An unusual stem rust infestation occurred in German wheat fields in summer 2013. This study analysed 48 isolates derived from 17 Puccinia graminis f. sp. tritici (Pgt) samples and six races were identified: TKTTF, TKKTF, TKPTF, TKKTP, PKPTF and MMMTF. Infection type and genotypic data confirmed that none of these races belonged to the TTKS (Ug99) race group. German isolates of race TKTTF are phenotypically different to the ones responsible for the stem rust epidemic in Ethiopia in 2013–2014. Forty isolates were genotyped using a custom SNP array. Phylogenetic analysis showed that these 40 isolates represented two distinct lineages (clade IV and clade V). Thirty‐eight isolates clustered into clade IV, which previously was defined by Ethiopian isolates of race TKTTF. Race TKKTP is of special concern due to its combined virulence to stem rust resistance genes Sr24, SrTmp and Sr1RSAmigo. The vulnerability to race TKKTP in US and international winter wheat was confirmed as 55% of North American and international cultivars and breeding lines resistant to race TTKSK (Ug99) became susceptible to TKKTP. Races identified in Germany in 2013 confirmed the presence of virulence to important resistance genes that are effective against race TTKSK. This information should be useful for breeders to select diverse and effective resistance genes in order to provide more durable stem rust resistance and reduce the use of fungicides.  相似文献

The wastes and the by-products of food industrial technologies are suitable for bioenergy generating because of the high organic matter content. Anaerobic digestion is the eldest technology for waste stabilization and however by controlled decomposition a high value and marketable energy source can be produced. Whey is normally used as a component of dairy products or as an additive for food product. In our work we focused on another utilization method: biogas generating from membrane separated fractions i.e.: permeate and concentrate of whey. The effect of the pH, thermal, microwave pre-treatment and their combinations on the biogas yield were investigated. Our results showed that the applied pre-treatments had significant effect on biogas production. In consequence of the hydrolysis of large molecules the biodegradability of the pre-treated whey fractions was enhanced, therefore the biogas and methane production yield increased significantly.  相似文献

The disease development and population structure of Puccinia graminis f. sp. avenae, which causes stem rust on oats, were studied to investigate if sexual reproduction plays an important role in the epidemiology of the disease. The genetic population structure of P. graminis f. sp. avenae in Sweden was investigated by sampling 10 oat fields in July and August 2008 and seven fields during the same period in 2009. Nine single‐pustule isolates were first used to test simple sequence repeat (SSR) markers developed for P. graminis f. sp. tritici. Eleven of the 68 tested SSR markers were useful for genotyping P. graminis f. sp. avenae. For the main study, DNA from single uredinia was extracted and the SSR markers were used to genotype 472 samples. Both allelic and genotypic diversity were high in all fields, indicating that P. graminis f. sp. avenae undergoes regular sexual reproduction in Sweden. No significant relationship between genetic and geographic distances was found. Disease development was studied on two farms during 2008 and 2009. The apparent infection rates ranged between 0·17 and 0·55, indicating the potential for rapid disease development within fields. The incidence of oat stem rust has increased recently in Sweden. One possible explanation is a resurgence of its alternate host, barberry (Berberis spp.), after the repeal of the barberry eradication law in 1994. Barberry is present in several grain‐producing areas in Sweden, which supports the conclusion that P. graminis f. sp. avenae undergoes regular sexual reproduction there.  相似文献

The pathogenicity and identity was studied of a field sample (PcE) of crown rust fungus Puccinia coronata collected in Hungary on wild couch grass (Elymus repens) and of a field sample (Psb) of stripe rust (P. striiformis) collected in the Netherlands on California brome (Bromus carinatus). We focused on the analysis of the host susceptibility of cultivated barley (Hordeum vulgare) to both pathogen samples, in direct comparison with an isolate of P. striiformis f.sp. tritici (Pst) and P. striiformis f.sp. hordei (Psh). At the seedling stage 83 % of the barley accessions were susceptible to PcE. At the adult plant stage, cultivated barley was predominantly resistant to this crown rust sample. Morphology, pathogenicity on Hordeum and Bromus and ITS DNA sequence analysis determined that PcE represents a European specimen of P. coronata f.sp. hordei. Sample Psb was pathogenic on 47 % of the cultivated barley at the seedling stage. This is intermediate between the isolates Psh (90 %) and Pst (10 %). Psb and Psh were pathogenic to grasses representing several genera. The host range of the Pst isolate was more narrow. We conclude that Psb is unlikely to represent a genotype of Psh that happened to have colonized B. carinatus, and is likely a distinct form of P. striiformis.  相似文献

Many legume plants benefit from the tripartite symbiosis of arbuscular mycorrhizal fungi (AMF) and rhizobia. Beneficial effects for the plant have been assumed to rely on increased P supply through the mycorrhizas. Recently, we demonstrated that P does not regulate the establishment of the tripartite symbiosis. Flavonoids appear to play a role as early signals for both rhizobia and AMF. Four soybean lines known to express different concentrations of the isoflavones genistein, daidzein and glycitein in the seed were used to test three hypotheses: (i) The establishment of the tripartite symbiosis is not dependent of a nutrient mediated effect; (ii) There is a positive correlation between seed isoflavone concentrations of different soybean lines and the progress of the tripartite symbiosis; (iii) Specific flavonoids control the establishment of the tripartite symbiosis in that a change in flavonoid root accumulation resulting from the development of one microbial partner can stimulate colonization of soybean roots by the other. Disturbed versus undisturbed soil treatments were produced to vary the potential for indigenous AMF colonization of soybean. In contrast, the potential for Bradyrhizobium was kept identical in both soil disturbance treatments. The uptake of P and Zn and the concentration of flavonoids in mycorrhizal soybean roots at 10 d after emergence were analysed either separately of Bradyrhizobium or in context of the tripartite symbiosis. Zinc nutrition did not differ between AMF treatments which supports the first hypothesis. The concentration of daidzein was at least four times greater in the root than in the seed reaching 3958±249 μg g−1 dry across soybean lines. Coumestrol, which was absent in the seed, was synthesized to reach 2154±64 μg g−1 dry. Conversely, the concentration of genistein was approximately three times smaller in the root that in the seed (301±15 μg g−1 dry), while glycitein and formononetin were never detected. The establishment of the tripartite symbiosis was identical across soybean lines which does not support the second hypothesis. Concentrations of flavonoids were significantly greater in roots under disturbed soil, for which both symbioses were not as developed as in plants from undisturbed soil. This clearly supports the third hypothesis. This research provides the first data linking the function of different flavonoids to the establishment of the tripartite symbiosis, and suggests that these compounds are produced and released into the rhizosphere as a function of the colonization process.  相似文献

Uranium-series dating of corals from marine deposits of the U.S. Atlantic Coastal Plain coupled with paleoclimatic reconstructions based on ostracode (marine) and pollen (continent) data document at least five relatively warm intervals during the last 500,000 years. On the basis of multiple paleoenvironmental criteria, we determined relative sea level positions during the warm intervals, relative to present mean sea level, were 7 +/- 5 meters at 188,000 years ago, 7.5 +/- 1.5 meters at 120,000 years ago, 6.5 +/- 3.5 meters at 94,000 years ago, and 7 +/- 3 meters at 72,000 years ago. The composite sea level chronology for the Atlantic Coastal Plain is inconsistent with independent estimates of eustatic sea level positions during interglacial intervals of the last 200,000 years. Hydroisostatic adjustment from glacial-interglacial sea level fluctuations, lithospheric flexure, and isostatic uplift from sediment unloading due to erosion provide possible mechanisms to account for the discrepancies. Alternatively, current eustatic sea level estimates for the middle and late Quaternary may require revision.  相似文献

Fluid inclusions in uranium series-dated calcitic veins from the southern Great Basin record a reduction of 40 per mil in the deuterium content of ground-water recharge during the Pleistocene. This variation is tentatively attributed to major uplift of the Sierra Nevada Range and the Transverse Ranges during this epoch with attendant increasing orographic depletion of deuterium from inland-bound Pacific storms.  相似文献

One oral dose of pyrazole caused necrosis of rat thyroid follicular epithelial cells but spared the parafollicular (C) cells and the parathyroid glands. Serum thyroxine (T4) and triiodothyronine (T3) were significantly decreased on day 3 after pyrazole administration and were immeasurable on day 5. At day 5 the thyroid was enlarged and the concentration of thyroid-stimulating hormone in the serum was increased, indicating an appropriate pituitary response to a primary lesion in the thyroid. Doses of pyrazole which produced no morphologic change in the thyroids also significantly depressed the concentrations of T4 and T3 in the serum.  相似文献

Wheat stem rust, caused by Puccinia graminis f. sp. tritici, is a re-emerging disease, posing a threat to wheat production. In Spain, stem rust has been rarely detected since the 1970s, but infection was observed in wheat fields in 2018. We analysed six stem rust samples collected in Rota, Cádiz province and one from Monteagudo del Castillo, Teruel province. All the samples from Rota were typed as race TKTTF, whereas the sample from Monteagudo del Castillo, collected in a wheat field adjacent to barberry bushes, was typed as race TKHBK. This race has a unique and significant virulence combination that includes virulence to Sr31, Sr33, Sr53 and Sr59, and is avirulent to Rusty, a durum line developed for universal susceptibility to the wheat stem rust pathogen. TKHBK is the first race outside the Ug99 race group with virulence to Sr31 and the first known race with virulence to Sr59. Genotyping studies indicate that race TKHBK does not belong to the Ug99 or TKTTF race groups and constitutes a previously unknown lineage. Two hundred bread and durum wheat cultivars and breeding lines from Spain were evaluated against TKHBK, TKTTF, and six additional races. Resistance was observed to all the races evaluated. Molecular markers confirmed the presence of Sr7a, Sr24, Sr31, Sr38 and Sr57 in bread wheat, and Sr13 in durum wheat. The re-emergence of wheat stem rust in Spain and the occurrence of unique virulences underscore the need to continue surveying and monitoring this disease.  相似文献
