The present study was performed to investigate genetic diversity of Kenyan landraces of the white-flowered gourd (Lagenaria siceraria), which exhibits tremendous morphological variation. RAPD analyses were performed on 53 landraces of the cultivated species L. siceraria and 42 accessions of three wild species (40 L. sphaerica, 1 L. abyssinica, and 1 L. breviflora). A total of 432 polymorphic bands were detected using 54 primers. The four species were clearly differentiated from one another. Intra-specific variations were investigated with L. siceraria and its wild relative L. sphaerica. Landraces of the cultivated species collected from different ethnic communities or regions were differentiated. Morphological variations were not associated with RAPD variations. Bitter landraces collected in Maasai communities showed two specific RAPD bands. In the wild species, accessions collected from the eastern and western sides of the Great Rift Valley were genetically differentiated from each other. In both species, genetic and geographical distance matrices computed among all pairs of accessions were significantly correlated, implying that the observed geographical variation can be explained by the 'Isolation by distance model'. Progeny plants derived from a common mother in L. siceraria showed a low level of segregation in RAPD pattern, suggesting that collected landraces are cultivated, maintaining their inherent traits although they are monoecious and insect-pollinated, whereas the wild relative L. sphaerica showed a higher level of segregation. The morphological diversity observed among landraces of L. siceraria is the result of human selection and their genetic identities are maintained by inbreeding probably resulting from frequent self-pollination.  相似文献

Fat tissue produces a variety of secreted proteins (adipocytokines) with important roles in metabolism. We isolated a newly identified adipocytokine, visfatin, that is highly enriched in the visceral fat of both humans and mice and whose expression level in plasma increases during the development of obesity. Visfatin corresponds to a protein identified previously as pre-B cell colony-enhancing factor (PBEF), a 52-kilodalton cytokine expressed in lymphocytes. Visfatin exerted insulin-mimetic effects in cultured cells and lowered plasma glucose levels in mice. Mice heterozygous for a targeted mutation in the visfatin gene had modestly higher levels of plasma glucose relative to wild-type littermates. Surprisingly, visfatin binds to and activates the insulin receptor. Further study of visfatin's physiological role may lead to new insights into glucose homeostasis and/or new therapies for metabolic disorders such as diabetes.  相似文献

To evaluate the effectiveness in adsorbing organochlorine compounds such as chloroform, dichloromethane, or benzene by lees materials, the determination of spherosomes from different lees materials was established by using a hemacytometer under an optical microscope. Rice bran, wheat bran, rapeseed, linseed, okara, and sakekasu were used for this investigation, and activated carbon was also used as a standard adsorbent material. The number of spherosomes varied from 1.82 x 10(10) particles/g for sakekasu to 4.95 x 10(10) particles/g for wheat bran. There was a high correlation between the removal efficiency in adsorbing organochlorine compounds such as chloroform, dichloromethane, or benzene by lees materials and the number of spherosomes from different lees materials.  相似文献

Antifungal activities of seven compounds, taxinine (1), paclitaxel (2), phenylisoserine methyl ester (3), sciadopitysin (4), ginkgetin (5), isorhamnetin (6), and quercetin (7), isolated from the leaves of kyaraboku, Taxus cuspidata var. nana, against five plant pathogenic fungi, Gibberella fujikuroi, Cladosporium cucumeninum, Fusarium oxysporum, Colletotrichum fragariae, and Corynespora cassiicola, were investigated for utilization of extractives from trees of the genus Taxus. Also, the amounts of compounds 2 and 3 on the leaf surface was measured in relation to the antifungal activities of compounds. Taxinine (1) showed antifungal activity against G. fujikuroi, C. cucumeninum, F. oxysporum, and C. cassiicola. The minimum inhibitory concentration of taxinine for the four fungi was 0.4mol. In addition, from the results of antifungal tests, it may be concluded that paclitaxel on the leaves and stem of T. cuspidata var. nana does not play an important role as an antifungicide in the resistance of trees to plant pathogenic fungal attack.  相似文献

Application of sewage sludge on agricultural land becomes more and more common in many parts of the world in order to recycle the nutrients from the sludge. A range of sewage sludge stabilization techniques are available to make the sludge more stable prior to storage, transportation, and application. These stabilization techniques include dewatering, drying, anaerobic digestion, composting, and reed bed sludge treatment. However, very few studies have investigated the effect of these techniques after the sludge has been applied to agricultural land. The objective of the current study was therefore to investigate the effect of sewage sludge stabilization techniques on the C and N mineralization and gaseous emissions from soil. A soil incubation was conducted to determine the rate of C and N mineralization and N2O and CH4 emissions of sewage sludge stabilized using different techniques. Unstabilized sludge released up to 90% of their C content as CO2, part of which could be caused by release of CO2 from carbonates. Compared with this, sludge stabilization including anaerobic digestion and drying resulted in a reduction of the C mineralization rate of about 40%. Liming reduced C mineralization with around 29%, while treatment in a reed bed system reduced it by 74%. The current study thus clearly demonstrated that stabilization techniques resulted in sludge that was more stable once they were applied to agricultural land. Stabilization also reduced the N immobilization phase, potentially improving the value of the sludge as a fertilizer. Emissions of CH4 were also reduced through sludge stabilization and mainly occurred after application of easily degradable sludge types, which is likely to have enhanced the creation of anaerobic microsites. The stabilization processes also decreased emissions of N2O. The results for both CH4 and N2O indicate that the stabilization tends to reduce the chance of developing conditions where these gases could be produced.  相似文献

Mice have become more important as genetically-modified model animals for analysis of physiological functions. The establishment of a frequent blood sampling system in conscious mice would provide a powerful tool for a better and more detailed understanding of the physiological status of circulating hormonal changes, such as pulse or surge modes of luteinizing hormone (LH) secretion. Frequent blood sampling, however, is considered problematic in mice because of the limited blood volume for their small body size. The present study, therefore, aims to establish a blood sampling protocol to determine the pulse and surge modes of LH secretion using intra-atrial cannulation and frequent blood sampling in free-moving conscious mice. Ovariectomized mice were bled every 3 min for 1.5 h to detect LH pulses. Blood glucose levels, an indicator of stress, were kept constant throughout the 1.5-h sampling period, suggesting that sampling can be performed under stress-free conditions. Obvious LH pulses were observed in ad lib-fed ovariectomized mice, whereas they were significantly suppressed after a 24-h fast. This indicates that the present sampling protocol is suitable for detecting physiological changes in pulsatile LH secretion. In addition, 1-h-interval blood collections in proestrous mice between 1300 and 2200 h revealed that individual preovulatory LH surges occur in the evening of proestrous days. Thus, the present study has developed a blood sampling protocol to detect individual profiles of pulse and surge modes of LH secretion in mice.  相似文献

We studied the prevalence of avian Plasmodium in 509 mosquitoes of 9 species collected from the Ishigaki and Iriomote islands in the Yaeyama Archipelago, located southwest from the mainland of Japan. Two identical avian Plasmodium lineages were detected from Culex (Culiciomyia) nigropunctatus. Detected lineages were phylogenetically classified into different clade to avian Plasmodium lineages from birds and mosquitoes in the mainland of Japan but identical to a lineage detected from a resident bird, White-breasted Waterken (Amaurornis phoenicurus). This is the first detection of avian Plasmodium DNA from mosquitoes in the Yaeyama Archipelago and suggested that resident birds might have been infected with an avian Plasmodium lineage specific to the studied area and C. nigropunctatus could be the candidate vector mosquito species.  相似文献
