Plasmatic concentrations of von Willebrand Factor (vWF) increase during pregnancy in humans and dogs; however the mechanism of such increase is still not well defined. The aims of this study were: (i) to evaluate changes in vWF concentration during pregnancy and during the subsequent oestrous cycle in bitches affected and unaffected by von Willebrand Disease (vWD); (ii) to correlate the vWF levels and cortisol levels in both groups. Seven vWD affected (GI) and nine unaffected (GII) bitches were used. The animals were assessed during pregnancy, parturition, lactation and non‐gestational oestrous cycle in 11 moments (Pregnancy 1, Pregnancy 2, Parturition, Lactation 1, Lactation 2, Lactation 3, Anestrus, Proestrus, Oestrus, Diestrus 1, and Diestrus 2). The following tests were performed; measurement of von Willebrand factor antigen (vWF:Ag), albumin and cortisol. In both groups, vWF concentration remained stable during the non‐gestational oestrous cycle, but increased during pregnancy, with the highest value observed at parturition. Increases of 70% and 124% in vWF were seen in GI and GII, respectively, compared to anestrus. No correlation was found between vWF and cortisol. Values of vWF:Ag changed during pregnancy, with a peak at parturition, both in vWD affected and unaffected animals. Values of vWF were not altered in the different phases of the oestrous cycle following pregnancy in both groups. Evaluation of vWF during pregnancy can cause false negative results for vWD, but assessment can be performed at any point in the oestrous cycle of non‐pregnant bitches.  相似文献

We describe the use of a capillary rise method to measure the wettability of 10 samples of agricultural soil from Rothamsted long-term experimental sites. The samples have very similar clay contents, but organic carbon (C) contents range from 11.5 to 31.2 g kg−1. Their wetting rates were interpreted by an improved method of data analysis, consistent with the Washburn equation, and showed an increase in the effective contact angle between the water meniscus and the soil with increasing C content. This corresponds to a decrease in wettability with increasing C content, and accords with other results reported in the literature. By contrast with water, we found that capillary rise for n -hexane into soil did not depend on the soil's bulk density or C content. A priori calculations of the expected wetting rates from fluid properties and an effective hydraulic radius estimated by other methods gave magnitudes and trends that agreed with our experimental data. The results show that estimates of effective hydraulic radius can provide a useful approximation for characterizing soil wetting, but that further modelling should be carried out.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) were measured in two wetland plant species grown outdoors in pots of sediment contaminated with up to 730,000 μg/kg total PAHs. After approximately 3 months, the plants were harvested, cleaned, and analyzed for an expanded suite of PAHs which included both the 16 priority PAHs and 22 alkyl PAH homologs. Sediment and air samples were also collected and analyzed. PAH compounds were detected in all of the samples, with the highest concentrations in the contaminated sediment. The root sample concentrations were generally about one order of magnitude lower than that of the sediment, and were strongly correlated with the concentration in the sediment in which they were grown. The concentrations in foliage were much lower and did not correlate with sediment concentration. Concentrations of low molecular weight PAH compounds detected in the foliage were not significantly lower in plants grown in control sediments, suggesting that the sediment is not the primary source of these PAHs. Several high molecular weight PAHs were detected only in plants grown in contaminated sediment. Plants of both species grown in control sediment were larger than plants grown in contaminated sediment.  相似文献

The aim of this study was to compare measurement of spermatozoal membrane status using computer‐assisted spermatozoal quantification (CASQ) and eosin‐nigrosin (EN) staining with manual counting after CFDA/PI staining. Analysis was performed on both fresh and thawed cryopreserved canine semen. Membrane‐disrupted spermatozoa (MDS) were counted using CASQ (n = 311) in an untreated sample and a completely membrane‐disrupted sample, and the percentage of membrane‐intact spermatozoa (MIS) calculated: (Total count ? Untreated sample count) ÷ Total count × 100. Spermatozoa were stained with a one‐step EN stain (n = 501), and then, at least 100 spermatozoa were manually examined under ×1,000 magnification and classified as MDS (stained with eosin) or MIS (non‐stained). Spermatozoa from the same samples were also stained with CFDA/PI, and then, at least 200 spermatozoa were manually examined under ×1,000 magnification and classified as MIS (completely stained by CFDA) or MDS. The percentage of progressively motile spermatozoa (PMS) was determined by both computer‐assisted semen analysis (CASA) and subjective methodologies, and the data were subsequently analysed to measure the agreement between the CASQ and EN methods with the CFDA/PI technique using Bland–Altman methodology. Pearson's correlation was measured between the MIS and PMS percentage samples and correlation coefficients compared. The mean MIS percentage was lower for CASQ and higher for EN than in CFDA/PI for all comparisons. The agreement of MIS percentage between CASQ and CFDA/PI was ?20.2% to 32.0%, and between EN and CFDA/PI was ?32.9% to 14.9%. In all methods, the MIS and PMS percentages were correlated (p < .001). Measurement of CFDA/PI appeared to be the most reliable and accurate method of determining MIS percentage in dogs. Further investigation is required to determine whether the CASQ technique can be improved. Eosin‐nigrosin staining also appeared to be unreliable at MIS <80% and overestimated the MIS percentage.  相似文献

Dairy Australia is the national service body for the Australian dairy industry. Its role is to help farmers adapt to a changing operating environment and achieve a profitable, sustainable dairy industry. Although the use of antibiotics in Australian agriculture is relatively low in global terms, Dairy Australia recognises important drivers for continuous improvement in antimicrobial stewardship (AMS). Dairy Australia’s first strategic priority is to support profitable farms. This priority has driven the development of a range of on‐farm change management programs in the animal health and welfare fields to optimise the unit cost of production and dairy cattle welfare. Dairy Australia’s third strategic priority is to further develop a ‘trusted dairy industry’. Previous and current work under these two strategies position the dairy industry favourably with respect to confronting the challenge of antimicrobial resistance (AMR) and supporting sound AMS with the guiding principle of ‘as little as possible, as much as necessary’. However, given an incomplete but ongoing threat of AMR, more work is needed. Supported by Dairy Australia, the dairy industry has developed an antimicrobial use strategy aligning with the Australian Animal Sector National AMR Plan 2018.  相似文献

The migration and evolution of a deep ocean hydrothermal event plume were tracked with a neutrally buoyant RAFOS float. The float remained entrained in the plume for 60 days, and the plume vorticity was calculated directly from the anticyclonic motion of the float. Concentrations of suspended particles, particulate iron, and dissolved manganese in the plume did not decay significantly during the 60 days, which indicates that event plumes would be easily detectable a year after formation.  相似文献

Invasive and/or opportunistic organisms can negatively influence the success of sea urchin culture operations. To our knowledge, pests associated with sea urchin aquaculture have not been reported. In this study, we report the predatory association of the amphipod, Elasmopus levis (Amphipoda: Melitidae), with the sea urchin, Lytechinus variegatus (Echinodermata: Echinoidea), within an intensive inland culture system. Wild‐caught Ly. variegatus broodstock were placed in an intensive culture system, and within their canopy of spines or within transport seawater, populations of epifauna (including amphipods) and microfauna were most likely transferred into the culture system. A growing population of the amphipod developed over time. Associations of the amphipod with the sea urchin were observed to negatively affect the health of the sea urchin, and in some cases, epithelial tissue from the sea urchin test appeared to be consumed by the amphipod. An infestation of these amphipods in commercial sea urchin cultures could have the potential to be costly, and we recommend quarantine and/or water purification procedures be considered to prevent the introduction of pests to inland culture systems.  相似文献

Four pairs of near-isogenic wheat lines, with and without the 1BL/1RS translocation, and differing at the Glu-1 loci (coding for high molecular weight [HMW] glutenin subunits) were evaluated for their dough mixing properties, dough stickiness, and baking performance. In all 1BL/1RS translocation lines, weakening of the dough consistency occurred within 2 min past peak time. The full-formula dough from every 1BL/1RS translocation line exhibited poor dough mixing characteristics and increased stickiness compared to the corresponding wheat control. The HMW glutenin subunits coded by the Glu-A1 locus had no apparent effect on mixing properties, but did have a slight effect on the dough stickiness at two of the four stages of dough mixing. Glu-B1 and Glu-D1 loci encoded glutenin subunits produced significant changes in dough mixing properties and dough stickiness, respectively. With respect to baking performance, there was no significant difference between loaf volumes of 1BL/1RS versus control wheats for three of four near-isogenic pairs. Within the 1RS-group, the translocation lines containing HMW glutenin subunits 5+10 produced bread with greater loaf volumes than the pairs containing its allelic counterpart 2+12. Loaf volume was not influenced by the subunits associated with the Glu-B1 loci. In general, the breads baked from 1BL/1RS translocation lines had a relatively poor crumb and crust quality and contained larger gas cells than the wheat controls. In comparing isogenic pairs, the magnitude of the difference in loaf volume between the control wheat and the corresponding 1BL/1RS translocation line was greater in the pair unique for HMW subunits 5+10; the difference was primarily due to the stronger mixing properties of the wheat control.  相似文献
