Native agars from Gracilaria vermiculophylla produced in sustainable aquaculture systems (IMTA) were extracted under conventional (TWE) and microwave (MAE) heating. The optimal extracts from both processes were compared in terms of their properties. The agars' structure was further investigated through Fourier transform infrared and NMR spectroscopy. Both samples showed a regular structure with an identical backbone, β-d-galactose (G) and 3,6-anhydro-α-l-galactose (LA) units; a considerable degree of methylation was found at C6 of the G units and, to a lesser extent, at C2 of the LA residues. The methylation degree in the G units was lower for MAE(opt) agar; the sulfate content was also reduced. MAE led to higher agar recoveries with drastic extraction time and solvent volume reductions. Two times lower values of [η] and M(v) obtained for the MAE(opt) sample indicate substantial depolymerization of the polysaccharide backbone; this was reflected in its gelling properties; yet it was clearly appropriate for commercial application in soft-texture food products.  相似文献

Traditional use of noncultivated vegetables has decreased with the development of agriculture and global supply chains. However, some species are still consumed as part of our traditional Mediterranean diet. Plants are among the most important sources of natural antioxidants for retarding lipid oxidative rancidity in foods or for pharmaceutical applications against chronic diseases related to free radicals production. The present study reports tocopherols composition and antioxidant activity of eight wild greens traditionally used in Spain. According to the edible part consumed, two groups were differentiated. Leafy vegetables whose young stems with leaves are consumed (Apium nodiflorum (L.) Lag., Foeniculum vulgare Mill., Montia fontana L. and Silene vulgaris (Moench) Garcke), and wild asparagus whose young shoots with leaf buds scarcely developed are eaten (Asparagus acutifolius L., Bryonia dioica Jacq., Humulus lupulus L. and Tamus communis L.). Among the leafy vegetables, Silene vulgaris and Apium nodiflorum presented the highest antioxidant capacity and antioxidants contents. Among the wild asparagus, the highest antioxidant capacity was obtained in Humulus lupulus.  相似文献

Ethanol is a principle ingredient of alcoholic beverages with potential neurotoxicity and carcinogenicity, and the ethanol-associated oxidative DNA damage in the central nervous system is well documented. The present work studied the possible protective effects of grape seed oligomer and polymer procyanidin fractions against ethanol-induced toxicity and compared these with resveratrol and other well-known antioxidants (ascorbic acid and vitamin E). By using the single cell gel electrophoresis (comet assay), a simple and sensitive technique for genotoxicity studies, the potential genotoxicity of acute and chronic ethanol administration in the different brain regions was investigated. Acute ethanol administration, at the dose of 2.5 or 5.0 g kg(-1) i.p., could induce significant DNA damage in cerebellum and hippocampus. Chronic administration of ethanol at the dose of 2.5 or 5.0 g kg-1 p.o. for 30 days could induce significant DNA damage in cerebellum, hippocampus, hypothalamus, and cortex, which could be auto-repaired at least 3 days after ethanol withdrawal. Oral administration of grape seed oligomer and polymer procyanidins and resveratrol (25, 50, and 100 mg kg(-1)) for 3 days before acute ethanol (5.0 g kg(-1), i.p.) or repeated administration of these substances together with ethanol (5.0 g kg(-1), p.o.) for 30 consecutive days could significantly inhibit DNA damage in brain cells induced by ethanol. As compared, ascorbic acid (50, 100, and 200 mg kg(-1)) and vitamin E (100, 200, and 400 mg kg(-1)) could also present protective effects on ethanol-induced DNA damage. Furthermore, the concentrations of ethanol and acetaldehyde in brain regions of the mice were detected by gas chromatography after administration of ethanol plus antioxidants. All of the results indicated that ethanol could induce region-specific oxidative DNA damage in which the cerebellum and hippocampus were more vulnerable, but intake of grape seed procyanidins or other natural antioxidants could protect the brain against ethanol-induced genotoxicity.  相似文献

One hundred and sixty-three wines from La Rioja, Utiel-Requena, and Tarragona were analyzed to determine if there were any differences in the concentrations of six biogenic amines that are found in these three regions. The influence of grape variety, type of vinification, wine pH, malolactic fermentation, and storage in bottle on biogenic amine concentrations was studied. Results show important differences in putrescine and histamine concentrations among regions, varieties of grape, and type of wine; differences were less appreciable for the remaining biogenic amines studied. Low pH prevented biogenic amine formation. Malolactic fermentation and short storage periods in bottle (3-6 months) showed increases in histamine concentration, whereas longer periods of storage led to a general decrease in histamine. Several strains of lactic acid bacteria were isolated in this work, and their ability to form biogenic amines was assayed in synthetic media, grape must, and wine. Grape varieties, different types of winemaking, pH, and lactic acid bacteria may be responsible for the differences observed in the biogenic amine concentrations of the wines analyzed.  相似文献

The effect of pH and hemoglobin on oxidation of the microsomal lipids of cod was determined in isolated microsomes and in washed cod muscle. An increase of hemoglobin concentration from 0.5 to 15 microM accelerated lipid oxidation in both systems. In cod microsomes the rate of lipid oxidation increased in the order pH 6.8 > pH 7.6 > pH 8.4 > pH 6.0 > pH 3.5. However, in washed cod muscle a decrease of pH from 7.8 to 6.8 greatly increased the lag phase and decreased the rate of lipid oxidation. A further decrease in pH to 3.5 decreased the lag phase and increased the rate of lipid oxidation further. A decrease of pH from 7.6 to 6.4 greatly reduced the affinity of hemoglobin for oxygen. Formation of methemoglobin due to autoxidation occurred more rapidly at pH 6.0 than at pH 7.5. Structural changes of the isolated microsomal membranes could be the reason for the unexpected slow lipid oxidation in microsomes at pH 6.0 and below.  相似文献

Purpose

Mining wastes may cause important environmental impacts in soil, water, and air due to their high metals and arsenic contents. The aim of this work was the assessment of the mobility of arsenic and several heavy metals in soils located near different types of tailing heaps in the town of Zimapán, México.

Materials and methods

One hundred twenty soil samples were collected nearby to three tailing heaps, one oxidized presenting a red color (RT), and two with gray wastes (OSM and NSM) but with different age, during the dry and rainy seasons at the surface and to 40 cm depth, as well as to different distances from the deposits. Arsenic, Cd, Cu, Fe, Mn, Pb, V, and Zn total concentrations were determined; in addition, geochemical phase distribution of As, Cu, Mn, and Zn in selected samples was determined by sequential extraction. Concentrations were measured by graphite furnace atomic absorption spectrometry and inductively coupled plasma. To interpret the results, statistical analyses were performed.

Results and discussion

All samples presented high As concentrations reaching more than 50,000 mg kg?1 close to OSM tailings, although the highest concentrations in the available fractions were measured in NSM impacted soils. Arsenic and metals concentrations exceeded the screening limits recommended for industrial sites. In samples influenced by OSM tailings, most of the elements analyzed were in the residual fraction, whereas in NSM and RT they were mostly in the organic and sulfide fractions and in the Fe and Mn oxides fractions, respectively. Larger concentrations of As and metals than those allowed by the screening values in Canada and the Netherlands were measured in the residential area representing a health threat for the inhabitants and the environment.

Conclusions

Acid mine drainage, water, and wind erosion of tailings have polluted nearby soils. Higher concentrations of As and metals were measured during the rainy season in gray tailings impacted soils and during the dry season in red tailings, showing both deposit types’ different mobility. Elements fractionation in soils depends mainly on tailings characteristics. Low metals and As proportions were found in the fraction with the highest mobility. Metals and arsenic are more stable in soils impacted by gray tailings, mainly in the organic and sulfides and residual fractions, while in RT, most are linked to Fe and Mn oxyhydroxides. Polluted soils in residential areas constitute a health hazard. Remedial actions must be taken to stop the population exposure.

The effects of hyperoxygenation on Chardonnay white musts and the influence of subsequent storage on the corresponding wines have been evaluated. Attention was focused on the color characteristics, phenolic and volatile composition, and sensorial analysis, not previously reported in conjunction. On the one hand, the hyperoxygenation treatment provoked a significant decrease in the concentration of virtually all phenolic compounds in musts, young wines, and one-year-stored wines. In addition, a higher resistance to browning was observed in stored wines derived from hyperoxygenated musts. Different storage conditions (light and dark) produced significant differences in the 2-S-glutathionylcaftaric acid derivatives amounts. On the other hand, significant differences were observed in the volatile composition of wines due to the hyperoxygenation treatment, such as a decrease in the isoamylic alcohols concentration, acetaldehyde, and β-damascenone, even after storage under different conditions. Finally, Chardonnay white wines derived from hyperoxygenated musts had higher banana odor and lower herbaceous and flowery notes.  相似文献

Benzoic acid-derived phenolics (p-hydroxybenzoic acid, 3,4-dihydroxybenzoic acid, vanillic acid, syringic acid, and gallic acid) and the polyphenols epicatechin and epigallocatechin gallate (EGCG) were evaluated for their efficiency in regenerating alpha-tocopherol from alpha-tocopheroxyl radical in comparison with ascorbyl palmitate, which is known to regenerate alpha-tocopherol. Ethanolic solutions of phenolic compounds were added to a homogeneous hexane medium containing alpha-tocopheroxyl radical generated by reaction of alpha-tocopherol in molar excess with 1,1-diphenyl-2-picrylhydrazyl radical, and the alpha-tocopheroxyl radical was monitored by electron spin resonance spectroscopy. p-Hydroxybenzoic acid, vanillic acid, and syringic acid (400 microM) did not exhibit a significant effect on alpha-tocopheroxyl radical concentration (0.6-0.7 microM). In contrast, 3,4-dihydroxybenzoic acid and gallic acid were able to reduce the concentration of alpha-tocopheroxyl radical by 16 and 64%, respectively. Epicatechin showed a reduction of alpha-tocopheroxyl radical similar to gallic acid, and EGCG and ascorbyl palmitate were the most effective, reducing alpha-tocopheroxyl radical completely at a much lower phenolic concentration (66.7 microM). The moles of alpha-tocopheroxyl radical reduced per mole of ascorbyl palmitate (0.93), EGCG (0.066), gallic acid (4.3 x 10(-4)), and epicatechin (4.5 x 10(-4)) were determined, and the logarithm of these stoichoimetric ratios showed a negative linear relationship with the bond dissociation enthalpies of the O-H bond of the phenolics. The relative capacity to reduce alpha-tocopheroxyl radical was found to be ascorbyl palmitate (2142) > EGCG (151) > gallic acid (1) approximately epicatechin (1).  相似文献

The effects of processing and cooking practices on the chemical composition and antioxidant activity of Portuguese wild edible mushroom species (Lactarius deliciosus, Macrolepiota mastoidea, Macrolepiota procera, and Sarcodon imbricatus) were investigated. Dried, frozen, and cooked samples were analyzed for proximate constituents (moisture, fat, crude protein, ash, and carbohydrates) and nutritional value. Fatty acid and sugar profiles were also obtained by gas-liquid chromatography/flame ionization detection and high-performance liquid chromatography/refraction index, respectively. The antioxidant properties were evaluated by several biochemical assays: 2,2-diphenyl-1-picrylhydrazyl radical scavenging capacity, reducing power, inhibition of beta-carotene bleaching, and inhibition of lipid peroxidation in brain tissue using thiobarbituric acid reactive substances. Results of this study show that mushroom species and processing and cooking practices are all effective determinants for either chemical composition or antioxidant properties. Cooked samples proved to have lower nutrient concentrations and lower antioxidant activities than either dried or frozen samples. In what concerns fatty acids and sugar individual profiles, only cooking proved to be relevant: The cooked samples presented higher monounsaturated fatty acid and lower polyunsaturated fatty acid and sugars contents.  相似文献

Purpose

The use of municipal solid wastes (MSWs) as a low-cost source of organic matter for soils should be considered after discarding the environmental risks related to their metal(loid) load. The goal of this work was to assess the employment of a MSW as an organic amendment in two types of soil (an agricultural soil, A, and a metal(loid)-enriched mine tailings soil, T) attending to changes in soil properties and in plant growth, nutrition and metal(loid) translocation from roots to aerial parts of Zea mays L. (stalk, leaves, tassel, husk, cob and kernel).

Materials and methods

After a comprehensive characterisation of each soil treatment (A, A + MSW, T, T + MSW), a pot-designed experiment was carried out. Soil solution was monthly monitored throughout the experiment, and metal(loid) concentrations were measured.

Results and discussion

The MSW improved some fertility-related parameters in both soils, A and T: increased total and dissolved organic carbon, total nitrogen and soil microbiology. However, an increase in 0.01 M CaCl2-extractable metal(loid) concentration was also observed. No differences in dry biomass were found between amended and not amended treatments. A fractionation of metal(loid) concentrations among plant organs occurred. For instance, the highest Cu and Pb concentrations were found in roots, while for Zn occurred in the stalk and the cob. The amended treatments favoured the accumulation of Mn in all plant organs. Kernels showed in general the lowest metal(loid) concentrations.

Conclusions

The addition of municipal solid wastes as organic amendment could be a suitable tool to increase soil fertility. However, due to the high metal(loid) content of this particular MSW, its use on agricultural soils would not be appropriate. By other hand, along with the improvement of soil fertility, the MSW was useful to promote plant development in the mine tailings soil which should be then considered as a potential tool to promote plant establishment in those metal(loid)-impacted soils.
