This study was carried out to assess the in vitro quality of canine semen frozen in an ultrafreezer at -152 degrees C and to evaluate the male-to-male variation of frozen semen in five male dogs of the Canarian Mastiff breed. Four ejaculates of each dog were processed individually (5% glycerol and 0.5% Equex) to reach a final concentration of 100 x 10(6) spermatozoa/ml. Then, two freezing techniques were tested to assess the seminal quality (sperm motility, live spermatozoa and abnormal sperm cell percentages) at 1, 30, 60, 120 and 360 days after freezing: (i) semen was frozen and stored in liquid nitrogen; (ii) semen was frozen and stored in the ultrafreezer at -152 degrees C. After freezing-thawing, both freezing protocols showed no significant differences in sperm motility and the percentages of live and abnormal spermatozoa. On the other hand, the microscopic characteristics of spermatozoa in fresh semen were practically similar among males; however, after the semen processing and freezing, significant differences were observed (p < 0.05) among males, especially as regards sperm motility. This inter-individual variability was detected in both freezing protocols, showing that the male-to-male variation in the seminal quality post-freezing was independent of the freezing technique used. The in vitro results obtained in the Canarian Mastiff breed confirmed that the use of ultra-freezers at -152 degrees C is a potential alternative to liquid nitrogen for storing canine semen for long periods of time.  相似文献

Twenty-two pregnant goats of the Canarian breed were administered 10 mg of the prostaglandin analogue luprostiol intramuscular (IM) on day 145 of pregnancy; seventeen pregnant goats were injected IM with saline solution the same day. Twenty hours after luprostiol application, goats were continuously observed in order to record the following parameters: parturition, dystocia incidence, placental delivery and live births. Parturition was successfully induced in all goats; the mean interval to parturition (first kid delivery) was of 32.9 ± 1.0 (SEM) hours (h) and the range varied from 21 to 41 h; in addition, no significant differences (p > 0.1) were detected between nuliparous and multiparous goats (31.3 ± 1.3 h and 34.7 ± 1.2 h, respectively). In the control group, the interval from saline injection to parturition was 75.9 ± 12.5 h (mean ± SEM), showing a range between 34 and 138 h. All the goats expelled the fetal membranes in the first 6 h after the induced or natural parturition. The incidence of dystocia due to fetal maldisposition was not significantly different between induced and control goats (9.0% and 11.8%, respectively). In addition, the percentage of live kids was practically similar between both groups (90.7% and 88.0%, induced vs control goats). This study confirms the effectiveness of the luprostiol to induce the parturition in goats, showing a narrow range (30–37 h) in most of the induced females (86.3%; 19/22 goats). The administration of prostaglandin analogue is a basic tool to plan kidding, allowing a effective concentration of effort and resources at the time of parturition.  相似文献

The magnitude of relationships among different traits is important in plant breeding programs to identify the best selection criteria and improve the efficiency of selection. This study was conducted to determine relationships between seed yield and seed oil content with other important agronomic traits among 36 diverse accessions of Vernonia (Vernonia galamensis variety ethiopica), a potentially novel industrial oilseed crop. Field evaluations were conducted during 2005, 2006 and 2007 at the Limpopo Province in South Africa using a partially balanced lattice design. Simple correlation and path analysis were performed to identify the best selection criteria for increased seed yield and seed oil content. Simple correlation and path analyses revealed that the formation of productive primary heads strongly associated with increased seed yield (rg = 0.81, p < 0.001). Furthermore, path analysis indicated selection for increased number of primary heads would bring about simultaneous and favorable change towards reduced days to maturity and shorter plant height. Further associational study of traits with seed oil content showed a significant (p < 0.05) correlation between oil content with 1000 seed weight (rg = 0.4). The path analysis, however, exposed seed yield followed by 1000 seed weight with significant direct effect on seed oil content. The study demonstrated that selection for increased number of productive primary heads is the principal selection criterion to improve seed yield. Whereas selection for 1000 seed weight and increased seed yield serve as major selection criteria to achieve increased oil content in V. galemanesis.  相似文献

The diagnostic challenge presented by an amelanotic uveal cyst with an atypical appearance in a 9‐year‐old Yorkshire terrier dog is reported. The dog was presented with a peculiar cystic neoformation adherent to the edge of the pupil of the right eye. The cyst wall was attached to the pupillary margin and it was bean‐shaped, measuring approximately 4.5 × 2.5 mm. It was white in colour with several red striations and a small brown spot in the middle, which conferred on it a peculiar appearance. The cyst could not be transilluminated and partially impaired vision. Apart from that, the ophthalmic exam revealed no other abnormalities and the eye showed no signs of inflammation. Ocular ultrasound revealed the cystic nature of the neoformation. During paracentesis of the anterior chamber, the cyst was deflated and both the cyst wall and fluid were aspirated. The tissue obtained was sent for a histological examination and was considered as corresponding to a uveal cyst. The dog improved from the post‐operative uveitis without any complication and after 24 months of follow‐up showed no recurrences.  相似文献

A 3-year irrigation trial provided basic information on the response of persimmon (Diospyros kaki cv. Triumph) water use and development to irrigation levels. Constant experimental factors applied to recommended “baseline” crop factors resulted in ratios of irrigation (I) to FAO56 reference crop evapotranspiration (ET0) ranging from 0.35 to 1.14. Vegetative and reproductive growth, sap flow, stem water potential (SWP), and local climate were monitored. An overall increase in yield and vegetative growth in response to irrigation was found, which suggests a potential yield increase for higher irrigation levels (40 tons/ha for annual irrigation of 1,000 mm). At high irrigation, the yield response curve levelled off and the marginal contribution of additional water declined. The up to threefold increase in number of fruits with irrigation, with no influence on natural abscission, suggests that differences in fruit quantities stem from response to irrigation at the earlier growth stages. Mean fruit size and fruit quality, as indicated by the ratio of rejected fruit, increased with irrigation up to I/ET0 of ~0.8. Relative yield increased linearly with relative transpiration. However, post-harvest quality was not influenced. SWP, sap flow, and non-transpirable water fractions indicated that the seasonal irrigation tables were not well tuned. Initial adjustments were made during the final season of the experiment and a new table was developed based on our results. The new table should be a basis for further trials.  相似文献

1. A study was conducted to test the hypothesis that feed efficiency (FE) correlated with the expression of genes from the somatotropic axis and mitochondrial genes involved in energy production, and that the environment to which the birds are exposed influenced the expression of such genes.

2. Quails were divided into High-FE and low-FE groups and maintained in a comfortable or heat stress (HS) (38°C for 24 h) environment to evaluate changes in insulin-like growth factor I (IGF-I), growth hormone receptor (GHR), adenine nucleotide translocator (ANT), uncoupling protein (UCP) and cytochrome oxidase subunit III (COX III) messenger ribonucleic acid (mRNA) expression in liver and muscle tissues.

3. High-FE quails (0.28 g/g) presented a higher final body weight, greater weight gain and a better feed conversion ratio than low-FE birds (0.18 g/g). High-FE birds showed greater IGF-I mRNA expression in the liver and muscle and greater GHR mRNA expression in the muscle.

4. Environmental effects only affected GHR expression in the liver, with quails under comfortable conditions exhibiting greater GHR expression than quails subjected to HS.

5. There was a significant interaction between FE and environmental temperature on ANT mRNA expression in the liver. The greatest ANT mRNA expression was observed for high FE-birds that remained under comfortable conditions.

6. In the liver, UCP mRNA expression did not differ among the quails and was not affected by environment or efficiency. However, comparisons of the low- and high-FE birds revealed higher levels of UCP mRNA in the muscle of low-FE birds.

7. COX III mRNA expression in the liver was dependent on environmental temperature and FE. Higher COX III mRNA expression was observed in animals that remained under comfortable conditions, and high-FE birds exhibited higher expression levels compared to low-FE birds.

8. These results suggest a correlation between IGF-I, GHR, ANT, UCP and COX III gene expression and FE and that environmental temperature could affect the expression of some of these genes.  相似文献

Lectins are proteins or glycoproteins with the ability to link to carbohydrates at the cell surface in a specific and reversible manner. Studies have showed that lectins are demonstrate immunomodulatory and antitumor agents. This study aims to evaluate the effect of lectin extracted from the seeds of Canavalia brasiliensis (ConBr) on murine melanoma B16F10 cells by analyzing cell viability, apoptosis index, cell migration, production of cytokines and nitric oxide (NO). Results showed that ConBr was able to reduce cell viability and thwart apoptosis, which could be observed by decrease in cell migration. ConBr also induced NO and IL-12 synthesis. Altogether, these data demonstrate the potential of ConBr as a therapeutic agent for melanoma.  相似文献

This study aimed to evaluate sex, the number of lambs per birth, and the family effects on production traits in the Santa Ines breed of sheep by estimating the least square means and coefficient of variance for those traits. A total of 484 lambs were evaluated for the following traits: weight at birth, at weaning, and at 240 days of age; weight gain during the pre-weaning and post-weaning periods; height, width, and length of different body regions; and rib eye area and fat thickness between the 12th and 13th ribs. This study was performed to evaluate plasma concentrations of anti‐Mullerian hormone (AMH) and the ovarian antral follicle population (AFP) in different genetic groups. Cyclic heifers (13 Bubalus bubalis [Murrah]; 15 Bos taurus [Holstein] and 10 Bos indicus [Gyr]) were maintained under the same management and were synchronized with two doses of 150 μg IM d‐cloprostenol administered 14 days apart. After the second d‐cloprostenol treatment, heifers had their ovaries scanned daily by ultrasound to define the day of ovulation. On the same day, the AFP was determined and a plasma sample was collected to measure AMH. Murrah heifers had less AFP (25.6 ± 2.1 follicles; p = 0.01) and plasma AMH concentration (0.18 ± 0.03 ng/ml; p < 0.001) than Gyr (60.0 ± 12.2 follicles and 0.60 ± 0.12 ng/ml of AMH); however, data were similar when compared to Holstein (35.9 ± 6.8 follicles and 0.24 ± 0.06 ng/ml of AMH) heifers. Regardless of genetic background, there was a positive relationship between the AFP and plasmatic AMH concentration (Murrah [r = 0.62; p < 0.01], Holstein [r = 0.66; p < 0.001] and Gyr [r = 0.88; p < 0.001]). Also, when heifers were classified according to high‐ or low‐AMH concentration based on the average within each genetic group, high‐AMH heifers had greater (p < 0.0001) AFP than low‐AMH heifers. In conclusion, both Murrah and Holstein heifers presented lower plasma AMH concentration and AFP when compared to Gyr.  相似文献
