The agar well technique compares favorably with the cylinder plate assay in accuracy, sensitivity, and precision. It is more flexible and more rugged, and growth of seed organism is not inhibited. The wells are precision cut with Grafar gel punch assembly with sets (6) of 10, 7, 5, 4, and 3 mm cutters. The wells are easily and rapidly filled with short, disposable Pasteur pipets fitted with rubber bulbs. The smaller wells are filled with capillary pipets. The diameter of the well (independent of volume) appears to be a function of concentration. For every decrease in the diameter size of the well, concentration can be increased, at least to the next higher level of the standard response line. Extracts of chlortetracycline containing as much as 3.2 mu-g/ml can be analyzed if the 3 mm wells are used and with no sacrifice in accuracy or precision. This works especially well for antibiotic premixes. The technique has been used successfully for penicillin, streptomycin, chlortetracycline, oxytetracycline, oleandomycin, and tylosin.  相似文献

A new procedure is described for the determination of polybrominated biphenyls (PBBs) in dry animal feeds and developmental results are discussed. Finely ground feed is packed into a chromatographic column containing Celite and then eluted with methylene chloride. The concentrated extract is cleaned up by elution with petroleum ether through Florisil before gas-liquid chromatographic quantitation. Chromatograms thus obtained were essentially free of the interfering peaks encountered when using AOAC methods for pesticide residues in dry products. Results of feed analyses by the proposed procedure averaged 30% higher than those obtained by AOAC procedures. Recoveries of PBBs from samples fortified at levels of 0.04 to 0.4 ppm ranged from 90 to 104%, with an average of 98%.  相似文献

The strategic use of closantel, a narrow-spectrum salicylanilide anthelmintic against bloodsucking helminths, and of albendazole, a broad-spectrum benzimidazole anthelmintic, in the control of gastrointestinal nematodes of sheep was investigated on a farm in Nyandarua District in the highlands of Kenya. Thirty Corriedale female lambs aged between 9 and 12 months were assigned to three treatment groups of 10 lambs each. The three groups were set stocked on separate paddocks for 12 months. Lambs in group 1 (strategic treatment group) were treated with closantel and albendazole at the beginning and towards the end of the long rains (April and June, respectively) and towards the end of the short rains (December). During the intervening dry season, the lambs were treated with albendazole. Lambs in group 2 (suppressive treatment group) were kept worm free by regular deworming with albendazole at 3-weekly intervals for 12 months. The third group of lambs remained untreat ed (control group). Gastrointestinal nematode infections and pasture infectivity were well controlled in the case of the strategic treatment group. This resulted in higher weight gains, wool production, packed cell volume, and serum albumin and protein concentrations compared with the untreated control lambs. These parameters were comparable between the strategic treatment and the suppressive treatment groups of lambs. It was concluded that worm control strategies based on the epidemiology of the parasites and the sustained anthelmintic action of closantel in combination with broad-spectrum anthelmintics can provide effective control of gastrointestinal nematodes of sheep in the study area.  相似文献

Recently, a new implement for controlling weeds in cereals (CombCut) has been developed. It cuts weeds in growing cereals without damaging them by using the physical differences (in height, stem thickness, straw stiffness and branching pattern) between crops and weeds. To evaluate and compare the effects of selective cutting with different timings of herbicide application on Cirsium arvense in spring barley, a randomised block experiment was conducted in Sweden in 2015–2017, in a field with a naturally occurring C. arvense population. Treatments consisted of control (C), herbicide application at 4–5‐leaf stage of C. arvense (H1), herbicide application at 8–10‐leaf stage (H2) and selective cutting at 10‐leaf stage (S). The treatments were performed in 2015 and repeated in 2016 in the same plots, and a final evaluation was performed in 2017. Compared to the control, S, H1 and H2 were equally efficient in reducing above‐ground biomass production of C. arvense and increasing spring barley grain yield per unit area. The number of C. arvense shoots per area was, however, higher in S compared to H1 and H2. No differences in control effects on shoot number were observed between H1 and H2. Our study indicates that (i) selective cutting (S) reduces C. arvense equally efficient as herbicide application and (ii) early herbicide spraying is as efficient as spraying later in the season.  相似文献

This study determined in vitro anthelmintic efficacy of three plant species: Trema orientalis, Urtica dioica and Zanthozylum capense on nematode larvae of small ruminants. Dried leaf samples (40 g) were extracted in 70% ethanol, in portions of 10 g and concentrated to 100 ml. Half and one quarter of the original crude extract were both made to 100 ml. Rectal faecal material from 10 Merino sheep and 25 Nguni goats was pooled within species and thoroughly hand-mixed. Dung samples, each of 5 g were cultured for 12 days at 27 °C. On day 13, 4 plates were watered and 4 others treated with ethanol to correct for solvent effect on mortality. The design was 2 (animal species)?×?3 (plant species)?×?3 (extract concentrations). In each of three runs, three plates were treated with each crude extract in three incremental concentrations. Surviving L3 larvae were isolated, counted and mortalities became indices of anthelmintic efficacy. Data from nematode larval mortality were analysed to determine the effect of animal species, plant species, concentration and their interactions. In the present study, 500 steers were used to develop models for predicting the percentage of intramuscular fat (PIMF) in live beef cattle. Before slaughter, steers were scanned across the 11th and 13th ribs using Aloka 500V (AL-500) and Classic Scanner 200 (CS-200) machines. Four to five images were collected per individual steer using each machine. After slaughter, a cross-sectional slice of the longissimus muscle from the 12th rib facing was used for chemical extraction to determine actual carcass percentage of intramuscular fat (CPIMF). Texture analysis software was used by two interpreters to select a region for determination of image parameters, which included Fourier, gradient, histogram, and co-occurrence parameters. Four prediction models were developed separately for each of AL-500 and CS-200 based on images captured by the respective machines. These included models developed without transformation of CPIMF (Model I), models based on logarithmic transformation of CPIMF (Model II), ridge regression procedure (Model III), and principal component regression procedure (Model IV). Model R2 and root mean square error of AL-500 Models I, II, III, and IV were 0.72, 0.84%; 0.72, 0.85%; 0.69, 0.91%; and 0.71, 0.86%; respectively. The corresponding R2 and root mean square error values of CS-200 Models I, II, III, and IV were 0.68, 0.87%; 0.70, 0.85%; 0.64, 0.94%; and 0.65, 0.91%; respectively. Initially, AL-500 and CS-200 prediction models were validated separately on an independent data set from 71 feedlot steers. The overall mean bias, standard error of prediction, and rank correlation coefficient across the four AL-500 models were 0.42%, 0.84%, and 0.88, respectively. For the four CS-200 models, the corresponding overall mean values were 0.67%, 0.81%, and 0.91, respectively. In a second validation test, only Model II of AL-500 and CS-200 was evaluated separately based on data from 24 feedlot steers. The overall mean bias, absolute difference, and standard error of prediction of AL-500 Model II were 0.71, 0.92, and 0.98%. For CS-200 Model II, the corresponding values were 0.59, 0.97, and 1.03%. Both AL-500 and CS-200 equipment can be used to accurately predict PIMF in live cattle. Further improvement in the accuracy of prediction equations could be achieved through increasing the development data set and the variation in PIMF of cattle used.  相似文献

Objective

To evaluate the effect of hyaluronidase on uptake, duration and speed of elimination of xylazine–tiletamine–zolazepam administered in the subcutaneous fat over the dorsal lumbar region of swine.

Study design

Blinded, randomized, crossover study.

Animals

Six healthy Landrace/Large White pigs weighing 132 ± 24 kg (mean ± standard deviation).

Methods

Animals were administered xylazine (1 mg kg?1) and tiletamine–zolazepam (8 mg kg?1) (control treatment, CON), or xylazine–tiletamine–zolazepam at the same doses with hyaluronidase (400 IU) (treatment HYA). The treatments were administered into the dorsal lumbar adipose tissue, 2.5–3.0 cm laterally from the spinous process of the second lumbar vertebra. The latency, anesthesia and recovery periods were measured. Heart rate, noninvasive systolic, diastolic, and mean arterial pressures, respiratory rate, hemoglobin oxygen saturation and rectal temperature were recorded every 10 minutes for up to 50 minutes.

Results

One animal in CON and one animal in HYA were responsive to stimulation and did not allow safe handling. No significant difference was found between treatments for latency (CON 11.3 ± 5.9 minutes, HYA 7.4 ± 5.1 minutes) and anesthesia (CON 53 ± 53 minutes, HYA 49 ± 38 minutes) periods. Recovery period was shorter in HYA (9 ± 6 minutes) than in CON (32 ± 16 minutes) (p < 0.05). Physiological variables were not significantly changed over time and were within accepted normal clinical limits for the species in both treatments.

Conclusion and clinical relevance

Hyaluronidase (400 IU) administered into adipose tissue in pigs did not reduce the latency and duration of dissociative anesthesia, but was associated with faster recovery.  相似文献

A polyphenol-rich extract from the seeds of fenugreek was evaluated for its protective effect against hydrogen peroxide(H2O2)-induced oxidation in normal and diabetic human erythrocytes (RBCs). RBCs, preincubated with increasing amounts of fenugreek seed extract and challenged with H2O2, were analyzed for hemolysis and lipid peroxidation. RBCs from diabetic subjects were more susceptible to oxidative hemolysis and lipid peroxidation than those from normal subjects. However preincubation with the polyphenol-rich extract significantly reduced the oxidative modifications in both the groups. The inhibition of lipid peroxidation was concentration-dependent up to 100 l of extract, which contained 0.75mM gallic acid equivalent (GAE) of phenolic compounds. These findings demonstrate the potent antioxidant properties of the fenugreek seeds.  相似文献
