A male bovine linkage map was constructed using six half-sib families of the Norwegian Cattle population. The family material consisted of six sires and 285 sons, with the number of sons per sire ranging from 32 to 71. Of the 288 markers analysed, 261 were anonymous microsatellites and 27 were coding genes. Eleven of the coding genes were mapped by a two-step procedure involving intron/exon sequencing in the six sires and development of polymerase chain reaction-based methods for efficient genotyping in heterozygous families. Linkage analysis including both informative and non-informative sons was performed in order to obtain unbiased estimates of recombination fractions. The autosomal genome length of the Norwegian Cattle Map (NCM) was estimated as 2682 cM, with an average interval of 12.5 cM. The map provides sufficient marker density for scanning the whole bovine genome to locate quantitative trait loci. The large number of common markers between NCM and previously published maps will contribute to the comparison and integration of bovine maps.  相似文献

OBJECTIVE: Food-based dietary guidelines (FBDGs) are globally promoted as an important part of national food and nutrition policies. They are presented within policy as key features of the strategy to educate the public and guide policy-makers and other stakeholders about a healthy diet. This paper examines the implementation of FBDGs in four countries: Chile, Germany, New Zealand and South Africa - diverse countries chosen to explore the realities of the FBDG within policy on public health nutrition. DESIGN: A literature review was carried out, followed by interviews with representatives from the governmental, academic and private sector in all four countries. RESULTS: In all four countries the FBDG is mainly implemented via written/electronic information provided to the public through the health and/or education sector. Data about the impact of FBDGs on policy and consumers' food choice or dietary habits are incomplete; nutrition surveys do not enable assessment of how effective FBDGs are as a factor in dietary or behavioural change. Despite limitations, FBDGs are seen as being valuable by key stakeholders. CONCLUSION: FBDGs are being implemented and there is experience which should be built upon. The policy focus needs to move beyond merely disseminating FBDGs. They should be part of a wider public health nutrition strategy involving multiple sectors and policy levels. Improvements in the implementation of FBDGs are crucial given the present epidemic of chronic, non-communicable diseases.  相似文献

With this topical issue, we present the work of the Priority Program 1090 of the German Research Foundation (“Deutsche Forschungsgemeinschaft DFG”): “Soils as a source and sink for CO2 – mechanisms and regulation of organic matter stabilisation in soils”. This introduction gives an overview on the sites investigated and the major research approaches, including a glossary of major terms used in the field of soil organic matter research. We point out the advantages of integration of data from a broad field of different soil‐science disciplines and the progress achieved by application and combination of new analytical methods describing the quality and turnover of soil organic matter.  相似文献

Background

Campylobacter is the most commonly reported bacterial cause of enteritis in humans in the EU Member States and other industrialized countries. One significant source of infection is broilers and consumption of undercooked broiler meat. Campylobacter jejuni is the Campylobacter sp. predominantly found in infected humans and colonized broilers. Sequence analysis of the 16S rRNA gene is very useful for identification of bacteria to genus and species level. The objectives in this study were to determine the degree of intraspecific variation in the 16S rRNA genes of C. jejuni and C. coli and to determine whether the 16S rRNA sequence types correlated with genotypes generated by PFGE analysis of SmaI restricted genomic DNA of the strains.

Methods

The 16S rRNA genes of 45 strains of C. jejuni and two C. coli strains isolated from broilers were sequenced and compared with 16S rRNA sequences retrieved from the Ribosomal Database Project or GenBank. The strains were also genotyped by PFGE after digestion with SmaI.

Results

Sequence analyses of the 16S rRNA genes revealed nine sequence types of the Campylobacter strains and the similarities between the different sequence types were in the range 99.6–99.9%. The number of nucleotide substitutions varied between one and six among the nine 16S rRNA sequence types. One of the nine 16S rRNA sequence profiles was common to 12 of the strains from our study and two of these were identified as Campylobacter coli by PCR/REA. The other 10 strains were identified as Campylobacter jejuni. Five of the nine sequence types were also found among the Campylobacter sequences deposited in GenBank. The three 16S rRNA genes in the analysed strains were identical within each individual strain for all 47 strains.

Conclusion

C. jejuni and C. coli seem to lack polymorphisms in their 16S rRNA gene, but phylogenetic analysis based on 16S rRNA sequences was not always sufficient for differentiation between C. jejuni and C. coli. The strains were grouped in two major clusters according to 16S rRNA, one cluster with only C. jejuni and the other with both C. jejuni and C. coli. Genotyping of the 47 strains by PFGE after digestion with SmaI resulted in 22 subtypes. A potential correlation was found between the SmaI profiles and the 16S rRNA sequences, as a certain SmaI type only appeared in one of the two major phylogenetic groups.  相似文献

The aim of this experiment was to evaluate the significance of neonatal environment on feed efficiency. For that purpose, rabbits from a line selected for residual feed intake (RFI) during 10 generations (G10 kits) were cross‐fostered with non‐selected control does (i.e., G0 line), and reciprocally. In parallel, sibs were fostered by mothers from their original line. Nine hundred animals were raised in individual (N = 456) or collective (N = 320) cages. Traits analysed in this study were body weight at 32 days and at 63 days, average daily gain (ADG), feed intake between weaning and 63 days (FI), feed conversion ratio (FCR) and RFI. The maternal environment offered by does from the line selected for RFI deteriorated the FCR of the kits, independently of their line of origin, during fattening (+0.08 ± 0.02) compared to FCR of kits nursed by G0 does. The line, the type of housing and the batch were significant effects for all the measured traits: G10 kits were lighter than their G0 counterparts at 32 days (?82.9 ± 9 g, p < 0.0001) and at 63 days (?161 ± 16 g, p < 0.0001). They also had a lower ADG (?2.36 ± 0.36 g/day, p < 0.0001), RFI (?521 ± 24 g/day, p < 0.0001) and a lower FI (?855 ± 31 g, p < 0.0001), resulting in a more desirable feed efficiency (FCR: ?0.35 ± 0.02). There was no significant difference in the contrast of G10 and G0 performances between collective and individual/digestive cages (p > 0.22): ?2.35 g/day versus 2.94 g/day for ADG, ?0.39 versus ?0.40 for FCR, ?577 g versus ?565 g for RFI and ?879 g versus ?859 g for FI, respectively). Thus, no genotype‐by‐environment (housing) interaction is expected at the commercial level, that is, no re‐ranking of the animals due to collective housing.  相似文献

Objectives— To reclassify canine small intestinal and cecal leiomyoma (LM) and leiomyosarcoma (LMS) into smooth muscle and gastrointestinal stromal tumors (GIST) using histologic and immunohistochemical (IH) analysis and to report clinical findings and survival data.
Study Design— Retrospective review of cases.
Animals— Dogs (n=47) with small intestinal (40 LMS; 7 LM) and 25 dogs with cecal tumors (23 LMS; 2 LM).
Methods— Clinical and survival data were reviewed. Tissue sections were reevaluated for light-microscopic malignancy criteria and examined for expression of SMA, desmin, vimentin, S-100, and CD117 (KIT) by immunohistochemistry.
Results— Reclassification resulted in 2 LM, 9 LMS, 19 GIST, and 17 GIST-like tumors in the small intestine and 23 GIST and 2 GIST-like tumors in the cecum. GIST-like tumors were morphologic and IH identical to GIST but lacked KIT expression. No significant difference in survival was observed for tumor type, location, histologic, or IH characteristics; however, dogs with cecal tumors were significantly older in age, presented more commonly with intestinal perforation and peritonitis, and less commonly with weight loss. Cecal tumors had more histologic malignancy criteria than small intestinal tumors. After excision, 1 and 2 year recurrence-free periods were 80.1% and 67.2% for small intestinal and 83.3% and 61.9% for cecal tumors.
Conclusion— Prognosis for intestinal tumors with histologic smooth muscle appearance is good after excision and not related to tumor type, location, histologic, or IH characteristics.
Clinical Relevance— Clinical importance could not be demonstrated for reclassification, but may be for future treatment, of intestinal smooth muscle or stromal tumors.  相似文献

There is broad consensus that the main problem facing fisheries globally is too many boats chasing too few fish. Unfortunately it is also possible to argue that there are too many proposed solutions and not enough practical answers to improving fisheries management. There is a deepening divide between those who propose alternative regulatory controls on fishers, including establishing large areas permanently closed to fishing, and those who argue for better alignment of incentives combined with broad participation of resource users in fishery management decisions (in simple terms, between top down and bottom up systems of governance). However despite the choice of policy instruments used, a consistent outcome is that resource users behave in a manner that is often unintended by the designers of the management system. Hence whilst uncertainty is broadly recognized as a pervasive feature of fisheries management, to date most of the attention has focussed on only part of that uncertainty – scientific uncertainty about the status of exploited resources. The effect of uncertainty generated on the human side of fisheries science and management has received much less attention. However, the uncertainty generated by unexpected resource user behaviour is critical as it has unplanned consequences and leads to unintended management outcomes. Using empirical evidence of unexpected resource user behaviour and reviewing current responses to unexpected management outcomes, we identify different approaches that both improve prediction of human behaviour in fisheries systems and identify management measures that are more robust to these sources of uncertainty. However, unless the micro scale drivers of human behaviour that contribute to macro scale implementation uncertainty are communicated effectively to managers and considered more regularly and in greater depth, unanticipated responses to management actions will continue to undermine management systems and threaten the sustainability of fisheries.  相似文献

One approach to ongrow juvenile European lobster, Homarus gammarus, is to utilize land based rearing systems, incorporating automated feeding, individual culture and provision of stable pelleted feeds, preferably using sustainable ingredients. We initiated three feeding experiments to investigate the general suitability of ingredients produced from seafood by‐products as novel feeds for H. gammarus, in terms of promoting survival, development and growth of post‐larval lobsters from post‐larvae (PL) stage IV to the first juvenile stage (stage V). The first experiment was designed to screen an array of candidate, locally produced, novel protein sources on growth performance parameters. This initial experiment revealed that PL reared on a raw (i.e. wet, unprocessed shrimp) feed used as a reference showed superior performance to those reared on experimental feeds containing fishmeal, herring protein isolate or mussel meal; however, a novel type of shrimp meal, produced by flocculation from waste water, promoted the best PL performance of any experimental feed. A second experiment was designed to test the effect of drying method and to optimize the form of a wet shrimp reference feed used by lobster hatcheries. This showed that the performance of PL reared on experimental freeze‐dried shrimp feed was not significantly different to those reared on the wet, unprocessed shrimp used as a reference feed. However, lobsters offered experimental oven‐dried shrimp feed (with or without an immune supplement) resulted in significantly lower survival or growth performance. A third and final experiment was designed in an attempt to improve a candidate herring‐based protein source, by supplementing with nutrients found in shrimp. However, the results showed that PL reared on the wet reference shrimp feed still showed superior growth and survival than those reared on a herring feed alone, or supplemented with additives found in shrimp meal (either glucosamine, astaxanthin or both supplements combined). The high survival and growth, low incidence of moulting problems and high availability of waste shrimp material, suggest that non‐heat‐treated shrimp products are a promising feed ingredient for post‐larval European lobsters.  相似文献
