The objective of this study was to find out the impact of L‐carnitine (10 mM) on developmental regulation of preimplantation sheep embryos cultured in vitro when supplemented in maturation medium and post‐fertilization medium separately. Subsequent objective was to observe the L‐carnitine‐mediated alteration in expression of apoptotic genes (Bcl2, Bax, Casp3 and PCNA) in sheep oocytes and developing embryos produced in vitro. Oocytes matured with L‐carnitine showed significantly (p < .05) higher cleavage (67.23% vs 43.12%), morula (47.65% vs 28.58%) and blastocysts (32.12% vs 13.24%) percentage as compared to presumptive zygotes cultured with L‐carnitine during post‐fertilization period. So it is suggested to use L‐carnitine during maturation than post‐fertilization period. Antiapoptotic and proliferative effects of L‐carnitine were confirmed by inducing culture medium with actinomycin D (apoptotic agent) and TNFα (antiproliferative agent), respectively, with and without L‐carnitine. Oocytes and embryos cultured with actinomycin D and TNFα showed developmental arrest with significant (p < .05) decrease in morula and blastocysts percentage but s upplementation of L‐carnitine to actinomycin D and TNFα induced culture medium showed similar result as that of control . L‐carnitine supplementation during IVM significantly (p < .05) upregulated the expression of Bcl2 and PCNA genes in majority of the developmental stages. Although L‐carnitine upregulated the expression of Bax in initial developmental stages but downregulated at latter part, whereas the expression of Casp3 was upregulated upto 16‐cell stage but after that there was no difference in expression. Expression of GAPDH gene was not affected by L‐carnitine supplementation. In conclusion, L‐carnitine acted as an antiapoptotic and proliferative compound during embryo development and supplementation of L‐carnitine during IVM altered the expression of apoptotic genes in the developmental stages of embryos.  相似文献

Objective To assess biosecurity practices in the fancy poultry show sector that would influence the establishment and spread of exotic diseases in poultry in New South Wales. Design A cross-sectional survey of 105 fancy poultry exhibitors at seven agricultural shows. Procedure Exhibitors were interviewed about biosecurity practices on their farms and their knowledge of exotic diseases. Poultry stewards at 18 shows were interviewed about biosecurity practices at their shows. Results Although many exhibitors travelled only short distances to attend shows, some exhibitors attended up to 30 shows per year and travelled interstate to exhibit poultry. A network diagram revealed extensive connections and interactions of poultry throughout the eastern half of NSW. Five of 18 shows included cash sales without any record of purchasers; 46% of exhibitors reintroduced exhibited birds back into their flocks without a quarantine period; and 16% failed to wash cages used to transport the birds. There was a general awareness that exhibition of birds posed a risk to flock health, but knowledge of avian influenza and practices that could be adopted to minimise the risk of disease introduction was limited. Conclusions The factors that could assist the establishment and spread of exotic diseases in poultry in NSW include the mixing of birds at shows, inadequate recording of exhibitor details at shows, inadequate biosecurity practices when reintroducing exhibited poultry back into flocks, cash sales associated with shows that did not include the collection of purchaser details, and inadequate identification of birds.  相似文献

Objective To evaluate the effect of the number of days exposure to a prepartum transition diet on reproduction and health in dairy cows. Design A prospective cohort study. Procedures We enrolled 1008 dairy cows from three herds to examine the effects on reproduction and health of increased days exposure to prepartum transition diets that included ryegrass pasture, ryegrass silage, cereal hay, grain, by-products, oilseed meals, BioChlor®, rumen modifiers, minerals and vitamins. Diets provided 9.9 MJ metabolisable energy per kg dry matter (DM), a metabolisable protein balance of 286 g/day and a dietary cation anion difference of −150 meq/kg DM. Statistical models controlled for herd, calving day, age and gestation period. Results In two of the three herds increased days exposure to prepartum transition diets increased the hazards of submission for breeding, conception and clinical mastitis, and decreased the hazard of cow removal. The odds of pregnancy by 6 and 21 weeks after the mating start date tended to increase with increasing days of exposure to prepartum transition diets. Conclusions Increasing exposure to a prepartum transition diet improved the calving to conception interval, tended to improve the odds of pregnancy and reduced the risk of culling of cattle, but increased the hazard of clinical mastitis.  相似文献

Two anthelmintics with known insecticidal action (ivermectin and closantel) and one with no recorded effect on insects (levamisole) were tested to evaluate their effects on buffalo fly (Haematobia irritans exigua). Blood from animals given closantel or levamisole had no significant effect on mortality of buffalo flies in an in-vitro assay. In contrast, blood from animals given ivermectin showed a dose-dependent effect on the mortality of buffalo flies. At 24 h after one injection of the recommended dose of ivermectin, 98% of the flies applied to cattle in an in-vivo assay are killed. Blood from cattle injected with ivermectin killed 95% of flies 8 d after injection and still killed 15% of flies at 18 days after injection. Surviving flies laid almost no eggs and this effect on flies was significant up to 33 d after injection. The results indicate that ivermectin may be useful to control buffalo fly populations in the field.  相似文献

This paper provides an overview of the changes in the pasture-based dairy systems of New Zealand and Australia that may influence the health of cattle.

There are relatively few available data that can be used to quantify the effects of increased intensification of milk production on the health of cattle. There is evidence that increased production increases the risk of mastitis and culling for udder health. Increased risks of mastitis with treatment with somatotropin support these findings; however, the risk of mastitis may decrease with increased milking frequency. Larger herds with greater stocking density should increase the risk for infectious disease, but evidence to support this contention is sparse.

Very intensive grazing patterns associated with higher grass yields achieved using better cultivars and greater use of fertilisers favour nematode parasites. There is some evidence of anthelmintic resistance in both nematodes and liver fluke. Veterinarians will need to be aware of the potential for these to reduce the productivity of cattle.

There have been benefits of improved nutrition on the efficiency of energy use for dairy production. Diseases such as bloat and ketosis appear to be of lower prevalence. It also appears that mineral nutrition of pasture-fed cattle is being better addressed, with gains in the control of milk fever, hypomagnesaemia and trace-element deficiencies. However, acidosis is a condition with a high point prevalence in pasture-based dairy systems where cows are fed supplements; one study in Australia found a point prevalence of approximately 11% of cows with acidosis. There is evidence from this study that the neutral detergent fibre (NDF) in pasture-based diets may need to be higher than 30% of the diet to maintain rumen stability. Laminitis and acidosis are different conditions with a similar pathogenesis, specifically highly fermentable diets. The prevalence of lameness was 28% in herds in Australia, suggesting that this condition must be a focus for preventive medical approaches, including the design of laneways, feedpads and dairies.  相似文献

The aim of this study was to evaluate the effects of thyroxine administration on morphometric parameters, expression of vascular endothelial growth factor (VEGF) and vascularization in the uterus and placenta and reproductive parameters in gilts at 70 days of gestation. At 150 days of age, i.e., before first heat, 20 gilts were randomly divided into two experimental groups: treated (n = 10) and control (n = 10). The treated group received a daily dose of 400 μg of l ‐thyroxine (T4) in their diet until slaughter and the control group received only typical meals. Before artificial insemination, blood was collected to determine plasma total T4. The gilts were inseminated in the second oestrus and slaughtered at 70 days of gestation. The foetal thyroid follicular epithelium height, number, size and weight of foetuses; foetal myogenesis, corpora lutea number, embryonic mortality rate, uterine weight, placental weight and placental fluid volume were measured. Histomorphometric and immunohistochemical analysis of uterus and placenta were determined. The averages of all variables were compared by the Student’s t‐test. The gilts treated with thyroxine showed significant increase of plasma total T4. At 70 days of gestation, the heights of the trophoblastic epithelium, endometrial epithelium and endometrial gland epithelium were significantly higher in the group treated with T4. The expression of cytoplasmatic and nuclear VEGF in trophoblastic cells and the number of blood vessels per field in endometrial stroma were significantly higher in the gilts treated with T4. No other significant differences between groups were obtained with respect to other parameters (p > 0.05). We conclude that oral administration of T4 up to 70 days of pregnancy in gilts affects the morphometric parameters, the expression of placental VEGF and the uterine vascularization but does not affect reproductive parameters in gilts during early gestation.  相似文献

The effects of lactation on the fertility of dairy cows   总被引:1,自引:0,他引:1  
Lactation has been negatively associated with fertility because pregnancy rates in maiden heifers exceed those obtained after first or subsequent calvings. The extent of this difference is less in pasture-fed dairy cows (<10%) than in American Holsteins (>20%) fed grain and conserved forages. The latter cows have pregnancy rates to first insemination and oestrus detection rates of only 40 to 45%. This suggests that the subsequent fertility of inherently fertile Holstein heifers may be severely compromised by high levels of milk production. International comparisons show that pasture-fed dairy cows may experience extended periods of anovulatory anoestrum but have normal fertility (60% pregnancy rate to first insemination) once cycling. The high-producing American Holstein may ovulate within 4 weeks postpartum but is more likely to continue ovulating without being detected in oestrus. Both situations are associated with negative energy balances (NEB) during early lactation. The severity and duration of this NEB may vary with body condition at calving, age or parity, ration formulation, production level and environmental factors. Relative daily milk yield is not an absolute indicator of NEB, because some lower producing cows within a herd have lower feed intakes and more severe energy deficits. NEB is not simple to measure; nonetheless, it is correlated with genetic improvement for milk yield. A positive energy balance, greater weight gain and higher body condition score have all been shown to be positively correlated with plasma progesterone concentrations in early lactation. No studies have investigated the possibility that the rapid increase in metabolic rate at this time may also alter steroid concentrations with consequent effects on oestrous behaviour and fertility. Studies to more precisely define the effects of increasing milk yields in early lactation, especially in Holsteins, may need to be completed in Australia and New Zealand. Oestrus detection rates and pregnancy rates for American Holsteins of less than 50% are accepted widely in the USA. Such low detection rates confound studies on fertility. The objective should be to increase these 2 rates to at least 80% and 60% respectively. This may involve the use of controlled breeding, especially if oestrous behaviour is less overt in high-producing Holstein cows.  相似文献
