BackgroundCanine herpesvirus type 1 (CaHV-1) infects dogs and is associated with neonatal deaths and reproductive, ocular, neurological, and respiratory problems. In Brazil, reports of CaHV-1 have been restricted to the southeast and south regions, particularly in municipalities in the state of Rio Grande do Sul.ObjectivesTo assess the presence and variability of CaHV-1 in canine populations in the state of Pará, North Brazil.MethodsBiological samples from 159 dogs from 4 municipalities in the State of Pará were evaluated using polymerase chain reaction and phylogenetic analyses, with the target being the viral enzyme, thymidine kinase.ResultsCaHV-1 was detected in 13 dogs (8.2%), with 2 animals being from the municipality of Santa Bárbara do Pará, 8 from Algodoal Island, 2 from Salinópolis, and one from Capanema. The study sequences revealed 100% identity among themselves and 64% to 100% identity with the other nucleotide sequences from Australia, Brazil, United Kingdom, and United States, including 100% identity with the 2002 isolate from Australia. The 1996 isolate from France was grouped in a branch that was different from the sequence of this study.ConclusionsThis study presents the first molecular detection of CaHV-1 in dogs from the Amazon region in northern Brazil. The nucleotide identity between the strains and cytosine insertion in the sequences isolated in this study suggests at least 2 strains of CaHV-1 circulating in Brazil (Pará and BTU-1). 相似文献

The objectives of this work were to evaluate the genetic variability of Meloidogyne enterolobii by molecular markers, and develop species‐specific molecular markers for application in detection. Sixteen M. enterolobii isolates from different geographical regions (Brazil and other countries) and hosts were used in this study. The identification and purification of the populations were carried out based on isoenzyme phenotype. The DNA amplification of the intergenic region (IGS) of the rDNA and of the region between the cytochrome oxidase subunit II (COII) and 16S rRNA genes (mtDNA) produced specific fragments of the expected size for this nematode, i.e. 780 and 705 bp, respectively. Intraspecific variability among the isolates was evaluated with three different neutral molecular markers: AFLP, ISSR and RAPD. The results showed a low level of diversity among the isolates tested, indicating that M. enterolobii is a genetically homogeneous root‐knot nematode species. The RAPD method allowed the identification of a species‐specific RAPD fragment for M. enterolobii. This fragment was cloned and sequenced, and from the sequence obtained, a set of primers was designed and tested. The amplification of a 520‐bp‐long fragment occurred only for the 16 isolates of M. enterolobii and not for the 10 other Meloidogyne species tested. In addition, positive detection was achieved in a single individual female, egg‐mass and second stage juvenile of this nematode. This SCAR species‐specific marker for M. enterolobii represents a new molecular tool to be used in the detection of this nematode from field samples and as a routine diagnostic test for quarantine devices . 相似文献

Summary Sweet potatoes were micropropagated and then transplanted from axnic conditions to fumigated soil in pots in the greenhouse. Spores of Glomus clarum were obtained from Brachiaria decumbens or from sweet potatoes grown in soil infected with this fungus and with an enrichment culture of Acetobacter diazotrophicus. Three experiments were carried out to measure the beneficial effects of vesicular-arbuscular mycorrhizal (VAM) fungi-diazotroph interactions on growth, nutrition, and infection of sweet potato by A. diazotrophicus and other diazotrophs obtained from sweet potato roots. In two of these experiments the soils had been mixed with 15N-containing organic matter. The greatest effects of mycorrhizal inoculation were observed with co-inoculation of A. diazotrophicus and/or mixed cultures of diazotrophs containing A. diazotrophicus and Klebsiella sp. The tuber production was dependent on mycorrhization, and total N and P accumulation were increased when diazotrophs and G. clarum were applied together with VAM fungal spores. A. diazotrophicus infected aerial plant parts only when inoculated together with VAM fungi or when present within G. clarum spores. More pronounced effects on root colonization and intraradical sporulation of G. clarum were observed when A. diazotrophicus was co-inoculated. In non-fumigated soil, dual inoculation effects, however, were of lower magnitude. 15N analysis of the aerial parts and roots and tubers at the early growth stage (70 days) showed no statistical differences between treatments except for the VAM+Klebsiella sp. treatment. This indicates that the effects of A. diazotrophicus and other diazotrophs on sweet potato growth were caused by enhanced mycorrhization and, consequently, a more efficient assimilation of nutrients from the soil than by N2 fixation. Brazilian consumers prefer beans of the beige grain colour. Common beans help supply dietary requirements for iron and zinc. The objectives of this study were (i) to determine whether early selection for grain colour affects the iron and zinc content and the grain yield in common beans and (ii) if an association between these characteristics was found, to obtain lines of carioca‐type grains with a high iron and zinc content and good grain yield. We evaluated 96 F3:4, F3:5 and F3:6 progeny at two locations for iron and zinc content and grain yield. We selected 48 progeny for grain colour and chose 48 others at random. We found that early selection for grain colour does not affect the zinc content and grain yield. A positive and high association was found between iron and zinc content, and both iron and zinc content were slightly negatively correlated with grain yield. It is possible to obtain common bean lines combining a high iron and zinc content with good grain yield as long as a selection index is used. 相似文献

The saffron finch (Sicalis flaveola brasiliensis) is a Passeriforme of the Brazilian wildlife. There are scarcely any morphological studies on it, although it is frequently trafficked for its birdsong abilities. Its peculiarities, such as territorialism and developed syrinx that provides outstanding song, draw attention towards its domestication. Thus, this study aimed to morphologically describe the tongue and digestive tube organs of this species to furnish subsidies for nutritional, clinical and conservation studies. Forty‐one birds from the Wild Animal Screening Center (CETAS)/Brazilian Institute of Environment (IBAMA)/city of Cabedelo, state of Paraíba (PB) were used. Samples were collected, identified and sent to standard light microscopy; samples of proventriculus and gizzard were sent to Scanning Electron Microscopy (SEM). The samples showed stratification similar to that of other domestic and wild birds, confirmed in the scanning electron microscopy; however, they differed in the absence of dermal papillae in the tongue, lack of ingluvial glands and lack of muscular mucosa and sub‐mucosa in the large intestine. 相似文献

Nitrogen fertilization is a common practice for sustaining forage production in forage systems in southeastern United States. Warm-season annual legumes may be an alternative forage to warm-season perennial grasses that do not require N fertilization. Sunn hemp (Crotalaria juncea L.) is a fast-growing, warm-season annual legume native to India and Pakistan. The objective of this 2-year study was to assess the herbage accumulation (HA), atmospheric N2 fixation (ANF) and nutritive value of sunn hemp. Treatments were the factorial arrangement of two sunn hemp cultivars (“Crescent Sun” and “Blue Leaf”), three seeding rates (17, 28 and 39 kg seed/ha) and seed inoculation (inoculated or non-inoculated seeds), distributed in a randomized complete block design with four replicates. Crescent sun had greater HA (3,218 vs. 1764 kg DM/ha) and ANF (41 vs. 25 kg N/ha). Blue leaf had greater crude protein (CP) (188 vs. 176 g/kg) and in vitro digestible organic matter (IVDOM) concentrations (564 vs. 531 g/kg) than crescent sun. Non-inoculated seed had greater CP than inoculated seed, 188 and 177 g/kg, respectively, and inoculation did not affect HA. Intermediate seeding rate (28 kg/ha) decreased HA (2002 kg DM/ha), while HA from high and low seeding rates (17 and 39 kg/ha, respectively) did not differ (2,863 and 2,615 kg DM/ha respectively). Planting non-inoculated crescent sun at 17 kg/ha seeding rate is a feasible management practice to produce sunn hemp in subtropical regions; however, inoculation should always be recommended for proper establishment. 相似文献
