Minipigs are widely used to examine physiologic mechanisms under experimental settings. The purpose of our study was to localize the visual cortex of minipigs using visual stimulation in functional magnetic resonance imaging (fMRI). Five male G?ttinger minipigs were studied with fMRI during visual stimulation with series of light impulses conducted into the MR cabin via fibre optical wave guides (EPI sequences, block design, 1.5 T clinical routine MR-scanner). Statistics were performed using SPM'99. All minipigs showed activation of the visual cortex due to light stimulation. The activation could be superimposed to T2-weighted structural images with good spatial resolution revealing the exact location of the visual cortex. We were able to localize the visual cortex in minipigs for the first time utilizing fMRI on a routine scanner. However, this may be useful for precise positioning of electrodes or implants or be used as a further uncomplicated non-invasive method in physiologic experiments. 相似文献

This study investigated the hypothesis that supplementation of methionine (Met) to broiler diets increases muscle growth due to regulation of molecular pathways related to protein synthesis and degradation depending on the Met source. Day‐old male Cobb‐500 broilers (n = 240) were phase‐fed three different wheat–soya bean meal‐based basal diets during days 1–10, 11–21 and 22–35. Basal diets (Met‐ group, Met + Cys concentration 15% below NRC recommendations) were supplemented with 0.10% or 0.40% Met either as DL‐Met (DLM) or DL‐2‐hydroxy‐4‐(methylthio) butanoic acid (DL‐HMTBA) (equimolar comparison). Breast muscle weights were lower in the Met‐ group compared to all Met‐supplemented groups and were lower in broilers supplemented with 0.10% of DL‐HMTBA compared to the other groups fed Met‐supplemented diets. However, the expression of genes or relative phosphorylation and thus activation state of proteins involved in the somatotropic axis, the mammalian target of rapamycin (mTOR) pathway of protein synthesis, the ubiquitin–proteasome pathway (UPP) and autophagy–lysosomal pathway of protein degradation, the GCN2/eIF2a pathway involved in the inhibition of protein synthesis and in the myostatin–Smad2/3 pathway involved in myogenesis were not affected by Met source. Feeding diets with suboptimum Met + Cys concentrations, however, decreased expression of GHR and IGF1 in liver and muscle and increased that of MURF1 involved in the UPP in the broiler's muscle at day 10 and 21, while that of FOXO and atrogin‐1 and FOXO phosphorylation remained unaffected. Additionally, suboptimum dietary Met concentrations increased expression of the autophagy‐related genes ATG5 and BECN1 at day 35. Met supplementation neither affected gene expression nor phosphorylation of proteins involved in the GNC2/eIF2a and mTOR pathways. These data indicate that protein synthesis was not affected on the molecular level, while protein degradation was marginally affected by dietary Met dosage. 相似文献

The stability of lycopene in an olive oil/tomato emulsion during thermal processing (80-140 °C) was studied. Initially, the degradation of total lycopene (all-E plus Z-forms) occurred quickly at temperatures above 100 °C. However, a nonzero plateau value, depending on the processing temperature, was attained after longer treatment times. Besides degradation, the isomerization of total-Z-lycopene as well as the individual isomerization of all-E-, 5-Z-, 9-Z-, and 13-Z-lycopene was studied in detail. After prolonged heating, the isomer conversion reached a temperature-dependent equilibrium state. The degradation of total lycopene and the isomerization could be described by a fractional conversion model. The temperature dependency of the corresponding reaction rate constants was quantified by the Arrhenius equation. The activation energy of degradation was estimated to be 28 kJ/mol, and the activation energy of overall (all-E and total-Z) isomerization was estimated to be 52 kJ/mol. 相似文献

Serum samples from 4,907 cows from 93 dairy and five beef herds were analyzed for antibodies to Neospora caninum by commercially available ELISA. The herds were recruited from 20 states and the territory of Puerto Rico. A questionnaire assessed the herd's abortion history and exposure risks to N. caninum. Results showed that 788 (16%) cows tested positive for antibodies to N. caninum. At least one positive animal was detected in 90% of the herds tested. The prevalence of cows seropositive to N. caninum varied from 2% to 65% among herds. Abortion rates ranging from 2% to 19% were reported. The epidemiologic data collected from the questionnaire suggested that the presence of farm dogs does not increase the risk of exposure of cows to N. caninum. 相似文献

Sustainable agricultural land use requires an assessment of degradable soil organic matter (SOM) because of its key function for soil fertility and plant nutrition. Such an assessment for practical land use should consider transformation processes of SOM and its sources of different origin. In this study, we combined a 120‐day incubation experiment with thermal decay dynamics of agricultural soils altered by added organic amendments. The aim was to determine the abilities and limits of thermal analysis as a rapid approach revealing differences in the degradability of SOM. The carried out experiments based on two independent sampling sets. The first sample set consisted of soil samples taken from non‐fertilized plots of three German long‐term agricultural field experiments (LTAEs), then artificially mixed with straw, farmyard manure, sheep faeces, and charcoal equal to 60 Mg ha?1 under laboratory conditions. The second sample set based on soil samples of different treatments (e.g., crop type, fertilization, cultivation) in LTAEs at Bad Lauchstädt and Müncheberg, Germany. Before and after the incubation experiment, thermal mass losses (TML) at selected temperatures were determined by thermogravimetry indicating the degradability of organic amendments mixed in soils. The results confirmed different microbial degradability of organic amendments and SOM under laboratory conditions. Thermal decay dynamics revealed incubation‐induced changes in the artificial soil mixtures primarily at TML around 300°C in the case of applied straw and sheep faeces, whereas farmyard manure showed mainly changes in TML around 450°C. Charcoal did not show significant degradation during incubation, which was confirmed by TML. Detailed analyses of the artificial soil mixtures revealed close correlations between CO2‐C evolution during incubation and changes in TML at 300°C with R2 > 0.96. Results of the soils from LTAEs showed similar incubation‐induced changes in thermal decay dynamics for fresh plant residues and farmyard manure. We conclude that the practical assessment of SOM could be facilitated by thermal decay dynamics if modified sample preparation and evaluation algorithms are used beyond traditional peak analysis. 相似文献

The ever-increasing interest in keeping a young appearance and healthy skin has leveraged the skincare industry. This, coupled together with the increased concern regarding the safety of synthetic products, has boosted the demand for new and safer natural ingredients. Accordingly, the aim of this study was to evaluate the dermatological potential of the brown seaweed Carpomitra costata. The antioxidant, anti-enzymatic, antimicrobial, photoprotective and anti-inflammatory properties of five C. costata fractions (F1–F5) were evaluated. The ethyl acetate fraction (F3) demonstrated the most promising results, with the best ability to scavenge 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals (EC50 of 140.1 µg/mL) and the capacity to reduce reactive oxygen species (ROS) production promoted by UVA and UVB radiation in 3T3 cells, revealing its antioxidant and photoprotective potential. This fraction also exhibited the highest anti-enzymatic capacity, inhibiting the activities of collagenase, elastase and tyrosinase (IC50 of 7.2, 4.8 and 85.9 µg/mL, respectively). Moreover, F3 showed anti-inflammatory potential, reducing TNF-α and IL-6 release induced by LPS treatment in RAW 264.7 cells. These bioactivities may be related to the presence of phenolic compounds, such as phlorotannins, as demonstrated by NMR analysis. The results highlight the potential of C. costata as a source of bioactive ingredients for further dermatological applications. 相似文献

Interactions between plants and microorganisms can significantly affect plant health and productivity as well as ecosystem
functioning. Detailed knowledge of the tripartite relationships between plants, fungi, and bacteria, and their environment
is still limited. In the present study, the soils adjacent to three plant species (Cruciata laevipes, Mentha piperita, Equisetum arvense) in the Ljubljana Marsh and the bulk, plant-free soil were analyzed for their bacterial community structure in June and October
2006. The terminal restriction fragment length polymorphism analysis indicated a different bacterial community structure in
the rhizosphere and in bulk soil, however, with almost no seasonal changes between late spring and autumn samples and no apparent
impact of the three plant species. In addition, root colonization of the three plant species by arbuscular mycorrhizal fungi
(AMF) and dark septate endophytes (DSE) was microscopically assessed monthly from May until October 2006. A presumably accidental
correlation between monthly precipitation and the degree of arbuscule formation, with the latter lagging 1 month, was noted
for M. piperita, the most heavily colonized of the three plant species. With all three plants, the phosphorus content in roots correlated
positively with most AMF structures. Microsclerotia of DSE were mainly abundant in autumn samples. Fungal diversity in roots
was estimated using temporal temperature gradient gel electrophoresis separation of the fungal polymerase chain reaction products
obtained for both 18S-rDNA and the 5.8S-ITS2-28S rDNA segments. No specific effects of either plant species or seasonal changes
on mycorrhizal community structure were discernible. 相似文献
