The antimutagenic activity against Trp-P-1 of methanolic extracts of 118 samples (108 species) of edible Thai plants was examined by the Ames Test. The activity was evaluated by the amount of plant extracts which suppressed 90% of the mutagenesis (ED90). Five plants, Micromelum minutum, Oroxylum indicum, Cuscuta chinensis, Azadirachta indica, and Litsea petiolata, exhibited significant activity with antimutagenic ED90 values lower than 5 microL/plate (0.1 mg of dry plant material equivalent). The activity-guided fractionation of the extract of M. minutum, which exhibited the highest antimutagenic activity in the screening, resulted in the isolation of an active principle, (+)-mahanine (1) as confirmed by its physicochemical properties. Compound 1 showed a wide variety of biological activity, including antimutagenicity against heterocyclic amines such as Trp-P-1 with an IC50 of 5.2 microM, cytotoxicity against a tumor cell line HL60 with a MIC100 of 4.0 microg/mL, and antimicrobial activity against Bacillus cereus and Staphylococcus aureus with MIC100 values of 6.25 and 12.5 microg/mL, respectively. 相似文献

Many reports have been published about designing the strength of glulam using simulation methods. In simulation methods, one of the most important problems is how to deal with correlations among strength factors, i.e., modulus of elasticity (MOE), modulus of rupture (MOR), tensile strength (σT), and compression strength (σC). For example, in the case that the MOR criteria of glulam is σ ni /f ni + σ bi /f bi ≥ 1 (where σ ni and σ bi are the axial stress and the bending stress of the i-th lamina respectively, and f ni and f bi are the axial strength and the bending strength of the i-th lamina respectively), a correlation between f ni and f bi exists. How can we account for this correlation when calculating the strength of glulam, bearing in mind that it is very difficult to measure the correlation coefficients among MOR, σT, and σC? We developed a method by which these problems could be solved, and, using random variables generated by this method, the strengths of glulam were simulated. A low-swelling smectite exhibiting little intra-crystalline swelling even after saturation with Na was found in paddy soils derived from marine alluvium of Ariake Bay in Kyushu, Japan. The low-swelling smectite was considered to be beidellite-nontronite mineral containing as much as 10% Fe2O3. The low-swelling characteristics of this smectite are ascribed to the considerable substitution of Fe2+ for Al3+ in the ochtahedral layer which depresses the dissociation of unit layers of smectite. Transformation of low-swelling to high-swelling smectite is believed to be due to the oxidation of Fe2+ to Fe3+ in the octahedral layer. 相似文献

Glioma is the second most common intracranial neoplasia in dogs, but the pathogenic mechanisms remain unclear. In humans, isocitrate dehydrogenase 1 (IDH1) is frequently mutated in gliomas. Although almost all human IDH1 mutations have been identified as involving the Arg132 codon, few studies have reported structural, functional, and mutational information for canine IDH1. Therefore, in this study, we cloned the canine IDH1 homologue and used PCR mutagenesis to substitute the wildtype (WT) Arg132 with His (R132H) or Ser (R132S). WT and mutated IDH1 were overexpressed in HeLa cells, and their presence was confirmed by immunoblotting and immunocytochemistry using mutation-specific antibodies. The IDH1 activity between WT, R132H, and R132S transfectants was compared by measuring the production of NADH and NADPH. NADPH production in R132H and R132S transfectants was lower than that in WT, but NADH levels were not significantly different. Finally, we detected increased expression of hypoxia inducible factor 1 alpha (HIF-1α) in the R132H and R132S transfectants. These results indicated that the canine IDH1 Arg132 mutation has the potential to induce carcinogenesis in canine somatic cells. 相似文献

There are concerted efforts toward development of tick vaccines to replace current chemical control strategies that have serious limitations [Parasitologia 32 (1990) 145; Infectious Disease Clinics of North America (1999) 209-226]. In this study, monoclonal antibodies (mAbs) specific to Haemaphysalis longicornis midgut proteins were produced and characterized. Eight antibody-secreting hybridomas were cloned and the mAbs typed as IgG1, IgG2a and IgG2b. On immunoblots, all mAbs reacted with a midgut protein band of about 76 kDa. All mAbs uniformly immunogold-stained the surface or epithelial layers of H. longicornis midgut and endosomes. Adult ticks (50%) that fed on an ascitic mouse producing the IgGs developed a red coloration and did not oviposit. As such, the 76 kDa protein that reacted with the mAbs could, therefore, be a potential candidate for tick vaccine development. 相似文献

We aimed to clarify the genomic characteristics of porcine endogenous retroviruses (PERVs) in Vietnamese native pig (VnP) breeds. First, we investigated genetic polymorphisms in β‐ and γ‐like PERVs, and we then measured the copy numbers of infectious γ‐like PERVs (PERV‐A, B, and C). We purified genomic DNA from 15 VnP breeds from 12 regions all over the country and three Western pig breeds as controls, and investigated genetic polymorphisms in all known PERVs, including the beta (β)1–4 and gamma (γ)1–5 groups. PERVs of β1, β2, β3, and γ4 were highly polymorphic with VnP‐specific haplotypes. We did not identify genetic polymorphisms in β4, γ1, or γ2 PERVs. We then applied a real‐time polymerase chain reaction–based method to estimate copy numbers of the gag, pol, and env genes of γ1 PERVs (defined as A, B, and C). VnP breeds showed significantly lower copy number of the PERV genes compared with the Western pig breeds (on average, 16.2 and 35.7 copies, respectively, p < .05). Two VnP breeds showed significantly higher copy number compared with the other VnPs (p < .05). Our results elucidated that VnPs have specific haplotypes and a low copy number of PERV genes. 相似文献

Abstract The effects of exposing the taste receptors to two common aquaculture therapeutants, potassium permanganate (KMnO4) and formalin, on the gustatory responses in the Nile tilapia Oreochromis niloticus (Linnaeus) were studied using an electrophysiological approach. Electrical responses to the stimuli before and after perfusing the receptors with the toxicants were compared. Responses to 1.0 mM l ‐methionine (l ‐met) were significantly reduced when the receptors were perfused with 2.0 mg L?1 KMnO4 or 200.0 mg L?1 formalin. KMnO4 suppressed the responses to 56%, whereas formalin reduced them to 83%. Both toxicants reduced the responses to l ‐arginine to a similar extent to that of l ‐met, indicating that the action of the toxicants is not specific for the receptor sites for amino acids. On the other hand, the toxicants suppressed the responses to quinine significantly more than those to amino acids, which indicates that the transduction pathways for bitter substances are much more sensitive. The suppressions were reversible, suggesting that these toxicants do not cause any permanent damage to the physiological features of the taste receptors, but only interfere with their normal functioning. The possible mechanisms of suppression and their consequences on the feeding process are discussed. 相似文献

The spleen plays an important role in cellular immunity leading to parasite elimination, especially in hemo-protozoan parasite infection. In the present study, we quantified the serum IFN-gamma in splenectomized (SP) and non-splenectomized (NSP) cows infected with Theirelia sergenti (TS) to investigate whether the spleen plays a role in the Th1-type cytokine responses in cows following parasite infection. A transient increase in IFN-gamma was observed in the early stages of infection in the NSP cows, and the cows did not develop parasitemia. In contrast, the SP cows showed no IFN-gamma response at the early stage of infection, and the cows developed parasitemia following anemia. The NSP cows produced IgG1 and IgG2 antibodies to TS-specific, whereas the SP cows showed increases only in IgG1. Increased PBMC proliferation by cocanavalin A stimulation was observed concomitant with the IFN-gamma response. Pretreatment with IFN-gamma suppressed the propagation of TS in the SP cattle. These results suggest that the spleen plays an important role in the resistance to TS infection, leading to a Th1-type immune response. IFN-gamma might contribute to the activation of immunocytes and play an important role in the immune response to resist TS proliferation. 相似文献

Johne’s disease, caused by Mycobacterium avium subsp. paratuberculosis (MAP), is a chronic enteritis of ruminants. Previous studies have shown that programmed death-ligand 1 (PD-L1) is associated with the disease progression, and PD-L1 blockade activates MAP-specific Th1 responses in vitro. Here, we performed anti-PD-L1 antibody administration using 2 MAP-infected cattle at the late subclinical stage of infection. After administration, bacterial shedding was reduced or maintained at a low level. Additionally, MAP-specific Th1 cytokine production was upregulated, and CD69 expression was increased in T cells. Collectively, the treatment has a potential as a novel control method against Johne’s disease. 相似文献
