Turkeys given cell-free culture filtrate (CCF) of Pasteurella multocida strain R44/6 orally, via air sacs, or subcutaneously mixed 1:1 with incomplete Freund's adjuvant (IFA) at 6 and 9.5 weeks of age were compared with negative controls given bacteriologic medium and positive controls vaccinated with a commercial bacterin. At 13 weeks of age, serum antibody titers to P. multocida were detectable only in turkeys given CCF in IFA (low titers) and positive control turkeys (high titers), at which time turkeys were challenged orally with either the homologous strain or strain P-1059. Protection against challenge with strain R44/6 was provided by the commercial bacterin, CCF in IFA, and CCF given via air sacs. When turkeys were challenged with strain P-1059, protection was superior in turkeys given CCF via air sacs, intermediate in turkeys given commercial bacterin or CCF in IFA, and absent in negative control turkeys and turkeys given CCF orally. These results indicate CCF is an effective immunogen when administered via the lower respiratory tract for protecting turkeys against pasteurellosis.  相似文献

Abstract. Ammonia volatilization with and without gypsum incorporation was measured in Gujranwala soil (Udic Haplustalf) in an incubation study using different nitrogen fertilizers e.g. urea, ammonium sulphate (AS), calcium ammonium nitrate (CAN), and urea nitrophos (UNP). Nitrogen from different fertilizers was applied at the rate of 200 mg N kg−1 to two sets of soils in plastic bags (1.0 kg soil) and plastic jars (0.5 kg soil). Soil moisture was maintained at field capacity. Application of urea increased soil pH to 9, three hours after its addition. Ammonium sulphate and calcium ammonium nitrate had little effect on soil pH. Ammonium volatilization losses from fertilizers were related to the increase in soil pH caused by the fertilizers. Consequently maximum losses were recorded due to application of urea. Losses through ammonia volatilization were significantly lower with AS, CAN and UNP in descending order. Gypsum incorporation significantly reduced the losses. Therefore, application of gypsum to soil before urea may substantially improve N use efficiency for crop production by reducing N losses.  相似文献

Mushrooms (Lentinus edodes) were processed for hot water (HWE), methanolic (ME), and polysaccharide (PSE) extracts. Polysaccharides were isolated through ion exchange (DEAE cellulose) and size exclusion (Sephadex G-100) chromatography. Monosaccharides including maltose (0.282%), glucose (0.113%), and mannose (0.451%) were identified, qualitatively and quantitatively, from the isolated polysaccharides through high-performance liquid chromatography. The whole study was divided into two experiments. Experiment 1 was meant for the evaluation of HWE and ME; whereas, experiment 2 was meant for the evaluation of PSE for immunostimulatory and immunotherapeutic activities. The cellular and humoral immune responses were demonstrated through lymphoproliferative response to phytohemagglutinin-P (PHA-P) and anti-body response to sheep red blood cells (SRBCs), respectively. The immunotherapeutic effects of these extracts were demonstrated against eimeriasis in terms of lesion scoring, oocysts per gram of droppings, and percent protection. Cell-mediated immune responses observed at 24, 48, and 72 h post-PHA-P injection were significantly higher (P < 0.05) in chickens administered with any of the three extracts (PSE, ME, and HWE), when compared with the controls. Humoral immune response in terms of anti-SRBCs anti-body titers was also observed higher in chickens administered with mushroom extracts. In the challenge experiment, significantly higher (P < 0.05) OPG and lesion scores were observed in controls as compared to the groups administered with mushroom extracts (HWE, ME, and PSE). Significantly higher (P < 0.05) percent protection against eimeriasis was observed in all groups administered with different extracts of L. edodes as compared to controls. In conclusion, L. edodes extracts showed immunostimulatory potential which persisted against eimeriasis in chicken.  相似文献

The aim of the current study was to evaluate the effect of dietary supplementation of Pinus ponderosa leaves ( pine leaves) and α‐tocopherol acetate (vitamin E) powder on male reproductive system, serum metabolites and carcass characteristics of Japanese quails. A total of 360‐day‐old male quails were purchased from the open market and kept at poultry shed for ninety‐four days. After ten days of adaptation, all quails were randomly assigned into 4 groups, control (IC); supplemented with α‐tocopherol acetate (IE) at the rate of 150 mg/L; Pinus ponderosa leaves (IP) at the rate of 150 mg/L; and 70 mg α‐tocopherol acetate and 70 mg Pinus ponderosa leaves (IEP). Pinus ponderosa leaves and α‐tocopherol acetate supplementation had not significantly (p > .05) effected on final body weight gain, feed intake and feed conversion ratio of quails. The high‐density lipoprotein cholesterol (HDLC) and total cholesterol (TC) were significantly (p > .05) affected by IE and IP groups as compared to IC and IEP groups. Triglyceride (TG), glutathione peroxidase (GPx) and superoxide dismutase (SOD) significantly (p < .05) increased in all treatment groups except for the IC group. Aspartate transaminase (AST) significantly (p > .05) decreased in treatment groups as compared to control group. Overall, the mineral levels significantly (p < .05) increased in treatment groups as compared to control. Cloacal gland index values, the quantity of foam production and testis weight were significantly (p < .05) increased in treatment groups. It was concluded that the supplementation of Pinus ponderosa leaves and α‐tocopherol acetate improved the testis weight, foam production, serum antioxidant enzymes and mineral level especially zinc in Japanese quail considered an indicative characteristic of higher sperm production rate and improved sexual activity. Further, higher gametogenesis rate, sperm production or reproductive behaviour including different hormonal level will be analysed in future study.  相似文献

Soil compaction affects plant growth by causing increased resistance to root penetration and a decreased uptake of water and ions. A pot experiment was conducted to study the effect of soil compaction in conjunction with the soil salinity and waterlogging on root growth and leaf ionic composition of two wheat genotypes (Aqaab and MH-97). Compaction was achieved at a 10% soil moisture content by dropping 5 kg weight, controlled by a tripod stand for 20 times from 0.6 m height on a wooden block placed inside the soil-filled pots. Soil bulk density of non-compact and compact treatments was measured as 1.21 and 1.65 Mg m−3, respectively. The desired salinity level (15 dS m−1) was developed by mixing required amount of NaCl in the soil before filling the pots. Waterlogging was developed by flooding the pots for 21 days both at tillering and booting stages. Compaction significantly reduced the root length density (RLD) of both the wheat genotypes while the combined effect of compaction×salinity was more drastic on root length density than compaction alone. Waterlogging however, did not decrease the root length density, rather it mitigated the effect of compaction. Compaction decreased the concentration of K+ and K+:Na+ ratio in leaves. Salinity also decreased the concentration of K+ and K+:Na+ ratio, but increased the concentrations of Na+ and Cl in the crop leaves. Salinity and compaction interacted to cause a greater reduction in K+ concentration and the K+:Na+ ratio, while there was lesser increase in the concentrations of Na+ and Cl compared with salinity alone. Waterlogging also decreased the concentration of K+ and K+:Na+ ratio in leaves. It intensified the effect of salinity but decreased the effect of compaction on leaf ionic composition. Therefore, the effect of compaction on root growth and ion uptake is more severe under salt-affected soil conditions than under normal soil conditions while occasional waterlogging of a compact soil for a few days makes the soil conditions favorable for root growth both under non-saline as well as saline soil conditions. Also, the performance of a genotype in stressed environment is related to maintenance of higher root length density, leaf K+ concentration and K+:Na+ ratio and lower leaf Na+ and Cl concentrations.  相似文献

The research was focused on the synthesis and characterization of zinc (Zn)-coated urea fertilizer. Zinc oxide (ZnO) particles were coated on urea granules through two different coating techniques, i.e., slurry coating and wet coating, and the release rate of the coated Zn was measured up to 24 hours. Slurry coating technique showed better Zn release rate and morphology of coated urea granules as compared with wet coating technique using different binders, i.e., water, molasses, paraffin oil, and the combined mixture of honey bee wax and gum arabica. Urea granules synthesized through wet coating technique have a low amount of Zn as compared with slurry-coated urea granules. Urea granules coated through slurry coating technique and molasses, paraffin oil, and water (MPW) as a binder were the best in term of coating efficiency, uniform size distribution of ZnO-coated particles, granule's morphology, and release rate of Zn from coated formulations.  相似文献

The onset of pregnancy may be associated with hormonal changes and decline in milk yield of buffaloes. To investigate this, forty lactating buffaloes from 1st to 23rd weeks post-conception were selected. The animals were assigned to three treatments: PRT, PRS, NPRT and three milk yielding groups HMY, 66 to 75 l/week, n = 12; MMY, 56 to 65 l/week, n = 16; LMY, 46 to 55 l/week, n = 12). Milk samples were collected on alternate weeks and analyzed with ultrasonic milk analyzer. EIA was used for milk progesterone concentrations. Group means were compared and correlation analysis was conducted. Progesterone concentrations increased in almost similar pattern with the advancing weeks post-conception. The high and low yielder showed greater progesterone concentrations in the supplemented than the animals on traditional ration (P < 0.001). Progesterone concentrations correlated positively with fat (%), negatively with milk yield, protein (%) and lactose (%). Decline in milk yield became drastic when progesterone concentrations rose above 6.44 ng/ml. The pregnant animals on traditional ration exhibited a sharper decline in milk yield with the increasing progesterone concentrations as compared to pregnant animals with supplemented ration. It is concluded that concentrates supplementation induced a raise in progesterone levels. Progesterone concentrations and milk yield showed an inverse relationship.  相似文献

The protein profiles and phagocytic ability of Sephadex-elicited chicken peritoneal macrophages were examined following heat-shock exposure. Macrophage cultures were exposed to various temperatures, time exposures and recovery periods. Densitometric analysis of SDS-PAGE autoradiographs revealed that heat-induced macrophages synthesized three major (23, 70 and 90 kD) heat-shock proteins (HSPs). The optimal temperature and time for induction of these HSPs was 45-46 degrees C for 1 h, with a variable recovery period for each HSP. Macrophages exposed to 45 degrees C for 30 and 60 min were significantly depressed in phagocytosis of uncoated sheep erythrocytes (SE) under 45 degrees C incubation conditions. However, phagocytosis of antibody-coated SE was not affected when compared to 41 degrees C control cultures. Macrophages allowed to recover at 41 degrees C following heat-shock exhibited no alterations in their phagocytic ability for either antibody-coated or uncoated SE. This study suggests that heat shock induces three major HSPs in chicken peritoneal macrophages in addition to maintaining their Fc-mediated phagocytic function while significantly depressing their nonspecific phagocytosis.  相似文献

Background: Forests are fundamental in maintaining water supplies, providing economic goods, mitigating climate change, and maintaining biodiversity, thus providing many of the world’s poorest with income, food and medicine.Too often, forested lands are treated as "wastelands" or "free" and are easily cleared for agricultural and infrastructure expansion.Methods: In this paper, the sustainability of two forest ecosystems(state and community/private owned) was evaluated using SPOT-5 satellite images of 2005 and 2011. This study was conducted in a sub-watershed area covering 468 km2, of which 201 km2 is managed by the state and 267 km2 by community/private ownership in the Murree Galliat region of Punjab Province of Pakistan. A participatory approach was adopted for the delineation and demarcation of forest boundaries. The Geographic Object-Based Image Analysis(GEOBIA) technique was used for identification and mapping of ten Land Cover(LC) features.Results: The results show that between the years 2005 to 2011, a total of 55 km2(24 km2 in state-owned forest and 31km2 in community/private forest) was converted from forest to non-forest. The conclusion is that conservation is more effective in state-owned forests than in the community/private forests.Conclusions: These findings may help to mobilize community awareness and identify effective initiatives for improved management of community/private forest land for other regions of Pakistan.  相似文献
