AIM: To determine the suitability of measuring faecal oestrone sulphate (OS) by enzymeimmunoassay as a means of determining pregnancy status in mares bred under New Zealand conditions. METHODS: An antibody-coated microtitre plate-based enzymeimmunoassay was used to determine the concentration of OS in faecal and plasma samples obtained from pregnant and non-pregnant mares. RESULTS: In non-pregnant mares, the mean faecal OS concentration was 34 ng/g, and the value three standard deviations above this was 80 ng/g. None of 427 faecal samples collected from 116 non-pregnant mares over a l-year period had an OS concentration >80 ng/g. Only five samples from three mares had an OS concentration >65 ng/g, the value two standard deviations above the mean non-pregnant value. Analysis of faecal OS concentrations in 532 faecal samples collected from 39 pregnant mares showed that as pregnancy progressed, an increasing proportion of faecal samples had OS concentrations >80 ng/g. None of the mares 150 days or more pregnant had faecal OS concentrations <50 ng/g, and 204/220 samples obtained from these mares had faecal OS concentrations >80 ng/g. Following foaling or foetal death, elevated faecal OS concentrations returned quickly to non-pregnant levels. The mean +/- s.e.m. plasma level of OS in five mares bled daily throughout one oestrous cycle was 1.7 +/- 0.2 ng/ml. Sixty-eight blood samples from pregnant mares bled up to five times between 92 days after mating and foaling all had plasma OS concentrations >30 ng/ml, with 64/68 being >50 ng/ml. CONCLUSIONS: This study shows that measuring faecal OS concentrations by enzymeimmunoassay offers a convenient, accurate, non-invasive means of determining pregnancy status in mares from 150 days after mating onwards. Mares with faecal OS concentrations <50 ng/g can be considered not pregnant, while mares with faecal OS concentrations >80 ng/g can be considered pregnant. Those few mares returning a faecal OS concentration between 50 and 80 ng/g should be retested to obtain a conclusive result. Measuring plasma OS concentrations allows pregnancy status to be determined earlier (from 100 days after mating). Moreover, the discrimination between non-pregnant and pregnant levels is greater for OS in plasma than in faeces. CLINICAL RELEVANCE: Measurement of OS concentrations in faeces provides an alternative and non-invasive means of determining pregnancy status in mares from 150 days after mating.  相似文献

Primary porcine endothelial cells have a limited life span in culture. After four to five passages, they tend to de-differentiate and eventually reach senescence. The aim of this work was to establish immortalized porcine aortic endothelial cell lines (AOCs) to facilitate in vitro studies of different pathological process involving the endothelium. Primary porcine aortic endothelial cells (PAECs) were transfected with a plasmid containing the SV40 genome and selected on the basis of morphological and phenotypical features. Flow cytometry analysis demonstrated uptake of acetylated low density lipoproteins (Ac-LDL) and constitutive expression of SLA class I, CD29, CD31, CD41/61, CD80/86, CD46, SWC3, and LAMP-1 antigens by all analyzed lines and showed little differences to primary cells. The functional similarity between primary and immortalized endothelial cells was demonstrated in a cytotoxicity assay using a human natural killer cell line (NKL) as effector. The AOCs cell lines should be valuable tools for in vitro study of the human immune response against pig endothelial cells. In addition, they would be very useful to gain insight in the pathogenesis of some viral haemorrhagic diseases of pig such as African swine fever (ASF) or classical swine fever (CSF).  相似文献

Catalytic antibodies, or artificial enzymes, act as catalysts by lowering the energy of the transition state of a reaction. In his Perspective, Brauman highlights the study by Simeonov et al., who have applied similar principles to photochemical processes and shown that intense fluorescence can be achieved. The approach provides a way of measuring internal molecular motion within the antibody on a very fast time scale. The ultrastructural cytology and reproduction of Amphiacantha longa Caullery et Mesnil, 1914 is described. Merogonial reproduction was not observed. The sporogony follows two lines: free disporoblastic and enveloped, polysporoblastic, involving sporoblast mother cells. The enveloped sporogony is endogenous in spore sacs of sporont origin, daughter cells are formed by vacuolation. Probably all stages have coupled nuclei. Both free and enveloped spores are equipped with an extrusion apparatus composed of a flat polar sac, a straight polar filament of manubrium type, and a posterior globular appendix. Manubrium and appendix are enclosed in a membraneous coat. Circular elements of coat material occur in the proximity of the extrusion apparatus. The membraneous coat and the surface layer of the manubrium penetrate the polar sac. The extrusion apparatus is located at the wide pole of the spore, the nuclei at the narrower pole. Hosts are gregarines of the genus Lecudina Mingazzini, living in the gut of the polychaete Lumbrinereis fragilis (O. F. Müller). The cytology and reproduction are discussed and compared to other genera of metchnikovellideans, to the chytridiopsid genera, and to microsporidia expressing the typical cytology for the group. Metchnikovellideans and chytridiopsids exhibit cytological and reproductive similarities. The species is redescribed, the diagnosis of the genus Amphiacantha Caullery et Mesnil, 1914 is emended, and the new family Amphiacanthidae, comprising the genera Amphiacantha and Amphiamblys Caullery et Mesnil, 1914, is established.  相似文献
