increased (P <0.05). A nucleotide sequence in the 3'-UTR of NLRP3 mRNA might be targeted by miR-223-3p. In the brain of rat BM model, compared with antagomir control group, the expression of NLRP3 was increased in miR-223-3p antagomir group with resveratrol treatment (P <0.05). CONCLUSION Resveratrol may reduce the inflammatory death of cortical neurons in BM model of infant rats through miR-223-3p/NLRP3 pathway, thus playing a protective role for the neurons. 相似文献
