Enset, Ensete ventricosum, is a crop that contributes to food security for more than 20 % of Ethiopia’s population. One clone never fulfills all the ecological and social requirements and hence farmers maintain a diverse range of enset cultivars on their farm. The objective of this work was to assess morphological and use value related characterization and document the indigenous knowledge of farmers on classification, cultivation and utilization of enset. A total of 280 farm households in seven zones were surveyed using individual household interviews. The observed traits showed low to high levels of variability among enset clones with a Shannon–Weaver diversity index (H′) value of 0.154–0.827 for bulla (extracted starch) quality and midrib color, respectively. Moreover, the overall H′ mean of 0.399 confirmed the existence of low to medium levels of phenotypic variation. The mean squares due to genotypes were highly significant (P ≤ 0.01) for all the quantitative traits studied, suggesting the presence of substantial genetic variability among the 165 enset clones. Kocho (fermented starch) yield had significant positive correlation with bulla quality and plant vigor. Knowledge of farmers’ practices is currently used to validate agronomic innovations and inform the setting up of a network of phenotype collections managed by farmers.  相似文献

Cryptosporidium parvum oocysts can be transported in overland flow from faecal deposits of grazing animals causing potential water contamination problems. Deposition of oocysts can occur when overland flow encounters vegetative buffers and its transport capacity is decreased in the upslope backwater region. Deposition of oocysts was investigated using a large rainfall simulator, with and without a vetiver buffer strip. Oocysts-spiked slurries were applied to two contrasting soils and the resultant oocyst/sediment depositions from the flow were compared. The buffers substantially reduced suspended sediment loads from the two soils and increased the oocyst concentration in the soil with >99% of the deposited oocysts measured upslope of the buffer.  相似文献

The aim of this study was to identify and characterize thermophilic Campylobacter species in faecal samples from goats in Khartoum State, Sudan, by application of multiplex polymerase chain reaction. Campylobacteriosis is a zoonotic disease of global concern, and the organisms can be transmitted to human via food, water and through contact with farm animals and pets. There are five clinically related Campylobacter species: Campylobacter jejuni (C. jejuni). Campylobacter coli, Campylobacter lari, Campylobacter upsaliensis and Campylobacter fetus. Conventional cultural methods to diagnose campylobacteriosis are tedious and time consuming. Wide ranges of genes have been reported to be used for PCR-based identification of Campylobacter spp. We used a multiplex PCR assay to simultaneously detect genes from the major five clinically significant Campylobacter spp. The genes selected were hipO (hippuricase) and 23S rRNA from glyA (serine hydroxymethyl transferase) from each of C. jejuni. C. coli, C. lari, and C. upsaliensis; and sapB2 (surface layer protein) from C. fetus subsp. fetus. The assay was used to identify Campylobacter isolates recovered from 336 cultured faecal samples from goats in three localities in Khartoum State. C. coli was the most predominant isolate (234; 69.6%), followed by C. jejuni (19; 5.7%), C. upsaliensis (13; 3.9%), C. fetus subsp. fetus (7; 2.1%) and C. lari (6; 1.8%). Twenty-nine goats showed mixed infection with Campylobacter spp., 21 of which harbored two Campylobacter spp., while eight animals were infected with three species. Ten out of twelve goats that displayed diarrhea harbored C. coli only. C. coli, C. jejuni and C. upsaliensis showed significant variation with localities. The prevalence of C. coli was significantly higher (87; 25.9%) in goats from Omdurman, whereas C. jejuni and C. upsaliensis were significantly higher (11; 3.3%, 9; 2.7%) in goats from Khartoum. The multiplex PCR assay was found to be rapid and easy to perform and had a high sensitivity and specificity for characterizing the isolates, even in mixed cultures. The study demonstrated the significance of goats as reservoirs in the dissemination of Campylobacter spp. which could be considered as potential agent of caprine enteritis and abortion as well as contamination of the wider environment posing serious public health concern in Khartoum State.  相似文献

This study determined the combining ability and gene action controlling yield and yield‐related traits in wheat under drought‐stressed and nonstressed conditions. Twelve parents possessing Rht‐B1b and Rht‐D1a genes and their 66 half‐diallel crosses were evaluated under field and glasshouse conditions. Plant height (PH), productive tiller number (TN), kernels per spike (KPS), thousand seed weight (TSW) and grain yield (GY) were recorded. Analysis of variance, heritability, correlation and combining ability analyses were performed. Heritability estimates ranged from 53.00% (TN) to 63.07% (KPS). Yield showed positive correlations with all other traits under all test conditions. Significant GCA effects were observed for all traits recorded across test conditions, except for yield in the glasshouse. All Baker's ratios were less than a unit, indicating predominance of nonadditive gene action. Consistently high positive GCA effects were observed on LM02 for GY; LM02 and LM23 for KPS; and LM04 and LM09 for TSW, while LM17 and LM21 had negative effects for PH. Good general and specific combiners will be used for further breeding and selection.  相似文献

Understanding the genetic relationship of crop ideotypes is essential for genetic analysis and breeding. The objective of this study was to determine genetic differentiation present among 20 selected tepary bean (Phaseolus acutifolius A. Gray) genotypes using morphological traits and simple sequence repeat (SSR) markers in order to identify genetically unique parental lines to develop breeding populations. Phenotypic diversity was estimated using Shannon-Weaver diversity index (H’), principal component analysis (PCA) and cluster analysis (CA). Genotypic diversity was estimated using Jaccard's genetic distances and CA. Shannon-Weaver diversity index values for the studied morphological traits ranged from 0.89 to 0.99, with a mean of 0.95 revealing high phenotypic differences among test genotypes. PCA identified four useful principal components (PC's) which contributed to 73% of the total phenotypic variation of collections. PC1 accounted for 28% of the total variation correlated to dry shoot mass and number of pods per plant. PC2 correlated with number of seeds per pod, grain yield and harvest index and contributed to 21% of total variation. The mean observed (Ho) and expected (He) heterozygosity values were 0.45 and 0.51, respectively revealing moderate genetic differentiation among genotypes. The mean polymorphic information content was 0.52, suggesting efficient discriminatory power of the SSR loci useful in future tepary bean genetic diversity analysis. The current study revealed moderate genetic differentiation among the studied tepary bean genotypes. Morphological traits and SSR markers well-correlated in allocating the tepary bean genotypes. The following genotypes were genetically distinct: G40201, G40237, G40068, G40033 and G40063 which are recommended for further crosses, selection and population development.  相似文献

Purpose: Sorghum [Sorghum bicolor (L.) Moench] has been cultivated for centuries as a staple food crop for millions of people in sub-Saharan Africa and Asia. In Ethiopia a total of 4.34 million tons of sorghum is being produced per annum, predominantly in the arid and semi-arid areas. However, the yield and quality of sorghum is affected by a wide array of production constraints, notably severe and recurrent drought stress. The aim of this study were to determine the impact of drought on sorghum production and productivity over time and space, and to identify farmers’ trait preferences, production constraints and coping strategies when dealing with drought in north eastern Ethiopia.

Materials and methods: Participatory rural appraisal (PRA) was employed in three administrative zones of north eastern Ethiopia. One Woreda from each Zone and two Kebeles from each Woreda were selected on the basis of sorghum area coverage, production, consumption and prior information on the intensity, duration and spatial coverage of drought. In each kebele, 30 respondents were selected for interview and 12 key informants were selected for group discussions and transect walk personal observations. Data collected from 180 respondents was subjected to statistical analysis and the information gathered from 72 key informants through focus group discussions and transect walk observations were used in discussing the results.

Results and discussion: The present study found that productivity of sorghum was challenged by recurrent droughts, Striga infestation, insects, birds, diseases, a lack of varieties with farmers-preferred traits and high yield potential, limited policy support, a lack of improved seed system, poor sorghum production practices and application of crop input and poor soil fertility, in a decreasing order of importance. Among the listed sorghum production constraints, severe drought in the post-flowering stage was identified by most interviewed farmers as the leading constraint across the three study zones. Focus group discussions and transect walk observations held in each Kebele revealed that farmers’ had lost numerous valuable local landrace varieties due to extreme drought conditions over the years. A significant number of interviewed farmers preferred to grow high grain and biomass yielder medium-maturing sorghum varieties which can be sown at the normal planting time but which would escape post-flowering drought.

Conclusion: Overall, sorghum breeding programme should be directed at developing farmers’ ideal sorghum varieties with high grain and biomass yield, adequate level of drought and Striga tolerance. In addition, development of farmer preferred medium-maturing sorghum varieties suitable for April planting should be strengthen to boost its productivity and to increase varietal adoption rate in the area.  相似文献

Knowledge on farmers’ perspectives is of paramount importance in order to design appropriate agricultural research and development interventions attuned to local farming systems. A participatory rural appraisal was conducted in order to understand perceived causes and indicators of soil acidity under multiple land-uses in three Districts of northwest Ethiopia. Soil samples were also collected from five dominant land-uses. The samples were analysed for soil pH, exchangeable acidity and other physico-chemical properties. The result indicated that the pH(H2O) of most of the soils in the study sites were in a strongly acidic range (4.6–5.5). Gashena Akayita of the Banja District was the most acidic of all. Among the land-uses, eucalyptus fields were the most acidic followed by crop outfields and grazing lands in that order. At all the study sites, exchangeable Al was detected in soils having a pH of less than 5.0. Overall, the nutrient dynamics showed variation across land-uses and study sites. Farmers’ perceived causes of soil acidity included: soil erosion; contending use of fertility replenishing local resources; abandoning traditional fertility management practices and minimal use of external inputs. The farmers attributed the exclusive use of acid-forming inorganic fertilizers to exhaustion of the soil. Various land and soil characteristics, plant growth attributes, changes in genetic diversity were mentioned as indicators of soil acidity. Particularly, the farmers used prevalence of acidophilic weed species on crop fields and grazing lands as marker of strongly acidic soil. Farmers’ perceived causes and indicators were in agreement with scientific facts and can be utilized as input in designing sustainable acid soil management strategies. Decline in genetic diversity of the once widespread crop species and land races, and expansion of newly introduced soil acidity tolerant species, suggests the need to undertake rescue collections in these areas.  相似文献

Information on the nutrient contents of newly developed orange-fleshed sweetpotato (OFSP) varieties is required for recommendations to growers and the food industry. Therefore, the objectives of this study were to determine the nutritional value of newly developed OFSP clones and to establish the associations between β-carotene content and micro-nutrients for targeted large scale production to alleviate nutrient deficiencies. Seventeen OFSP and eight white/cream-fleshed sweetpotato clones were evaluated across six diverse environments (Halaba, Kokate, Areka, Arbaminch, Hawassa and Dilla) in southern Ethiopia in 2014 using a 5?×?5 simple lattice design. Nutritional traits data were collected on dry-weight basis and subjected to analysis of variance and correlation analyses. Environment, genotype, and genotype?×?environment interaction effects were highly significant (p??1), protein (7.08%), iron (2.55 mg 100?g?1), zinc (1.42 mg 100?g?1), fructose (4.45%), glucose (5.34%) and sucrose (16.20%). Genotypes G15 and G19 also performed relatively well for the above nutritional traits. The three genotypes, G8, G15 and G19 had mean fresh root yield of 23.5,13.7 and 21.3?tha?1, respectively. These genotypes had root dry matter content of 26.99%, 25.23% and 33.09%, respectively. Β-carotene content had significant positive correlations with iron, zinc, fructose, glucose and sucrose content. This reflects the potential to breed for OFSP varieties enriched with the important micro-nutrients. Overall, the candidate OFSP clones, G8 (Resisto?×?PIPI-2), G15 (Resisto?×?Temesgen-23) and G19 (Resisto × Ogansagen-23) were good sources of nutritional traits such as vitamin A, iron, zinc, protein, sucrose, glucose and fructose. The selected genotypes can be recommended for large-scale production, food processing or further sweetpotato improvement to alleviate nutrient deficiencies in Ethiopia or similar environments in sub-Saharan Africa.  相似文献

本研究根据鸡Apo VLDL-Ⅱ基因内含子Ⅰ的基因序列设计3对引物,分别采用PCR-RFLP、PCR-SSCP技术对文昌鸡、如皋黄鸡、丝羽乌骨鸡、安卡肉鸡及红色原鸡5个鸡种进行多态性研究。结果表明,鸡Apo VLDL-Ⅱ基因内含子Ⅰ存在4处突变,分别为:1950处G/A、2522处A/C、2705处A/C及2793处C/T。统计结果显示:安卡肉鸡基因型频率的分布与如皋黄鸡、红色原鸡存在显著差异,文昌鸡基因型频率的分布与乌骨鸡差异不显著,除安卡肉鸡外所有鸡种均处于Hardy-Weinberg平衡状态。  相似文献

The greater wax moth Galleria mellonella L. (Lepidoptera: Pyralidae) is occasionally found in beehives and is a major pest of stored wax. Entomopathogenic fungi have recently received attention as possible biocontrol elements for certain insect pests. In this study, 90 isolates of Beauveria bassiana and 15 isolates of Metarhizium anisopliae were screened for proteases and lipases production. The results showed significant variations in the enzymatic action between the isolates. In the bioassay, the selected isolates evinced high virulence against the 4th instar of the G. mellonella larvae. The isolates BbaAUMC3076, BbaAUMC3263 and ManAUMC3085 realized 100% mortality at concentrations of 5.5 × 106 conidia ml−1, 5.86 × 105 conidia ml−1, and 4.8 × 106 conidia ml−1, respectively. Strong enzymatic activities in vitro did not necessarily indicate high virulence against the tested insect pest. The cuticle of the infected larvae became dark and black-spotted, indicating direct attack of fungus on the defense system of the insects. The LC50 values were 1.43 × 103, 1.04 × 105 and 5.06 × 104 for Bba3263AUMC, Bba3076AUMC and Man3085AUMC, respectively, and their slopes were determined by computerized probit analysis program as 0.738 ± 0.008, 0.635 ± 0.007 and 1.120 ± 0.024, respectively.  相似文献
