Improved large ruminant productivity is increasingly acknowledged as a pathway for the alleviation of rural poverty and food insecurity in smallholder communities in Southeast Asia; yet, in much of Laos, bovine reproductive management is practically absent. Large ruminant reproduction skills were studied, using face-to-face surveys (n=60) of the knowledge, attitudes, and practices (KAP) of farmers, plus an extension of an examination of parameters of reproductive efficiency (n?=?1786 cattle and 434 buffalo) in the northern provinces of Luang Prabang and Xieng Khouang. The surveys particularly involved female farmers to provide gender-disaggregated data, with females making up 38.3 % of participants. Results confirmed that KAPs of smallholder farmers on bovine reproductive management were low (34–46 %) with trends toward higher KAP scores in male survey respondents. Poor reproductive parameters were identified in both provinces, with low calving percentages of 54–75 and 45–54 % in cattle and buffalo groups, respectively, and prolonged inter-calving intervals of 14.1–19.8 and 26.0 months for the cattle and buffalo groups, respectively. Improving the reproductive efficiency of large ruminants in the northern upland regions would enable smallholder farmers to be more effectively engaged in the dramatic economic growth of the Southeast Asia region, although these findings indicate that intensive training and supportive interventions are required to improve large ruminant reproductive outcomes in communities that have low-level large ruminant husbandry skills. 相似文献

Urbanisation is increasing tremendously in some parts of the world. Consequently, many rural forests may become depleted, although many opportunities exist for urban forests to increase. However, few studies have quantified the carbon (C) sequestration capacities of urban and rural forests in specific climatic zones. The present study compared carbon sequestration in two temperate deciduous forests located in Nagoya and Toyota, central Japan. The Nagoya University forest represented an urban forest, and a site in Toyota represented a rural forest. The urban forest at Nagoya University had comparatively smaller areas of green space and larger areas of buildings and roads. Land uses for building and road, which are typical of urban areas, result in smaller diurnal temperature ranges but higher air temperature, vapour pressure deficit, and atmospheric carbon dioxide (CO2) concentration. The urban forest in this study exhibited higher gross ecosystem exchange (GEE), especially in the active growing season from May to September, suggesting the possible effect of CO2 fertilisation. However, higher air temperatures caused comparatively smaller net ecosystem exchange (NEE) because of higher ecosystem respiration (RE). Although both forests functioned as CO2 sinks at annual time scales, the rural Toyota forest site (5.43 t C ha–1 yr–1) had 36% higher net ecosystem production (NEP=–NEE; the negative sign indicates uptake by the forest ecosystem from the atmosphere) than that at the urban forest. The higher normalised respiration (i.e., RE/GPP ratio; GPP=–GEE where GPP represents gross primary production) at the Nagoya University forest might be attributable to factors associated with the degree of urbanisation. Thus, in temperate forests, factors associated with urbanisation may reduce the atmospheric carbon sink function by accelerating respiration. The hypothesis that apoptotic factors play some roles in the denucleation of erythroblasts has been confirmed by the immunohistological detection of both phosphatidylserine and thrombospondin as phagocytosis-inducing factors in general apoptotic events. Both phosphatidylserine and thrombospondin were detected on the surface of cell membrane of mature erythroblasts, while thrombospondin was also detected in more immature erythroblasts. The intensities of their immune reactions increased as the erythroids matured. During denucleation, the positivities of both phosphatidylserine and thrombospondin were restricted on the surface of the cell membrane surrounding the protruding nuclei. Thus, the apoptotic process involves denucleation of erythroblasts and phosphatidylserine, and thrombospondin acts as phagocytosis-inducing factors in the denucleation event. 相似文献

The aim of the present study was to clarify the overall efficiency of porcine somatic cell nuclear transfer (SCNT) by incorporating cryopreservation of the cloned embryos before transfer. The SCNT embryos reconstructed with preadipocytes and in vitro-matured (IVM) oocytes were cultured to harvest morula stage embryos; they were then subjected to delipation (removal of cytoplasmic lipid droplets) and vitrification. After warming and culture, the embryos developing to blastocysts were transferred to recipients to obtain cloned piglets. From 372 reconstructed embryos, 188 (50.5%) reached the morula stage and 117 (31.5%) developed to blastocysts after vitrification. Transfer of 98 (26.3%) morphologically normal blastocysts gave rise to 6 (1.6%) piglets, including 1 stillborn. The efficiency of the cloned piglet production was comparable with that obtained using SCNT embryos without cryopreservation (2.7%, 17/635). Here, we demonstrate that porcine somatic cell cloning can be performed without a significant reduction in efficiency even when the SCNT embryos are cryopreserved before transfer. 相似文献

We examined the effects of low-dose fish oil ingestion on hepatic lipid accumulation caused after high cholesterol feeding in C57BL/6J mice. The mice were fed purified experimental diets consisting of 20 energy % (en%) safflower oil (SO or SO/CH), 2 en% fish oil + 18 en% safflower oil (2FO or 2FO/CH), or 5 en% fish oil + 15 en% safflower oil (5FO or 5FO/CH) with or without 2 weight % (wt %) cholesterol for 8 weeks. Hepatic triglyceride and total cholesterol contents were significantly lower in groups that were fed diets containing fish oil and cholesterol than in those that were fed safflower oil and cholesterol. The hepatic mRNA levels of fatty acid synthase (FAS) were lower in groups fed cholesterol or fish oil. Fatty acid oxidation-related hepatic gene expressions were higher in fish oil-fed groups. Fecal cholesterol excretion was higher in all cholesterol-fed groups; cholesterol excretion was high in groups fed fish oil and cholesterol. These results suggest that low-dose fish oil diets improve lipid metabolism by modifying the expression of lipid metabolism-related genes in the liver and increasing fecal cholesterol excretion. 相似文献

Uptake and elimination of aflatoxins (AFs) by rainbow trout ( Oncorhynchus mykiss ) during a long-term (21 days) dietary exposure were studied to assess contamination by AFs in aquaculture fish fed AF-containing feed. The uptake factor (UF) of aflatoxin B(1) (AFB(1)) in muscle ranged from 0.40 × 10(-3) to 1.30 × 10(-3). AFB(1) concentrations in liver were 165-342 times higher than in muscle. AFs from feed were more highly accumulated in liver than in muscle. Aflatoxicol (AFL) and aflatoxin M(1) (AFM(1)) were detected in muscle and liver and also in the rearing water. AFL concentrations were higher than AFM(1) by 2 orders of magnitude in muscle, and AFL was a major metabolite of AFB(1). The elimination rate constants (α) of AFB(1) and AFL in muscle (1.83 and 2.02 day(-1), respectively) and liver (1.38 and 2.41 day(-1), respectively) were very large. The elimination half-life (t(1/2)) of AFB(1) was 0.38 days (9.12 h) in muscle and 0.50 days (12.00 h) in liver. The elimination half-life of AFL in muscle and liver was 0.34 day (8.16 h) and 0.29 day (6.96 h), respectively. These data show that AFs are eliminated rapidly and are not biomagnified in fish. Thus, AFB(1) concentration in muscle of fish fed AFB(1)-containing feed (ca. 500 μg/kg) decreased to below the detection limit (20 ng/kg) of the most sensitive analytical method at 1.54 days (36.96 h) after the change to uncontaminated feed. 相似文献

The in vitro reactivity of capsanthin (1) and fucoxanthin (2) with peroxynitrite was investigated, and the reaction products produced by scavenging with peroxynitrite were analyzed. (14'Z)-Nitrocapsanthin (3) and 12-nitrocapsanthin (4) were isolated from the products of the reaction of capsanthin with peroxynitrite. Similarly, (14Z)-15-nitrofucoxanthin (5), (11Z)-11-nitrofucoxanthin (6), and (14Z,9'Z)-15-nitrofucoxanthin (7) were obtained from the reaction of peroxynitrite reaction with fucoxanthin. Capsanthin and fucoxanthin inhibited the nitration of tyrosine by peroxynitrite. Furthermore, nitrocapsanthins (3 and 4) and nitrofucoxanthins (5 and 6) exhibited an inhibitory effect on Epstein-Barr virus early antigen activation in Raji cells and an antiproliferative effect on human pancreatic carcinoma. The critical Na levels in soil which restricted seed germination and growth of radish, and effect of the amending materials on reducing the salinity hazard were determined by performing petri dish and pot experiments. The plants were grown in amended salinized soil in a phytotron for 21 days. Excess accumulation of Na in cells suppressed seed germination and plant growth. The critical Na level in control soil which restricted germination and growth was 12.5mg Na/g soil. At the same salinity level, amending materials promoted seed germination and plant growth mainly by inhibiting the excess flow of Na into the cell through ion holding mechanism. Among the materials used, the commercial manure of 4 months old held greater amount of Na(3.7mg/g) and thus more efficiently reduced the ion damage. Plants grown under such treatment had the highest dry matter yield (28.Gmg/plant), water content was normal(84%), and the ratio for Na in root to Na in plant's shoot was 2.4. We examined arsenic (As) accumulation and speciation in the major cultivars currently grown in Japan, because differences in grain As levels among cultivars may influence dietary As exposure in Japanese people. Ten major cultivars (Oryza sativa L.) were grown under flooded conditions in a paddy field with a background level of As (low-As soil) or in pots filled with soil containing a high level of As (high-As soil). In the low-As soil, the total grain As ranged from 0.11 to 0.17?mg?kg?1, with a mean concentration of 0.14?mg?kg?1, and inorganic As was the major species in all cultivars. There were few genotypic differences in the levels of either total As or inorganic As in the grain. In the high-As soil, total grain As increased to a mean level of 2.4?mg?kg?1 in the 10 cultivars, with markedly increased levels of dimethylarsinic acid. The genotypic variations among cultivars in the levels of both total As and dimethylarsinic acid were statistically significant. However, the genotypic variability of inorganic As levels was quite small, and these levels remained low (at about 0.2?mg?kg?1) even when total As levels increased markedly. These results suggest that differences in grain As levels among Japanese cultivars may not influence dietary As exposure, because there is little genotypic difference in the accumulation of inorganic As, which is considered more toxic than organic As to humans. We discuss the possible mechanism of As accumulation in Japanese paddy rice, in the context of the accumulation of As species in the developing grain and in other plant tissues. 相似文献
